Localization of Divalent Cation-Binding Site in the Pore of a Small Conductance $Ca^{2+}$-activated $K^+$ Channel and Its Role in Determining Current-Voltage Relationship

  • In our previous study (Soh and Park, 2001), we proposed that the inwardly rectifying current-voltage (I-V) relationship of small-conductance $Ca^{2+}$-activated $K^{+}$ channels (S $K_{Ca}$ channels) is the result of voltage-dependent blockade of $K^{+}$ currents by intracellular divalent cations. We expressed a cloned S $K_{Ca}$ channel, rSK2, in Xenopus oocytes and further characterized the nature of the divalent cation-binding site by electrophysiological means.(omitted)

전기투석 막여과의 이온제거 특성 및 지하염수의 담수화효율 (Selectivity of cations in electrodialysis and its desalination efficiency on brackish water)

  • In this study, desalination by electrodialysis with ion exchange membranes was applied to synthetic waters with various ion concentrations and also for ground waters from coastal areas in Korea. Electrodialysis performance on the synthetic solutions showed the similar tendency in operation time and current curves, i.e., shorter operation time and higher maximum current with increasing applied voltages. The ED results of synthetic waters with different ion compositions, i.e., $Na_2SO_4$, $MgSO_4$, $CaSO_4$, at the similar conductivity condition, i.e., $1,250{\mu}s/cm$ revealed that effects of mono- and divalent ions on water quality and performance in electrodialysis were different. The divalent ions had less efficiency in the ED compared to monovalent sodium ions and also divalent calcium ions showed better performance than Mg ions. The electrodialysis on the ground waters produced high quality of drinking water. The groundwater from SungRoe however showed a buildup of membrane resistance. Organic matter concentrations and great portions of divalent ions in the groundwater were possible causes of the deteriorated performance.

2가 양이온이 Thiamine Pyrophosphate에 의한 Group I Intron Ribozyme의 Splicing 억제에 미치는 영향 (Effects of Divalent Cations on the Self-splicing Inhibition of Group I Intron by the Coen-zyme Thiamine Pyrophosphate)

  • 2가 양이온($Mg^{2+}$, $Mn^{2+}$, $Zn^{2+}$)이 조효소 thiamine pyrophosphate에 의한 T4파지 티민생합성 유전자 (td) 인트론 RNA의 splicing에 미치는 영향을 조사하였다. $Mg^{2+}$를 30 mM까지 증가시켰을 때 splicing 활성은 농도에 비례하여 증가하였다. 그러나 $Mg^{2+}$이 존재하지 않는 상태에서 0.1-4 mM 농도에 걸쳐 $Zn^{2+}$를 사용한 결과 약 20% 정도의 splicing이 일어났다. 이때 대부분의 splicing product는 인트론-엑손2 및 엑손2였고 엑손1-엑손2는 검출되지 않았다. 그리고 4 mM 농도에서는 RNA가 대부분 가수분해되는 현상이 나타났다. $Mn^{2+}$ 이온은 사용한 농도 범위 (0.1-8 mM)에서 $Zn^{2+}$ 보다는 전반적으로 약간 증가된 splicing 활성을 보였으며 8 mM 농도에서도 약 30% 정도의 splicing 활성을 보였다. $Zn^{2+}$ 이온처럼 splicing product는 인트론-엑손2 및 엑손2였고 엑손1-엑손2는 검출되지 않았 다. 반면에 splicing반응에 10 mM $Mg^{2+}$ 를 첨가했을 때 $Zn^{2+}$$Mn^{2+}$ 이온은 평균 약 35-40% 정도 splicing활성을 촉진시키는 것으로 나타났다. 실험한 2가 양이온 중에서 특히 $Mg^{2+}$ 은 가장 낮은 농도에서 thiamine pyrophosphate 에 의한 억제반응을 극복하는 최고의 활성효과를 나타냈다. 이와 같은 억제 회복 효과는 $Mg^{2+}$에 의한 리보자임의 td intron 구조적 안정성에 기인하는 것으로 추정된다.

Effects of Divalent Cations on the Spicing of Phage T4 Thymidylate Synthase Intron RNA

  • Effects of divalent cations such as $Mg^{2+}$, $Mn^{2+}$, $Ca^{2+}$, and $Zn^2$ on splicing activity of phage T4 thymidylate synthase intron RNA have been investigated. At the concentration of 0.5 mM, $Mn^{2+}$ in the absence of $Mg^{2+}$, a very small amount of pre-RNA was cleaved into ligation products (El-E2) but no circular or linear intron was produced. As the concentration of $Mn^{2+}$ was increased from 1 to 5 mM the pre-RNA was completely hydrolyzed. In the presence of 5 mM $Mg^{2+}$, both the linear intron and circular intron were produced but no El-E2 ligation product was produced. At both 3 and 5 mM $Mn^{2+}$ the RNA was hydrolyzed completely as observed with no $Mg^2+$ being present. In the case of $Zn^{2+}$, even at 0.5 mM concentration, the pre-RNA was completely hydrolyzed. This observation suggested that $Zn^{2+}$ facilitates RNA hydrolysis more rapidly than $Mn^{2+}$ does. at 5mM $Ca^{2+}$, the RNA was not hydrolyzed and remained intact as a primary transcript.

ATP Hydrolysis Analysis of Severe Acute Respiratory Syndrome (SARS) Coronavirus Helicase

  • Severe acute respiratory syndrome coronavirus (SARS-CoV) helicase separates the double-stranded nucleic acids using the energy from ATP hydrolysis. We have measured ATPase activity of SARS-CoV helicase in the presence of various types of nucleic acids. Steady state ATPase analysis showed that poly(U) has two-times higher turnover number than poly(C) with lower Michaelis constant. When M13 single-stranded DNA is used as substrate, the Michaelis constant was about twenty-times lower than poly(U), whereas turnover numbers were similar. However, stimulation of ATPase activity was not observed in the presence of double-stranded DNA. pH dependent profiles of ATP hydrolysis with the helicase showed that the optimal ATPase activities were in a range of pH 6.2 ~ 6.6. In addition, ATP hydrolysis activity assays performed in the presence of various divalent cations exhibited that $Mg^{2+}$ stimulated the ATPase activity with the highest rate and $Mn^{2+}$ with about 40% rate as compared to the $Mg^{2+}$.

  • Dependence of the flow behavior of aqueous suspension of Young-Il bentonite on the concentration and the type of electrolytes was studied. Viscosity measurements were made was Rion Viscotester, using No. 3 rotor at 62.5 R. P. M. at 25.deg.C. As electrolyte concentration increased, the apparent viscosity was observed to increase. Changes in viscosity were in general agreement with predicted results based on the Hofmeister sequence and the Schulze-Hardy rule. The observed electrolyte effect on the apparent viscosity was discussed in terms of the Verwey-Overbeek theory.

Characterization of Cation Exchange and Cesium Selectivity of Synthetic Beta-Dicalcium Silicate Hydrate

  • 240$^{\circ}C$와 Ca/Si=2 몰 비율의 열수상태에서 합성된 고체 베타-디칼슘 실리케이트 하이드레이트($\beta-C_2$SH)는 Fe, Cu, Zn, Cd, 및 Pb와 같은 2가 금속 양이온에 대한 양이온 교환 성질을 보여준다. 그 고체에 희한 금속 양이온 흡인력은 $Fe^{2+}$$Cu^{2+}$$Zn^{2+}$$Cd^{2+}$ = $Pb^{2+}$의 순서로 됨이 밝혀졌다. 고체에 세슘 선택성은 $Li^+$, $Na^+$$K^+$와 같은 1가 양이온이나 $Ca^{++}$, $Mg^{++}$$Ba^{++}$와 같은 2가 양이온이 $Cs^+$보다 백배이상 진한 상태에서 나타내었다. $Cs^+$의 흡인력은 $Mg^{++}$의 존재하에서 최대치를 보여주었고, 반면에 $K^+$의 존재하에서 최소치를 보여주었다. 2가 금속이온에 대한 $\beta-C_2$SH의 다른 친화도는 이들 이온을 분리하는데 사용 할수 있다. 또한 $\beta-C_2$SH에 희한 금속 양이온 교환에 대한 반응경로 및 세슘 선택성이 연구되었다.

알긴산 코팅에 의한 골판지 상자 제조용 라이너 원지의 수분저항성 증진 (Improvement of Water Resistant Properties of a Linerboard for Corrugated Fiberboard Box by Coating with Na-alginate)

  • 골판지 상자의 수분저항성을 증진시키기 위해 골판지 상자 제조에 사용되는 라이너지의 표면을 알긴산으로 표면코팅을 하기 위한 최적 조건을 결정하기 위하여 농도, 가소제의 종류와 사용량, 알긴산의 불용화를 위한 다가 금속이온의 종류와 최적농도 및 침지시간 등을 결정하였다. 라이너지의 표면코팅을 위한 알긴산의 농도는 2.5%(w/w)가 적합하였으며, 가소제인 글리세롤은 알긴산 대비 35%의 농도로 사용하는 것이 적합하였고, 알긴산 필름의 불용화를 위해서는 2% $CaCl_2$ 용약을 사용하여 3분간 처리하는 것이 라이너지의 수분에 대한 저항성 증진에 최적조건을 결정되었다. 다가 금속이온으로 $CA^{2+}$ 이외에 $Cu^{2+}$를 사용했을 때 낮은 투습계수와 수분용해도를 나타냈다. 가소재의 종류에 따라서는 수분 흡습에 대한 저항성이 큰 솔비톨을 첨가 했을 때 라이너지의 수분저항성이 유의적인 차이를 보이며 증가하였다.

Ion Chromatography에 의한 혈액중에서 양이온의 분석에 관한 연구 (A Study on the Analysis of Cation in Blood by Ion Chromatography)

  • There are many pretreatment and detection methods for divalent cations in blood. But our purpose was to study the pretreatment of blood for the determination of cations by Ion Chromatography. We compared with recovery of Mg$^{++}$, and Ca$^{++}$ contained in plasma according to four pretreatment methods, that is, add of trichloroacetic acid and perchloric acid, dilution with distilled water and membrane filter method. As a result, add of trichloroacetic acid was found to be the most suitable method for good recovery of Mg$^{++}$ (98.0%) and Ca$^{++}$(96.0%) in plasma, and the Mg$^{++}$ and Ca$^{++}$ contents in plasma was 20 and 102 ($\mu$g/ml)

Design and Synthesis of Novel Rhodamine-based Chemosensor Probe Toward Cu2+ Cation

  • Nowdays, fluorescent rhodamine chemosensors have attracted a worldwide interest due to its ability to selectively detect heavy and transition metal cations. Due to the importance in environmental and biological toxic effects, the developments of fluorescent chemosensors have been received considerable attention in recent. Especially, a rhodamine-based chemosensor probes have been proved to be useful by exhibiting the efficient "off-on" fluorescence switching toward selected metal cations. This fluorophore can undergo the transformation from non-fluorescent and colorless spirolactam derivative to fluorescent ring-open form. In this study, a new fluorescent chemosensor was synthesized using rhodamine B through two-step procedures, and its selectivity and related optical property were characterized. Selectivity and sensitivity was found toward $Cu^{2+}$ guest molecules and then related optical properties of rhodamine B based fluorescent chemosensor compound were characterized using discussed. In addition, computational calculation was used to determine the HOMO/LUMO values.
