나프록센 함유 방출제어형 패취의 제제설계 및 평가 (Formulation and Evaluation of Controlled Release Patch Containing Naproxen)

  • The purpose of this study is to prepare the controlled release adhesive patch containing naproxen. Pressuresensitive adhesive (PSA)-type patch was fabricated by casting of polyisobutylene (PIE.) and mineral oil in toluene. Membrane-controlled release (MCR)-type patch was prepared by the attachment of the controlled release membrane on the PSAtype patch. The membrane was mainly composed of Eudragit, polyethylene glycol(PEG) and glycerin. The drug release profile and skin permeation test with various patches were evaluated in vitro. The release of naproxen from PIE-based PSAtype patch with various loading doses fitted Higuchi's diffusion equation. However, the permeation of naproxen through hairless mouse skin from PSA-type patch followed zero-order kinetics. In MCR-type patch, thickness of controlled release membrane affected on the drug release rate highly. In the composition of membrane, the release rate was decreased as the ratio of Eudragit increased. The drug release from the MCR-type patch followed zero order kinetics. The permeation of naproxen through hairless mouse skin from MCR-type patch showed lag time for the intial release period and didn't fit the zero-order kinetics

Hydroxypropylmethylcellulose로부터 염산슈도에페드린의 방출조절에 관한 연구 (A Study on the Control of Pseudoephedrine Hydrochloride Release from Hydroxypropylmethylcellulose Matrices)

  • Hydroxypropylmethylcelluloses (HPMC)는 친수성 cellulose 계통의 중합체로서 독성이 적고 가격이 저렴하여 경구용 서방출성 제제에 널리 이용되고 있다. 본 연구에서는 매트릭스로서 HPMC를 이용하여 새로운 서방출성제제인 HPMC/염산슈도에페드린계를 설계하고, 직타법에 의하여 정제를 제조하였으며, 이 팽윤될 수 있는 친수성 약물전달계를 이용하여 약물의 방출에 영향을 주는 여러 인자들이 약물의 방출속도에 미치는 영향에 대하여 조사하였다. 결과로서, HPMC/염산슈도에페드린계에서 매트릭스인 HPMC의 분자량이 클수록, HPMC의 함량이 많을수록 약물의 방출속도가 더 느려짐을 알 수 있었으며, 정제의 경도나 용출시 pH 변화에는 무관한 것으로 나타났다. 특히, 음이온 계면활성제인 sodium laurylsulfate는 HPMC로부터 염산슈도에페드린의 방출속도를 지연시키는데 큰 영향을 미치는 것으로 나타났으며, 이러한 현상은 음이온 계면활성제인 sodium laurylsulfate가 반대의 이온성을 갖는 약물인 염산슈도에페드린과 착물을 형성함으로써 약물의 용해성이 떨어져서 약물방출이 지연되는 것으로 생각된다.

Precise ultrasonic coating and controlled release of sirolimus with biodegradable polymers for drug-eluting stent

  • In the current study, a drug-eluting stent coated with biodegradable polymers and sirolimus was developed by using an ultrasonic nanocoater and characterized in aspects of surface smoothness and coating thickness. In addition, in vitro release profiles of sirolimus by changing top coating layer with different biodegradable polymers were investigated. Smooth surfaces with variable thickness could be fabricated by optimizing polymer concentration, flow rate, nozzle-tip distance, gas pressure, various solvents and ultrasonic power. Smooth surface could be generated by using volatile solvents (acetone, chloroform, and methylene chloride) or post-treating with solvent vapor. Coating thickness could be controlled by varying injection volume or polymer concentration, and higher concentration could reduce the coating time while obtaining the same thickness. The thickness measurement was the most effectively performed by a conventional cutting method among three different methods that were investigated in this study. Release profiles of sirolimus were effectively controlled by changing polymers for top layer. PLGA made the release rate 3 times faster than PDLLA and PLLA and all top layers prevented burst release at the initial phase of profiles. Our results will provide useful and informative knowledge for developing drug-eluting stents, especially coated with biodegradable polymers.

염산에틸에프린의 마이크로캅셀에 관한 약제학적 연구 (Pharmaceutical Studies on Microencapsulated Etilefrine Hydrochloride)

  • Etilefrine hydrochloride was microencapsulated with ethylcellulose by phase separation method to develop a sustained release dosage form. The results of dissolution test carried out with various microcapsules showed that the drug release was decreased with increasing the particle size of microcapsules at a constant core to wall ratio, and with decreasing the core to wall ratio. Also ethylcellulose 50 cps and fast stirring rate (900 rpm) was better in decreasing the drug release than ethylcellulose 22 cps and slow stirring rate (300 rpm), respectively.

  • To explore the potential of shell crosslinked micelle (SCM) as a drug carrier, the drug release behavior of poly($\varepsilon$-caprolactone)-b-poly(acrylic acid) (PCL-b-PAA) SCMs was investigated. PCL-b-PAA was synthesized by ring opening polymerization of $\varepsilon$-caprolactone and atom transfer radical polymerization of tert-butyl acrylate, followed by selective hydrolysis of tert-butyl ester groups to acrylic acid groups. The resulting amphiphilic polymer was used to prepare SCMs by crosslinking of PAA corona via amidation chemistry. The drug release behavior of the SCMs was studied, using pyrene as a model drug, and was compared with that of non-crosslinked micelles, especially below the critical micelle concentration (CMC). When the shell layers were crosslinked, the drug release behavior of the SCMs was successfully modulated at a controlled rate compared with that of the non-crosslinked micelles, which showed a burst release of drug within a short time.

  • In order to study drug release from the suppository, three types of hollow suppositories and one conventional suppository were prepared using indomethacin(IDM) as a model drug and Witepsol H-15 as a base. The 4 types of suppository prepared are as follows: type I, conventional suppository containing 50 mg of IDM powder, type II, hollow supository containing 50 mg of IDM powder in the cavity, type III, hollow suppository containing 25 mg of IDM powder in the base and IDM microcapsules (25 mg as IDM powder) in the cavity, and type IV, hollow suppository containing IDM microcapsules (25 mg as IDM powder) in the base and 0.5 ml of 5%(w/v) IDM-PEG 300 solution in the cavity. The drug amount released(%) from type II and I within 24 hrs was 46.7% and 66.9%, respectively. Comparing with the drug amount released from four types of suppository within initial 2 hrs and 24 hrs, that of type IV was high as 32.7% and 76.6%, respectively. IDM-ethycellulose microcapsules passed through 270 mesh sieve and the IDM content was 20.95%.

압축코팅법에 의한 3단계 약물방출형 지속성제제의 제조 및 용출특성 (Preparation and Dissolution Characteristics of the Compression-Coated Controlled Release Tablet Exhibiting Three-step Release)

  • A novel oral controlled release tablet which may offer more uniform drug level in the body than simple zero-order was developed. The tablet is composed of three layers; outer film layer, middle part compression-coated hydroxypropylmethylcellulose (HPMC) matrix layer, and inner core layer. Each layer contains nicardipine HCl as a model drug. In vitro dissolution test showed that the tablet released the drug in clear three steps; a rapid initial release, followed by a constant rate of release, and then a second phase of fast release of drug. The dissolution characteristics could be modified easily by changing the grade of HPMC, thickness of matrix layer, content of methylcellulose in matrix layer, content of active ingredient in each layer. The pH of dissolution medium did not affect the release profile. This three-step release system is expected to raise the blood concentration rapidly to effective level and to maintain effective blood level longer than simple slow-release systems.

알긴산나트륨 마트릭스로부터 플루오로우라실의 제어 방출 (Controlled Release of Fluorouracil from Sodium Alginate Matrices)

  • The applicability of sodium alginate as a carrier of 5-fluorouracil as an oral delivery system was investigated. Hydrophobicity of sodium alginate was controlled by introducing cetyl group to this polymer. The effects of degree of esterification for n-cetyl partial ester on the rate of release of 5-fluorouracil in artificial gastric juice and artificial intestinal juice were examined. The release rete of the drug in the gastric juice was mainly affected by the diffusion of the drug. The release rate of the drug in the intestinal juice could be controlled by the degree of esterification. The alginate matrices may be a valuable addition as the carrier of 5-fluorouracil for an oral delivery system.

  • Tamsulosin or a salt thereof such as its hydrochloride salt has been known to have an adrenaline ${\alpha}$ receptor blocking action for urethra and prostate areas. It has been widely used as a drug which lowers the prostate pressure and improves urinary disturbance accompanied by prostate-grand enlargement, thus for the treatment of prostatic hyperplasia. To avoid dose-dependent side effects of tamsulosin upon oral administration, the development of sustained-release delivery system is essentially required, that can maintain therapeutic drug levels for a longer period of time. The aim of this study was therefore to formulate sustained-release tamsulosin granules and assess their formulation variables. We designed entric coated sustained-release tamsulosin granules for this purpose. Nano-pores in the outer controlled release membrane were needed in order to obtain initial tamsulosin release even in an acidic environment such as gastric region. In our sustained release osmotic granule system, hydroxypropylmethylcellulose in a drug-containing layer was used as a rate controller. The drug-containing granules were coated with hydroxypropylmethylcellulose phthalate (HPMCP) and Eudragit, along with glycerol triacetate as an aqueous nano-pore former. The release of tamsulosin depended heavily on the type of Eudragit such as RS, RL, NE 30D, used in the formulation of controlled release layer. These results obtained clearly suggest that the sustained-release oral delivery system for tamsulosin could be designed with satisfying drug release profile approved by the Korean Food and Drug Administration.

Effect of Crosslinking on Release of Model Drug from Electrospun Poly(vinyl alcohol) Fiber Mats

  • Electrospun fibers of poly(vinyl alcohol) (PVA) were successfully prepared and applied as drug carriers for transdermal drug delivery system. Sodium Salicylate (SS) was the model drug and it was incorporated in the PVA fibers by adding 20 % of SS in a PVA solution prior to electrospinning. Electrospinning of SS-containing PVA solution resulted in the formation of beaded fibers. In order to control the rate of SS release and decrease water solubility of PVA, the SS-loaded electrospun PVA mat was cross-linked by either glutaraldehyde or glyoxal vapor. The morphology, thermal behavior, swelling behavior, release characteristic, kinetics of drug release and also toxicity of the cross-linked sample were investigated.
