  • 1986년(年)에 전국적(全國的)으로 수집(蒐集)한 피(Echinochloa crusgalli (L.) Beauv.)를 1987년(年)에 2회(回)에 걸쳐 증식(增殖) 및 선발(選拔)하였으며 선정(選定)된 피 17종(種)을 1980년(年)에 공시(供試)하였다. 시험(試驗)은 P.E. film house내(內)의 1/5000 a의 Wagner pot에 의하여 수행(遂行)되었으며, 피 수집종(蒐集種)에 대한 잡초(雜草)로서의 생태적(生態的) 특성(特性)을 파악(把握)할 목적(目的)으로 이루어진 결과(結果)는 다음과 같다. 수집종(蒐集種) 피의 양적(量的) 및 질적형질(質的形質) 차이(差異)를 보면 57종목(種目)의 조사형질(調査形質) 가운데서 종내변이(種內變異)가 컸던 형질(形質)은 종실(種實) 100 입중(粒重), 이식후(移植後) 30일(日)까지 분벽증가속도(分蘗增加速度) 및 분벽수(分蘗數) 및 파종후(播種後) 15일(日)부터 22일(日) 사이의 건물중증가속도(乾物重增加速度)는 변이계수(變異係數)가 30---- 이상(以上)이었으며, 제(第)1엽(葉)의 크기, 22일묘(日苗)의 생체중(生體重)과 건물중(乾物重), 유효경(有效莖) 비율(比率), 주당수수(株當穗數) 및 분벽개각도(分蘗開角度)는 25---- 내외(內外)였다. 반면(反面)에 종내변이(種內變異)가 극(極)히 작았던 형질(形質)은 파종(播種)이 늦었을 때 (6월(月) 6일(日))의 출수소요일수(出穗所要日數), 22일묘(日苗)의 건물중(乾物重) 생체중비(生體重比) 및 초장(草長)으로서 변이계수(變異係數)가 5---- 내외(內外)였다. 질적형질(質的形質)로서도 엽(葉), 엽초(葉鞘), 중조(中助), 절(節), 수(穗) 및 종실(種實)의 색깔과 입상(立狀), 엽록(葉綠)의 거치장단(鋸齒長短), 종보형상(種寶形狀), 엽면(葉面)의 자색(紫色)띠 유무(有無)는 종(種) 특성(特性) 판별(判別)에 유용(有用)할 것으로 판단(判斷)되었다.

버섯의 갈변 및 색도에 대한 전리에너지의 영향 (Browning end Color Characteristics in Mushrooms (Agaricus bisporus) As Influenced by ionizing Energy)

  • 백색종 양송이의 보존기간 연장을 위한 전리에너지 처리가 저장중 버섯의 갈변현상과 색도에 미치는 영향을 조사하였다. 버섯의 갈변현상과 관련된 polyphenol성 화합물은 저장 중 $(9{\pm}1^{\circ}C,\;80{\pm}7%\;RH$, 20일간) 감소하였고 갈변색소는 저장 후 3일경부터 크게 증가하였으며, 이들의 증감현상은 2-3kGy의 감마선 조사로서 유의적으로 감소되었다. 포장된(종이상자와 PE) 버섯의 갓 및 줄기 부위의 표피와 내부조직에 대한 기계적 색도 측정에서 감마선은 처리직후 Hurter L값(백색도)의 감소와 a 및 b값(적색도 및 황색도)의 증가현상을 초래하였으나 저장 후 15일까지 버섯의 변색방지 효과를 뚜렷이 나타내었다. 이 같은 효과는 줄기에 비해 갓 부분에서 높았으며 조사선량(1-3kGy)에도 비례하였다.

The Riparian Vegetation of Close-to-Nature River${\cdot}$Streams in Korea

  • The present study was undertaken to classify and describe the riparian vegetation of the 6 river${\cdot}$stream of Korea Peninsula. As a result the vegetation was divided into nineteen communities. The vegetation units obtained in the present study were as follow: A: Salix koreensis community, B: Salix gracilistyla community, C: Robinia pseudo-acacia community, D: Amorpha fluticosa community, E: Brousonnetia papyrifera community, F: Phyllostachys bambusoides community, G: Rubus corchorifolius community, H: Phramities japonica community, I: Phramites communis community, J: Miscanthus sacchariflorus community, K: Miscanthus sinesis var. purpurascens community, L: Artemisia princeps var. orientalis community, M: Humulus japonicus community, N: Zoysia japonica community, O: Inperata cylindrica var. koenigii community, P: Agropyron tsukushiense var. transiens community, Q: Juncos effusus var. decipiens community, R: Rumex crispus community, S: Persicaria hydropiper community. The vegetation characteristics of riversides was recovered in the surveyed results according to river${\cdot}$stream basin, in other words, 10 communities in the upper river${\cdot}$stream riparian, 15 communities in the middle river${\cdot}$stream riparian, 10 communities in the down river${\cdot}$stream riparian. The Phragmites japonica community in the upper and Phragmites communis community in the down was analyzed by common community of the 6 river${\cdot}$stream riparian, respectively, but none in the middle. The standing profile of vegetation across 6 river${\cdot}$stream was seen stepwise Phragmites japonica community, Salix gracilistyla community, Miscanthus sinensis var. purpurascens community, shrub community by natural waterway in the upper, Salix gracilistyla community, Phragmites communis community, Rumex crispus community, Miscanthus sacchariflorus community, Humulus japonicus community in the middle, Phragmites communis community in the down. The differences of distributional featurs of vegetation emerged from the riparian of the 6 river${\cdot}$stream, but don't from the 6 river${\cdot}$stream.

오매(烏梅)의 다성분 동시분석 및 항알러지 효과 (Simultaneous Analysis and Anti-allergic Effect of Mume Fructus)

  • The Mume Fructus (MF) has been used for relieves cough, arrests arrest chronic diarrhea, treat fluid depletion, and treat ascariasis in Korea. In this study, a high-performance liquid chromatography (HPLC) method was established for simultaneous determination of six main components of MF. Additionally, we were investigated the anti-inflammatory and anti-allergic effects of MF extract on lipopolysaccharide (LPS)-treated RAW264.7 cells and tumor necrosis factor (TNF)-${\alpha}$/interferon (IFN)-${\gamma}$-treated HaCaT cells. The analytical column for separation was used a Gemini $C_{18}$ column maintained at $40^{\circ}C$. The mobile phase consisted of 1.0% (v/v) acetic acid in water (A) and 1.0% (v/v) acetic acid in acetonitrile (B). The flow rate was 1.0 mL/min and the detector was a photodiode array (PDA) set at 280 nm and 320 nm. We evaluated the inhibitory effect of MF extract on the production of inflammatory markers, nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in LPS-stimulated RAW264.7 cells and thymus- and activation-regulated chemokine (TARC/CCL17) in TNF-${\alpha}$/IFN-${\gamma}$-treated HaCaT cells, respectively. We confirmed the genes expression related with TARC, macrophage-derived chemokine (MDC/CCL22) and regulated on activation, normal T cell expressed and secreted (RANTES/CCL5) in HaCaT keratinocyte cells by MF extract. The contents of the five compounds in MF were 0.22-1.01 mg/g. Also, the MF extract show inhibition of about 78% and 75% on NO and $PGE_2$ production at the concentration 1000 mg/mL in RAW264.7 cells. MF extract suppressed the hTARC level and genes expression such as TARC, MDC, and RANTES on TNF-${\alpha}$/IFN-${\gamma}$-treated HaCaT cells.

상업용 올리브 잎 추출물의 화합물 특성과 이들의 oleuropein 함량 비교분석 (Characterization of compounds and quantitative analysis of oleuropein in commercial olive leaf extracts)

  • 올리브 (Olea europaea L.) 잎은 풍부한 폴리 페놀을 함유하고 있으며, 이는 항산화, 항균, 항 바이러스, 항암 활성을 연관시키고 혈소판 활성화를 억제하는 것으로 보고 되어있다. 올리브 잎은 건강기능성식품 및 기능성 화장품의 원료로 상업적으로 사용되고 있으며, 이러한 건강상의 이점은 올리브 잎의 주요 생리활성 물질인 oleuropein와 관련이 있다. Oleuropein은 항산화, 항노화, 항염증, 항암, 항미생물, 항바이러스, 항죽상동맥혈증, 지질 저하 효과, 혈당 저하 효과, 피부 보호 효과가 보고되어 있다. oleuropein이 올리브 잎의 중요한 화합물 임에도 불구하고 상업용 제품에서 oleuropein 함량을 밝히는 정량적 접근 방법은 아직 없다. 본 연구에서는 UPLC-QTof/MS, PDA, CAD로 개발된 방법을 이용하여 10개의 상업용 올리브 잎 추출물의 성분 및 oleuropein 함량을 분석 하였다. Iridoids (1, 3, 4, 14, and 16-18), coumarin (2), phenylethanoids (5, 9, and 11), flavonoids (6-8, 10, 12, and 13), lignan (15)을 포함한 총 18종의 화합물이 올리브 잎에서 예상되었다. 총 10종의 올리브 잎 추출물 분석은 3종의 제품(A, G, H)에서 제품에서 제시한 oleuropein 함량보다 높게, 5종의 제품(B, E, H, I, J)에서 5-10%의 오차범위로 나타났고 C, D에서 함량 미달로 검출되었다. 본 연구에서 oleuropein의 함량분석이 올리브 잎의 품질 관리에 도움이 될 수 있음을 판단하였다.

소청룡탕(小靑龍湯) 물 및 에탄올 추출물의 염증 관련 질환 약리 효능에 관한 실험적 연구 (Experimental Studies on the inflammation-related diseases pharmacological effect of water and 70% ethanol extracts from Socheongnyong-tang)

  • Objectives : The present study aimed to investigate the pharmacological activity of water and ethanol (EtOH) extracts from Socheongnyong-tang (SCNT) on inflammation and its related disease. Methods : The cells were treated with nontoxic concentrations of water and EtOH extract from SCNT in BEAS-2B, HaCaT, RAW 264.7 and 3T3-L1 cells. These cells were stimulated by tumor necrosis facter (TNF)-${\alpha}$, TNF-${\alpha}$/interferon (IFN)-${\gamma}$, and lipopolysaccharide (LPS), respectively. 3T3-L1 cells were differentiated by insulin. After incubation, supernatant were collected and biological indicator measured by enzyme-linked immunosorbent assay. Results : Our results indicate that the water and EtOH extract of SCNT significantly inhibited the production of regulated on activation normal T-cell expression and secreted (RANTES) by treatment of TNF-${\alpha}$ in BEAS-2B cell, and significantly reduced the production of RANTES and macrophage-derived chemokine increased by treatment of TNF-${\alpha}$/IFN-${\gamma}$ in HaCaT cell. Moreover, those extracts significantly decreased the activity of nitric oxide and prostaglandin $E_2$ in LPS-induced RAW 264.7, and significantly inhibited the increased activity of glycerol-3-phosphate dehydrogenase and expression of leptin induced by differentiation in 3T3-L1 cell. Conclusions : These results indicate that both water and EtOH extract of SCNT has powerful effects on inflammation and its related disease. Therefore, SCNT can be developed as a potential pharmacological agent related various diseases. Although the significant effects were observed in both SCNT water and EtOH extract, the EtOH extract was more effective on most experiments than its water extract. Taken together, these findings indicate that the SCNT EtOH extract may have more potential pharmacological agent.

숙성에 의한 뽕나무 상지 내 옥시레스베라트롤 축적 (Accumulation of oxyresveratrol in Ramulus mori upon postharvest storage)

  • 뽕나무의 어린 가지인 상지에서 기능성 물질인 옥시레스베라트롤이 발견됨에 따라 상지를 고부가 건강 기능성 식품 소재로 개발을 위해 옥시레스베라트롤 함량 증대 연구가 필요하였다. 따라서 본 연구에서는 상지의 옥시레스베라트롤 함량을 증가시키기 위하여 국내산 상지의 품종, 숙성 기간, 숙성 온도, 미생물에 의한 접종 등의 숙성 방법에 따른 옥시레스베라트롤 함량 변화를 조사하였다. 국내에서 재배되는 뽕나무 9품종의 상지를 절단 후 7일 숙성하여 옥시레스베라트롤 함량을 비교한 결과, 품종들 간에 큰 차이가 있었으며, 그 중 수원 품종이 숙성에 의해 옥시레스베라트롤 축적이 가장 높게 나타났다. 상지의 옥시레스베라트롤 축적을 위한 최적 숙성 온도를 조사한 결과, $30^{\circ}C$가 최적 온도이었으며 $70^{\circ}C$의 고온 숙성 시에는 옥시레스베라트롤의 축적 효과가 없었다. 뽕나무 9품종 중에서 선발된 수원 품종과 청일 품종에 대한 숙성 기간에 따른 옥시레스베라트롤 함량 변화를 확인한 결과, $30^{\circ}C$, 6일 숙성 시 수원 품종은 약 14배, 청일 품종은 약 7.8배 증가하였으나 8일 이후는 급속히 감소하였다. GRAS 등급의 L. acidophilus, B. coagulans, R. capsulatus, S. cerevisiae, E. faecium 등을 상지에 접종한 후 숙성 기간에 따른 옥시레스베라트롤 함량 변화를 조사한 결과, 미생물이 접종된 실험군들은 대조군에 비해 옥시레스베라트롤 함량이 약 1.7-4.0배 증가하였다. 특히 5균주 중에서 B. coagulans가 옥시레스베라트롤 축적에 가장 뛰어난 효과를 보였다. 따라서 상지 내 미생물 접종에 의한 생물학적 스트레스는 옥시레스베라트롤 축적에 매우 효과적인 방법으로 확인되었다. 결과적으로 상지 내 축적되는 옥시레스베라트롤 함량은 건조 온도, 건조 시간, 수분의 상태 등의 물리적인 요인뿐만 아니라 감염 미생물의 종류에 의해서도 큰 차이가 있음을 확인하였다. 또한 상지 내 옥시레스베라트롤의 함량을 증대시키기 위해서 자외선, 햇빛 등의 물리적 스트레스, 오존, 락스, 재스몬산메틸 등의 화학적 스트레스에 의한 효과도 검토해야 할 것이다. 게다가 숙성동안 옥시레스베라트롤 합성의 주효소인 스틸벤 합성효소 발현량이 조사된다면 숙성에 의한 옥시레스베라트롤 함량 증대 원인을 보다 명확하게 규명할 수 있을 것이다.

Ecotoxicological effects of ballast water effluent teated by an electrolytic method on marine environment

  • Ballast water effluent treated by an electrolytic method contains reactive chlorine species and disinfection by-products (DBPs). In this study, we conducted whole effluent toxicity (WET) testing and ecological risk assessment (ERA) to investigate its ecotoxicological effects on marine environment. WET testing was carried out for three marine pelagic organisms, i.e., diatom Skeletonema costatum, rotifer Brachionus plicatilis and fish Paralichthys olivaceus. The biological toxicity test revealed that S. costatum was the only organism that showed apparent toxicity to the effluent; it showed no observed effect concentration (NOEC), lowest observable effect concentration (LOEC) and effect concentration of 50% (EC50) values of 12.5%, 25.0% and 83.3%, respectively, at brackish water condition. In contrast, it showed insignificant toxicity at seawater condition. B. plicatilis and P. olivaceus also showed no toxicities to the effluent at the both salinity conditions. Meanwhile, chemical analysis revealed that the ballast water effluent contained total residual oxidants (TROs) below $0.03{\mu}g/L$ and a total of 20 DBPs including bromate, volatile halogenated organic compounds (VOCs), halogenated acetonitriles (HANs), halogenated acetic acids (HAAs) and chloropicrin. Based on ERA, the 20 DBPs were not considered to have persistency, bioaccumulation and toxicity (PBT) properties. Except monobromoacetic acid, the ratio of predicted environmental concentration (PEC) to predicted no effect concentration (PNEC) of the other 19 DBPs did not exceed 1. Thus, our results of WET testing and ERA indicated that the ballast water effluent treated by electrolysis and subsequently neutralization was considered to have no adverse impacts on marine environment.

HPLC-DAD-MS/MS를 이용한 금은화 생리활성 물질의 동시분석 (Simultaneous Analysis of Bioactive Metabolites from Lonicera japonica Flower Buds by HPLC-DAD-MS/MS)

  • A high-performance liquid chromatography (HPLC) with diode array detector (DAD) and electrospray ionization mass spectrometry (ESI-MS) was established for the simultaneous determination of chlorogenic acid (1), sweroside (2), luteolin-7-O-glucoside (3), (E)-aldosecologanin (4) and 3,5-dicaffeoylquinic acid (5) from Lonicera joponica flower buds. The optimal chromatographic conditions were obtained on an ODS column (5 ${\mu}m$, 4.6${\times}$150 mm) with the column temperature $25^{\circ}C$. The mobile phase was composed of (A) water with 0.1% formic acid and (B) acetonitrile with 0.1% formic acid using a gradient elution, the flow rate was 0.3 ml/min. Detection wavelength was set at 250 nm. All calibration curves showed good linear regression ($r^2$>0.994) within test ranges. The developed method provided satisfactory precision and accuracy with overall intra-day and inter-day variations of 0.05${\sim}$1.95% and 0.15${\sim}$2.26%, respectively, and the overall recoveries of 97.71${\sim}$103.65% for the five compounds analyzed. The verified method was successfully applied to quantitative determination of the three types (phenolic compounds, iridoids and flavonoids) of bioactive compounds in 21 commercial L. japonica flower buds samples from different markets in Korea and China. The analytical results demonstrated that the contents of the five analytes vary significantly with sources.

Effects of Aging and Aging Method on Physicochemical and Sensory Traits of Different Beef Cuts

  • Wet and dry aging methods were applied to improve the quality of three different beef cuts (butt, rump, and sirloin) from Hanwoo cows (quality grade 2, approximately 50-mon-old). After 28 d of wet aging (vacuum packaged; temperature, $2{\pm}1^{\circ}C$) and dry aging (air velocity, 2-7 m/s; temperature, $1{\pm}1^{\circ}C$; humidity, $85{\pm}10%$), proximate composition, cooking loss, water holding capacity, shear force, color, nucleotides content, and sensory properties were compared with a non-aged control (2 d postmortem). Both wet and dry aging significantly increased the water holding capacity of the butt cuts. Dry aging in all beef cuts induced lower cooking loss than that in wet-aged cuts. Shear force of all beef cuts was decreased after both wet and dry aging and CIE $L^*$, $a^*$, and $b^*$ color values in butt and sirloin cuts were higher in both wet and dry aging (p<0.05) groups than those in the non-aged control. Regardless of the aging method used, inosine-5'-monophosphate content among beef cuts was the same. The sensory panel scored significantly higher values in tenderness, flavor, and overall acceptability for dry-aged beef regardless of the beef cuts tested compared to non- and wet-aged cuts. In addition, dry-aged beef resulted in similar overall acceptability among the different beef cuts, whereas that in wet-aged meat was significantly different by different beef cuts. In conclusion, both wet and dry aging improved the quality of different beef cuts; however, dry aging was more suitable for improving the quality of less preferred beef cuts.
