남해지역 자연산 해산어의 Trichodina 감염 현황 (Survey of Trichodina infection in wild populations of marine fish caught from Namhae region, southen coast of Korea)

  • The ectoparasite ciliate Trichodina has been recorded from above 100 species of freshwater and marine fishes. In April 2009, we investigated the trichodia infection in 13 species of marine fishes captured by emplacement net and gill net in order to understand trichodina infection status among the natural fish populations along the Namhae-Gun coast area, the southen coast of Korea. Trichodina infection was observed in 10 out of 10 sea basses Lateolabrax japonicus, 2 out of 5 finespotted flounder Pleuronichthys cornutus and 1 out of a stone flounder Kareius bicoloratus. The sea bass infected with the parasite showed hypertrophy and exfoliation in gill epithelium. In addition, hyperplasia and lamellar fusion of gill filament were noticed among naturally affected sea bass.

Occurrence and Molecular Identification of Microcotyle sebastis Isolated from Fish Farms of the Korean Rockfish, Sebastes schlegelii

  • Microcotyle sebastis is a gill monogenean ectoparasite that causes serious problems in the mariculture of the Korean rockfish, Sebastes schlegelii. In this study, we isolated the parasite from fish farms along the coasts of Tongyeong, South Korea in 2016, and characterized its infection, morphology and molecular phylogeny. The prevalence of M. sebastis infection during the study period ranged from 46.7% to 96.7%, and the mean intensity was 2.3 to 31.4 ind./fish, indicating that the fish was constantly exposed to parasitic infections throughout the year. Morphological observations under light and scanning electron microscopes of the M. sebastis isolates in this study showed the typical characteristics of the anterior prohaptor and posterior opisthaptor of monogenean parasites. In phylogenetic trees reconstructed using the nuclear 28S ribosomal RNA gene and the mitochondrial cytochrome c oxidase I gene (cox1), they consistently clustered together with their congeneric species, and showed the closest phylogenetic relationships to M. caudata and M. kasago in the cox1 tree.

  • The head louse Pediculus humanus capitis (De Geer) is a hematophagous ectoparasite that inhabits the human scalp. The infestations are asymptomatic; however, skin irritation from scratching occasionally may cause secondary bacterial infections. The present study determined the presence and frequency of the knockdown resistance (kdr) mutation T929I in 245 head lice collected from Mexico, Peru, and Canada. Head lice were collected manually using a comb in the private head lice control clinic. Allele mutation at T9291 was present in 100% of the total sampled populations (245 lice) examined. In addition, 4.89% of the lice were homozygous susceptible, whereas 6.93% heterozygous and 88.16% homozygous were resistant, respectively. This represents the second report in Mexico and Quebec and fist in Lima.

Ectoparasites: immature Japanese hard ticks (Ixodes nipponensis; Acari: Ixodidae) on Korean lizards

  • Although lizards are important hosts for hard ticks (Ixodidae), very few studies have been conducted in South Korea. To determine whether or not hard ticks can infest lizards endemic to South Korea, we examined 77 lizards of four species (Eremias argus, Sincella vandenburghi, Takydromus amurensis, and Takydromus wolteri) that were collected at 22 different sites between April and October 2010. We confirmed that all four lizard species can be infested by Ixodes nipponensis larvae or nymphs. Of the 62 E. argus examined, we found an average of 12.5 larvae on two lizards and an average of one nymph on one lizard. We found seven nymphs on one S. vandenburghi. We found an average of two nymphs on two of the five T. amurensis and an average of one nymph on four of the nine T. wolteri. Ixodes nipponensis larvae and nymphs were found most frequently on the foreleg axillae (87.8%), followed by the forelegs (7.3%), the eyelids (2.4%), and the ears (2.4%) of the lizards. To the best of our knowledge, this is the first report of I. nipponensis infestations of lizards endemic to South Korea.

한국 박쥐에 기생하는 Exoparasites와 Rickettsia, Hantaan Virus 감염에 관한 연구 (Study of Exoparasites, Rickettsia and Hantaan virus in Bats)

  • To study the relation between ectoparasite, Rickettsia and hanntan virus in bats, four order of Athropoda were collected from Rhinolophus ferrumequinum captured in Cheju and Eptesicus serotinus captured in Chungnamin from July 1989 to March 1998. Also antibody of Rickettsia and hanntan virus were detected by immunofluoroscent antibody technique and RT-PCR. The results are as follows. 1. Five species of Acarina were identified from E. serotinus: Leptotrobidium subakamushi of Trombiculidae, Macronyssus coreanus, Steatonyssuss spinosuss and Steatonychus superans of Macronyssidae, Argas vespertilionis of Metastigmata. 2. Ischnopsyllus needhami of Siphonaptera and Cimex of Hemiptera were identified from E. serotinus. 3. Cycteribia uenoi and Brachytarsina kanoi of Diptera were identified from R. ferrumequinum. 4. The positive rate of rickettsial antibodies in E. serotinus were 17.58%, 15.15%, 22.22%, 52.73% against R. tsutsugamushi, R. typhi, R. sibirica and R. thai tick typhus, respectively. The high positive rate of antibody related to the high content of Arthropoda. 5. The Positive rate of hantaan virus IFA antibodies were 3.4% (27 of 802) and hanntan virus infection rate 36.7% (22 of 60) by RT-PCR in bats. According these result, we showed that certain species of Athropoda isolated playa role as vector of Rickettsia in E. serotinus. Also bats playa role as a reservoir of hantaan virus in nature.

진드기 체항원을 이용한 새응애 감염증에 대한 면역효과 (Immune effects on the somatic antigens against Dermanyssus gallinae and Dermatophagoides pteronyssinus in chicken)

  • Fowl red mite, Dermanyssus gallinae, is the most important ectoparasite affecting egg layers worldwide. More than 35 compounds have been used for fowl red mite control. Although some of them are efficient, several compounds are unsuitable in terms of food safety and environmental problems. Some compounds are efficient in theory but inadequate in practice. It is also expensive in material and labor to control effectively. Effective doses are very close to toxic doses and repeated treatment is required. Repeated, long term treatment of compounds on fowl red mite populations, may cause heritable resistance against the mites. In this study, antigenicity of fowl red mite and house dust mite, Dermatophagoides pteronyssinus, were identified by SDS-PAGE, silver staining, Western blotting and ELISA to investigate immune effects against fowl red mite using somatic antigens of fowl red mite and house dust mite. By SDS-PAGE, silver staining and Western blotting, several common antigens (110, 60, 56, 49, 46 kDa) of both fowl red mite and house dust mite were recognized. To identify immune effect of somatic antigens of fowl red mite and house dust mite, sixty white leghorn broilers(1 week old) were used. Among sixty white leghorn broilers, twenty were immunized with fowl red mite somatic antigens(Group I), twenty immunized with house dust mite antigens(Group II), and twenty were control group without antigen(Group III), respectively. After immunization, it was identified that antibody titers were increased both in group and II. Then all groups were challenged with fowl red mites. After 2 months, measurements of body weights, packed cell volume(PCV), ELISA OD values and numbers of mites were significant(p<0.05). These results suggest that fowl red mite and house dust mite, which are easy to collect and maintain, can be good vaccine candidates against fowl red mite in chicken.

국내 유기견의 개 옴 감염 상황 (Prevalence of canine scabies in the Korean stray dogs)

  • Recently, the number of stray dogs is proportionate to the increase of the number of the companion dogs. Sarcoptic scabiei var canis, causing scabies, is one of the most important canine zoonotic arthropods in Korea and around the world. Thus, we have tried to know the prevalence of canine scabies in the stray dogs in Korea. A total of 565 stray dogs were collected from the rescue centers all over the country from Jan. 2006 to Dec. 2006. They were grouped with euthanasia or natural death and examined for the gender and age estimated by dental formula. To identify the lesions, the whole body was grossly examined and tested pathologically. Thirty two (5.66%) of 565 dogs were diagnosed as canine scabies. Dogs from urban areas had fewer scabies (0.62%) than those of rural areas (12.5%). Prevalence of scabies in male and female dogs was no difference as 5.96% and 5.25%. Euthanasia group showed higher prevalence (6.48%) than natural death group (2.44%) in scabies. Old dogs over five years showed lower infestration (1.82%) in scabies. In histopathological findings, there were mites in the burrows formed in the subcorneal space. Acanthosis, hyperkeratosis with crust, and vascular dilatation were main findings. One hundred thirteen (20%) of 565 stray dogs were diagnosed to have skin disease. Among them, canine scabies is the most prominent ectoparasite as 5.66 %. With previous reports on human infection in Korea, canine scabies must be regarded as the important zoonotic canine skin disease. Accordingly, for the human and canine hygiene it is imperative that stray dogs with skin problems are segregated and tested for the parasites to treat properly as soon as arriving at rescue shelter.

고차기생봉(高次寄生蜂) Tetrastichus sp.의 생태(生態) (Biology of the hyperparasite Tetrastichus sp. (Hymenoptera : Eulopidae))

  • 고차기생자(高次寄生者) Tetrastichus sp.는 단기생성(單寄生性) 외부기생자(外部寄生者)로 일차기생자(一次寄生者) Aneristus ceroplastae, Microterys flavus, Coccophagus hawaiiensis의 종령유충(終齡幼蟲)이나 용에 산란(産卵)하였다. 란(卵)부터 성충(成蟲)까지의 발육한계온도(發育限界溫度)와 유효적산온도(有效積算溫度)는 각(各) $9.8^{\circ}C$와 272일도(日度)로 계산(計算)되었다. $25^{\circ}C$의 항온(恒溫), 16시간(時間) 조명조건(照明條件)에서 봉밀원액(蜂蜜源液)을 급여(給與)하며 사육(飼育)한 자(雌) 성충(成蟲)의 평균수명(平均壽命)은 40일(日) 이었다. 성충(成蟲)은 우화(羽化) 24시간후(時間後)부터 산란(産卵)을 시작하여 일생(一生)동안 평균(平均) 220란(卵)을 낳았으며, 50% 누적산란율(累積産卵率)은 산란개시(産卵開始) 14일(日) 후(後)에 달성(達成)되었다. 순번식율(純繁殖率)(Ro)과 세대기간(世代期間)(T), 내적자연증가율(內的自然增加率)(r)은 각(各) 99.6/세대(世代), 32일(日), 0.142/♀/일(日) 이었다.

집먼지진드기에서 분리한 용출단백질의 개옴진드기 감염증에 대한 항원효과 (Antigenetic effects of the eluted proteins from house dust mite (Dermatophagoides pteronyssinus) in dogs infested with sarcoptic mite (Sarcoptes scabiei var. canis))

  • Canine sarcoptic mite (Sarcoptes scabiei var. canis) is ectoparasite which burrow usually in the stratum corneum of the skin of dogs. Antigens from the burrowing mites induce humoral and cellmediated immune responses in the hosts. The effect of antigenecity induced by somatic antigens of house dust mite (Dermatophagoides pteronyssinus) isolated by continuous elution has been evaluated in canine sarcoptic mites infestation. Continuous elution was carried out in 7.5% SDS-PAGE to isolate proteins of common antigens from somatic antigens of house dust mite. These eluted proteins from somatic antigens of house dust mite were confirmed by Western blotting in 7.5% SDS-PAGE, and eluted proteins (65, 60 kDa) were isolated. To evaluate the antigenetic effect of eluted proteins, eight dogs were divided as 4 groups such as non-vaccinated and non-challenged control (Group I), challenged control (Group II), vaccinated (Group III), and vaccinatedandchallenged (Group IV) groups. Group II and IV were artificially infested canine sarcoptic mites. Group III and IV were immunized with eluted proteins (65, 60 kDa). At the 6th week of the vaccination, the antibody titers of Group of IV were statistically significant higher than those of Group II (p<0.05). And antibody titers of Group III were also statistically significant higher than those of Group I (p<0.05). From these result, it is possible to replace somatic antigens of canine sarcoptic mites with eluted proteins from somatic antigens of house dust mites in order to diagnose and prevent the canine sarcoptic mite infestations.

Chewing Lice of Swan Geese (Anser cygnoides): New Host-Parasite Associations

  • Chewing lice (Phthiraptera) that parasitize the globally threatened swan goose Anser cygnoides have been long recognized since the early 19th century, but those records were probably biased towards sampling of captive or domestic geese due to the small population size and limited distribution of its wild hosts. To better understand the lice species parasitizing swan geese that are endemic to East Asia, we collected chewing lice from 14 wild geese caught at 3 lakes in northeastern Mongolia. The lice were morphologically identified as 16 Trinoton anserinum (Fabricius, 1805), 11 Ornithobius domesticus Arnold, 2005, and 1 Anaticola anseris (Linnaeus, 1758). These species are known from other geese and swans, but all of them were new to the swan goose. This result also indicates no overlap in lice species between older records and our findings from wild birds. Thus, ectoparasites collected from domestic or captive animals may provide biased information on the occurrence, prevalence, host selection, and host-ectoparasite interactions from those on wild hosts.
