미나리 분말을 첨가한 만두피의 품질특성 (Quality characteristics of dumpling shell added with dropwort powder)

  • 본 연구는 생리 활성 기능을 지닌 미나리를 이용한 다양한 가공제품의 개발 및 미나리의 활용성 증대의 일환으로 미나리 분말 첨가에 따른 만두피의 제조 및 품질특성에 대하여 살펴보았다. 밀가루 대체 미나리분말 0, 1, 3, 5, 7%를 첨가하여 만두피를 제조한 결과 수분결합력은 미나리 분말이 첨가될수록 증가하였으며 수분함량은 미나리 분말 첨가량에 따른 유의적인 차이는 없었다. 만두피의 조리 특성에서는 무게와 수분흡수율과 부피는 대조군에 비하여 미나리 분말 첨가구가 높게 나타났으며 미나리 분말 첨가량이 많아짐에 따라 유의적으로 증가하는 경향을 보였다(p<0.001). 탁도는 미나리 분말 7% 첨가 만두피가 가장 높게 나타났으며 미나리 분말 첨가량이 증가할수록 용출성분이 유의하게 증가하였다(p<0.001). 미나리 분말 첨가 만두피의 DPPH radical 소거능은 미나리 분말 첨가량이 증가할수록 높게 나타났으며 색도에서는 미나리 분말 첨가량이 증가할수록 L값과 a값은 감소하였으며 b값은 증가하였다. Texture에서 경도는 대조군에 비해 미나리 분말 첨가구가 높게 나타났으며 탄력성과 응집성은 미나리 분말 첨가량이 증가할수록 유의적으로 높게 나타났다(p<0.001). 외관의 기호도에서는 미나리 분말이 많이 첨가될수록 선호도가 높았으며 향미, 맛, 질감, 전체적인 기호도에서 미나리 분말 3% 첨가 만두피가 기호도면에서 우수하게 평가되어 기능성과 기호적인 측면에서 도움을 줄 수 있는 만두피로 사용 될 수 있을 것이라 사료된다.

수경미나리의 영양성분 분석에 관한 연구 (Study on the Nutrition Composition of Hydroponic Water Dropwort)

  • This study was conducted to investigate the nutrient composition of hydroponic water dropwort and the effect of blanching condition on ascorbic acid content. Nutrient composition of hydroponic water dropwort of mid harvest on the 30th day was measured in three portions of leaves, petioles and stems, and was compared with that of late harvest on the 40th day. Hydroponic water dropwort was obtained from the Department of Horticultural Science, Seoul National University. The results were as follows. The nutrients content of leaves were significantly higher in ash and ascorbic acid and lower in moisture, crude fat and crude fiber than those of stems and petioles. There was no significant difference in total vitamin A and thiamin contents between three portions. Ascorbic acid content of leaves harvested on the 30th day was the highest, 57mg/100g, and decreased to 18mg/100g in the leaves harvested on the 40th day. Marked increase in crude fiber and vitamin A content of leaves was observed in late harvest may be due to the maturation. The results of nutrient composition analysis suggest that the leaves of hydroponic water dropwort is important in ascorbic acid and ash. It is recommended that shorter blanching time and addition of 0.5% NaCl to the blanching water are better for higher ascorbic acid retention of hydroponic water dropwort. In conclusion, as hydroponic water dropwort has high content in ash, calcium, vitamin and free sugar with alkalinity, leaves as well as stems and petioles can be recommonded as a vegetable of high nutritional quality.

Monitoring of Fasciola Species Contamination in Water Dropwort by COX1 Mitochondrial and ITS-2 rDNA Sequencing Analysis

  • Fascioliasis, a food-borne trematode zoonosis, is a disease primarily in cattle and sheep and occasionally in humans. Water dropwort (Oenanthe javanica), an aquatic perennial herb, is a common second intermediate host of Fasciola, and the fresh stems and leaves are widely used as a seasoning in the Korean diet. However, no information regarding Fasciola species contamination in water dropwort is available. Here, we collected 500 samples of water dropwort in 3 areas in Korea during February and March 2015, and the water dropwort contamination of Fasciola species was monitored by DNA sequencing analysis of the Fasciola hepatica and Fasciola gigantica specific mitochondrial cytochrome c oxidase subunit 1 (cox1) and nuclear ribosomal internal transcribed spacer 2 (ITS-2). Among the 500 samples assessed, the presence of F. hepatica cox1 and 1TS-2 markers were detected in 2 samples, and F. hepatica contamination was confirmed by sequencing analysis. The nucleotide sequences of cox1 PCR products from the 2 F. hepatica-contaminated samples were 96.5% identical to the F. hepatica cox1 sequences in GenBank, whereas F. gigantica cox1 sequences were 46.8% similar with the sequence detected from the cox1 positive samples. However, F. gigantica cox1 and ITS-2 markers were not detected by PCR in the 500 samples of water dropwort. Collectively, in this survey of the water dropwort contamination with Fasciola species, very low prevalence of F. hepatica contamination was detected in the samples.

미나리가 마우스의 카드뮴 중독에 미치는 병리조직학적 관찰 (Histopathological Studies on Effect of Dropwort on Cadmium Poisoning in Mice)

  • This study was undertaken to find out the effect of dropwort on histopathological changes of cadmium toxicity in mice. Thirty two mice of BALB/c strain were divided into a control group (A) and three experimental groups (B, C, D). Each group was consisted of four males and four females : group A received tap water and basal diet, group B received tap water and diet supplemented with 100 mg/kg of cadmium, group C received dropwort juice and diet supplemented with 100 mg/kg of cadmium, group D received dropwort juice and basal diet. All mice were dissected on the 35th day. Pathological changes in liver, kidney, spleen, heart, lung, stomach and muscle were observed. Group D showed no significant change as the control group. But group B showed many cytoplasmic vacuoles and necrosis of hepatocytes, glomerular swelling, and degeneration and necrosis of convoluted tubules in kidney, marked congestion and hemorrhage, and a large number of variably sized alveolar macrophages appearance in lung and swelling in some part of muscle fiber. On the other hand, group C showed a little convalescent changes and maintained their normal architectures in liver, kidney, lung and muscle.

Phytol과 들미나리추출물이 Sarcoma 180마우스의 T Subset에 미치는 효과 (Effects of Phytol and Small Water Dropwort Extract on the T Subset in the Sarcoma 180-Transplanted Mice)

  • 본 연구는 녹황색채소류에서 추출되어 항암효과를 나타내는 활성 물질로 보고되어 있는 phytol과 들미나리 추출물을 sarcoma 180마우스에 주사한 후 적출한 비장세포내 T임프구와 T subset, 그리고 asialo GM1$^{+}$세포를 정량하여 다음과 같은 결과를 얻었다. 1) 종양 마우스에 phytol 을 투여하였을 때 비장세포내의 T cell과 T-subset은 종양세포이식에 의해서 상승된 치를 더욱 증가시켰다. 그러나 들미나리추출물의 경우는 대동소이하였다. 2) Asialo GM1$^{+}$세포는 종양마우스에 phytol이나 들미나리추출물을 주사하였을 때 모두 상승하였으며 정상마우스에 phytol 을 투여하였을 때도 대조군에 비하여 상승했지만 들미나리추출물을 작용시켰을 때는 저하되었다. 3) L3T4$^{+}$/Lyt-2$^{+}$세포비는 종양마우스에 phytol을 주사하였을 때 감소를 보였지만 정상 마우스에 투여하였을 때는 더욱 크게 낮아졌다. 그러나 들미나리추출물을 정상마우스에 투여 하였을때는 크게 감소하던 것이 종양마우스에 적요 시켰을 때는 증가현상을 보였다. 이상의 결과로 미루어 볼 때 phytol이나 들미나리추출물은 종양마우스에서 작용자 세포인 자연살해세포(natural killer cell)의 활성 인자로서 작용할 가능성이 높을 것으로 사료된다.

미나리 프락토올리고당 발효액의 발효기간에 따른 품질특성 및 간암세포 증식 억제 효과 (Quality Characteristics and Anti-proliferative Effects of Dropwort Extracts Fermented with Fructooligosaccarides on HepG2 Cells)

  • 발효기간에 따른 미나리 발효추출액의 품질 특성을 검토하고 간암세포주에 대한 증식 억제 효과를 알아보았다. 발효 숙성 기간 중에 총 플라보노이드 함량, 색도 및 생균수는 감소하였으며, 포도당과 과당과 같은 환원당 함량은 증가하였다. 정상 간세포주와 간암 세포주를 이용하여 농도별로 시료 처리를 한 후 각 세포의 증식에 대한 효과를 알아 본 결과, 미나리 발효액이 정상 간세포의 증식에는 영향을 미치지 않았으며, 인간유래 간암 세포주인 HepG2의 증식을 농도 의존적으로 억제하였다. 특히 미나리발효액 1년에서 현저한 억제 효과를 확인하였다. 본 연구 결과는 미나리 발효액의 발효 기간에 따른 품질의 변화 및 간암세포 성장 억제 효능을 확인함으로서, 천연의 미나리 발효 추출액을 다양한 식품 산업에 활용하기 위한 기초적 데이터를 처음으로 제시했다.

국내산 미나리 가루 첨가량에 따른 머핀의 품질 특성 (Quality Characteristics of Muffins Containing Domestic Dropwort Powder (Oenanthe stolonifera DC.))

  • In this study, muffins were produced using dropwort powder and to the quality characteristics such as volume, height and moisture content The results showed that the height of the muffin was 5 cm and there were no significant differences across samples (p<0.05). The volume of the muffin increased as the amount of dropwort powder increased. With respect to the moisture content, a comparison group showed a moisture content of 26%, and the 3%-added and the 6%-added groups did not show any significant difference. The 9%-added group, however, did show a significant difference (p<0.05). The results of texture measurements showed that hardness increased as the amount of added dropwort powder increased. There were also significant differences in adhesiveness and springing across samples (p<0.05). With respect to cohesiveness, gumminess and chewiness, these characteristics tended to increase as the amount of added dropwort powder increased, and there were significant differences across samples (p<0.05). Sensory test results of added showed that the 3%-added group scored the highest for color, flavor and taste. For texture, the 6%-added group showed a value of 6.2, which was the highest texture. In terms of the overall acceptability, the 3% dropwort powder group showed a score of 7.2, which was the highest acceptability. These results suggest that the 3%-added group was the most desirable and appropriate.

흑설탕, 과당, 올리고당을 이용한 미나리 추출물의 제품화 (Manufacture of Dropwort Extract Using Brown Sugar, Fructose Syrup and Oligosaccharides)

  • 당의 종류에 따라 추출 및 발효 숙성된 미나리 액상 추출물들의 $CO_2$ 생성 억제 및 미나리 액상 추출물의 규격화를 수행하였다. 흑설탕, 올리고당 및 과당으로 침출된 미나리액상 추출물은 적정산도 $0.74\%{\~}1.12\%$를 나타냈으며, 과당을 사용한 경우에 환원당이 $50\%$로 가장 높은 값을 보였다. 미나리 액상추출물의 초기 생균수는 흑설탕을 이용한 경우에 $1.6{\times}10^6$ CFU/mL의 생균수를 보였으며, 올리고당, 과당의 순서로 생균수가 낮았다. 미나리 액상추출물의 $CO_2$ 생성량은 올리고당을 사용한 경우에 가장 높았으며 과당을 이용한 경우에 가장 낮았다. 흑설탕 또는 올리고당 침출 미나리액상 추출물은 $85^{\circ}C$에서 15분 열처리에 의해서 미나리 액상추출차의 규격에 맞는 생균수를 보였으며, $3\%$ 구연산의 첨가에 의해서 당의 종류에 관계없이 미나리 액상추출물에 존재하는 미생물을 사멸시킬 수 있었다. 흑설탕, 과당, 올리고당으로 제조된 미나리 액상 추출물에 $3\%$ 구연산을 첨가하여 열처리하여 액상추출차로 제품화한 후에 성분 규격을 검사한 결과, 미나리 액상 추출물은 성상, 대장균군, 세균수, 타르색소, 납 성분 검사에서 모두 적합하였다.

Phylogenetic rind Taxonomic Status of the Phytoplasmas Associated with Water Dropwort (Oenanthe javanica DC) Disease in Korea and Japan

  • To evaluate the phylogenetic and taxonomic status of the phytoplasmas associated with water dropwort (Oenanthe javanica DC) disease in Korea and Japan, their 16S rDNA was analyzed. DNAs extracted from water dropworts collected in Korea (Kyongnam province) and Japan (Chiba prefecture) affected by witches' broom and yellows were subjected to PCR using phytoplasma-specific primers, which amplified a 1.4-kbp fragment that included the 16S rDNA. Phytoplasmas were characterized by RFLP analysis using AluI, HaeIII, HhaI, KpnI, MseI, and RsaI restriction enzymes and by sequence analysis of the PCR products. The mater dropwort witches'broom (WDWB) and water dropwort yellows (WDY) 16S rDNA sequences were identical and closely related to onion yellows (OY, 99.9% identity), which belong to the aster yellows (AY) 16S-subgroup. However, the KpnI RFLP analyses clearly distinguished the WDY and WDWB phytoplasmas from the OY phytoplasma. The phylogenetic analysis based on 16S rDNA showed that WDWE and WDY phytoplasmas are members of a relatively homogeneous group that evolved from a common ancestor.

돌미나리 메탄올 추출물의 항돌연변이 작용과 암세포증식 억제효과 (The Antimutagenic Activity and the Growth Inhibition Effect of Cancer Cells on Methanol Extracts from Small Water Dropwort)

  • The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.
