향나무 심재 추출물로 염색된 직물의 방미성과 집먼지 진드기 기피효과 (Antifungal activity and house dust mite repellent effect of fabric dyed with Juniperus chinensis heartwood extracts)

  • This study was carried out to investigate the antifungal activity and house dust mite repellent effect of cotton and silk fabrics dyed with Juniperus chinensis heartwood extracts by hot-water and methanol. The chemical composition of Juniperus chinensis heartwood extracts by hot-water and methanol was analyzed by GC-MS. In the case of antifungal activity, dyed fabrics with hot water extract had no effect against chaetomium globosum fungus. But showing no growth in fabrics dyed with methanol extract, dyed fabrics with methanol extract showed very excellent antifungal activity. Silk fabric dyed with methanol extract showed excellent house dust mite repellent effect of 94.3%~96.0% against dermatophagodes farinae. The composition of Juniperus chinensis heartwood extracts from hot-water and methanol was different. There were no terpene and its analogue peaks in hot-water extract. However there were terpene and its analogue peaks in methanol extract. From these GC-MS results, terpene was identified in methanol extract of Juniperus chinensis heartwood. Efficient ingredient of antifungal activity and house dust mite repellent effect was assumed terpene.

Effect of Asthma Management Education Program on Stress and Compliance of Patients with Allergic Asthma to House Dust Mite

  • Purpose. This study was designed to examine the effect of asthma management education program applied to allergic asthma patients receiving immunotherapy due to house dust mite on their stress and compliance with health care regimens. Methods. A quasi experimental design with non-equivalent control group and non-synchronized design was used. The subjects of this study were 61 patients who were receiving immunotherapy at intervals of a week after their symptoms were diagnosed as house dust mite allergic asthma at the pulmonary department of a university hospital in Seoul. They were divided into an experimental group of 29 patients who received asthma management education and a control group of 32 patients. The asthma management education pro-gram was composed of group education (once) and reinforcement education (three times) with environmental therapy and immunotherapy to house dust mite. Results. Stress significantly decreased in the experimental group compared to that in the control group. Compliance with health care regimens significantly increased in the experimental group compared to that in the control group. Conclusions. The results suggested that the asthma management education program is effective for the management of stress and the improvement of compliance in patients with allergic asthma to house dust mite.

집먼지진드기에 대한 침엽수 정유의 기피효과 (Repellent effect of essential oils from coniferous trees against the house dust mites (Dermatophagoides farinae and D. pteronyssinus))

  • The avoidance of the allergen of the house dust mite is one of the challenges to reduce and treat the symptoms of allergic diseases. Accordingly, various acaricidal agents are being used to kill the mite, but just killing it leaves the remains of it, which still act as the allergen. Therefore expelling the mite is thought as best policy to avoid the mite allergen. For this, some materials have been applied to repellent agents against the mite. Among them, a material with natural origin, known as the phytoncide, is being used for its repellent activity, as well as for its benefits for health. In this experiment, essential oils extracted from Korean white pine (Pinus koreaiensis S. et Z.) and hinoki cypress (Chamaecyparis obtusa), which are widely used as the source of phytoncide products, are studied for demonstration of the repellent effect against the house dust mites, Dermatophagoides farinae and D. pteronyssinus. Direct contact method was used to evaluate the repellent effect (%). And the results suggest the oils have a significant effect enough to be used as a source of repellent agent. For the repellent effect, the most effective concentration was $0.5{\mu}l/cm^2$ at 45 minutes both in the white pine and the hinoki oil.

아토피 아동 가정내 집먼지 진드기 농도와 환경요인: 환자 대조군 연구 (House Dust Mites and Associated Environmental Factors in Homes of Atopic Children: a Case-Control Study)

  • Objectives: The purpose of this study was to determine house dust mite concentrations in living rooms and bedding materials in atopic case-control groups. Methods: Fifty four homes with children suffering atopic diseases as cases and fifty one homes without atopic diseases as controls were selected after diagnosis at three elementary schools in Ulsan City. Dust samples were collected from the living rooms and bedding materials in each home during summer and winter with a vacuum cleaner and analyzed for house dust mites using enzyme-linked immunosorbent assay (ELISA). Environmental factors and family history were surveyed during sampling. Results: Both species of house dust mite Dermatophagoides farina (Der f 1) and Dermatophagoides pteronyssinus (Der p 1) were found in all homes. Derf1 concentrations were much higher than Der P1 concentrations, both in atopic homes (1518.9 vs. 27.0 ng/g. dust, respectively) and non-atopic homes (810.8 vs. 44.3 ng/g. dust, respectively). Der f 1 concentrations were significantly higher in the atopic group than in the non-atopic group (atopic: 1518.9 ng/g.dust, non-atopic: 810.8 ng/g.dust, p = 0.035). However, total house dust mite Der p 1 concentrations were significantly higher in the non-atopic group than in the atopic group (atopic: 27.0 ng/g.dust, non-atopic: 44.3 ng/g.dust, p = 0.035). Multiple regression implied that mothers with family history of atopic diseases (OR = 4.79, 95% CI = 1.81-12.69), Der f 1 concentrations (OR = 1.74, 95% CI = 1.07-2.81), and air freshener use (OR = 4.60, 95% CI = 1.72-12.34) had significant associations with atopic children. Conclusion: House dust mite Der f 1 concentrations were associated with atopic children. This study suggests that parents should reduce house dust mite Der f 1 concentrations through environmental controls.

집먼지진드기에서 분리한 용출단백질의 개옴진드기 감염증에 대한 항원효과 (Antigenetic effects of the eluted proteins from house dust mite (Dermatophagoides pteronyssinus) in dogs infested with sarcoptic mite (Sarcoptes scabiei var. canis))

  • Canine sarcoptic mite (Sarcoptes scabiei var. canis) is ectoparasite which burrow usually in the stratum corneum of the skin of dogs. Antigens from the burrowing mites induce humoral and cellmediated immune responses in the hosts. The effect of antigenecity induced by somatic antigens of house dust mite (Dermatophagoides pteronyssinus) isolated by continuous elution has been evaluated in canine sarcoptic mites infestation. Continuous elution was carried out in 7.5% SDS-PAGE to isolate proteins of common antigens from somatic antigens of house dust mite. These eluted proteins from somatic antigens of house dust mite were confirmed by Western blotting in 7.5% SDS-PAGE, and eluted proteins (65, 60 kDa) were isolated. To evaluate the antigenetic effect of eluted proteins, eight dogs were divided as 4 groups such as non-vaccinated and non-challenged control (Group I), challenged control (Group II), vaccinated (Group III), and vaccinatedandchallenged (Group IV) groups. Group II and IV were artificially infested canine sarcoptic mites. Group III and IV were immunized with eluted proteins (65, 60 kDa). At the 6th week of the vaccination, the antibody titers of Group of IV were statistically significant higher than those of Group II (p<0.05). And antibody titers of Group III were also statistically significant higher than those of Group I (p<0.05). From these result, it is possible to replace somatic antigens of canine sarcoptic mites with eluted proteins from somatic antigens of house dust mites in order to diagnose and prevent the canine sarcoptic mite infestations.

Association of Anti-apoptotic Mechanism Due to House Dust Mite in Neutrophils with Protein Synthesis and Bad

  • House dust mite is an essential allergen in the pathogenesis of allergic diseases. Abnormal regulation of neutrophil apoptosis is an important pathogenic process in allergic diseases. In the present study, we investigated the effects of house dust mites on spontaneous apoptosis of neutrophils and its associated mechanisms. Extract of Dermatophagoides pteronissinus (DP) inhibited neutrophil apoptosis in a time-dependent manner. Cycloheximide (CHX), an inhibitor of translation, increased apoptosis of DP-treated neutrophils as well as control cells. The pro-apoptotic effect of CHX was blocked by DP in neutrophils. In addition, DP increased the phosphorylation of Bad in a time-dependent manner, indicating that it exerted an inhibitory effect on the function of Bad. These results suggest that DP has anti-apoptotic effects of neutrophils and may regulate protein synthesis and activation of Bad. Moreover, these findings may shed light on elucidation of allergy pathogenesis due to house dust mites.

  • The exposure to house dust mites is closely connected with allergic diseases such as atopic dermatitis, allergic rhinitis and asthma. Control of house dust mite populations has been principally achieved by using chemical insecticides. But the risk to human health would be a potential problem. Moreover, house dust mite remain as allergens even after death. Therefore, repelling the house dust mites is more effective method than killing them. The use of plant-derived repellents has been considered as a promising alternative to chemical repellents. Eucalyptus oils are regarded as safe compounds, there have been attempts to commercialize and market the insecticides/repellent products containing Eucalyptus oil as such or based upon them. For that reason, we experimented repellent activity against these mites using laundry detergent with Eucalyptus oil in this experiment. House dust mites (Dermatophagoides farinae and Dermatophagoides pteronyssinus) were exposed at different concentrations (4, 8, 16, $32{\mu}L/mL$) in fabric treated laundry detergent with Eucalyptus oil for 24 hours. The repellent effects (%) of laundry detergent with Eucalyptus oil exposed at 4, 8, 16, and $32{\mu}L/mL$ were 48.16%, 74.62%, 52.67%, and 44.47%, respectively. These results suggest that laundry detergent with Eucalyptus oil have potential effect to repel the house dust mites.

Isolation of Human scFv Antibodies Specific for House Dust Mite Antigens from an Asthma Patient by Using a Phage Display Library

  • Background: In order to characterize human antibodies with specificity for mite allergens at the molecular level, a scFv phage display library was constructed using peripheral blood mononuclear lymphocytes from an asthma patient allergic to mite as Ig gene sources. Methods: Immunoglobulin $V_H$ and V gene fragments were obtained by polymerase chain reaction, and randomly combined in pCANTAB-5E vector. The resulting human scFv phage display library had $3{\times}10^4$ independent clones, and biopanning was performed with house dust mite extracts. Results: Four scFv clones specific for house dust mite extract were isolated. Immunoblot assay showed that our clones reacted to 25 kDa and 50~60 kDa proteins with unknown identity in mite extracts. Sequence analysis indicated that two clones (b7 and c15) are identical, and all clones belong to human $V_H3$ subgroup. On the other hand, light chain usage was different in that two clones (a2 and b7 / c15) belonging to V ${\kappa}4$ subgroup, but a4 used V ${\kappa}1$ light chain gene. Conclusion: Our approach should facilitate provision of useful information on the antibody responses against allergens at the molecular level in humans.

Effect of House Dust Mite and CCL2 on S100A8 and S100A9 Expression in Human Monocytes

  • The S100A8 and S100A9 proteins play important roles in inflammatory diseases. The house dust mite acts as a major allergen that induces allergic diseases. We investigated the effect of the house dust mite on S100A8 and S100A9 protein expression in monocytes. We also examined the effect of CCL2, a powerful monocyte chemoattractant, on the expression of both proteins. Extract of Dermatophagoides pteronissinus (DP), recombinant Der p 1 and Der p 2, or CCL2 had no effect on S100A8 and S100A9 expression in human monocytic THP-1 cells. Monocytes were isolated from healthy donors and treated with DP, Der p 1, and Der p 2. S100A8 expression in monocytes increased after a 24 h stimulation with DP, Der p 1, and Der p 2, and CCL2 also increased S100A8 production. However, S100A9 expression in monocytes was not altered by DP, Der p 1, Der p 2, or CCL2. These results indicate that house dust mite and CCL2 may trigger an inflammatory response by altering S100A8 expression.

알레르기 환자 가정에서 생물학적 유해인자에 대한 노출평가 -세균, 진균, 집먼지 진드기 알레르겐, 내독소를 대상으로 (Exposure Assessments on Biological Contaminants in Homes of Allergy Patients - Bacteria, Fungi, House Dust Mite Allergen and Endotoxin)

  • To assess exposure of allergy patients to a biological environment, measurements were made of levels of airborne bacteria and fungi, house dust endotoxin and mite allergens in homes of 7 allergy patients and 12 healthy families. Concentrations of airborne total bacteria and the ratio of indoor and outdoor concentrations(I/O) in allergy patient's homes were high compared to non-allergy houses. But no significant differences could be shown for the concentration of spores of viable fungi and staphylococcus in indoor air between the homes of allergic patients and healthy families. The results of investigation on house dust mites, Dermatophagoids farinae were detected in all mattress samples and the levels in the allergy patient's homes were generally high, with individual measurements exceeding $2{\mu}g/g$ dust found in $30\%$ samples. In contrast with, Dermatophagoids pteronyssinus were detected in only $60\%$ samples and the concentrations were very low. The levels of endotoxin in dust samples collected from the allergy patient's home mattresses were higher than those of control houses. There was a positive correlation between the endotoxin levels and the house dust mite allergens.
