Plasma Phospholipids, including Plasmalogens, after Consumption of Diets Enriched in Long-chain n-3 Fatty Acids

  • The level of long-chain n-3 fatty acids in chicken and pork can be increased by changing the diet of the animals. Increased levels of these essential fatty acids improve cardiovascular health in humans. The purpose of this study was to study the effects of the consumption of pork and chicken enriched in docosahexaenoic acid (DHA) on plasma lipids. The consumption of these products decreased the levels of two cardiovascular risk factors, LDL-cholesterol and triacylglycerols, in the plasma of female college students. The effect on LDL-cholesterol differed from that of fish oil, which does not affect the level of LDL-cholesterol. The proportions of DHA in the triacylglycerols and the glycerophospholipids were increased markedly. The greatest changes in the glycerophospholipids were in the ether types of the ethanolamine glycerophospholipids. Dietary DHA appears to be incorporated preferentially into the plasma ethanolamine plasmalogens, which can act as antioxidants. This agrees with our hypothesis that DHA stimulated the transcription of the genes for peroxisomal enzymes that are required for plasmalogen synthesis.

Lipase-Catalyzed Synthesis of DHA-Enriched Structured Lipid (Docosahexaenoic acid 함유 재구성지질의 효소적 합성 연구)

  • Structured lipid (SL) was synthesized by enzymatic interesterification of algae oil and corn oil in stirred tank batch reactor, The reaction, performed for 15hr at $65^{\circ}C$, was catalyzed by sn-1,3-specific lipase RM IM from Rhizonucor miehei without organic solvent. DHA, oleic acid, and linoleic acid contents of SL were 14.9, 17.3, and 31.8 mol%, respectively. ${\alpha}-,\;{\gamma}-,\;and\;{\delta}-tocopherol$ contents and physiochemical property of SL were evaluated. During 15 hr reaction, most reaction occurred within 6 hr, and highest relative production rate was observed between 3 to 6 hr.

  • This paper examines the process and evidence used to create the Dietary Reference Intake (DRI) of alpha-linolenic acid (ALA) and eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) for Koreans. ALA (18:3n3) is an essential fatty acid, and EPA and DHA are known to have beneficial effects on cardiovascular disease risk and reduction of triglyceride levels. Various international organizations have suggested dietary recommendations for n-3 polyunsaturated fatty acids (PUFAs), including ALA, EPA, and DHA. A DRI for Koreans was established for the first time in 2020, specifically for the adequate intake (AI) of ALA and EPA + DHA. This recommendation was based on the average intake of ALA and EPA + DHA from the Korea National Health and Nutrition Examination Survey 2013-2017. For Korean infants, the AI of ALA and DHA was based on the fatty acid composition of maternal milk. Estimated average requirement and a tolerable upper intake level have not been set for n-3 PUFA due to insufficient evidence. In addition, the intake level of n-3 PUFA for prevention of chronic disease has also not been determined. Future studies and randomized controlled trials are required to establish the UL and to define the level for disease prevention.

Variation in the Lipid Class and Fatty Acid Composition of Thraustochytrium aureum ATCC 34304 (Thraustochytrium aureum ATCC 34304의 지질 및 지방산 조성 변화)

  • The heterotrophic marine algae Thraustochytrium aureum ATCC 34304 produces substantial amount of polyunsaturated fatty acids (PUFAs), especially docosahexaenoic acid (DHA). In this study, changes in the lipid and fatty acid compositions of Thraustochytrium aureum ATCC 34304 were investigated according to the growth stage. The major lipids of Thraustochytrium aureum ATCC 34304 were found to be composed of triacylglyceride (TAG), phospholipid (PL), and sterol (ST). The content of triacylglyceride increased during the exponential phase of cell growth, but the content of phospholipid decreased. The composition of total polyunsaturated fatty acids decreased from 60.3% to 45.3% and that of docosahexaenoic acid from 42.1% to 33.9% in the triacylglyceride. The composition of total saturated fatty acids, however, increased from 24.9% to 27.8%. The content of total polyunsaturated fatty acids decreased greatly from 48.0% to 17.5% but the decrease in the content of saturated fatty acids was slight in phospholipid.

Chemical Changes of Low Salt-dried Yellow Corvenia Muscle During Frozen Storage (저염건 조기의 냉동저장 중 화학적 변화)

  • Changes of chemical and nutritional composition were investigated in low salt-dried Yellow corvenia muscle during frozen storage at $-20^{\circ}C$. Moisture contort of Yellow corvenia was not significantly changed until after 5 months frozen storage. Salinity was $0.28\%$ in fresh Yellow corvenia and $2.30\%$ in the salted sample. Acidity of the fish muscle was increased in excess of 5 months storage. TBA value was increased by a little amount untel 1 month of storage and significantly incraeased in 3 months of storage. Free fatty acid was increased during frozen storage with rapid increase up to 1 month storage. VBN was increased after 1month storage and amino type nitrogen was increased in 5 months of storage. Major free amino acids were glutamic acid, alanine, glycine and lysine, and composition ratio of most free amino acids were not changed untel after 5 months storage. Polyenoic acid contents were decreased after storage and major fatty acids were palmitic acid (16 : 0), stearic arid (18 : 0) and major fatty acids were palmitic acid (16 : 0), stearic acid (18 : 0) and docosahexaenoic acid (22 : 6). From the results of chemical experiments, low salt-dried yellow corvenia would not be stored at $20^{\circ}C$ for more than 5 months.

Effects of ${\alpha}-linolenic$, eicosapentaenoic and docosahexaenoic acids administration on lowering of triacylglycerol level in the hepatic and serum of rats (n-3계 지방산 투여가 성장기 흰쥐의 간장 및 혈청 Triacylglycerol 농도에 미치는 영향)

  • We studied the difference effects of dietary ${\alpha}-linolenic\;acid\;({\alpha}-LA,\;18:3\;n-3)$, eicosapentaenoic acid (EPA, 20:5 n-3) and docosahexaenoic acid (DHA, 22:6 n-3) on the lowering of triacylglycerol in the liver and serum on lipid metabolism in rats. Rats were fed semipurified diets containing 10% fat with constant polyunsaturated/monounsaturated/saturated fatty acids (1:1:1) and n-6/n-3 ratio (1:2). EPA (98%) and DHA (98%) were added in diets as the ethyl esters. The concentration of liver triacylglycerol was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. The concentration of liver phospholipid was significantly higher in rats fed DHA than in those fed ${\alpha}-LA$ and EPA. Both EPA and DHA reduced serum triacylglycerol concentration compared with ${\alpha}-LA$, but this effect was more pronounced in the EPA diet. The activity of phophatidate phosphohydrolase in the liver microsome was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. but, there was no significant difference on the activities of diacylglycerol acyltransferase among the three groups. The concentration of liver triacylglycerol were correlated with changes in the microsomal phosphatidate phosphohydrolase activity (r=0.84). Hepatic NADPH generating enzyme, glucose-6-phosphate dehydrogenase was more effective to reduce the activity in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. In conclusion, EPA or DHA reduced the hepatic triacylglycerol concentration by inhibiting microsomal phosphatidate phosphohydrolase, thereby inhibiting synthesis of triacylglycerol in the liver.

The Effect of Dietary Concentrated Oils of Eicosapentaenoic Acid, Docosahexaenoic Acid and Perilla Oil on lipid Metabolism in Rata Fed Low Fat Diet (Eicosapentaenoic Acid, Docosahexaenoic Acid 농축어유와 들깨유가 저지방 식이를 섭취한 흰쥐의 지질대사에 미치는 영향)

  • To compare the effect of three kinds of n-3 fatty acids-eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA) and perilla oil (PO)-on serum and liver lipid levels and fatty acid composition of liver phospholipid(PL) at low fat level(5%, w/w), 4-weeks old Sprague-Dawley rats were fed with one of five different oil diets for 4 weeks. Beef tallow(BT) and corn oil(CO) was used as control for sturated or n-6 fatty acid respectively. Se겨m concentrations of cholesterol(TC) and phospholipid(PL) were lower in PO DHA and EPA groups than in BT and CO groups. HDL-cholesterol levels were higher in CO and PO groups than in EPA, DHA and BT groups. Liver PL concentrations were higher in DHA and EPA groups than in CO, PO and BT groups, but liver TC and heal PL and TC concentrations did not show any significant difference among groups. Hepatic fatty acid composition of phosphatidylcholine (PC) and phosphatidylethanolamine (PE), two major phospholipids in liver, reflected their dietary fatty acid composition. In PC and PE, total percentage own-6 series was higher in CO group than in any other groups, and that own-3 series was higher in DHA and PO groups than in EPA, CO and BT groups. Moreover, the ratio of 20 : 4/18 : 2 was lower in PO and DHA groups than in EPA, CO and BT groups. On the contrary, the percentage of C22 : 6 was lower in EPA, CO and BT groups than in PO and DHA groups. These results revealed that n-3 series(EPA, DHA and PO) were more effective in lowering um lipids than n-6 fatty acids or saturated fatty acid. Based on the results of fatty acid composition of hepatic phospholipid, we suggest that the dietary effect of PO and DHA on antiatherogenic characteristics seems to be similar extent. In addition, the effect of EPA might not be significantly different from that of BT or CO in the view of eicosanoids production from the precursor fatty acid. These difference of hepatic fatty acid composition might come from other characteristics of dietary oil as well as the type of unsaturation, not from the carbon chain length or the degree of unsaturation of n-3 fatty acid.

Fatty Acid Composition of Canned Fish and Shellfish Products on Korean Market (국내시판 수산물통조림의 지방산조성)

  • The fatty acid composition of lipids separated from 14 kinds of canned fish and shellfish products from the market were examined. The crude lipid contents of canned fish and shellfish products showed wide difference (1.3-15.2%) depending on raw materials and kinds of products. Fatty acid composition of products were mainly consisted of polyenoic acid(34.5-61.4%), and followed by saturated acid(24.5-47.8%), monoenoic acid(12.4-32.0%). The major fatty acids were palmitic acid, palmitoleic acid oleic acid, eicosapentaenoic acid and docosahexaenoic acid. In case of canned tuna in oil and canned smoked oyster in oil, linoleic acid was the most predominant component(48.2%, 34.6%). Judging from the results, canned fish and shellfish products were abundant of the polyunsaturated fatty acids in spite of the high temperature sterilization and lone storage periods.

The Base Catalyzed Synthesis of Sucrose Ester Containing Omega-3 Fatty Acids (오메가 3 지방산을 함유한 Sucrose Ester의 합성)

  • Sucrose esters were synthesized by transesterification of sucrose with docosahexaenoic acid ethylester mixture (DHAEE). Potassium carbonate as a base catalyst was used in the presence of dimethylsulfoxide (DMSO) for the reactions. The reactions were performed with the different reaction times and molar ratios of substrates in the presence of surfactant in vacuum. Among the reaction conditions in this study, SE#4~7 showed the relatively high conversion rate (>96%) of DHAEE, leading to the high yield of sucrose esters. In addition, the product composition was changed from sucrose mono ester to di/tri/polyesters after the prolonged reaction time while the increased molar ratio of DHAEE also resulted in the composition changes of sucrose mono ester to the sucrose di/tri/polyesters. From the reaction (SE#7), conversion ratio was 98.5% in which 87.3% mono ester and 13.7% di/tri/polyester were found, resulting in the highest content of mono ester. Therefore, the sucrose ester containing various rates of mono and di/tri/polyesters, which effects on hydrophilic lipophilic balance (HLB) values, can be manipulatively synthesized using the reaction conditions reported in this study.

  • Marine fungus, Thraustochytrium aureum ATCC 34034, was incubated in artificial sea water media in order to produce docosahexaenoic acid(DHA). Cultures were performed at $24^{\circ}C$ in light for 13 days with orbital shaker at 100rpm. Maximum cell productivity of 1.34g/L and DHA yield of 41.4 mg/L were obtained by using this method, which is almost the twice level of DHA yield obtained for the strains reported previously. This strain did produce much more DHA after sporangium disruption.
