Application of Gel-based Proteome Analysis Techniques to Studying Post-mortem Proteolysis in Meat

  • This study was conducted to evaluate the possible application of 2 D-SDS-PAGE (2 DE)-based proteome analysis techniques to the assessment of extreme proteolysis in postmortem skeletal muscle. Eight Hanwoo longissimus muscles were incubated immediately after slaughter for 24 h at 5$^{\circ}C$, 15$^{\circ}C$ or 36$^{\circ}C$. Warner Bratzler (WB)-shear force and ultrastructural configuration were determined at 24 h, and rate of proteolysis to 24 h was determined by 1 D-SDS-PAGE (1 DE) and 2 DE. In addition, tentative protein identification was performed from peptide mass fingerprints of MALDI-ToF analysis of major protein groups on 2 DE profiles. The result showed that although ultrastructural configuration was similar between the 5$^{\circ}C$ and 36$^{\circ}C$ treatments, meat at 5$^{\circ}C$ had higher WBshear force (approximately 5 kg greater). A higher rate of protein degradation at 36$^{\circ}C$ was observed based on Troponin-T degradation, 1 DE, and 2 DE analysis. This indicates that proteolysis during the early postmortem period was a significant determinant of shear force at 24 h. Little difference in proteolysis between 5$^{\circ}C$ and 15$^{\circ}C$ treatments was found based on classic 1 DE profile assessment. Meanwhile, considerable differences in the 2 DE profiles between the two treatments were revealed, with substantially higher rate of proteolysis at 15$^{\circ}C$ compared to 5$^{\circ}C$. Nuclease treatment improved 2 DE profile resolution. 400 ${\mu}$g and 600 ${\mu}$g of sample loading appeared to be appropriate for 24 cm pH 3-10 and pH 5-7 IPG strips, respectively. Protein detection and quantification of the 5$^{\circ}C$, 15$^{\circ}C$ and 36$^{\circ}C$ 2 DE profiles revealed 78, 163 and 232 protein spots respectively that were differentially modified in terms of their electrophoretic properties between approximately pI 5.3-7.7 with the molecular weight range of approximately 71-12 kDa. The current results demonstrated that 2 DE was a superior tool to 1 DE for characterising proteolysis in postmortem skeletal muscle.

항공라이다의 결측률 산출을 위한 영역확장 기법 (Region Growing Method for Calculating Unmeasured Rate of Aerial LiDAR Data)

  • 2000년 초반 도입된 항공라이다는 높은 정밀도를 가지고 있으며, 기존의 항공측량과 다른 형태의 포인트 데이터로 자료를 획득하기 때문에 이를 검증하기 위한 새로운 방법이 필요하다. 2009년에 제정된 항공레이 저측량 작업규정에 의하면 크게 결측률과 점밀도, 정밀도 이 3가지 항목으로 검증을 실시하고 있는데, 이 중 결측률은 수계지역과 같은 미반사 지역을 제외한 일정한 격자대의 포인트 유무에 대한 비율을 수치화하는 것이다. 결측률 산출에 있어서 미반사 지역은 반드시 제거해야 하는데, 일반 라이다 자료의 경우 수계지역에 대한 난반사로 인해 포인트가 매우 적어서 추가적인 공간정보자료를 통해 수계지역을 구분해야 한다. 따라서 본 연구에서는 0.3m급의 고해상도 CIR 영상에서 영역확장 기법으로 수계지역을 구분하여 결측률을 산출하였으며, 결측률의 정확도를 비교해보기 위해 수치지도를 이용한 방법과 비교 분석하였다. 그 결과 결측률 값이 미반사 지역을 제외하기 전보다 매우 낮아진 것을 확인 할 수 있었으며, 영상확장 기법과 수치지도를 이용한 값은 유사하다는 것을 알 수 있었다.

Evaluation of the clinical and radiographic effectiveness of treating peri-implant bone defects with a new biphasic calcium phosphate bone graft: a prospective, multicenter randomized controlled trial

  • Purpose: Biphasic calcium phosphate (BCP), a widely used biomaterial for bone regeneration, contains synthetic hydroxyapatite (HA) and β-tricalcium phosphate (β-TCP), the ratio of which can be adjusted to modulate the rate of degradation. The aim of this study was to evaluate the clinical and radiographic benefits of reconstructing peri-implant bone defects with a newly developed BCP consisting of 60% β-TCP and 40% HA compared to demineralized bovine bone mineral (DBBM). Methods: This prospective, multicenter, parallel, single-blind randomized controlled trial was conducted at the periodontology departments of 3 different dental hospitals. Changes in clinical (defect width and height) and radiographic (augmented horizontal bone thickness) parameters were measured between implant surgery with guided bone regeneration (GBR) and re-entry surgery. Postoperative discomfort (severity and duration of pain and swelling) and early soft-tissue wound healing (dehiscence and inflammation) were also assessed. Data were compared between the BCP (test) and DBBM (control) groups using the independent t-test and the χ2 test. Results: Of the 53 cases included, 27 were in the test group and 26 were in the control group. After a healing period of 18 weeks, the full and mean resolution of buccal dehiscence defects were 59.3% (n=16) and 71.3% in the test group and 42.3% (n=11) and 57.9% in the control group, respectively. There were no significant differences between the groups in terms of the change in mean horizontal bone augmentation (test group: -0.50±0.66 mm vs. control groups: -0.66±0.83 mm, P=0.133), postoperative discomfort, or early wound healing. No adverse or fatal complications occurred in either group. Conclusions: The GBR procedure with the newly developed BCP showed favorable clinical, radiographic, postoperative discomfort-related, and early wound healing outcomes for peri-implant dehiscence defects that were similar to those for DBBM.

외국인환자와의 의료분쟁에 관한 연구 (A Study of the Medical Disputes with Foreign Patients)

  • Although the number of foreign patients visiting Korea for medical treatments or plastic surgery is rapidly increasing, countermeasures against unforeseen medical disputes involving foreign patients are adequate. To date, the record shows that most foreign patients have visited doctors at the departments of family medicine, internal medicine, dermatology (incl. plastic surgery), and healthcare centers, which, fortunately, indicates that there are not many severe, high risk patients. However, if the current growth rate continues to rise and the number of foreign patients visiting each department continues to grow, more diverse medical practices will be likely to take place in the future, and consequently, it is expected that the possibilities of medical malpractice and the costs of dispute resolution will also rise dramatically. When a medical dispute occurs, in general, a lawsuit is ultimately settled by the court. However, since this can damage the creditworthiness of medical institutions and also incur significant litigation costs, which is a typical characteristic of a medical lawsuit, medical professionals or institutions will be heavily burdened. Furthermore, an adequate policy or countermeasure against a medical dispute with a foreign patient has not yet been established, and it would be difficult to resolve a dispute by finding the middle ground, due to relative standards and policies between countries. Now, we need to improve the existing policies and prepare for countermeasures that will allow us to precisely predict the nature of such disputes, which have been increasing, and resolve them peacefully. Based on such knowledge, this study aims to establish countermeasures against medical disputes with foreign patients, and examine ways to promptly and reasonably resolve them at an early stage.

PET-MRI에 대한 최적의 정보 제공에 대한 연구: 문헌 보고 중심으로 (A Study on the Optimal Information Provision for PET-MRI: Focused on Literature Article)

  • Currently, state-of-the-art devices such as SPECT, PET/CT, and PET/MRI are rapidly spreading nationwide, and the penetration rate of nuclear medical devices is also ranked fifth in the world. However, PET/MRI's system is slower and less common because it is more complex than PET/CT. The purpose of this study is to provide optimal information on PET/MRI according to the patient's disease. The subjects obtained information on head and neck cancer, pediatric patients, breast cancer patients, heart disease patients, lung cancer patients, and rectal cancer patients. We tried to accumulate protocols by obtaining a lot of information about each disease. In diagnosing head and neck cancer, it is believed that it is highly likely to be used in evaluating preoperative stage determination, recurrence and remote metastasis after treatment, and unclear primary cervical lymph node metastasis. Diagnosis and continuous follow-up of pediatric patients can increase patient benefits by minimizing radiation exposure. Breast cancer provides a comprehensive evaluation of the clinical need to determine the extent of disease in breast and local lymph nodes and the systematic stages of early diagnosis or recurrence. In diagnosing heart disease patients, MR-based PET motion correction helps to realize the full potential of PET images. For lung cancer patients, the clinical value and usefulness of the resolution and detection ability of integrated PET/MRI for soft tissues such as lung cancer will be sufficient. In diagnosing rectal cancer patients, the detection of missing residual diseases can change the clinical response evaluation for rectal cancer patients treated with TNT, and both the initial stage and treatment response evaluation are possible. Therefore, this literature study provided basic clinical data for PET/MRI tests.

컴퓨터 단층촬영의 조직간 밀도차이에 대한 노이즈 영향 (Effect of Noise on Density Differences of Tissue in Computed Tomography)

  • 현재 우리나라의 암 사망률 중 가장 높은 암은 폐암이며 조기발견이 어려운 대표적인 암이다. 조기 발견을 위하여 저선량 흉부 CT를 활용하고 있으며 이는 일반 흉부 X선 사진에 비해 약 3배 정도 폐암 진단율이 높다. 그러나 저선량 흉부 CT는 영상 해상도가 크게 저하될 뿐 아니라 신호가 약해 잡음에 민감한 단점이 있다. 또한 공기로 채워져 있는 폐는 밀도가 낮은 장기로 잡음의 유무가 암의 조기 진단에 영향을 크게 줄 수 있다. 본 연구는 Visual C++을 이용하여 2.0밀도를 가진 큰 원 내부에 물의 밀도인 1.0을 값을 갖는 원을 설정하고 그 안에 각각 밀도가 다른 작은 5개의 원을 수학적 팬텀화하고 가우시안 노이즈를 1%, 2%, 3%, 4% 각각 발생시켜 밀도차에 의한 노이즈의 영향을 평균 값과 표준편차 값, 신호대잡음비(SNR)로 확인하였다. 1% 노이즈 발생 시 큰 원과 작은 원의 밀도차가 가장 큰 영역의 SNR은 4.669로 노이즈의 영향이 작게 나타났으며 밀도차가 가장 낮은 영역의 SNR은 1.183으로 노이즈의 영향이 크게 나타났다. 또한 음의 밀도차에서도 같은 결과 값을 얻었으며 양의 밀도와 음의 밀도 모두 큰 원과 작은 원의 밀도차이가 높은 경우에 SNR 값이 높은 것을 확인 할 수 있다. 화질 또한 밀도차가 크게 나타났을 때 확연하게 육안으로 확인할 수 있었으며 노이즈레벨이 증가하는 경우에는 SNR이 감소하여 잡음의 영향이 크게 나타났다. 이는 밀도차가 적은 장기 또는 암과의 밀도가 비슷한 영역의 장기는 노이즈 영향이 크게 나타날 것이며 노이즈의 발생 확률에 따른 밀도차이의 영향이 진단에 영향을 끼칠 것으로 사료된다.

HEVC 고속 부호화를 위한 효율적인 CU 분할 조기 결정 알고리즘 (An Early Termination Algorithm for Efficient CU Splitting in HEVC)

  • 최근 ITU-T/VCEG과 ISO/IEC MPEG은 공동으로 차세대 비디오 압축 표준 기술인 High efficient video coding (HEVC)를 제정하고 있다. HEVC는 H.264/AVC 표준 기술과 비교해 보면 매우 향상된 압축 효율을 보여 주고 있으며, 특히 Full HD 이상의 매우 큰 사이즈의 동영상 및 그에 따른 고화질 비디오 서비스를 주요 목표로 하고 있다. 그러나 품질과 압축 효율 향상을 위해 새로운 구조와 압축 도구들이 추가됨으로써 인코더의 연산 복잡도가 매우 증가되어 있으며, 이에 화질의 열화를 최소한으로 유지하면서 계산량을 감소시키는 연구가 필요해 지고 있는 상황이다. 본 연구에서는 화면 간 예측 시에 부호화 단위의 분할을 효율적으로 조기 종료하는 방법을 제안한다. 제안된 방법은 현재 코딩 단위 (Coding unit)의 움직임 벡터 정보, 율-왜곡 비용, 그리고 상위 코딩 단위에서의 개별 사이즈에 대한 평균 율-왜곡 비용값을 활용하여 분할을 조기에 중단한다. 실험 결과를 통하여 제안된 방법이 기존의 ECU 방법보다 약 10% 정도로 속도 향상이 있으며, HM 참조 소프트웨어와 비교 시 BD-rate 증가는 1.975% 정도로 매우 적게 나타남을 보인다.

정부 R&D 지원사업의 공공 기술사업화 정책 사례연구: 기술정보 유통 확산을 중심으로 (Government R&D Technology Commercialization Policy Case Study: Focusing on Technical Information Distribution)

  • Purpose - National scientific technology R&D investment is exceeding 60 trillion won per year, and the results of patent applications and technology transfers are visually improving. However, despite the improving research results of national R&D, the practical results of technology startups are mediocre. It is now time to expand the construction of the technology commercialization ecosystem, where the expansion of national R&D leads to the results of technology startups. Therefore, this study discussed the measures to increase the competitiveness of technology startups through the factual survey of the companies that benefitted from R&D support programs. Research design, data, and methodology - This study targeted 996 companies that benefitted from the R&D projects of the Technology Transfer Center for National R&D Programs, and deducted itemized issues through the survey replies. Survey questions were prepared to estimate the national R&D results, and the technology recognition path, the purpose of detailed introduction of the technology, investment of the commercialization fund, economic results, and the factors of success and failure were analyzed. Results - As for the recognition rate of technology during the process of corporate technology commercialization through the technology transfer, recognition through project participation showed a high response rate, and diverse implications of technology commercialization were deducted through the analysis of economic results. As for the resolution alternatives, the proliferation of technology commercialization platform that can create excellent technology for the companies in early stages and the measure of expanding the distribution of technology infrastructure were suggested. In this study, public technology commercialization strategy is established, and the innovative marketing strategy is presented. Conclusions - This study reveal that the result of creating scientific technology jobs should be deducted, in order to produce the revolutionary results of job creation by suggesting the success models of technology commercialization based on domestic scientific technology. In particular, even though the support systems for public research results are being diversely suggested, accurate studies on their actual conditions are currently lacking. Therefore, this study suggest realistic political alternatives to assure results in the process of public technology commercialization, by examining the current state of public research results of R&D support institutions and diagnosing the issues.

지구온난화에 따른 우리나라 벼농사지대의 생산성 재평가 (Evaluation of Site-specific Potential for Rice Production in Korea under the Changing Climate)

  • 본 연구에서는 실측 일기상자료 대신 예측 기후평년 값을 적용하여 기후변화와 그에 상응한 벼 작황의 지리적 분포양상을 복원함으로써 지구온난화에 따른 우리나라 벼농사지대의 생산성을 재평가하였다. 기상청 56개 지점 종관자료(일 최고/최저 기온의 월별 평균값)를 1971-2000년 30년 단위로 수집하여 270m 해상도의 수치기후도를 작성하고, 벼논픽셀에 해당되는 기후자료를 추출하였다. 동일한 시군에 속하는 벼논픽셀의 기후자료를 평균함으로써 시군단위의 '벼논맞춤형 기후자료'를 준비하였다. 같은 방법으로 기상연구소에서 제작한 2011-2100년 기간의 3개 평년(2011-2040, 2041-2070, 2071-2100) 기후시나리오에 근거하여 해당 평년의 기후자료를 추정하였다. 농촌진흥청의 정밀토양도로부터 해당 픽셀의 토성과 토심정보를 검색하고 이를 토대로 유효수분 조견표에 의해 토양자료를 준비하였다. 자포니카형 벼의 특성을 갖도록 개조한 벼 생육모형(CERES-Japonica)에 이들 자료를 입력하고 조생종(오대벼), 중생종 (화성벼), 만생종 (동진벼)의 생육을 모의하였다. 시군 공간평균을 기준으로 3품종 모두 가까운 미래(2011-2040년)에는 출수기가 일주일 정도 빨라지고, 먼 미래(2071-2100년)에는 최대 20일 까지 단축될 수 있다. 생리적 성숙기는 3품종 모두 가까운 미래(2011-2040년)에는 15일 정도 단축되고, 먼 미래(2071-2100년)에는 최대 한달까지도 빨라질 수 있어 출수기에 비해 단축정도가 심하다. 평야지 수량의 경우 조생종인 오대벼는 10a당 6-25%, 중생종 화성벼는 3-26%, 만생종 동진벼는 3-25%까지 감소하였다. 하지만 산간지역에서는 발육속도가 빨라지고 수량이 증가하거나 큰 변화가 없는 곳도 많아 온난화조건에서도 지역별 정밀기후 추정과 이에 근거한 최적품종의 선택, 이앙기 및 수확기 등 생육기간의 조절이 온난화 대응기술로서 유효할 것으로 기대된다.

Detection Method for Bean Cotyledon Locations under Vinyl Mulch Using Multiple Infrared Sensors

  • Purpose: Pulse crop damage due to wild birds is a serious problem, to the extent that the rate of damage during the period of time between seeding and the stage of cotyledon reaches 45.4% on average. This study investigated a method of fundamentally blocking birds from eating crops by conducting vinyl mulching after seeding and identifying the growing locations for beans to perform punching. Methods: Infrared (IR) sensors that could measure the temperature without contact were used to recognize the locations of soybean cotyledons below vinyl mulch. To expand the measurable range, 10 IR sensors were arranged in a linear array. A sliding mechanical device was used to reconstruct the two-dimensional spatial variance information of targets. Spatial interpolation was applied to the two-dimensional temperature distribution information measured in real time to improve the resolution of the bean coleoptile locations. The temperature distributions above the vinyl mulch for five species of soybeans over a period of six days from the appearance of the cotyledon stage were analyzed. Results: During the experimental period, cases where bean cotyledons did and did not come into contact with the bottom of the vinyl mulch were both observed, and depended on the degree of growth of the bean cotyledons. Although the locations of bean cotyledons could be estimated through temperature distribution analyses in cases where they came into contact with the bottom of the vinyl mulch, this estimation showed somewhat large errors according to the time that had passed after the cotyledon stage. The detection results were similar for similar types of crops. Thus, this method could be applied to crops with similar growth patterns. According to the results of 360 experiments that were conducted (five species of bean ${\times}$ six days ${\times}$ four speed levels ${\times}$ three repetitions), the location detection performance had an accuracy of 36.9%, and the range of location errors was 0-4.9 cm (RMSE = 3.1 cm). During a period of 3-5 days after the cotyledon stage, the location detection performance had an accuracy of 59% (RMSE = 3.9 cm). Conclusions: In the present study, to fundamentally solve the problem of damage to beans from birds in the early stage after seeding, a working method was proposed in which punching is carried out after seeding, thereby breaking away from the existing method in which seeding is carried out after punching. Methods for the accurate detection of soybean growing locations were studied to allow punching to promote the continuous growth of soybeans that had reached the cotyledon stage. Through experiments using multiple IR sensors and a sliding mechanical device, it was found that the locations of the crop could be partially identified 3-5 days after reaching the cotyledon stage regardless of the kind of pulse crop. It can be concluded that additional studies of robust detection methods considering environmental factors and factors for crop growth are necessary.
