  • Translationally controlled tumor protein (TCTP) is one of the most abundant proteins in various eukaryotic organisms. TCTPs play important roles in cell physiological processes in cancer, cell proliferation, gene regulation, and heat shock response. TCTP is also considered an important factor in the resistance to oxidative stress induced by dithiothreitol or hydrogen peroxide (H2O2). Arctic calanoid copepods have a variety of antioxidant defense systems to regulate the levels of potentially harmful reactive oxygen species generated by ultraviolet radiation in the Arctic marine ecosystem. However, information on the antioxidant activity of TCTP in the Arctic Calanus glacialis is still scarce. To understand the putative antioxidant function of the Arctic copepod C. glacialis TCTP (Cg-TCTP), its gene was cloned and sequenced. The Cg-TCTP comprised 522 bp and encoded a 174-amino acid putative protein with a calculated molecular weight of ~23 kDa. The recombinant Cg-TCTP (Cg-r TCTP) gene was overexpressed in Escherichia coli (BL21), and Cg-rTCTP-transformed cells were grown in the presence or absence of H2O2. Cg-rTCTP-transformed E. coli showed increased tolerance to high H2O2 concentrations. Therefore, TCTP may be an important antioxidant protein related to tolerance of the Arctic copepod C. glacialis to oxidative stress in the harsh environment of the Arctic Ocean.

  • Twenty years have passed since a prenatal screening for Down syndrome and neural tube defect was applied to obstetric field. The Quad test (AFP, hCG, uE3, Inhibin-A) of the second trimester and the combination test (PAPP-A, hCG, NT) of the first trimester became popular now. The recent trend of prenatal screening is to combine these two screening tests together in order to increase a detection rate of Down syndrome. Three types of screening methods are introduced as follows; integrated test, sequential test and contingent test. In addition to combination of each test, an incorporation of characteristic ultrasound findings of Down syndrome is suggested for its risk calculation. The absence of fetal nasal bone would be a very useful marker especially in the first trimester screening test. According to a change of way calculating risk of Down syndrome, obstetrician's role will be more increased not by passive participation, but by active participation using ultrasound in risk calculation.

  • Special attention has been paid to human chorionic gonadotropin (hCG) for athlete doping control because it stimulates the endogenous production of testosterone and epitestosterone without increasing the T/E ratio which is a doping indicator for the exogenous administration of testosterone. Even though the IOC banned the use of hCG. a detection method has not been decided upon since there are a variety of immunoassay kits available on the market. We evaluated three kits in terms af their performance characteristics. (omitted)

Transfer Rate of Cross Contamination of Listeria monocytogenes between Pork Meat and Workers' Hands during Pork Meat Processing (포장돈육 가공공정 중 돈육과 작업자 손과의 Listeria monocytogenes의 교차오염 전이율)

  • This study was performed to determine the transfer rates of each foodborne pathogen from pork meat packaging during the processing. We analyzed the transfer rate of Listeria monocytogenes from contaminated pork meat to worker's hands (wearing polyethylene gloves, PEG; cotton gloves, CG; and bare hands), cutting boards and knives, and vice versa. Transfer rate of CG 100.00% was higher than that of bare hands 2.513% and PEG 1.511%. In particular, when wearing Co, the transfer rate from the CG to bare hands with CG was 0.08%. Also, the range of transfer rates from contaminated pork meat to cutting board and knife was 0.352-3.791%. In contrast, transfer rates from the workers' hands (with PEG/CG and bare hands) to cutting board, knife, and pork meat ranged from 0.001 to 0.141%. There was a lower transfer rate from workers' hands than from pork meat. These findings indicate that use of PEG could effectively reduce or prevent the cross-contamination compared to CG and provide important information concerning the consecutive transfer of L. monocytogenes during food processing.

  • 본 연구에서는 온라인 다중 사용자 환경의 eBook annotation 시스템 개발에서 데이터를 의미 기반으로 관리하고, 데이터에 대하여 상호 공통적인 이해를 표현하며, 그리고 데이터에 대한 무결성 검사 등을 지원하기 위해서 eBook annotation 온톨로지를 설계하였다. eBook annotation 데이터에 대한 상호 공통적인 이해의 표현을 위해서 한국 전자책 문서 표준인 EBKS(Electronic Book of Korea Standard)를 기반으로 설계 하였으며 설계 된 온톨로지는 Conceptual Graph(CG)를 사용하여 표현하였다. 의미 기반의 처리를 위해서 본 온톨로지에서는 동의어(synonym) 관계와 다국어(multilingua) 관계를 고려하였으며 또한 annotation 데이터 생성시 오류 방지와 중요도를 표현 하기 위해서 무결성 검사, 중요성 axiom을 고려 했다. 제안된 온톨로지는 annotation 데이터의 재사용성을 높일 수 있고 의미 정보를 활용함으로써 eLearning, cyberclass과 같은 다중사용자 환경에서 효과적인 협업을 가능하게 한다. 본 연구에서 구현한 eBook annotation 시스템은 설계한 온톨로지를 이용함으로써 의미 기반의 데이터 관리가 가능하다. 또한 annotation 생성 시 온톨로지 구조를 모르더라도 annotation을 생성할 수 있는 인터페이스를 구현하였다.

Mutational Analysis of the Region between vir Box and -35 Sequence in virE Promoter of pTiA6

  • To investigate how the dyad symmetry region (DSR) and the distance between vir box and -35 sequence of the virE promoter plays a role in virE gene expression, two mutants were constructed by base substitution and insertional mutagenesis. The base substitutional mutation, a AAlongrightarrowCG substitution at positions -39 and -40 on the DSR, showed the level of $\beta$-galactosidase activity approximately 91% of the wild type virE promoter activity. Therefore, the native structure of the DSR seems to be not essential for virE expression. The insertional mutation, constructed by inserting 8 bp ClaI linker between -49 and -50, displayed the $\beta$-galactosidase activity at 12% of the native virE promoter activity. However, this striking reduction appears to be not caused by destruction of the native DSR structure, but by shifting the vir box far from putative -35 sequence.

Reproductive Response of Ewes Fed with Taiwan Grass Hay (Pennisetum purpureum Schum.) Supplemented with Duckweed (Lemna sp. and Spirodela sp.)

  • The effect of duckweed (DW) supplementation was evaluated on dry matter intake (DMI), presence and duration of estrus, percentage of ewes repeating estrus and pregnancy rate, as well as the concentration of progesterone ($P_4$) in multiparous crossbred ewes from Pelibuey, Dorper, and Katahdin breeds, fed with Taiwan grass hay (TWH). Eighteen ewes with $39.7{\pm}4kg$ mean body weight, kept in individual pens, were randomly assigned to one of the following treatments: $T_1$: TWH, $T_2$: TWH plus 200 g DW, $T_3$: TWH plus 300 g DW. The ewes were synchronized with 40 mg fluorogestone acetate (FGA) and 400 UI equine chorionic gonadotropin (eCG). Data were analyzed as a completely randomized design using the GLM procedure. DW supplementation had no effect on dry matter intake (p>0.05); however, a slight decrease of TWH intake was observed as DW supplementation increased. No differences (p>0.05) were found in the beginning of estrus, percentage of ewes presenting it, its duration, or pregnancy rate. There were no differences (p>0.05) on $P_4$ concentration among treatments, or $treatment{\times}period$ interaction (p>0.05). However the period was significant (p<0.01), since the $P_4$ levels increased as time increased after the removal of the FGA device and eCG application.

Characterization of ovarian culture in vitro and sex steroids in vivo by recombinant eel gonadotropin treatments in the eel Anguilla japonica

  • In the present study, we investigated the effects of recombinant eel gonadotropins (rec-GTHs) on maturation induction in immature ovarian culture in vitro and sex steroid hormones in vivo in the Japanese eel Anguilla japonica. To study the in vitro effects of rec-GTHs on estradiol-17β (E2) production in immature ovarian tissues, ovarian tissues were incubated with different doses of rec-follicle-stimulating hormone (rec-FSH) or rec-luteinizing hormone (rec-LH). The results revealed that the E2 levels in the rec-FSH (0.1, 0.5, or 1 ㎍/mL)- and rec-LH (0.1 or 0.5 ㎍/mL)-treated groups were significantly higher than those in the female eels from the control group. Furthermore, to investigate the in vivo effects of rec-GTHs on the gonadosomatic index (GSI) and plasma sex steroid hormone levels, the eels were injected intraperitoneally with eel's ringer (control), salmon pituitary extract (SPE; for female eels), human chorionic gonadotropin (hCG; for male eels), rec-FSH, rec-LH, and rec-FSH + rec-LH once a week. The results revealed that except for the SPE and the hCG groups, none of the groups exhibited a significant difference in GSI values. However, in vivo plasma E2 levels increased at the end of 4 weeks after rec-FSH treatment in female eels. Based on these results, it is suggested that rec-GTHs may have a positive effect on sexual maturation in female eels; however, further studies on complementary rec-protein production systems and additional glycosylation of rec-hormones are needed to elucidate hormone bioactivity in vivo and in vitro.
