송이버섯과 공생하는 소나무 세근으로부터 분리된 내생균의 다양성 (Diversity of Endophytic Fungi Isolated from the Rootlet of Pinus densiflora Colonized by Tricholoma matsutake)

  • 송이버섯 생육지의 소나무세근으로부터 내생균이 분리되었다. 분리된 내생균을 rDNA-ITS 영역으로 동정한 결과 18종이 확인 되었으며, 이중에는 자낭균류 15종, 털곰팡이 아문 3종이 포함되어 있음이 확인 되었다. 본 연구의 내생균 중에는 Penicillium속 균주가 가장 높은 빈도로 생육하고 있음이 확인되었다.

Cloning and Expression Analysis of Phenylalanine Ammonia-Lyase Gene in the Mycelium and Fruit Body of the Edible Mushroom Flammulina velutipes

  • Phenylalanine ammonia-lyase (PAL) gene is known to be expressed in plants, and is involved in the differentiation, growth and synthesis of secondary metabolites. However, its expression in fungi remains to be explored. To understand its expression in mushroom fungi, the PAL gene of the edible mushroom Flammulina velutipes (Fvpal) was cloned and characterized. The cloned Fvpal consists of 2,175 bp, coding for a polypeptide containing 724 amino acids and having 11 introns. The translated amino acid sequence of Fvpal shares a high identity (66%) with that of ectomycorrhizal fungus Tricholoma matsutake. Distinctively, the Fvpal expression in the mycelium was higher in minimal medium supplemented with L-tyrosine than with other aromatic amino acids. During cultivation of the mushroom on sawdust medium, Fvpal expression in the fruit body correspondingly increased as the mushroom grew. In the fruiting body, Fvpal was expressed more in the stipe than in the pileus. These results suggest that F. velutipes PAL activity differs in the different organs of the mushroom. Overall, this is first report to show that the PAL gene expression is associated with mushroom growth in fungi.

First Report of Eight Milkcap Species Belonging to Lactarius and Lactifluus in Korea

  • Lactarius and Lactifluus are milkcaps that are characterized by the secretion of latex. These two genera are part of a globally distributed cosmopolitan group of ectomycorrhizal fungi that is an important food resource in various ecosystems. Recently, the taxonomy of Lactarius and Lactifluus has been revised based on molecular phylogenetics. Despite the importance of these taxa, Korean species of both genera are poorly understood. In an effort to describe milkcap species that are indigenous to Korea, a long-term study has been initiated. During a recent survey, eight species of milkcaps that were previously unrecorded in Korea were detected based on morphological observation and sequence analysis of the internal transcribed spacer region: five Lactarius species (Lactarius atromarginatus, L. austrotorminosus, L. kesiyae, L. tabidus, and L. vietus) and three Lactifluus species (Lactifluus acicularis, Lf. pilosus, and Lf. pinguis). Detailed morphological descriptions and phylogenetic relationships of these species are provided in this article.

Comparison of Soil Higher Fungal Communities between Dead and Living Abies koreana in Mt. Halla, the Republic of Korea

  • To improve our understanding of the relationship between soil higher fungi (belonging to Ascomycota and Basidiomycota) and Abies koreana, we surveyed A. koreana soil fungal communities in a forest in Mt. Halla, Jeju Island, Korea by next-generation sequencing (Illumina Miseq). To confirm the soil higher fungal communities, we collected two types of soils from a defined plot: soils with dead (AKDTs) and living A. koreana (AKLTs), respectively. Soil fungi were classified into 2 phyla, 19 classes, 64 orders, 133 families, 195 genera, and 229 OTUs (895,705 sequence reads). Nonmetric multidimensional scaling (NMDS) showed significantly different soil higher fungal communities between AKDTs and AKLTs (p < .05). In addition, the saprophyte composition was significantly affected by A. koreana status (p < .05). The proportion of the mycorrhizal Clavulina spp. was different between soils with AKDTs and AKLTs, suggesting that Clavulina spp. may be a crucial soil fungal species influencing A. koreana. This study will lead to a better understanding of the ecological status of A. koreana in Mt. Halla. In addition, this study could be useful for the conservation and management of A. koreana habitats.

Taxonomic Revision of the Genus Lactifluus (Russulales, Basidiomycota) of South Korea

  • Lactifluus (Pers.) Roussel is an ectomycorrhizal genus that was recently recognized to be distinct from the genus Lactarius. To date, 226 Lactifluus species have been reported worldwide. Misidentification of Lactifluus species is common because of intraspecific morphological variation, cryptic diversity, and the limited number of taxonomic keys available. Molecular data are indispensable for species delimitation; a multilocus phylogenetic analysis showed that most Asian Lactifluus species are not conspecific with morphologically similar species present on other continents. In particular, Korea has misused European and North American Lactifluus names. In this study, we evaluated the taxonomy of Lactifluus in Korea using both morphological and multilocus molecular (ITS, nrLSU, rpb1, and rpb2) data. We examined 199 Lactifluus specimens collected between 1980 and 2016, and a total of 24 species across the four Lactifluus subgenera were identified. All Korean species are distinct and clearly separated from European and North American species. Five taxa corresponded to previously described species from Asia and the remaining 19 taxa are confirmed as new species. Herein, we provide keys to the Korean Lactifluus species within their subgenera, molecular phylogenies, a summary of diversity, and detailed description of the new species.

Promotion of Tricholoma matsutake mycelium growth by Penicillium citreonigrum

  • Tricholoma matsutake has been the most valuable ectomycorrhizal fungi in Asia because of its unique flavor and taste. However, due to the difficulty of artificial cultivation, the cultivation of T. matsutake has relied on natural growth in forests. To cultivate the T. matsutake artificially, microorganisms in fairy rings were introduced. In this study, we isolated 30 fungal species of microfungi from the soil of fairy rings. Among them, one single fungal strain showed a promoting effect on the growth of T. matsutake. The growth effect was confirmed by measuring the growth area of T. matsutake and enzyme activities including a-amylase, cellulase, and b-glucosidase. In comparison with control, microfungal metabolite increased the growth area of T. matsutake by 213% and the enzyme activity of T. matsutake by 110-200%. The isolated fungal strain was identified as Penicillium citreonigrum by BLAST on the NCBI database. The Discovery of this microfungal strain is expected to contribute to artificial cultivation of T. matsutake.

Unveiling the Diversity of Hydnum in the Republic of Korea with One New Species, Hydnum paucispinum

  • Hydnum is a genus of ectomycorrhizal fungi belonging to the Hydnaceae family. It is widely distributed across different regions of the world, including North America, Europe, and Asia; however, some of them showed disjunct distributions. In recent years, with the integration of molecular techniques, the taxonomy and classification of Hydnum have undergone several revisions and advancements. However, these changes have not yet been applied in the Republic of Korea. In this study, we conducted an integrated analysis combining the morphological and molecular analyses of 30 specimens collected over a period of approximately 10 years in the Republic of Korea. For molecular analysis, the sequence data of the internal transcribed spacer (ITS) region, the large subunit of nuclear ribosomal RNA gene (nrLSU), and a portion of translation elongation factor 1-a (TEF1) were employed as molecular markers. Through this study, we identified eight species that had previously not been reported to occur in the Republic of Korea, including one new species, Hydnum paucispinum. A taxonomic key and detailed descriptions of the eight Hydnum species are provided in this study.

Antioxidant, anti-inflammatory, and cytotoxic properties of fruiting bodies and their mycelia as sources of Cordyceps

  • Mushrooms play crucial roles as reservoirs of naturally occurring bioactive compounds. Among these, Cordyceps militaris is significant because of its well-established reputation for organoleptic excellence and positive health effects, which have led to its widespread commercialization. In contrast, the key properties of Paecilomyces variotii, an ectomycorrhizal symbiont, has received limited attention. In alignment with current research trends, the study of the mycelia and culture media of these mushrooms hold promise in identifying potential sources of valuable bioactive compounds. In the present study, we investigated C. militaris and P. variotii for their phenolic acids and sterols, assessing antioxidant capacity, anti-inflammatory effects, and anti-proliferative activity. Interestingly, P. variotii mycelia exhibited higher concentrations of ergosterol and phenolic compounds, with comparable levels observed in the fruiting bodies, along with superior antioxidant activity compared to that of C. militaris. In contrast, C. militaris mycelia demonstrated anti-inflammatory effects (which were absent in P. variotii mycelia) and cytotoxicity comparable to, and at times exceeding, that of its fruiting bodies (in contrast to P. variotii). In addition, the species analyzed in this study displayed variations in growth rates and mycelial production, which merit consideration for potential future applications and further study.

송이균 접종으로 외생균근을 형성한 소나무 묘목의 PDMP 및 Tween 용액 처리에 의한 순화 (Acclimation of Ectomycorrhizal Pine (Pinus densiflora) Seedlings Inoculated with Tricholoma matsutake by the Treatment of PDMP and Tween Solutions)

  • 송이균을 접종하여 균근을 형성시킨 소나무 묘목을 실외로 가지고 나갈 경우 송이 균근이 이탈되는 현상을 극복하기 위한 방법을 모색하였다. 실내에서 형성한 송이균 접종 소나무 배지의 초기 토양구조를 유지하며 수분을 공급하고자 배양병의 아래에 구멍을 뚫어 화분형태로 변형을 시킨 후 8종류의 급수방식을 시도하였다. 증류수만을 공급한 경우에는 송이 균근이 이탈되었지만, 2.5% PDMP(24 g potato dextrose broth, 3 g malt extract, 1 g peptone) 용액이나 0.5% Tween 80 용액을 2주 간격으로 관수한 경우에는 3개월의 순화처리 이후에도 송이 균근이 강한 활성을 띠고 있음을 확인하였다. 순화기간동안 송이 균근 형성 비율이 증가하였으며, 순화 성공률은 70% 수준에 이르렀다. 금후 송이를 접종한 소나무 묘목은 이러한 순화처리 과정을 거쳐서 균근을 안정시킨 후 현지에 이식하는 방법을 사용하기를 추천한다.

한국 산 송이에서 분리된 균사 (The Mycelia Isolated from the Basidiocarps of Tricholoma matsutake in Korea)

  • 송이는 동양권 문화에서는 중요하며, 값비싼 버섯으로 알려져 있다. 채집된 송이의 자실체에 분리된 송이균사에서 추출된 DNA를 RAPD 방법을 통하여 비교하였다. 비교된 송이균사들은 각각 분리된 송이자실체에서 분리되었으며, PDA에서 성장이나 균총의 형태가 다른 것들을 취하여 비교하였다. 그런데, 이 실험을 통하여, PCR 조건에서 만들어진 RAPD-밴드를 집괴분석한 것을 비교하였으며, 또 다른 방법으로는 Southern blotting 통하여 확인하였다. 그결과, 두 방법에서 송이균사로 추정되는 균사를 확인하여, 이들의 균사에 대한 실험실내의 성장과 균총을 확인하였다. 균사의 성장은 매우 느리며 (10 cm per month), 균총은 반투명한 것이며, 현미경관찰하에 어떤 후막포자와 갈고리모양의 균사가 발견되지 않았다.
