홍미삼과 홍삼정박의 건열처리가 Fusarium owsporum의 균사 생장과 승홍에 대한 내성에 미치는 영향 (Effect of Dry Heat Treatment of Red Ginseng and Red Ginseng Residue on Mycelial Growth and on Induced Tolerance of Fusarium oxysporum to Mercury Chloride)

  • Korean red ginseng and water extract residue of red ginseng roots were treated with dry heat and incorporated in PDA medium to examine the effect of the materials on induced tolerance against mercury chloride and mycelial growth of Fusarium oxysporum. Ginseng residue was not effective in the inducement of tolerance to mercury chloride regardless of dry heat treatment. However, the heat treatment of ginseng and ginseng residues stimulated the mycelial growth of the fungus. The materials responsible for the detoxification appeared to be water-soluble. The stimulation of the fungal mycelial growth on the media by the heat treatment was highest in the water extract of ginseng. Due to the heat treatment, the mycelial growth was also slightly increased in n-hexane and methanol extracts of ginseng, compared with the ginseng fractions without dry heat treatment.

벼 밀양 23호$\times$기호벼의 재조합 자식계통에서 수발아성,건열 저항성 및 종자수명간 상관관계 (Relationships among Viviporous Germination, Dry-heat Tolerance and Seed Longevity in Milyang 23/Gihobyeo RILs)

  • 밀양 23호/기호벼 유래 164 RILs($F_{17}$)을 이용하여, 종자의 휴면과 관련이 있는 수발아 저항성, 건열 저항성 및 종자수명을 검정하고, 이들 상호간 상관관계를 조사하였다. 출수 40일째에 밀양23호와 기호벼의 수발아율은 0%, 7%이었고, M/G RILs은 0$\sim$50.9%의 범위를 보였으며, 평균 3.2%였다. $90^{\circ}C$, 24시간 건열처리후 밀양23호와 기호벼는 99.2%, 37.6%의 발아율을 보였고, $T_{50}$은 2.7일, 12.9일로 유의한 차이를 보였으며, M/G RILs의 평균 발아율은 72.4%였다. 상온저장 한 종자수명은 밀양23호는 수확후 3.5년까지도 80% 이상의 종자 발아율을 유지하였으나, 기호벼는 18개월 후부터 급격히 발아력이 감소하여(30%이하), 42개월 후 발아력을 완전히 상실하였다. M/G RILs의 종자활력은 수확후 42개월까지 71계통이 90%이상의 발아율을 보였다. 수발아 저항성, 건열 저항성 및 종자수명은 밀양 23호와 기호벼 간에 뚜렷한 품종간 차이를 보였으며, M/G RILs은 양친의 중간값에 가까운 평균값을 보였다. 건열저항성과 종자수명 간에는 고도로 유의한 정의 상관관계를 보였고, 수발아와 종자수명, 수발아와 건열저항성 간에는 부의 상관관계가 성립하였다.

열, 살균소독제, 항생제에 의한 분리 Cronobacter spp. (Enterobacter sakazakii) 제어 (Biocontrol of Isolated Cronobacter spp. (Enterobacter sakazakii) by Heat, Sanitizer, and Antibiotic)

  • 국내의 건조분말 식품에서 분리한 Cronobacter spp.의 건조내성에 따른 열, 살균소독제, 항생제에 대한 내성특성 분석하여 Cronobacter spp.을 제어하는 효과적인 방법을 연구하고자 하였다. 분리된 37개의 건조저항 균주, 7개의 건조민감 균주, 그리고 68개의 중간그룹의 균주 총 112개에 대하여 분석한 결과 건조내성을 가지고 있는 Cronobacter spp.는 열에 대해서도 저항성이 큰 것으로 나타났다. 따라서 조제분유등의 건조 스트레스에 노출되어 살아남은 Cronobacter spp.는 열 스트레스에서도 높은 내성을 보일수 있을 것으로 사료된다. 살균소독제를 이용한 Cronobacter spp.의 제어효능은 sodium hypochlorite는 15-25 ppm와 benzalkonium chloride는 5-15 ppm 농도에서 5 log CFU/mL 살균 효과가 있었다. Albumin으로 3%(w/v)오염시킨 부유세균의 살균력은 건조내성 균주와 건조민감 균주 모두에서 오염전과 비슷하여 유기물 오염상태에서도 살균소독제 효능에는 큰 영향을 주고 있지 않음을 알 수가 있었다. 그리고 항생제 내성은 $\beta$-lactam 계열의 ampicillin에 대하여 약 90% 분리주가 64-128 ppm의 농도에서 최소저해 농도를 보여 주었다. Aminoglycoside 계열의 kanamycin과 Gram(-) spectrum의 nalidixic acid에서 4 ppm의 가장 낮은 내성을 보였다. 건조민감 균주과 건조내성 균주의 최소저해농도 경우 전반적으로 모든 항생제에 대해서 건조 내성이 높은 균주가 더 큰 항생제 내성을 지니고 있는 것으로 나타났다. 따라서 분리 Cronobacter spp.의 건조내성이 열과 항생제의 저항성과 관련이 있는 것으로 보인다. 그러므로 조제분유와 같은 분말식품과 그 원료의 오염 Cronobacter spp.의 살균제어에 더 강력한 처리가 필요할 것으로 보인다.

Etching characteristics of Al-Nd alloy thin films using magnetized inductively coupled plasma

  • For advanced TFT-LCD manufacturing processes, dry etching of thin-film layers(a-Si, $SiN_x$, SID & gate electrodes, ITO etc.) is increasingly preferred instead of conventional wet etching processes. To dry etch Al gate electrode which is advantageous for reducing propagation delay time of scan signals, high etch rate, slope angle control, and etch uniformity are required. For the Al gate electrode, some metals such as Ti and Nd are added in Al to prevent hillocks during post-annealing processes in addition to gaining low-resistivity($<10u{\Omega}{\cdot}cm$), high performance to heat tolerance and corrosion tolerance of Al thin films. In the case of AI-Nd alloy films, however, low etch rate and poor selectivity over photoresist are remained as a problem. In this study, to enhance the etch rates together with etch uniformity of AI-Nd alloys, magnetized inductively coupled plasma(MICP) have been used instead of conventional ICP and the effects of various magnets and processes conditions have been studied. MICP was consisted of fourteen pairs of permanent magnets arranged along the inside of chamber wall and also a Helmholtz type axial electromagnets was located outside the chamber. Gas combinations of $Cl_2,{\;}BCl_3$, and HBr were used with pressures between 5mTorr and 30mTorr, rf-bias voltages from -50Vto -200V, and inductive powers from 400W to 800W. In the case of $Cl_2/BCl_3$ plasma chemistry, the etch rate of AI-Nd films and etch selectivity over photoresist increased with $BCl_3$ rich etch chemistries for both with and without the magnets. The highest etch rate of $1,000{\AA}/min$, however, could be obtained with the magnets(both the multi-dipole magnets and the electromagnets). Under an optimized electromagnetic strength, etch uniformity of less than 5% also could be obtained under the above conditions.

국내에 도입된 Sedum album L.의 생육 특성 및 저토심 옥상 녹화 시스템에 관한 연구 (Studies on Growth Characteristics and Shallow Green-Roof Systems of Sedum album L. Introduced in Korea)

  • These studies were carried out (1) to investigate the growth characteristics of Sedum album L. in the field, (2) to propose a suitable shallow peen roof system for this plant, and (3) to evaluate plant growth in the proposed system over the long term. The growth characteristics, such as morphological properties, growth habit, shade tolerance, and flowering, were surveyed. In experimental shallow green-roof systems, the effects of drainage type, substrate type, and soil depth on plant growth were investigated. Then drought tolerance was investigated. After planting Sedum album L. in the proposed system survival rate, cover, and resistance to insects, heal and cold were evaluated for about 2 years. The results of these studies are summarized below. 1. In the field, the aboveground part of Sedum album L. did not die back during the winter. Plant height was 4$\sim$7 cm. Roots were distributed to a depth of 5$\sim$7 cm. Sedum album L. is a compact ground-cover plant that spreads vigorously. Shading condition of less than $30\%$ of full sunlight didn't cause any trouble, but shading conditions above $87\%$ made the shape of the shoots and leaves abnormal. The plant bloomed from June to August and had a rather large compound umbel of white, star-shaped flowers. 2. Two systems, a drainage-blend-10 cm soil depth and a reservoir$\cdot$drainage-blend-15 cm soil depth, performed best in terms of cover, fresh weight, and dry weight. The first has an advantage for green roofs because it is lighter than the latter. 3. In drainage-blend-10 m soil depth and modified reservoir · drainage-blend-10 cm soil depth system no plants died for about 4 months after stopping the irrigation. The visual quality of the latter system was above 5 for 4 months and that of the former was under 5 after 2 months. In the field, however, the drought tolerance of Sedum album L. grown in the former would be enough to withstand the dry season. Considering the urban ecosystem and the importance of healthy growth the modified reservoir $\cdot$ drainage-blend-10 cm soil depth system was finally recommended. This system was composed of a 4 cm thick drainage layer and drain outlets placed at a height of 2.5 cm. 4. In the proposed system, the survival rate was $100\%$, and there was no injury induced by insects and heat. The leaf density decreased a little in winter. Cover increased throughout the year. Sedum album L. was planted with a cover of 72$cm^{2}$ on 3 April 2003; on 16 June 2003 and 15 June 2004, cover was $132.66\pm$5.87 $cm^{2}$(1.8 times) and $886.98\pm$63.51 $cm^{2}$(12.3 times), respectively.

Hybridization system을 이용한 유산균의 장용성 건식 피복 (Dry Enteric Coating Process of Lactic Acid Bacteria by Hybridization System)

  • 유산균의 활용성을 증진시키기 위한 방안으로서 건식 Hybridization system을 이용하여 장용성 피복재로 유산균분말의 표면처리를 실시하였다. 전자현미경 관찰 결과 표면처리는 유산균의 표면을 매끄러운 구형의 모양으로 변화시켰으며 표면처리를 통한 분체복합화 과정에서 유산균의 활력은 유의적인 변화없이 유지되었다. 또한 유산균의 내염성은 처리여부에 따른 유의적 변화를 보이지 않음으로써 표면처리 과정 중 유산균 세포막의 물리적 손상은 일어나지 않은 것으로 판단된다. 표면처리된 유산균의 내산성은 처리전후에 차이를 나타내지 않았으나 이는 균주 자체의 높은 내산성에 기인한 것으로 판단된다. 표면처리 후의 유산균은 유의적으로 높은 열저항성을 보여 표면처리가 유산균의 내열성 향상을 위하여 활용될 수 있는 가능성을 나타냈다. 조사된 장용성 피복재 중 Sureteric은 다른 피복재에 비하여 우수한 표면 처리 효과를 보였으며 유산균분말의 표면처리를 위한 적합한 처리조건은 유산균의 초기입도가 $100{\sim}200\;{\mu}m$, 유산균:피복재의 혼합비율(w/w)은 9 : 1, 처리속도는 15,000 rpm, 3분이었다.

Heat tolerance of goats to increased daily maximum temperature and low salinity of drinking water in tropical humid regions

  • Objective: The daily maximum temperature and seawater level continuously increase as global warming continues. We examined the adaptability and production performance of heat-stressed goats with a supply of low-saline drinking water. Methods: Twelve Kacang and Kacang Etawah cross goats were exposed to two climatic conditions (control, 25℃ to 33℃, 83% relative humidity [RH], temperature humidity index [THI]: 76 to 86; and hot environment, 26℃ to 39℃, 81% RH, THI: 77 to 94) and two salt levels in drinking water (0% and 0.4% NaCl). The experimental design was a Latin Square (4×4) with four treatments and four periods (28 days each). Results: Temperature of the rectal, skin, and udder, and respiration rate rose, reached a maximum level on the first day of heat exposures, and then recovered. Plasma sodium rose at 0.4% NaCl level, while the hot environment and salinity treatments increased the drinking water to dry matter (DM) intake ratio. Water excretion was elevated in the hot environment but lowered by the increase in salinity. Total lying time increased, whereas change position frequency decreased in the hot condition. Lying and ruminating and total ruminating time increased and explained the enhanced DM digestibility in the hot conditions. Conclusion: The goats exhibited a high level of plasma sodium as salinity increased, and they demonstrated physiological and behavioral alterations while maintaining their production performances under increasing daily maximum temperatures.

Effective Heat Treatment Techniques for Control of Mung Bean Sprout Rot, Incorporable into Commercial Mass Production

  • Seedlot disinfection techniques to control mung bean sprout rot caused by Colletoricum acutatum and C. gloeosporioides were evaluated for commercial production scheme. Soaking seedlots in propolis (100 X) and ethanol (20% for 30 min) appeared promising with control values of 85.5 and 80.8 respectively, but still resulted in up to 20% rot incidence. None of the C. acutatum conidia survived through hot water immersion treatment (HWT) for 10 min at temperatures of 55, 60 and $65^{\circ}C$, whereas the effective range of the dry heat treatment (DHT) was $60-65^{\circ}C$. Tolerance of mung bean seedlot, as estimated by hypocotyl elongation and root growth, was lower for HWT than for DHT. Germination and growth of sprouts were excellent over the range of $55-65^{\circ}C\;at\;5^{\circ}C$ intervals, except for HWT at $65^{\circ}C$ for 5 min. At this marginal condition, heat damage appeared so that approximately 2% of seeds failed to sprout to normal germling and retarded sprouts were less than 5% with coarse wrinkled hypocotyls. These results suggested that DHT would be more feasible to disinfect mung bean seedlots for commercial sprout production. Heat treatment at above ranges was highly effective in eliminating the epiphytic bacterial strains associated with marketed sprout rot samples. HWT of seedlot at 55 and $60^{\circ}C$ for 5 min resulted in successful control of mung bean sprout rot incidence with marketable sprout quality. DHT at 60 and $65^{\circ}C$ for 30 min also gave good results through the small-scale sprouting system. Therefore, we optimized DHT scheme at 60 and $65^{\circ}C$ for 30 min, considering the practical value of seedlot disinfection with high precision and accuracy. This was further proved to be a feasible and reliable method against anthracnose incidence and those bacterial strains associated with marketed sprout rot samples as well, through factory scale mung bean sprout production system.

하절기 홀스타인 젖소의 행동 및 체온에 미치는 환경 효과 (The Environmental Effects on the Activities and Rectal Temperatures of Holstein Cows in a Summer Season)

  • 본 연구는 하절기 홀스타인 젖소의 행동과 체온(내서성)에 관하여 환경효과를 추정하기 위하여 1999년 7월 6일부터 13일까지 수행하였다. 행동지수를 조사하기 위하여 Holstein 젖소 77두를 공시하였으며 사양은 NRC 사양표준에 의거 농후사료와 옥수수 사일리지 위주로 관리하였다. 체온은 스탄촌에 계류된 상태에서 오후 1시부터 3시 사이에 직장내 온도를 측정하였으며 이때 기온은 $29.0^{\circ}C$ 이었다. 건유 및 착유 상태, 산차에 따라 체온이 달랐으나(p<0.01), 행동지수에 대하여 착유우와 건유우는 오전 9시부터 정오까지, 오후 1시부터 4시까지는 차이가 없었다. 착유우의 직장내 체온은 39.0${\pm}0.03^{\circ}C$으로서 건유우의 체온(38.6 ${\pm}0.04^{\circ}C$)보다 높았다. 오후 1-4시까지는 3산차 이하의 개체들이 4산차 이상의 개체들에 비하여 행동지수가 높았고, 산차의 증가에 따라 감소하는 경향이었다. 우유 성분과 체온과의 상관에 대하여 유당을 제외한 지방, 단백질, 무지고형분율 및 총고형분 등과는 부의 표현형 상관을 나타내었다.
