  • Former studies dealing with combined effects caused by chemical compounds in the metabolically competent hepatoma cell line Hep G2 indicated that Hep G2 cells are useful and sensitive indicators for the identification of synergisms of promutagens, comutagens and antimutagens which are relevant in eukryontic (human) cells. In the present study we examined the modulation of DNA damages by the suspected antimutagens ascorbic acid, beta-carotene, alpha-naphthoflavone and beta-naphthoflavone in Hep G2 cells.(omitted)

  • The mutagenic and antimutagenic activities of catechin, hamamelitannin and two proanthocyanidin fractions prepared from the bark of Hamamelis virginia L. - a commonly used medicinal herb - were investigated in a human derived hepatoma (Hep G2) cell line using the single cell gel electrophoresis (SCGE, syn. Comet assay) for the detection of DNA-migration. The cells possess different phase I and phase II enzymes involved in the biotransformation of xenobiotics.(omitted)

Acute Toxicity of Four Agrochemicals on Larval and Juvenile Oriental Weatherfish(Misgurnus anguillicaudatus) (미꾸리(Misgurnus anguillicaudatus)의 치어 및 자어에 대한 4가지 농약의 급성독성)

  • 미꾸리(Misgurnus anguillicaudatus)에 대한 4가지 농약(diazinon, carbaryl, fenpropathrin, myclobutanil)의 급성독성을 평가하기 위하여, 치어 및 자어 단계(postlarva I, postlarva II, 8 WPH(weeks posthatch), 16 WPH, 20 WPH, 32 WPH, 40 WPH)의 미꾸리를 사용하여 급성독성실험을 실시하였다. 각 생육단계 별로 구해진 각 실험농약의 96-hr $LC_{50}$값을 비교하여 미꾸리에 대한 급성독성이 높은 순서를 정리한 결과는 Fenproparthri>Diazinon>Carbaryl, Myclobutanii 순으로 나타났다. 또한, 본 연구에서 사용된 4가지 농약의 독성은 미꾸리의 생육단계에 따라 차이를 나타냈다. 농약에 대한 감수성이 민감한 미꾸리의 생육단계 순으로 정리하면 Postlarva I>Postlarva II, 8 WPH>16 WPH, 20 WPH, 32 WPH, 40 WPH 나타났다. 그러므로 환경관리 및 규제목적으로 미꾸리 성체를 사용하여 독성실험을 할 경우에는 독성을 과소평가 할 수도 있음을 보여주었다.

Molecular/biochemical Biomarkers for Exposure to Hazardous Chemicals in the Water Environment and their Application to Freshwater Fish (유해물질 노출로 인한 분자.생화학적 바이오마커와 담수 어류에 대한 현장 적용성)

  • As concerns regarding water pollution grow, the need increases for a fast and accurate assessment of ecological risk. In this context, many studies have been conducted to identify biomarkers which can sensitively indicate exposure to and effects of various contaminants in a water environment. However, the utility of most such biomarkers in the real water environment is not yet validated. In this paper, we conducted a thorough review of publications that were related to developing or evaluating molecular and biochemical biomarkers of freshwater fish in ecological risk assessment, and evaluated whether these biomarkers of interest could link to the effects on higher biological levels, such as histopathology and above. Biomarkers of interest included those associated with metabolism, oxidative stress, reproduction and endocrine disruption, genotoxicity, and defense against heavy metal exposure. We found that, when used alone, most molecular and biochemical biomarkers are not sufficient to understand the effects of toxic substances in higher biological levels, due to defense or acclimation mechanisms of organisms. Moreover, some biomarkers respond not only to hazardous substances but also to the changes in water quality and disease outbreak. Molecular and biochemical biomarkers may be most useful in understanding the potential biological effects of toxic compounds when used in parallel with relevant endpoints of higher biological levels.

Use of Stable Isotope Probing in Selectively Isolating Target Microbial Community Genomes from Environmental Samples for Enhancing Resolution in Ecotoxicological Assessment

  • In this study we attempted to develop a novel genomic method to selectively isolate target functional microbial genomes from environmental samples. For this purpose, stable isotope probing (SIP) was applied in selectively isolating organic pollutant-assimilating populations. When soil microbes were fed with $^{13}C-labeled $ biphenyl, biphenyl-utilizing cells were incorporated with the heavy carbon isotope. The heavy DNA portion was successfully separated by CsCl equilibrium density gradient. And the diversity in the heavy DNA was sufficiently reduced, being suitable for the current DNA microarray techniques to detect biphenyl-utilizing populations in the soil. In addition, we proposed a new way to get more genetic information by combining this SIP method with selective metagenomic approach. The increased selective power of these new DNA isolation methods will be expected to provide a good quality of new genetic information, which, in turn, will result in development of a variety of biomarkers that may be used in assessing ecotoxicology issues including the impacts of organic hazards, and antibiotic-resistant pathogens on human and ecological systems.

Ecotoxicological Studies Using Aquatic Oligochaetes: Review (수생 지렁이를 이용한 생태 독성 평가 연구에 대한 고찰)

  • Oligochaetes distribute widely in freshwater ecosystem, and some species are used as bioindicators for water quality assessment because they are tolerant to organic enrichment. They are acknowledged for potential for environmental health recovery of organic polluted environment. There are a lot of studies on ecology and toxicity assessment using oligochaetes in aquatic environment. In this study, we reviewed literature on ecotoxicology of aquatic oligochaetes. We searched literature from a database 'google scholar' by using keywords such as aquatic, oligochaete, and toxicity. The literature were summarized according to publication years, species, test methods, and chemicals. We obtained 133 articles published from 1953 to 2015 from the database. Among them, 58 papers(43.6% of total) have been published in 1990s. Three species(Lumnbriculus variegatus, Tubifex tubifex, and Limnodrilus hoffmeisteri) have been used most frequently in the study. Different species displayed different toxicological responses to different toxic chemicals. The results on the ecotoxicological study with aquatic oligochaetes revealed the possibility of the development for early warning system using aquatic oligochaetes to monitor aquatic ecosystem disturbance.
