Characterization of a Positive Regulatory cis-Element and Transacting Factors for the Hepatitis B Viral Pregenomic Promoter

  • Transcription of hepatitis B viral pregenomic promoter is known to be regulated mainly by the combined interaction of enhancers I, II and the intervening regulatory sequences between the two enhancers. A positive regulatory element was identified by serial deletion and measuring the linked chloramphenicol acetyltransferase (CAT) activities, which overlapped with the 5' region of the X open reading frame. When the positive regulatory element was inserted upstream of the SV40 early promoter, it elevated SV40 promoter activity in HepG2 cells. Two cellular proteins of 110 (p110) and 33 (p33) kDa interacted with the positive element and both of them were present in the nucleus, but p110 also existed in the cytoplasm in phosphorylated form. Dephosphorylation of p110 by acid phosphatase enhanced the DNA-binding activity of p110. The p33 could bind to single-strand DNA specifically as well as to double-strand DNA.

Expression of GRP78 Enhance-CAT Fusion Constructs Microiniected into Xenopus Iceuis Oocytes (Xenopus 난자에 미세주입된 GRP78 Enhancer-CAT 이형접합자의 발현)

  • Microiniection of genes Into Xenopus laeuis oocvtes in highly useful in the annvsis of gene regulation, since a large number of oocvtes can be injected in a relatively short time. The GRP78 enhancer has been identified to a 291-bp fragment that spans a region of GRP78 promoter between -378 and -87 (Lin et at., 1986: Kim and Lee, 1989). We examined whether this GRP78 enhancer is effective in directing expression of heterologous gene in Xenopus laeuis oocytes. The chloramphenicol acetvltransferase (CAT) fusion constructs containing the GRP78 promoter and the SV4O early promoter were constructed and were injected into nuclei of Xencpus laeuis oocvtes. The recipient oocvtes were then assayed for CAT activity. The fusion constructs exhibited higher activity as compared to SV40 promoter tested here. The GRP78 enhancer showed 8.5- to 9.2-fold enhancement over that of the SV4O promoter. The orientation of GRP78 enhancer with respect to the direction of CAT transcription unit had no significant effect. Thus, the GRP78 enhancer is a viable candidate for the construction of expression system for use in Xenopus laevss oocvtes and will be important for the studY of a gene expression throughout development.

The Brassica rapa Rubber Elongation Factor Promoter Regulates Gene Expression During Seedling Growth in Arabidopsis thaliana and Brassica napus

  • A tissue-specific and developmentally expressed gene was isolated from Chinese cabbage (Brassica rapa L. ssp. pekinensis), designated BrREF (B. rapa Rubber elongation factor). BrREF transcripts were expressed at high levels in seedlings and at low levels in flower buds and roots. To study the activity of this promoter, the 2.2 kb upstream sequence of BrREF gene was fused to a β-glucuronidase (GUS) reporter gene and was introduced into Arabidopsis thaliana and B. napus by Agrobacterium-mediated transformation. Strong expression of GUS driven by the BrREF promoter was detected in the cotyledons and hypocotyls of transgenic plant seedlings, but GUS expression was weak in roots, excluding the root tips. GUS expression in the cotyledons and hypocotyls decreased dramatically as the seedlings matured and was not detected in the tissues of mature plants. During floral development, GUS expression was observed in immature anthers. These findings suggest that the BrREF promoter can modulate the tissue-specific and developmental expression of gene at the early stages of growth and development.

MGMT-B Gene Promoter Hypermethylation in Patients with Inflammatory Bowel Disease - A Novel Finding

  • Inflammatory bowel disease (IBD) is a disease strongly associated with colorectal cancer (CRC) as a well-known precancerous condition. Alterations in DNA methylation and mutation in K-ras are believed to play an early etiopathogenic role in CRC and may also an initiating event through deregulation of molecular signaling. Epigenetic silencing of APC and SFRP2 in the WNT signaling pathway may also be involved in IBD-CRC. The role of aberrant DNA methylation in precancerous state of colorectal cancer (CRC) is under intensive investigation worldwide. The aim of this study was to investigate the status of promoter methylation of MGMT-B, APC1A and SFRP2 genes, in inflamed and normal colon tissues of patients with IBD compared with control normal tissues. A total of 52 IBD tissues as well as corresponding normal tissues and 30 samples from healthy participants were obtained. We determined promoter methylation status of MGMT-B, SFRP2 and APC1A genes by chemical treatment with sodium bisulfite and subsequent MSP. The most frequently methylated locus was MGMT-B (71%; 34 of 48), followed by SFRP2 (66.6 %; 32 of 48), and APC1A (43.7%; 21 of 48). Our study demonstrated for the first time that hypermethylation of the MGMT-B and the SFRP2 gene promoter regions might be involved in IBD development. Methylation of MGMT-B and SFRP2 in IBD patients may provide a method for early detection of IBD-associated neoplasia.

Analysis of Promoter Strength of Autographa californica Nuclear Polyhedrosis Virus IE1 Gene by Using Rreconmbinant Baculovirus

  • To analysis a promoter strength of Atographa californica nucler polyhedrosis virus (AcNPV) IE1 gene, an immediate viral gene, ${\beta}$-glactosidase gene as a reporter gene was introduced under the control of the IE1 promoter. The restriction fragment containing IE1 promoter and ${\beta}$-galctosidase gene from pAcIE1-gal were inserter into pBacPAK9 to yield transfer vector pAcNPV-IE1-gal. The pAcNPV-IE1-gal was cotransfected with AcNPV genomic DNA BacPAK6 into Sf9 cells to produce recombinant baculovirus AcNPV-IE1-gal. In addition, recombinant bacvulovirus AcNPV-gal, which express ${\beta}$-galac-tosidase under the control of the polyhedrin promoter, was constrer, was constructed to compared with AcNPV-IE1-gal. The recombinant viruses were respectively infected into Sf9 cells and characterized by the virus titer and expression of ${\beta}$-galactoxidase in Sf9 cells. The promoter strength of IE1 and polyhedrin promoters was determined by the amount of ${\beta}$-galactosidase secreted into medium by viral infection. The titer of AcNPV-IE1-Gal determined by plaque assays in Sf9 cells was similar to that of AcNPV-gal. However, expression level of ${\beta}$-galactosidase by AcNPV-IE1-gal was significantly lower than that by AcNPV-gal. In conclusion, promoter strength of IE1 was approximately 25-fold lower than that of polyhedrin.

Identification of the Negative Regulatory Element on the Caprine $\beta$ Lactoglobulin Promoter (염소의 베타-락토글로불린 유전자 프로모터의 음성 조절 인자 규명)

  • Mammary tissue-specificity of the caprine $\beta$-lactoglobulin promoter appears to be secured by repression in non-expressing cells. In order to identify the mechanism of the negative regulation, the upstream promoter sequence of the caprine $\beta$-lactoglobulin gene was analyzed in detail. The repression was mediated by the upstream flanking sequence from -47O to -205. The sequence could repress the promoter activity of $\beta$-lactoglobulin in either orientation. The effect of the putative negative regulation element of caprine $\beta$-lactoglobulin on heterlogous promoters, however, varied: the promoter activity of herpes simplex virus thimidine kinase was either repressed or activated by the sequence depending on its orientation, while the SV4O early promoter was activated rather than repressed. The regulatory sequence involving the putative negative regulatory element was strongly shifted with the nuclear extract from non-mammary HeLa and CV-1 cells, while only weak shift was observed with that of mammary HC11 cells. Such correlation between repression and factor binding suggests that the protected regions in foot-printing assay may be the negative regulatory elements of $\beta$-lactoalobulin that serve tissue-specific repression.

T $\alpha$ 1 $\alpha$ -tubulin promoter directs neuron-specific expression of green fluorescent protein in loach embryo

  • A DNA construct containing rat T $\alpha$ 1 $\alpha$ -tuulin gene 5'-flanking sequence and GFP reporer gene was microinjected into 1-cell loach embryos. Neuron-specific FGP expression was observed in developing loach embryos and early stage fry. The results demonstrated that rat T $\alpha$ 1 $\alpha$ -tubulin gene promoter may be sufficient to specify gene expression to neurons in loach embryos. Thus, the use of GFP reporter controlled by T $\alpha$ 1 $\alpha$ -tubulin gene promoter may facilitate visualization of the dynamic processes of neural tissue development.

Effects of DME/Diesel as an ignition promoter on combustion of hydrogen homogeneous charge compression ignition (수소-예혼합 압축착화 엔진에서 착화제인 DME/diesel이 엔진 연소에 미치는 영향)

  • Hydrogen-dimethy ether (DME) and hydrogen-diesel compression ignition engine combustion were investigated and compared each other in a single cylinder compression ignition engine. Hydrogen and DME were used as low carbon alternative fuels to reduce green house gases and pollutant. Hydrogen was injected at the intake manifold with an injection pressure of 0.5 MPa at fixed injection timing, $-210^{\circ}CA$ aTDC. DME and diesel were injected directly into the cylinder through the common-rail injection system at injection pressure of 30 MPa. DME and diesel inejction timing was varied to find the optimum CI combustion to reduce CO, HC and NOx emissions. When DME was injected early, CO and HC emissions were high while NOx emission was low. Fuel consumption, heat release rate, and exhaust emissions were measured to analyze each combustion characteristics of each ignition promoter. Fuel consumption was decreased when diesel was used as an ignition promoter. This is due to the lower volatility of diesel which created more stratified charge than DME.

  • construct containing reporter gene(pFV4CAT) regulated by carp $\beta$-actin promoter was microinjected into the one-cell stage egg of mud loach (Misgurnus mizolepis), and was successfully expressed, possibly by the integration into the genome. Both mean hatching success and early survival of the microinjected groups were not significantly different with those of control groups (P>0.05). The incidence of transgene was ranged from 7 to 48% based on the PCR and/or Southern blot analyses with the DNA prepared from fin or blood tissue. The spatial and temporal patterns of expression of the pFV4CAT gene, measured by in situ immunohistochemical analysis peroxidase-conjugated anti-CAT antibody, were variable among the experimental individuals. These results suggest that carp $\beta$-actin promoter is effective to express other transgene in mud loach, such that this promoter can be useful in the generation of valuable transgenic mud loach.

Expression of Gal4-VP16 and Gal4-DNA binding domain under the control of the T lymphocyte-specific lck proximal promoter in transgenic mice

  • Thymocyte-specific transcriptional regulatory systems can be used to better understand the relationship between transcription and V(D)J recombination during early T cell development. In this study, we generated transgenic mice expressing the transactivator Gal4-VP16 or the Gal4 DNA binding domain (Gal4-DBD) under the control of the lck proximal promoter, which is only active in immature thymocytes. From these studies Gal4-VP16 and Gal4-DBD expression was shown to significantly alter thymic cellularity and differentiation without significantly changing the $CD3^+$ thymocyte distribution. Furthermore, the presence of Gal4-VP16 or Gal4-DBD in the transgenic thymocytes retarded the mobility of the Gal4 DNA binding motif as determined by a gel mobility shift assay, suggesting that the developmental alteration did not affect the functional property of the transgenic proteins. These results indicated that lck promoter-driven Gal4-VP16 or Gal4-DBD expression did not affect $CD3^+$ mature thymocytes, thus this system can be applied to study transcriptional regulation of transresponder genes in bigenic mouse model thymocytes.
