Effects of ${\alpha}$-Chymotrypsin Modification on the Functional Properties of Soy Protein Isolates

  • Effects of ${\alpha}$-chymotrypsin modification on degree of hydrolysis (DH), solubility, emulsifying capacity and thermal aggregation of laboratory-purified soy protein isolate (SPI) using a lipoxygenase-defected soybean (Jinpum-kong) and commercial soy protein isolate (Supro 500E) were compared. SPIs were hydrolyzed by ${\alpha}$-chymotrypsin at pH 7.8 and $37^{\circ}C$ for 30 min. DHs of Supro 500E and Jinpum-kong SPI were increased by ${\alpha}$-chymotrypsin modification, and DH of Supro 500E was higher than that of Jinpum-kong SPI. DH of ${\alpha}$-chymotrypsin treated Jinpum-kong SPI was similar with untreated Supro 500E and DH of treated Supro 500E was the highest. Solubility, emulsifying capacity and thermal aggregation of SPIs were increased by ${\alpha}$-chymotrypsin modification, and these changes were highly related to changes in DH. Functional properties of Supro 500E were higher than Jinpum-kong SPI in both of untreated and ${\alpha}$-chymotrypsin treated SPIs.

  • This paper presents a robust discrete MRAC with the discrete e/sub 1/ -modification. Discrete e/sub 1/ -modification is proposed by extending the Narendra's e/sub 1/ -modification to the discrete version. In this case the adaptive controller was reformed by Lyapunov's direct metbod. And the improved robustness by using this algorithm was showed by sector theory.

  • Four turbulent $k-{\varepsilon}$ models (i.e., standard model, modified models with streamline curvature modification and/or preferential dissipation modification) are applied in order to analyze the turbulent flow of wall-attaching offset jet. For numerical convergence, this paper develops a method of slowly increasing the convective effect induced by skew-velocity in skew-upwind scheme (hereafter called Partial Skewupwind Scheme). Even though the method was simple, it was efficient in view of convergent speed, computer memory storage, programming, etc. The numerical results of all models show good prediction in first order calculations (i.e., reattachment length, mean velocity, pressure), while they show some deviations in ·second order (i.e., kinetic energy and its dissipation rate). Like the previous results obtained by upwind scheme, the streamline curvature modification results in better prediction, while the preferential dissipation modification does not.

Study on the Transmission Error Prediction for a Spur Gear Pair (스퍼기어의 전달오차에 관한 연구)

  • Nowadays, lower gear vibration and noise are necessary for drivers in automotive gearbox, which means that transmission gearbox should be optimized to avoid noise annoyance and fatigue before quantity production. Transmission error (T.E.) is the excitation factor that affects the noise level known as gear whine, and is also the dominant source of noise in the gear transmission system. In this paper, the research background, the definition of T.E. and gear micro-modification were firstly presented, and then different transmission errors of loaded torques for the spur gear pair were studied and compared by a commercial software. It was determined that the optimum gear micro-modification could be applied to optimize the transmission error of the loaded gear pair. In the future, a transmission test rig which is introduced in this paper is about to be used to study the T.E. after gear micro-geometry modification. And finally, the optimized modification can be verified by B&K testing equipment in the semi-anechoic room later.

The Effects of Exercise.Behavior Modification Therapy on the Obesity Control and Self-esteem of the Obese Female College Students (운동.행동수정요법이 비만여대생의 비만도, 혈중지질 및 자아존중감에 미치는 효과)

  • Purpose: To examine the effect of the exercise behavior modification therapy one the obesity control and self-esteem of the obese female college students. Method: Data was collected from March 10, 2006 to June 10, 2006. The research design was adopted randomized control group (EG=exercise group) pretest-posttest experimental ($E{\cdot}BG$=exercise${\cdot}$behavior modification therapy group) design. The subjects were nursing students at T college. A total of 37 obese female college students (BMI: over $27mg/m^2$) were selected for this research. The exercise program was executed for 12 weeks and 4 days a week, and the behavior modification was therapy performed for 12 weeks and 60 minutes per week. The data were analyzed with the SPSS Win 12.0 program through the Mann-Whitney test. Result: BMI (Z=-3.049, p=.002) of the $E{\cdot}BG$ was significantly different from the EG. The Total Cholesterol(Z=-1.162, p= .250) of the $E{\cdot}BG$ was not significantly different from the EG. The self-esteem(Z=-3.196, p= .001) of the $E{\cdot}BG$ was significantly different from the EG. Conclusion: The exercise behavior modification therapy was more effective than the exercise therapy in improving the obesity and self-esteem of the obese female college students.

The Effects of Exercise Therapy and Exercise-Behavior Modification Therapy on Obesity, Blood Lipids, and Self-esteem of the Obese Middle-aged Women (운동요법, 운동.행동수정요법이 중년 비만여성의 비만도, 혈중지질 및 자아존중감에 미치는 효과)

  • To examine the effect of the exercise therapy, and exercise-behavior modifi- cation therapy on obesity, blood lipids and self-esteem of the obese middle-aged women. Method: A total of 35 middle-aged women (BMI: over 30) were selected for this research. Walking at a 50% intensity was administered 4 days a week for 12 weeks, while the behavior modification therapy performed for 60~90 minutes per week for 12 weeks. Result: Body weight and BMI has significantly reduced in the case of EG and E BG. The result of comparing body weight between groups showed significant difference between EG and CG, and E BG and CG whereas BMI showed significant difference between EG and CG only. TC, TG, LDL-C, %TC/HDL-C have shown significant decrease in EG and E-BG, while HDL-C displayed significant increase in EG and E BG. And HDL-C showed significant decrease in CG. As for comparison between groups, significant difference was noted in EG and CG, and E BG and CG at TC, HDL-C, LDL-C, and in EG and CG at %TC/HDL-C. Self-esteem displayed significant increase in EG and E BG; however, there was no significant different in CG. As for comparison between groups, there was significant difference noted in E BG and CG only. Conclusion: The results showed that the exercise therapy and the exercise-behavior modification therapy were effective in changing obesity, blood lipids and self-esteem of the obese middle-aged women.

Characterization of the active site and coenzyme binding pocket of the monomeric UDP- galactose 4'- epimerase of Aeromonas hydrophila

  • Aeromonas hydrophila is a bacterial pathogen that infects a large number of eukaryotes, including humans. The UDP-galactose 4'-epimerase (GalE) catalyzes interconversion of UDP-galactose to UDP-glucose and plays a key role in lipopolysaccharide biosynthesis. This makes it an important virulence determinant, and therefore a potential drug target. Our earlier studies revealed that unlike other GalEs, GalE of A. hydrophila exists as a monomer. This uniqueness necessitated elucidation of its structure and active site. Chemical modification of the 6xHis-rGalE demonstrated the role of histidine residue in catalysis and that it did not constitute the substrate binding pocket. Loss of the 6xHis-rGalE activity and coenzyme fluorescence with thiol modifying reagents established the role of two distinct vicinal thiols in catalysis. Chemical modification studies revealed arginine to be essential for catalysis. Site-directed mutagenesis indicated Tyr149 and Lys153 to be involved in catalysis. Use of glycerol as a cosolvent enhanced the GalE thermostability significantly.

A Study on Optimization of Tooth Micro-geometry for a Helical Gear Pair (헬리컬 기어의 치형최적화에 관한 연구)

  • Nowadays, modern gearboxes are characterized by high torque load demands, low running noise and compact design. Also durability of gearbox is specially a major issue for the industry. For the gearbox which used in wind turbine, gear transmission error(T.E.) is the excitation that leads the tonal noise known as gear whine, and radiated gear whine is also the dominant source of noise in the whole gearbox. In this paper, tooth modification for the high speed stage is used to compensate for the deformation of the teeth due to load and to ensure a proper meshing to achieve an optimized tooth contact pattern. The gearbox is firstly modeled in Romax software, and then the various combination analysis of the tooth modification is presented by using Windows LDP software, and the prediction of transmission error under the loaded torque for the helical gear pair is investigated, the transmission error, contact stress, root stress and load distribution are also calculated and compared before and after tooth modification under one torque condition. The simulation result shows that the transmission error and stress under the loads can be minimized by the appropriate tooth modification.

Study on aerodynamic shape optimization of tall buildings using architectural modifications in order to reduce wake region

  • One of the most important factors in tall buildings design in urban spaces is wind. The present study aims to investigate the aerodynamic behavior in the square and triangular footprint forms through aerodynamic modifications including rounded corners, chamfered corners and recessed corners in order to reduce the length of tall buildings wake region. The method used was similar to wind tunnel numerical simulation conducted on 16 building models through Autodesk Flow Design 2014 software. The findings revealed that in order to design tall 50 story buildings with a height of about 150 meters, the model in triangular footprint with aerodynamic modification of chamfered corner facing wind direction came out to have the best aerodynamic behavior comparing the other models. In comparison to the related reference model (i.e., the triangular footprint with sharp corners and no aerodynamic modification), it could reduce the length of the wake region about 50% in general. Also, the model with square footprint and aerodynamic modification of chamfered corner with the corner facing the wind could present favorable aerodynamic behavior comparing the other models of the same cluster. In comparison to the related reference model (i.e., the square footprint with sharp corners and no aerodynamic modification), it could decrease the wake region up to 30% lengthwise.

  • One of the interesting trends in genome research is the study about epigenetic modification above single gene level. Epigenetics refers study about heritable change in the genome, which accompany modification in DNA or Chromatin besides DNA sequence alteration. We used to have the idea that the coding potential of the genome lies within the arrangement of the four bases A, T, G, C; However, additional information that affects phenotype is stored in the distribution of the modified base 5-methylcytosine. This form of information storage is flexible enough to be adapted for different somatic cell types, yet is stable enough to be retained during mitosis and/or meiosis. Epigenetic modification is a modification of the genome, as opposed to being part of the genome, so is known as "epigenetics"(Greek for "upon" genetics). This modification could be methylation on Cytosine base or post translational modification on histone protein(methylation, acetylation, phosphorylation, Sumoylation)($Dimitrijevi\hat{E}$ et al 2005). In this review, we would like to focus on the relationship of DNA methylation and cancer.
