Virulence, Resistance Genes, and Transformation Amongst Environmental Isolates of Escherichia coli and Acinetobacter spp.

  • The association of verotoxic E. coli and Acinetobacter spp. with various antibiotic-resistant, diarrhogenic, and nosocomial infections has been a cause for concern worldwide. E. coli and A. haemolyticus isolated on a number of selective media were screened for virulence factors, antibiotic resistance, and transformation of resistance genes. Out of 69 E. coli isolates obtained, 25 (35.23%), 14 (20.30%), and 28 (40.58%) were positive for Vtx1&2, Vtx1, and Vtx2, respectively, 49 (71.015%) for extendedspectrum beta-lactamases (ESBLs), 34 (49.28%) for serum resistance, 57 (82.61%) for cell surface hydrophobicity, 48 (69.57%) for gelatinase production, and 37 (53.62%) for hemolysin production. For the 14 A. haemolyticus isolates, only 2 (14.29%) in each case from all the samples investigated were positive for Vtx1, Vtx2 and Vtx1&2 respectively, 8 (57.14%) for ESBLs, 7 (50.00%) for serum resistance, 11 (78.57%) for cell surface hydrophobicity, 4 (28.57%) for gelatinase production, and 8 (57.14%) for hemolysin production. Although transformation occurred among the E. coli and Acinetobacter isolates (transformation frequency: $13.3{\times}10^{-7}-53.4^{-7}$), there was poor curing of the plasmid genes, a confirmation of the presence of stable antibiotic-resistant genes (DNA concentration between 42.7 and 123.8 ${\mu}g$) and intragenetic transfer of multidrug-resistant genes among the isolates. The isolates were potentially virulent and contained potentially transferable antibiotic resistance genes. Detection of virulence factors, antibiotic resistance genes, and transformation among these isolates is a very significant outcome that will influence approaches to proactive preventive and control measures and future investigations. However, continued surveillance for drug resistance among these bacteria and further investigation of the mechanism of action of their virulence factors are a necessity.

  • The aim of this research is comparing the existing approximation models (e.g. Affine Transformation and Direct Linear Transformation) with Rational Function Model as a substitute of rigorous sensor model of linear array scanner, especially push-broom sensor. To do so, this research investigates the mathematical model of each approximation method. This is followed by the assessments of accuracy of transformation from object space to image space by using simulated data generated by collinearity equations which incorporate or depict the physical aspects of linear array sensor.

Development of a new Model and Methodology for the Analysis of the Performance Evaluation of G2B Systems in e-government: EEM (전자정부 G2B 시스템의 성과평가 분석을 위한 새로운 평가 모델 및 방법론 개발)

  • It is very difficult problem to estimate and evaluate the performance of e-government system which scope and size are large and its effectiveness can not be seen shortly but reveals after several years. It is because the previous offline processes can not be transformed to online ones fully and shortly. For such e-transformation cases, the performance evaluation model should be adjusted and modified gradually as time passes. This paper propose new EEM(E-transformation Evaluation Model) model and methodology to evaluate G2B system that is one of large e-government project. EEM model can derive monetary value of e-transformatized business process areas(online areas). It also estimate the expected effect of offline area that is not yet transformed to online. EEM model consists of standard model, verification model and estimation model with some variables such as evaluation year, evaluation area and data type. By using survey data and database data together it can validate the correctness of the model and derive the effect of the system introduction. This paper also propose EEM evaluation methodology consisting of 5 stages and 10 sub processes to evaluate online and offline effect efficiently. To show the usefulness of this study, we evaluate the performance of Korea G2B system named KONEPS which is famous as a successful e-government case in the world by using the proposed model and methodology. The proposed model and methodology can be applied to different similar areas including e-government projects and large scale information system introduction in private sectors. This study can be also used for establishing appropriate policies about e-government project and informatization issues.

Flood Frequency Analysis by the Box-Cox Transformation

  • Abstract This study was conducted to pursue the normalization of frequency distribution by making an approach to the coefficient of skewness to nearly zero through the Box-Cox transformation, to get probable flood flows can be calculated by means of the transformation equation which has been derivated by Box-Cox transformation in the annual maximum series of the applied watersheds. It has been concluded that Box-Cox transfromation is proved to be more efficient than logarithmic, square root and SMEMAX transformation which is based on the trigonometric solution of a right triangle whose three verteces repesent the smallest, median and largest observed values of a population in making the coefficient of skewness nearer to zero. Consequently it is shown that probable flood flows according to the return period based on Box-Cox transformation are closer to the observed data as compared to other methods including SMEMAX transformation and fitted probability distributions such as the three parameter lognormal and the type I extremal distribution for the applied watersheds.

  • Recombinant chimeric plasmid constructed with Xba I digested pUBl10 and -pE194 was transformed by polyethylene glycol induced protoplast transformation system into Bacillus subtilis BR 151 on the mannitol regeneration media, and two genes of antibiotics resistance were expressed simultaneously in the transfromant. Transformation frequency of the recombinant plasmid was 6.5 $\times$ 10$^{-5}$ on the mannitol regeneration agar plate containing neomycin and erythromycin. The replication of recombinant plasmid in the recipient cells was confirmed by the alkaline extraction method and agarose gel electrophoresis.

  • Translation initiation factor 4E (eIF4E) is a key regulator of protein synthesis. Abnormal regulation of eIF4E is closely linked to oncogenic transformation. Several regulatory mechanisms affecting eIF4E are discussed, including transcriptional regulation, phosphorylation and binding of an inhibitor protein. However it is not clear how the level of eIF4E protein is regulated under basal conditions. Here we demonstrate that Diap1 (Drosophila Inhibitor of Apoptosis Protein), a cell death inhibitor, binds directly to eIF4E and poly-ubiquitinates it via its E3 ligase activity, promoting its proteasome-dependent degradation. Expression of Diap1 caused a reduction of Cyclin D1 protein level and inhibited the growth stimulation induced by overexpression of eIF4E. Taken together, our results suggest that the level of eIF4E protein is regulated by Diap1, and that IAPs may play a role in cap-dependent translation by regulating the level of eIF4E protein.

  • The purpose of this study is to investigate the research trends and the present conditions of real digital texts on "Visual Transformation." For the purpose of this study adopted two different methods: meta analysis and case study. The research trends on visual transformation of digital text were investigated through analyzing the total of 167 literature by means of synthetic meta analysis. Relevant literature was categorized into three types of research: functional, dynamic, and interactional transformation. The type of literature and research methods in each literature were analyzed. The present conditions of real digital texts on visual transformation were investigated by means of case study. The well designed 12 e-learning contents selected and analyzed in terms of the analysis framework which was drawn by the research trends. The results suggested problems as follows in designing e-learning contents. Firstly, there were some cases that did not follow the basic design principles related to typography. Secondly, the content was just provided in each learning steps without consideration of design to enhance text comprehension in many cases. Thirdly, web technology adequately was not applied to design e-learning contents.

  • As a part of efforts to reduce the global emissions of greenhouse gases, the Kyoto Protocol was signed by major developed countries ("Annex I" countries). According to the Kyoto protocol, the Emission Trading Scheme that derives a trading market of the $CO_2$ emission rights is appeared. It causes that business institutions give lots of efforts to reduce $CO_2$ by using new environmentally sound technologies or increasing efficiency in production. On the while there have been several studies trying to develop a methodology to measure the effect of $CO_2$ reduction and its monetary value. In this research we suggest ECRE (Evaluation of $CO_2$ Reduction in E-transformation) model which can measure the $CO_2$ reduction effect through the introduction of G2B system. ECRC model was developed based on the IPCC methodology. ECRC model measures the two major effects of the $CO_2$ reduction which are '$CO_2$ reduction effect from transportation' and '$CO_2$ reduction effect from the decrease of paper use'. In this paper, we calculate the economic effect of $CO_2$ reduction with the case of the G2B system in Korea. This research suggests a basic methodology to measure the $CO_2$ reduction performance for the e-transformed institution.

Agrobacterium-Mediated Co-transformation of Multiple Genes in Metarhizium robertsii

  • Fungi of the Metarhizium genus are a very versatile model for understanding pathogenicity in insects and their symbiotic relationship with plants. To establish a co-transformation system for the transformation of multiple M. robertsii genes using Agrobacterium tumefaciens, we evaluated whether the antibiotic nourseothricin has the same marker selection efficiency as phosphinothricin using separate vectors. Subsequently, in the two vectors containing the nourseothricin and phosphinothricin resistance cassettes were inserted eGFP and mCherry expression cassettes, respectively. These new vectors were then introduced independently into A. tumefaciens and used to transform M. robertsii either in independent events or in one single co-transformation event using an equimolar mixture of A. tumefaciens cultures. The number of transformants obtained by co-transformation was similar to that obtained by the individual transformation events. This method provides an additional strategy for the simultaneous insertion of multiple genes into M. robertsii.

Critical Success Factors for Implementation of e-Business in the Public Sector : A Case Study of the Korean ‘Onbid’ Asset-Management System

  • The purpose of this study is to identify the critical factors for the successful implementation of e-business in the public sector. The paper reports on a case study of the 'Onbid' asset-management system developed by the Korean Asset Management Corporation (KAMCO). 'Onbid' system is an e-marketplace for trading in public assets, including the disposition by public sale of real estate. Through this case study, the paper: (i) explores the changes in organizational culture that are required for successful e-transformation in organizations of public sectors; and (ii) identifies the critical success factors for the implementation of e-business in the public sector.
