Effects of Anonaine on Dopamine Biosynthesis in PC12 Cells.

  • The effects of anonaine, an aporphine isoquinoline alkaloid, on dopamine biosynthesis and L-DOPA-induced neurotoxicity in PC12 cells were investigated. Treatment of PC12 cells with 0.05 ${\mu}$M anonaine showed a significant inhibition of dopamine content. The IC$\sub$50/ value of anonaine was 0.05 ${\mu}$M. Under the same conditions, 0.05 ${\mu}$M anonaine also inhibited tyrosine hydroxylase (TH) activity at 24 h (62.0% inhibition of the control level). TH mRNA levels were also decreased by the treatment with anonaine. (omitted)

  • In vivo aromatic ${\beta}$-carbolines, such as harman and norharman, may easily be formed by cyclization of indoleamines with e.g. aldehydes. Because of the structural similarity to MPTP, ${\beta}$-carbolines have been proposed as endogenous toxins. In this study, we have investigated the effects of harman and norharman on dopamine biosynthesis and L-DOPA-induced cytotoxicity in PC12 cells. Treatment of PC12 cells with harman and norharman showed 48.8% and 49.5% inhibition of dopamine content at a concentration of 20 ${\mu}$M and 100 ${\mu}$M for 48h. (omitted)

  • The effects of aporplline isoquinoline alkaloids such as liriodenine. anonaine and asimilobine on dopamine biosynthesis in PC12 cells were investigated. Treatment of PC12 cells with liriodenine (10 ${\mu}$M), anonaine (0.05 ${\mu}$M) and asimilobine (0.15 ${\mu}$M) showed 33.6%, 37.7% and 35.1 % inhibition of dopamine content for 12 h. The IC$\sub$50/ values of liriodenine. anonaine and asimilobine were 8.4 ${\mu}$M. 0.05 ${\mu}$M and 0.13 ${\mu}$M. respectively. (omitted)

Experimental Study on the Expression of Striatal Dopamine Receptors in the Rat Model of Parkinsonism (파킨슨씨병 모델 흰쥐에서 줄무늬체 도파민 수용체의 발현에 대한 연구)

  • This study was designed to investigate the underlying mechanisms for the temporal changes of the striatal dopamine D2 receptors in the rat model of parkinsonism. After injection of the 6-hydroxydopamine into the substantia nigra of adult rats, we measured the receptor binding capacity(Bmax), mRNA and protein of the D2 receptor at 2, 4 and 8 weeks. Following the lesion, mRNA and protein were elevated simultaneously on both sides of the striata. They showed more increase on the normal side at 2 and 4 weeks, and then they were almost equally abundant on both sides at 8 weeks. We also observed their increased production in the diffuse cortical and subcortical regions. The Bmax value also increased bilaterally in both striata, and was higher on the normal side at 2 weeks and then on the lesioned side at 4 and 8 weeks. These findings suggest that production of the striatal D2 receptor is regulated at the transcriptional level in this animal model. They also imply that this control may be mediated through a pathway which can have influence on the whole brain, rather than the local control of the dopamine content alone. The measured functional activity(Bmax) of the D2 receptor was not proportional to the amount of the receptor mRNA and proteins produced. This difference may be explained by the post-translational modification of the receptor proteins, which may be controlled by such factor as the local concentration of dopamine.

  • Objective : The study was aimed to evaluate the anti-stress effect of KYT on the cold-stressed Mice. Methods: The experimental animals were stressed in cold room $(4{\pm}0.5^{\circ}C)$ for one hour in a day during two weeks, and administred 131mg/100g KYT extract for two weeks before stress. For two weeks, while the experiment was taking place, we conducted a research about the change of weight and content of norepinephrine, epinephrine, dopamine, cortisol and GOT, GPT in the mice serum. Results : KYT extract significantly inhibited the decrease of body weight induced by stress, compared with the control group. KYT extract had no significant decreasing effect on the change of content of the serum GOT but had significant decreasing effect on the change of content of the serum GPT compared with the control group. KYT extract had significant decreasing effect on the change of content of the serum cortisol compared with the control group. KYT extract had decreasing effect on the change of content of the serum dopamine compared with the control group. KYT extract had no decreasing effect on the change of content of the serum norepinephrine compared with the control group. KYT extract had significant decreasing effect on the change of content of the serum epinephrine compared with the control group. Conclusions : According to the above results, it is concluded that KYT will be useful as a preventive and remedy against stress disease.

  • The present study was aimed at establishing what changes occur in the dopamine levels and pattern of TH-immunoreactive neurons of certain areas of rat brain during food intake suppression produced by intraperitoneally administration of CCK-8. CCK-8 in dose of $10\;{\mu}g/kg$ was injected intraperitoneally to 48 h food-deprived rats. In the fasted group, the contents of dopamine were decreased in the frontal, striatum, hypothalamus and amygdala as compared to those of the fed control group. The administration of CCK-8 showed significant decrease on the dopamine levels of the hypothalamus, in comparison to those of the sated and starved group. During deprived condition, the density and number of TH-immunoreactive neurons in the paraventricular nucleus, arcuate nucleus, median eminence and substantia nigra were lower than those of the fed control group. After administration of CCK-8, the pattern and distribution of TH-positive neouons in the hypothalamic areas and substantia nigra were increased when compared to those of the starved group. It is concluded that the results demonstrate the partial involvement of hypothalamic dopamine-containing neurons in the feeding inhibition of CCK-8. Furthermore, the results indicate that TH-immunoreactive neurons play on important role in the hypothalamus and substantia nigra for eating behavior

Enantio-Selective Inhibition of (1R,9S)- and (1S,9R)-$\beta$-Hydrastines on Dopamine Biosynthesis in PC12 Cells

  • The inhibitory effects of (1R, 9S)- and (1S, 9R)-enantiomers of $\beta$-hydrastine (BHS) on dopamine biosynthesis in PC12 cells were investigated. (1R, 9S)-BHS decreased the intracellular dopamine content with the $IC_{50}$ value of 14.3 $\mu\textrm{M}$ at 24 h, but (1S, 9R)-BHS did not. In these conditions, (1R, 9S)-BHS inhibited TH activity mainly in a concentration-dependent manner(33% inhibition at 20 $\mu\textrm{M}$) and decreased TH mRNA level. (omitted)

  • This study aimed to evaluate the anti-stress effect of Yangsimtang and Yangsimtang+Siyup on the rats in immobilization stress.The experimental animals were immobilized in the stress box(5${\times}$5${\times}$20cm) for 12 hours in a day suring 3 days, and administered 1g/100g of Yangsimtang and Yangsimtang+Siyup and Siyup extract for 12 days before stress. The norepinephrine, epinephrine, dopamine and serotonin contents were measured by HPLC method in various part of rat brain.The following results were observed.1. In frontal cortex the norepinephrine content of control group was 561.${\pm}$24.46 ng/g brain tissue, that of saple 1 group was 430.8$\pm$41.2 ng/g brain tissue, and that of sample 2 group was 417.2$\pm$38.5 ng/g brain tissue. The differences was statistically significant.2. In corpus striatum, the norepinephrine content of control group was 561.3$\pm$27.3 ng/g brain tissue, and that of sample 1 group was 422.1$\pm$21.2 ng/g brain tissue, the dopamine content of control group was 1205.1$\pm$75.9 ng/g brain tissue, that of sample 2 group was 685.6$\pm$41.5 ng/g brain tissue. The differences was statistically significant.3. In hypothalamus, the norepinephrine content of control group was 1165.1$\pm$162.6 ng/g brain tissue, that of sample 2 group was 947.2$\pm$35.7 ng/g brain tissue. The differences was statistically significant.4. In hippocampus, the norepinephrine content of control group was 931.6$\pm$82.2 ng/g brain tissue, that of sample 1 group was 652.1$\pm$47.5 ng/g brain tissue, and that of sample 2 group was 627.4$\pm$31.2 ng/g brain tissue, the dopamine contrnt of control group was 315.4$\pm$28.4 ng/g brain tissue, that of sample 2 group was 208.5$\pm$23.7 ng/g brain tissue. The differences were statistically significant.Base on the above results, it may be concluded that Yangsimtang and Yangsimtang+Siyup are effective to reduce stress.

Effects of Berberine on 6-hydroxydopamine-induced Parkinsonism in Rats (Berberine이 백서의 6-hydroxydopamine-유도 파킨슨병 모델에 미치는 영향)

  • Many isoquinoline alkaloids including berberine lower dopamine content by reducing tyrosine hydroxylase (TH) activity and aggravate L-DOPA-induced cytotoxicity in PC12 cells. In this study, the effects of berberine on 6-hydroxydopamine (6-OHDA)-induced cytotoxicity in PC12 cells and on unilateral 6-OHDA-lesioned rat models were investigated. Berberine at 10-30 ${\mu}M$ did not affect cell viability in PC12 cells. However, berberine at concentrations higher than $50{\mu}M$ caused cytotoxicity at 24 h. Berberine (10-50 ${\mu}M$) also enhanced 6-OHDA (10-50 ${\mu}M$)-induced cytotoxicity at 24 h compared to 6-OHDA alone with an apoptotic process. In addition, treatment with berberine (5 and 30 mg/kg, i.p.) for three weeks showed a dopaminergic cell loss in substantia nigra of 6-OHDA-lesioned rats: 30 mg/kg berberine was more intensive cytotoxic. The levels of dopamine were also decreased by berberine (5 and 30 mg/kg) in the ipsilateral substantia nigra of 6-OHDA-lesioned rats. These results suggest that berberine aggravated 6-OHDA-induced cytotoxicity in PC12 cells and treatment with berberine (5 and 30 mg/kg) in 6-OHDA-lesioned rats also enhanced the degeneration of dopaminergic cell death and the decrease in dopamine levels in substantia nigra. Therefore, the long-term L-DOPA therapeutic patients with isoquinoline compounds including berberine may need to be checked for the adverse symptoms.
