Morphological Comparison of Four Brown Frogs (Amphibia; Ranidae) (산개구리류 4종(양서강, 개구리과)의 형태적 비교)

  • To clarify the key characteristics of Korean brown frogs (R. dybowskii, R. huanrenesis and R. amurensis coreana), this study employed a morphological and morphometrical comparisons among brown frogs found in Korea and Japanese endemic species, R. tsushimensis. As a result, it was considered that the key characteristics were the shape of the finger and toe tips, the markings of the lower jaw, the presence or absence of an upper lip line, the development of toe webbing, and the shape of the vomerine teeth series by morphological comparisons of brown frogs. Also, there were significant differences in tympanum diameters and foot lengths of the four species of brown frogs. Therefore, a comparison between morphological and morphometric characters is an effective way to clarify the identification of brown frogs.

Assessment of Characteristics and Functions of Abandoned Rice Paddy Wetlands as Habitats for the Amphibia within Land Development Districts (택지개발지구내 양서류 서식처로서 묵논습지의 특성과 기능평가)

  • The current study analyzed appropriateness of abandoned rice paddy wetlands as habitats for the Amphibia by assessing functions and value of abandoned rice paddy wetlands within land development districts as a habitat for living creatures and researching into the Amphibia living in the subject districts. For this purpose, the study designated abandoned rice paddy wetlands within Hwaseong A District and Pangyo B District under land development as subject lands. Those wetlands went through succession after abandonment of farming. Detailed study results are as follows. From classification of wetland types and an analysis of their characteristics, it was found that herbaceous plants such as smartweed communities, horsetail communities and reed communities, and woody plants such as pussy willow communities appeared in both districts. These abandoned rice paddy wetlands shows typical characteristics of palustrine wetlands. As a result of a vegetation structure research, it was observed that succession has been progressed for at least over 2~3 years. In assessment of wetland functions by item, it was found that they are developing into fine wetlands in terms of functions because they are ranked over Moderate Class except separate distance from streams. Also from a research into animals living in the abandoned rice paddy wetlands, it was found that main inhabitants are Rana nigromaculata,Rana dybowskii, Hyla japonica, Bufo bufo gargarizans, etc. It can be confirmed that the abandoned rice paddy wetlands are providing appropriate conditions as habitats to the Amphibia in a view that those species have life cycles of inhabitation (shores and wetlands) - egg-laying (rice paddy, puddles and swampy places) - hibernation (rice field banks and swampy places). From this point of view, it was found that abandoned rice paddy wetlands that developed because of abandonment of farming can be used as means to improve structure and functions of habitats.

Quality of Fried Fish Paste Prepared with Sand-lance (Hypoptychus dybowskii) Meat and Propolis Additive (프로폴리스 첨가 양미리 연육 튀김 어묵의 품질특성)

  • We made sand-lance meat paste containing 0.1-0.5% propolis to improve the quality of the product. The quality characteristics were analyzed by the acid value, peroxide value, volatile basic nitrogen, pH, viable cell count, and sensory evaluation. The fried sand-lance meat paste made with added propolis had a lower acid value, peroxide value, and volatile basic nitrogen content after frying compared to the control prepared with potassium sorbate. The antioxidant and antispoiling effect of propolis on the fried fish meat paste increased with the amount of propolis added. The fried sand-lance meat paste made with 0.2% added propolis was very acceptable. In addition, the bitter taste of the sand-lance meat paste containing 0.2% propolis was blocked by adding 2% sweet amber powder to the paste.

Occurrence of Hymenoptera (wasps and bees) and their foraging in the southwestern part of Jirisan National Park, South Korea

  • The aim of this study was to assess the occurrence and foraging of social and other wasps and bees in Jirisan National Park (JNP, South Korea), in particular in an apiary. Sixty one traps were placed in the southwestern part of JNP to identify social wasps from July to September 2014, and the damage to the apiary caused by wasps or bees was observed once a month from May to December 2014 between 10 a.m. and 5 p.m. In total, 10 species of Vespidae were collected by trapping. Vespa crabro was the most abundant (245 individuals, 28.3%), followed by Vespa velutina (162 individuals, 18.7%). In the apiary, however, V. velutina was the most frequent species. V. velutina visited a maximum of 167 times a day in September, which corresponded to one visit in 2.5 min. Accordingly, these data are in line with the most serious impact of V. velutina on the apiaries in South Korea. V. simillima was the second most frequent species; both Vespa species hawked honeybees. Even though the occurrence of V. mandarinia was low, this species caused serious damage by mass slaughter of honeybees. The occurrence of V. crabro, V. analis and V. ducalis was quite low and their impact on honeybees was negligible. There have been few reports of V. dybowskii foraging for honeybees, but they are considered to be a new pest because their impact on apiaries is considerable. Most Vespa species attacked the apiary from June to October, with a maximum in September. However, V. velutina visited until November to early December. Vespula species are not more serious pests than Vespa species, but many adults were observed stealing honey from beehives. Polistes, Orancistrocerus, and Bombus species had no impact on honeybee colonies in the apiary.

Amphibian Fauna in the Mt. Myungji (명지산 일대의 양서류)

  • Amphibian biodiversity, community structure and habitats were surveyed in the Mt. Myungji area from April 2002 to March 2003. During the survey period 2orders, 4 families, 7 species of amphibians were observed. Bombina orientalis (DI =45.1%) was dominant and Bufo stejnegeri (DI = 12.8%), Rana rugosa and Rana dybowskii (DI = 9.9% each) Rana nigromaculata and Rana huanrenensis (DI 1.4% each), and Onycodactylus fisheri (DI = 7.0%) followed. Frequency of occurrence of O. fisheri and B. stejnegeri was largely different according to the altitude and human residence in their habitats.R. nigromaculata and R. huanrenensis showed low frequency of occurrence (<10%) suggesting the decrease in habitation density in this area. The species richness (R'), general diversity (H'), and evenness (E') of amphibians in this area were 0.98, 2.12, and 0.76, respectively, suggesting relatively healthy condition of amphibian community in this area.

Comparative Studies on the Developmental Capacity by Interspecific Nuclear Transplantation (이종간의 핵치환에 의한 발생수행능력의 비교 연구)

  • In order to investigate the role of nuclei and cytoplasm in early embryogenesis, interspecific nuclear transplantation was carried out Nuclei of one species of amphibian was transplanted into enucleated egg of another species. In interspecific hybrids between Rana species, development was arrested before or right after the dorsal lip formation. In intergenetic hybrids between Rana, Xenopus and axolod, developmental arrest took place at late blastula stage. Comparing the developmental capacity of each nudeocytoplasmic hybrid, the more distantly related the species, the earlier does development arrest. The general rule is that nuclear transfers between any amphibian species will form a regulary cleaved late blastula. Two plausible factors relate to limitation of tite developmental capacity of nudeocytaplasmic hybrids. One is an irreversible change in grafted nuclei and another is tunctional incompatibility between the recipient cytoplasm and transplanted nucleus. The later is postulated a more possible cause of the arrestrnent.

Localization of gonadotropin Releasing Hormone(mGnRH, sGnRH and cGnRH II) in the Brain of three species of Frog, R. nigromaculata, R. dybowskii and R. rugosa (참개구리, 북방산개구리, 옴개구리 뇌에서 3가지 생식소 자극 호르몬 분비 호르몬(mGnRH, sGnRH, cGnRH II )의 분포)

  • 면역효소법을 이용하여 3종의 한국산 개구리 참개구리(Rono nigromaculuto), 옴개구리(R. rugosa), 북방산개구리(R. 겨대bowskii)의 뇌에서 GnRH 뉴우런의 분포 부위와 GnRH의 종류 등을 연구하였다. 1차 항체로는 anti-rat GnRH, anti-salmon GnRH anti-chicken 11 GnRH 항체를 사용하였다. 3종의 개구리에서 mGnRH cGnRH 11와 sGnRH가 이둔 동정되었으나 3가지 항체에 대한 각 종의 면역 반응성은 종에 따라 달리 나타났다 mGnRH는 옴개구리와 참개구리에서, sGnRH는 북방산개구리에서 강한 면역 반응을 나타냈으며 cGnRH 11는 3종의 개구리에서 중간 정도의 면역 반응을 나타냈다. 각각의 GnRH의 상대적인 양에는 차이가 있으나 일부 경우를 제외하고는 뇌의 동일한 지역에 분포하였다. 참개구리에서는 GnRH가 중격 내측핵(NMS), Broca band 핵(NDB)에 집단으로 분포하였다. 북방산개구리에서는 GnRH가 중격 내측핵, Broca bnad 핵에서 등쪽에서 배쪽으로 길게 선상으로 가장 협소하게 분포하였으며, 번식기와 직전(1월-3월)에만 면역 반응을 나타냈다. 옴개구리의 뇌에서 가장 광범위한 지역, 즉 종뇌의 중격 내측핵, Broca band 핵, 아래 교차 지역(SCA)과 간뇌에 GnRH 신경세포가 분포하였으며. 제3뇌실 맥락얼기에서 mGnRH 신경세포가 처음으로 동정되었다. 3종에서 공통적으로 중격 내측핵과 Broca band 핵에서 유래한 신경섬유는 복측 시상하부를 거쳐 정중융기에 이르렀다. 이러한 결과는 GnRH가 뇌하수체에서 생식소 자극 호르몬의 분비 조절에 밀접한 관계가 있음을 뜻한다.

Induction and Inhibition of Amphibian(Rana dybowskii) Oocyte Maturation by Proteolytic Enzymes In vitro. (단백질분해효소들의 양서류 난자에 대한 성숙유도와 억제작용에 관하여)

  • Fully grown amphibian oocytes undergo their maturation (germinal vesicle breakdown, GVBD) during in vitro follicle culture when they are stimulated with frog pituitary homogenate (FPH) or progesterone. Present experiments were designed to determine whether proteolytic enzymes are involved in the regulation of the matunation process. Treatment of a $\alpha$ -chymoiyypsin inhibitor, N a -tosyl-L-phenylalanine-chloromethyl-ketone(TP) to the oocytes exhibited a biphasic phenomenon, the induction of the maturation without added hormone at relatively low doses (0.001-1 $\mu$M) and inhibition of the hormone induced oocyte maturation at a high dose (100 $\mu$M). Treatment of a trypsin inhibitor, N a -tosyl-L-lysine-chloromethyl ketone(TLCK) to the oocytes did not induce the maturation, but rather suppressed the hormone induced oocyte maturation in a high dose(100 $\mu$ M). Treatment of exogenous iyypsin to the oocyte induced their maturation without added hormone in a dose dependent manner (0.001-1 $\mu$ M). The data presented here indicate that some proteolytic enzymes play a role in the regulation of the maturation(meiotic arrest or reinitiation) in amphibians.

  • The census of Amphibia fauna in Tokyusan National Park was carried out during April 10, 1993 and August 8 - August 9, 1993 at the areas of construction field of golf course in Muju Resort and the Chibong. There were 6 species of 5 genera on the 4 families, 2 orders of Amphibia in Tokyusan National Park including Hynobius leechii. Onychodactylus fischeri, and Bufo bufo gargarizans which were announced the specific wild animals with the object of conserving by The Ministry of Environment. At the area of construction field of golf course in Muju Resort, there were 5 species of Amphibia observed as Hynobius leechii, Bombina orientalis, Bufo bufo gargarizans, Rana nigromaculata, and Rana dybowskii. Onychodactylus fischeri and Rana dyhowskii were observed at the Chibong area. The Chibong inwhich was observed 9 relatively large number of individuals of Onychodactylus fischeri was revealed as an environmentally sound habitat to Onychodactylus fischeri. Whereas in the construction field of golf course, there were intended to diminishing species and population size rapidly by progress with construction works. Therefore the large construction works such as golf couse and ski slopes which were concerning to deteriorate the natural environment should be restrain in National Park for animal diversity conservative.

Comparison of the Bacterial and Fungal Colonies from Rana dybowskii which Collected from Inside and Outside Frog Farms and Identification of the Bacteria from the Tadpoles (개구리 증양식장 내·외부에서 채집된 북방산개구리(Rana dybowskii)로부터 검출된 세균과 곰팡이 콜로니 수의 비교 및 유생으로부터 확인된 세균 규명)

  • There are many ongoing studies of infectious diseases as the major factor responsible for global declining of the amphibian population. Although some point out the amphibian rearing facilities like frog farms as one of the important sources of harboring and spreading amphibian infectious pathogens in the wild, there have been few related studies in South Korea. In this study, we investigated the bacterial and fungal colonies on the skin and in the internal organs of frogs and tadpoles collected inside and outside of Dybowski's brown frog farms in Inje, Goesan, and Gongju to compare the difference according to the region and between inside and outside the farm. We also intended to classify the bacteria collected from the tadpoles into species by analyzing 16s rDNA gene sequences. The result showed that the number of bacterial colonies found in the skin and gut of frogs and the number of fungal colonies found in the skin and liver of frogs collected in Goesan was significantly greater than those in the frogs in Inje. However, there was no difference between the frogs collected inside and outside of farms in both regions. In the case of tadpoles, the number of fungal colonies in the tadpoles collected from Gongju was greater than that in the tadpoles collected from Inje. The comparison of inside and outside frog farms showed that there were more bacterial colonies on the skin of the tadpoles collected from inside than outside the frog farm in Inje and more bacterial colonies in the organs of the tadpoles collected from outside than inside the farm in Gongju. The frogs with higher condition factor (body weight/snout-vent length*100) showed fewer bacterial colonies on the skin and fewer fungal colonies in the heart, but there were no significant relationships in tadpoles. We identified the total of 15 genera and four phyla of bacteria, but the difference according to regions and between inside and outside farm was not evident. The result of this study indicates that the different conditions according to the locality of farm and between inside and outside farm cause the difference in the population sizes of bacterial and fungal colonies and that it can affect the overall health condition of Dybowski's brown frogs in the farm. Moreover, the result suggests that effective disease control in the facility is greatly necessary to ensure successful operation of amphibian rearing facility and to prevent the possible spread of diseases from the facility to the wild.
