An Investigation of Microbial Contamination of Ready-to-Eat Products in Seoul, Korea (서울지역 유통판매 중인 즉석섭취.편의식품의 위해 미생물 오염도 조사)

  • This study was carried out to examine microbiological contamination of ready-to-eat products and to propose a draft-standard and specifications according to food types. RTE foods were classified into 6 groups including fish products, meat products, breads, rices, salads, and fresh cut foods. The prevalence rates of pathogens detected from all samples were compared among food categories. The pH ranges for all RTE samples were between 3.8 and 7.3. Total aerobic cell counts ranged from 2 to 6 log CFU/g. Bread, rice and fresh-cut foods showed significantly higher counts, which ranged above 4.0 log CFU/g among the samples. Two kinds of rice were above the level of the KFDA Food Code standard for Eschrichia coli. The prevalence rate of E. coli in the rice was 6.7%. For Staphylococcus aureus, one fish product and one bread-product had levels above 2 log CFU/g. Bacillus cereus counts for all samples were below the level of 3 log CFU/g. Listeria monocytogenes was not detected in the samples. Therefore, these data suggest that the primary microbial hazard factors for ready-to-eat foods and risk assessments should focus on E. coli, S. aureus, and B. cereus.

Benthic Foraminiferal Assemblage and Sedimentary Environment of Core Sediments from the Northern Shelf of the East China Sea (북동중국해 대륙붕 코아 퇴적물의 저서유공충 군집 특성과 퇴적환경 연구)

  • Benthic foraminiferal assemblage and AMS radiocarbon dating of core sediments from the northern shelf of the East China Sea were analyzed in order to understand the paleoenvironment and sedimentary environmental changes around the Korean marginal seas since the last glacial maximum (LGM). The core sediments, containing continuous records of the last 16,000 years, reveal a series of well-defined vertical changes in number of species (S), P/T ratio and species diversity (H) as well as foraminiferal assemblage. Such down-core variations display a sharp change at a core depth of approximately 240 cm, which corresponds to ca. 10,000 year B.P. The sediments of the lower part of the core (240${\sim}$560 cm, Zone I), including the well-developed tide-influenced sedimentary structures, are characterized by high abundances of Ammonia beccarii and Elphidium clavatum (s.l.) and low values in number of species, P/T ratio and diversity. These tide-influenced signatures and foraminiferal assemblage characters suggest that the sediments of Zone I were deposited in a coastal environment (water depths of 20${\sim}$30 m) such as tidal estuary with an influence of the paleo-rivers (e.g., old-Huanghe and Yangtze rivers) during the early phase of the sea-level rise (ca. 16,000 to 10,000 years) since the LGM. In contrast, the upper core sediments (0${\sim}$240 cm, Zone II) are characterized by abundant Eilohedra nipponica and Bolivina robusta with a minor contribution of A. ketienziensis angulata and B. marginata. and high values in number of species, P/T ratio and diversity. Based on relative abundance of these assemblage, Zone II can be divided into two subzones (IIa and IIb). Zone IIa is interpreted to be deposited under the inner-to-middle shelf environment during the marine transgression in the early Holocene (after ca. 9,000 yr B.P.) when sea level rapidly increased. The sediments of zone IIb most likely deposited after 6,000 yr B.P. under the outer shelf environment (80${\sim}$100 m water depth), which is similar to modem depositional environments. The muddy sediments of zone IIb were probably transported from the old-Huanghe and Yangtze Rivers during the late Holocene. We suggest that the present-day oceanographic conditions over the Yellow and the East China Seas have been established after ca. 7,000${\sim}$6,000 yr B.P. when the Kuroshio Current began to influence this area.

Characterization of Korean Cattle Keratin IV Gene

  • Keratins, the constituents of epithelial intermediate filaments, are precisely regulated in a tissue and development specific manner. There are two types of keratin in bovine. The type I is acidic keratin and the type II is neutral/basic keratin. 1.5 kb of 5' flanking sequence of Korean cattle Keratin IV gene, type II keratin (59 kDa), was cloned and sequenced. A symmetrical motif AApuCCAAA are located in a defined region upstream of the TATA box. Proximal SP1, AP1, E-box and CACC elements as the major determinants of transcription are identified. When it was compared to the bovine sequence from -600 bp to ATG upstream, the homology was 97% in nucleotide sequence. Several A and T sequences, located in the promoter region, are deleted in the Korean cattle. An expression vector consisted of Korean cattle Keratin IV gene promoter/SV40 large T antigen was transfected to HaCaT cell (Epithelial keratinocyte). The transformed HaCaT cells showed active proliferation when treated with PDGF (Platelet-derived growth factor) in 0.3% soft agar compared to control cells. These results indicate that Korean cattle Keratin IVgene promoter can be used as a promoter for transfection into epithelial cell.

Rheological and Baking Studies of Composite Flour from Wheat and Naked Barley (밀-쌀보리 복합분의 물리적 성질 및 제빵 시험)

  • Barley (variety, Bangsa No. 6) was milled on a stone mill with 60% flour extraction. Rheological and baking properties of composites containing 5 and 10% of the barley flour with wheat flour (11.4% protein) were investigated. As the barley flour level was increased, amylograph paste viscosities increased at all reference points and farinograph stability decreased. However, no significant differences were noted in the proportional number measured with extensigraph with the various flours. Loaf volume and the staling rate of bread decreased and increased respectively as the barley flour level was increased.

  • Plantaginis seed has been applied in Chinese medicine a as well as in folk remedy. It was advocated that Plantaginis S Semeη exerts good therapeutic effects as anti-inflammatory, antitussive, obstipant and diuretic agent in some cases of alimentary, respiratory a and renal disorders. This study was carried out in order to r re-evaluate the pharmacological action, especially the hypotensive a action of Plantaginis Semen and to elucidate the mechanism of its a action, making use of Plantaginis Semen methanol extract (PME), because its basic pharmacological action, i. e., hypotensive action is n not clear. 1) PME, when administered into intravenous route, elicited the h hypotensive response dependent on the dose of PME given to the rabbit anesthetized with urethane. 2) This hypotensive response of P PME was inhibited by atropine and potentiated by physostigmine, but not influ$\varepsilon$need by vagotomization. 3) Depressor effect of PME was blocked by chlorisondamine, phentolamine, and bethanicline, while not altered by cyproheptadine, diphenhydramine and propran¬olol. 4) The secondary pressor response after blocking the depressor e effect of PME by chlorisondamine was produced, but this pressor response was deminished by atropine. 5) PME augmented the pressor e effect of norepinephrine and angiotensin, on the other hand, reduced b blood pressure elevated by carotid occlusion reflex. 6) These observa¬t tions suggest that PME may induce the hypotensive response via dual mechanisms of parasympathomimetic and sympatholytic action, that the positions of this action are cholinergic peripheral site and sympathetic ganglia respectively, and that PME may possess the pressor activity caused by stimulation of "atropine-sensitive site" which seems to existsin the sympathetic ganglia.

Effects of Heat Treatment on the Characteristics of Royal Paulownia (Paulownia tomentosa (Thunb.) Steud.) Wood Grown in Korea (국산 참오동나무재의 열처리 특성)

  • Effects of heat treatment on the characteristics of Royal paulownia (Paulownia tometosa) wood were investigated. The results were compared with those of Suwon silver poplar (Populus tementiglandulosa) and Korean red pine (Pinus densiflora) woods. The wood samples of the three species were treated at $160^{\circ}C$, $180^{\circ}C$, $200^{\circ}C$ and $220^{\circ}C$ in an electric furnace for 2 hours. The changes of color, density, mass loss, and relative crystallinity were investigated before and after heat treatment. The lightness ($L^*$) decreased rapidly from $200^{\circ}C$ in all species. There were no change in red-green chromaticity($a^*$) and yellow-blue chromaticity($b^*$) of Royal paulownia and poplar woods with increasing temperature. Whereas, yellow-blue chromaticity($b^*$) of Korean red pine wood decreased sharply from $200^{\circ}C$. Royal paulownia wood showed appreciable color change(${\Delta}E^*$) after heat treatment above $180^{\circ}C$. Poplar and pine woods, however, presented significant color change from $160^{\circ}C$. Color change of the three wood species increased rapidly with increasing temperature. Mass loss of the three wood species by heat treatment was the highest in the Royal paulownia wood and the lowest in the pine wood. Mass loss and relative crystallinity increased and density decreased slightly with increasing temperature. Consequently, it is revealed that Royal paulownia wood showed considerable differences in the temperature of color change, weight loss and change of relative crystallinity compared to the other wood species.

Heavy Metal Concentrations of Sediment and Ruditapes philippinarum Inhabited in the Intertidal Zone of Kwangyang Bay (광양만 조간대의 퇴적토 및 바지락(Ruditapes philippinarum)내 중금속 분포)

  • The objectives of this study were to evaluate the extent of heavy metals(As, Pb, Cd, Hg)accumulated in sediments of the Kwangyang Bay, and to investigate bioaccumulation of heavy metals(As, Pb, Cd, Hg) in shellfish(Ruditapes philippinarum) commonly found in the intertidal zone of the Kwangyang Bay. The data was also compared with that of Namhae and Kohung intertidal zones, which were regarded as control stations in this study. Substantial geographical variations in heavy metal concentrations in the sediment samples were found. However, heavy metal concentrations in the sediment collected from the intertidal zones of Yochon(stations I, J) and Myodo(stations G, H) close to Yochon Industrial Area exhibited relatively higher heavy metal concentrations, compared to those from other intertidal zones(stations A, B, C) adjacent to POSCO at Kwangyang Bay. In addition, stations A, B, C showed lower heavy metal concentrations in the sediments than controls(K, L). The annual mean concentrations of the heavy metals in the shellfish from the stations(G, H, I, J) were significantly higher than those from other stations(A, B, C, D, E, F). Nevertheless, heavy metal accumulation in sediments was not reflected in bioaccumulation of heavy metals in shellfish, probably indicating that interactions between the heavy metals in sediment and shellfish might be poor in the current study area, Kwangyang Bay.

Characterization of a Revertant that Restroes the Export of Ribose-Bnding Potein to the Priplasm in Echerichia coli (리보스 결합 단백질을 페리플라슴으로 수송하는 복귀변이주의 분석)

  • A spontaneous revertant of mutation rbsB103 that is ribose taxis-positive was characterized. This revertant was found to be export-competent in the export of ribose-binding protein shown by the disappearance of accumulated mutant precursor protein and the export of mature ribose-binding protein to the periplasm. The reversional change was shown to be in the region of risB gene that codes for the amino terminal portion of ribose-binding protein. Analysis by high-performance liquid chromatography of peptide patterns of ribose-binding proteins confirmed the relationship between the wild-type and the revertant proteins as shown for the mutant previously (Iida et al., 1985). When the processing rate of presursor proteins from the wild type and the revertant strain in vivo was compared by pulse-chase experiment, it was found that processing is less efficient than normal in the revertant. Purified mature proteins from both wild-type and revertant were subjected to amino acid sequencing. The results confirmed the amino acid changes deduced from the DNA sequencing and showed that processing of the revertant precursor occured in the correct position even though there are two different amino acids present in the signal sequence.

Pharmacological Activity of Chaga Mushroom on Extraction Conditions and Immunostimulating Polysaccharide (추출조건에 따른 차가버섯 생리활성 및 면역활성 다당)

  • To investigate the pharmacological activity of chaga mushroom (Inonotus obliquus) on extraction conditions, chaga was extracted using water (reflux at $50^{\circ}C$, decoction over $90^{\circ}C$, pressure at $121^{\circ}C$) or ethanol (reflux at 50, 70, or $90^{\circ}C$). When water extract was further fractionated into crude polysaccharide (IO-CP), yields of IO-CP (4.8~16.8%) were higher than those of ethanolic extracts (IO-E, 1.9~2.7%) at increased temperature. For antioxidant activity, crude polysaccharide (IO-CP-121) obtained by pressurized extraction showed the highest polyphenolic and flavonoid contents (35.10 mg TAE/g and 18.48 mg QE/g, respectively) as well as DPPH and ABTS free radical scavenging activities (26.08 and 27.99 mg AEAC/100 mg, respectively). Meanwhile, IO-CP-D (decoction) and IO-CP-50 (reflux) had more potent mitogenic effects (2.10- and 1.95-fold of saline control at 100 ${\mu}g/mL$) as well as intestinal immune system modulating activities (6.30- and 5.74-fold) compared to IO-CP-121, whereas ethanolic extracts showed no activity. Although no IO-CP showed cytotoxicity against RAW 264.7 cells at 0.1 mg/mL, IO-CP-121 significantly inhibited TNF-${\alpha}$ and NO production as pro-inflammatory factors in LPS-stimulated RAW 264.7 cells (29.2 and 63.5%, respectively). Ethanolic extracts also showed no cytotoxicity at 0.1 mg/mL, whereas inhibition of TNF-${\alpha}$ and NO production was significantly low compared to that of IO-CP-121. In addition, active IO-CP-D was further fractionated into an unadsorbed (IO-CP-I) and seven adsorbed fractions (IO-CP-II~VIII) by DEAE-Sepharose CL-6B column chromatography in order to isolate immunostimulating polysaccharide. IO-CP-II showed the most potent mitogenic effect and macrophage stimulating activity (4.51- and 1.64-fold, respectively). IO-CP-II mainly contained neutral sugars (61.86%) in addition to a small amount of uronic acid (2.96%), and component sugar analysis showed that IO-CP-II consisted mainly of Glc, Gal, and Man (molar ratio of 1.00:0.55:0.31). Therefore, extraction conditions affect the physiological activity of chaga, and immunostimulating polysaccharide fractionated from chaga by decoction is composed mainly of neutral sugars.

Feature Filtering Methods for Web Documents Clustering (웹 문서 클러스터링에서의 자질 필터링 방법)

  • Clustering results differ according to the datasets and the performance worsens even while using web documents which are manually processed by an indexer, because although representative clusters for a feature can be obtained by statistical feature selection methods, irrelevant features(i.e., non-obvious features and those appearing in general documents) are not eliminated. Those irrelevant features should be eliminated for improving clustering performance. Therefore, this paper proposes three feature-filtering algorithms which consider feature values per document set, together with distribution, frequency, and weights of features per document set: (l) features filtering algorithm in a document (FFID), (2) features filtering algorithm in a document matrix (FFIM), and (3) a hybrid method combining both FFID and FFIM (HFF). We have tested the clustering performance by feature selection using term frequency and expand co link information, and by feature filtering using the above methods FFID, FFIM, HFF methods. According to the results of our experiments, HFF had the best performance, whereas FFIM performed better than FFID.
