Development of GMAW Process with Twin Torch for Wide Overlay using Compound Filler Plate (분말 용가재판을 사용한 광폭 오버레이용 트윈토치 GMAW 공정개발)

  • Generally, wear plate is steel plate having improved surface contact strength and impact strength by surface hardening which is welded using materials with good corrosion resistance, wear resistance and thermal resistance property. CFP GMAW(Compound Filler Plate Gas Metal Arc Welding) is the cladding method using GMAW with the CFP, which is bound with waterglass, on the substrate. It has advantages of reducing compound powder loss, uniform penetration, and preventing hardness decrease. To develope mass production technique of CFP GMAW process for production of high quality wear plate, the method for controling shallow penetration and increasing productivity is required. In this study, twin torch method applied to CFP GMAW process for increasing productivity. And the method was developed by controling penetration control, CFP dry time, gas formation flux and water glass concentration. As a result, applying twin torch method to CFP GMAW process was possible and high quality wide bead could be made without overlap joint.

Studies on Production of High-Quality Cornus controversa Container Seedlings (층층나무 우량 용기묘 생산에 관한 연구)

  • This study was carried out to develop the technique for the production of high quality container seedlings of Comus controversa Hemsley. The seedlings were grown on the medium of peatmoss : perlite : vermiculite (1:1:1, v/v) in plastic net container and no-net plastic container as control for five months in the PE house. The seedlings grown in the plastic net container showed better root collar diameter growth, better development of long and fine roots, more increment of dry mass of roots and shoots than those grown in the no-net container. But the height growth of the seedlings in both container was similar. In particular the seedlings grown in plastic net container had no spiraling roots which were always observed in the control container seedlings. This result was induced by air-root pruning effect from the plastic net container.

  • A yeast strain utilizing styrene epoxide as a sole carbon and energy source was isolated from soil samples for the production of enantiopure of styrene epoxide by kinetic resolution. The strain, identified as a Rhodosporidium toruloides SJ-4, expressed an epoxide hydrolase which preferentially converted (R)-styrene epoxide into the corresponding diol. A maximum activity of 135 U/L was observed when biomass (dry cell mass) reached 6.7 g/L at 21 h of batch culture. Under the partially optimized reaction conditions ($35^{\circ}C$ and pH 8.0), the optically pure (S)-styrene epoxide was obtained with the yield of 21% when the initial substrate concentration was 100 mM. The reaction was completed at 9 h.

Citric Acid Production and Scale-up in Dual Hollow Fiber Bioreactor (이중실관 생물 반응기에서의 구연산 생산과 Scale-up)

  • A study on the citric acid production was performed in various size dual hollow fiber bioreactors with immobilized Aspergillus niger (KCTC 1232). The final dry cell mass density reached 300g/l based on the space volume available for cell growth. Under air and oxygen aeration the volumethe productivity reached 0.63 and 1.02g/l.h, which cormsponded to 10 and 16 fold over those of batch fermentation, respectively. The initial pH of the medium was a critical factor and the lower value resulted in higher citric acid yield. The increase in the feeding rate of medium or the number of reactor unit resulted in the improvement of the productivity due to higher consumption rate of substrate.

Manure Based Duckweed Production in Shallow Sink : Effect of Nutrient Loading Frequency on the Production Performance of Lemna purpusilla

  • Effect on the bio-mass and nutrient yield of Lemna perpusilla Torry was studied at two nutrient loading frequencies. Six polythene sinks ($6{\times}2{\times}0.5m^{3}$ area) randomly allocated to two treatments having three sinks in each treatment. Each sink containing $6{\times}2{\times}0.35m^{3}$ water body, was changed either daily with 12.6 kg anaerobically fermented cattle manure (effluent) or with 76 kg efluent at six-day interval. Media Kjeldhal-N (40 vs. 36 mg/L; p>0.05) and $NH_3$-N (9 vs 7 mg/L; p<0.05) concentration were higher in daily charged sinks than those charged at six-day interval. However, temperature ($28.48{^{\circ}C}\;vs.\;28.60{^{\circ}C}$) and pH (6.82 vs. 6.84) were sinilar in both groups of sinks. Biomass (93 vs. $80g/m^{2}/d$) and crude protein (1.24 vs. $1.11g/m^{2}/d$) yield were non-significantly (p>0.05) higher in the daily charged than 6-d interval charged ponds. However, dry matter (4.52 vs. 4.15%), organic matter (83 vs. 84%), crude protein (31.4 va. 29.6%) and organic carbon (47 vs. 47%) content were similar in both daily and 6-d interval charged ponds. There were no apparent relationships between the crude protein content of Lemna and the media Kjeldhal-N or $NH_3$-N concentration apparently due to saturated level of nutrient (N in particular). It was concluded that with the level of effluent used for two treatments, nutrient-loading frequency had no significant effect on the biomass and protein yield of Lemna. Technique described here can be used for year-round duckweed production from fermented cattle manure for feeding poultry/fish in smallholder production system.

Growth Characteristics of Microalgae Scenedesmus obliquus by LED Light Source (LED 광원에 따른 미세조류 Scenedesmus obliquus의 성장 특성)

  • Microalgae are independent organisms that perform photosynthesis and can alter the culture environment to increase accumulation of useful substances derived from microalgae. In this study, cell growth was measured by incubation for 39 days using MBBM, Neo medium, and seven light sources, which is the main factor affecting cell growth of microalgae S. obliquus. In the case of S. oliquus, which grew in MBBM and Neo medium, cell growth was highest under fluorescent light sources and Red2 LED (R660) light sources, and cell growth was lowest under Infra Red LED (R741) light sources. The average cell growth rate was 17.7% for MBBM and 15.4% for Neo. Comparing the effects of dry cell weight of Neo medium containing nutrients on the production of aquatic plants, MBBM and dry cell weight of Neo resulted in higher cell growth than Neo medium under all LED light sources except for Blue LED (B450). This proves that MBBM is more suitable for increasing the cell growth of microalgae than Neo medium and confirms that light source selection is important in the production of useful materials through mass cultivation of microalgae in the future.

Effects of cultivation methods on methane emission in rice paddy

  • Methane is the main greenhouse gas released from rice paddy field. Methane from paddy fields accounts for 11 % of the global total methane emission. The global warming potential (GWP) of methane is 25 times more than that of carbon dioxide on a mass basis. It is well known that most effective practice to mitigate methane in paddy is related to the water management during rice growing season and the use of organic matters. This study was conducted to investigate the effects of tillage and cultivation method on methane emission in paddy. Tillage (tillage and no-tillage) and cultivation methods (transplanting and direct seeding) were combined tillage-transplanting (T-T), tillage-wet hill seeding (T-W), tillage-dry seeding (T-D) and no-till dry seeding (NT-D) to evaluate methane mitigation efficiency. Daily methane emission was decreased on seeding treatments (T-W, T-D, NT-D) than transplanting treatment (T-T). Amount of methane emission during rice growing season is highest in T-T ($411.7CH_4\;kg\;ha^{-1}y^{-1}$) and lowest in NT-D treatment (89.7). In T-W and T-D treatments, methane emissions were significantly decreased by 36 and 51 % respectively compared with T-T. Methane emissions were highly correlated with the dry weight of whole rice plant ($R^2=0.62{\sim}0.93$). T-T treatment showed highest $R^2$ (0.93) among the four treatments. Rice grain yields did not significantly differ with the tillage and cultivation methods used. These results suggest that direct seeding practice in rice production could mitigate the methane emissions without loss in grain yield.

Growth of Strawberry Plantlets Cultured in Vitro in the Agar or Commercial Plug Medium as Affected by Ionic Strength (이온강도에 따른 Agar와 공정육묘용 상토에서 기내배양된 딸기 소식물체의 생육)

  • This study was conducted to investigate the effect of supporting material and ionic strength of the MS medium on growth of strawberry plantlets cultured in vitro for the rapid mass production. Explants of $Fragaria$ $ananassa$ 'Houkouwase' in vitro were planted in the agar or Tosilee (commercial plug medium) medium as the supporting material and supplied with 1/4 MS, 1/2 MS or basal (as the control) MS medium in an autotrophic micropropagation. Plant height and root length were significantly greater when they were cultured in 1/2 MS medium as compared to those grown in the agar medium. Also, shoot fresh and dry weights, and leaf area in the 1/2 MS medium were greater than in 1/4 MS or basal MS medium. When plantlets were cultured in Tosilee medium and fed with the basal MS medium, plant height, root length, shoot fresh and dry weights, root fresh and dry weights, and leaf area were promoted and greater than those in plantlets cultured in the agar medium.

Physical and Mechanical Properties of Wood Fiber-Polypropylene Fiber Composite Panel

  • This study was to find a way of reusing wood and plastic wastes, which considered as a troublesome problem to be solved in this age of mass production and consumption, in manufacturing wood fiber-polypropylene fiber composite panel. And the feasibility of this composite panel as a substitute for existing headliner base panel of automobile was also discussed, especially based on physical and mechanical performance. Nonwoven web composite panels were made from wood fiber and polypropylene fiber formulations of 50 : 50, 60 : 40, and 70 : 30, based on oven-dry weight, with densities of 0.4, 0.5, 0.6, and 0.7 g/$cm^3$. At the same density levels, control fiberboards were also manufactured for performance comparison with the composite panels. Their physical and mechanical properties were tested according to ASTM D 1037-93. To elucidate thickness swelling mechanism of composite panel through the observation of morphological change of internal structures, the specimens before and after thickness swelling test by 24-hour immersion in water were used in scanning electron microscopy. Test results in this study showed that nonwoven web composite panel from wood fibers and polypropylene fibers had superior physical and mechanical properties to control fiberboard. In the physical properties of composite panel, dimensional stability improved as the content of polypropylene fiber increased, and the formulation of wood fiber and polypropylene fiber was considered to be a significant factor in the physical properties. Water absorption decreased but thickness swelling slightly increased with the increase of panel density. In the mechanical properties of composite panel, the bending modulus of rupture (MOR) and modulus of elasticity (MOE) appeared to improve with the increase of panel density under all the tested conditions of dry, heated, and wet. The formulation of wood fiber and polypropylene fiber was considered not to be a significant factor in the mechanical properties. All the bending MOR values under the dry, heated, and wet conditions met the requirements in the existing headliner base panel of resin felt.

Cultural Characteristics and Scale-up for Submerged Cultivation of Hericium erinaceum Through Air-lift and Jar Fermenter System (Air-lift 및 Jar Fermenter에 의한 Hericium erinaceum 심부배양의 배양특성 및 Scale-up)

  • For the study of Hericium erinaceum as a useful functional foods and materials, liquid cultivation under two different bioreactors(air-lift fermenter and jar fermenter) which was not studied systematically until now, was conducted as a method of mass cultivation for H. erinaceum. A batch cultivation in an air-lift fermenter and a jar fermenter was examined for enhancing the productivity because of small amounts of mycelial weight and slow growth in case of a liquid culture for H. erinaceum. We found that air lift fermenter system was more effective than jar fermenter for mycelial production of H. erinaceum, and mycelial morphology was a critical factor of the growth. By scale-up and cultivation based on morphological analysis, the conditions for mass production with 30 L and 500 L jar fermenter was 200 and 150 rpm of agitation speed at 1 vvm of aeration rate, respectively, and mycelial dry weight under these conditions was enhanced to about $13{\sim}14g/L$.
