Effect of Mild Heat Treatments Prior to Air Dehydration of Dried Onions Quality (열풍건조 전 순한 열처리가 건조 양파의 품질에 미치는 영향)

  • The effects of immersion temperature $(20,\;40\;and\;60^{\circ}C)$ and immersion times (6. 12 and 18 min) in a distilled water prior to air dehydration upon the browning reaction and pyruvic acid content of air dried onions to a 4.071 moisture content (wet basis) were analyzed by a response surface methodology (RSM). Those values were also predicted by using a second degree polynomial regression model. Immersion temperature had more influence to browning reaction and pyruvic acid content than immersion time in these experimental ranges. The processing conditions to minimize the browning reaction of dried onions at $50^{\circ}C$ of air temperature (O.D.=0.071) were $60^{\circ}C$ of immersion temperature and 18 min of immersion time compared to control (O.D.=0.168) of air dehydration at $50^{\circ}C$ Pyruvic acid contents of dried onions at $50^{\circ}C$ of air temperature were maximized $(39.85{\mu}mole/g\;onion\;solid)$ at $60^{\circ}C$ of immersion temperature and 12 min of immersion time compared to control $(24.08{\mu}mole/g\;onion\;solid)$ of air dehydration at $50^{\circ}C$.

Effect of Hwanggumgung, a Natural Product, on Hair Growth Promotion in C57BL6 Mice (C57BL6 마우스에서 복합생약제인 황금궁의 육모 효과)

  • Hwanggumgung (HGG) is a hair-care product which is composed of several plant extracts used in oriental medicine. This study was carried out to investigate effect of HGG on hair regrowth in a shaving model of C57BL6 mice. Five-week-old mice were acclimated for 1 week under 23$\pm$3$^{\circ}C$, 50$\pm10\%$ relative humidity and 12 h of a light/dark cycle before beginning experiment. There were four experimental groups including distilled water (D.W., control), 10$\%$ ethanol (EtOH, vehicle control), a positive control of 3$\%$ minoxidil (MXD), and HGG for female and male mice, respectively; Six-weeks old mice were trimmed by electric clippers so as not to damage the skin. The next day; mice without visible scraches were selected, randomized and separated in groups of 11 mice. The test compounds were topically treated with 0.15ml per mouse per day for 21 days. The hair regrowth was photographically and histologically determined during the experimental period of 21 days. Enzyme activities of $\gamma$-glutamyl transpeptidase and alkaline phosphatase were also determined using a rate assay method. There were no clinical signs in all experimental groups. The topical application of 3$\%$ MXD and HGG in female mice promoted hair regrowth earlier and faster than the control groups. In male mice, the topical application of 3$\%$ MXD and HGG also accelerated hair growth compared with the controls. Ten percent ethanol also promoted hair growth faster than D.W group. The histology of hair growth in experimental groups was strongly associated with the hair regrowth. 3$\%$ MXD and HGG promoted elongation of hair follicles compared with the controls in both female and male mice. Activities of alkaline phosphatase and $\gamma$-glutamyl transpeptidase, enzymes related to hair growth, significantly increased after treatments of 3$\%$ MXD and HGG for 2 weeks in both female and male mice (p < 0.05). These results suggest that HGG has hair growth promoting activities and it can be for treatment for alopecia.

An Edible Alginate Microcapsulation of Entomopathogenic Nematode, Steinernema carpocapsae (알지닌캡슐을 이용한 곤충병원선충(Steinernema carpocapsae)의 섭식유도형 제제화 기술)

  • Field application of the entomopathogenic nematode, Steinernema carpncapsae, is limited by its susceptibility to UV irradiation and desiccation especially at leaf spray control. This study was conducted to develop the control technique using alginate biocapsulation of the nematodes against the beet armyworm, Spodoprera exigua and the tobacco cutworm, Sp. litura that are normally infesting hosts above ground level. The alginate capsules including infective juveniles gave significant feeding toxicities to the larvae of the two lepidopteran species. The lethality followed a typical sigmoid dose-mortality pattern with increase of the nematode densities embedded in the capsules. Moisture content in the capsule was critical to the survival of the infective juveniles. More than 80% nematodes could survive above 10% moisture content remained in the capsule. Remaining moisture content within the capsule was dependent on relative humidity, ambient temperature, and capsule size, but not on citric acid reaction time during capsule formation. More than 80% of infective juveniles in the alginate capsules could survive in distilled water at 15$^{\circ}C$ for 60 days. When these nematode capsules containing welsh onion extract as another phagostimulant were applied on the 3rd instar larvae of Sp. exigua infesting peanut plants, they resulted in about 90% control efficacy. These results indicate that the alginate capsulation can be used for leaf-spray agent of the entomopathogenic nematodes as well as for improved storage purpose.

Anti-oxidant and Anti-inflammatory Effects of Acanthopanacia Cortex Hot Aqueous Extract on Lipopolysaccharide(LPS) Simulated Macrophages

  • Objectives : This study is to investigate the effects of Acanthopanacis Cortex hot aqueous extract on nitric oxide(NO), prostaglandin E2(PGE2) production and DPPH(1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity in macrophages. Methods : Acanthopanacis Cortex(200 g) was heated at $100^{\circ}C$ with distilled water(2 L) for 4hrs. The extract was filtered and concentrated to 100 ml using a rotary evaporator and was frozen at $-80^{\circ}C$, then was freeze-dried. The RAW 264.7 macrophages were subcultured. In order to evaluate cytotoxicity, MTT assay was performed. Experimental groups were divided into five(control, AC 25, 50, 100 and 200 ${\mu}g/ml$) and we measured cytotoxicity. The concentrations of NO were preprocessed by Griess assay. The RAW 264.7 macrophages was pretreated by 10 ${\mu}g/ml$ LPS and experimental groups were divided into five and we measured NO production. The concentrations of $PGE_2$ were measured by enzyme immunoassay. The RAW 264.7 macrophages was pretreated by 10 ${\mu}g/ml$ LPS. Experimental groups were divided into five and we measured $PGE_2$ production. Antioxidant activity was measured by the DPPH method. experimental groups were divided into four(AC 25, 50, 100 and 200 ${\mu}g/ml$) and we measured DPPH radical scavenging activity. Results : 1. Viability of RAW 264.7 macrophages did not significantly decrease in 25, 50 and 100 ${\mu}g/ml$ Acanthopanacis Cortex hot aqueous extract compared to control group. 2. NO production in LPS-stimulated RAW 264.7 macrophages significantly inhibited in 100, 200 ${\mu}g/ml$ Acanthopanacis Cortex hot aqueous extract compared to control group. 3. $PGE_2$ production in LPS-stimulated RAW 264.7 macrophages significantly inhibited in 100, 200 ${\mu}g/ml$ Acanthopanacis Cortex hot aqueous extract compared to control group. 4. DPPH radical scavenging capability of Acanthopanacis Cortex hot aqueous extract in RAW 264.7 macrophages had the high level in 100, 200 ${\mu}g/ml$. Conclusion : According to the results, Acanthopanacis Cortexx hot aqueous extract has ability to suppress NO, $PGE_2$ production and improve DPPH free radical scavenging activity. So Acanthopanacis Cortex hot aqueous extract may have an anti-inflammation effect and antioxidant activity.

Browning inhibition of fresh-cut lotus roots by blanching in Glycyrrhiza glabra L. and Astragalus membranaceus Bunge extracts (감초, 황기 추출물로 블랜칭 처리한 신선편이 연근의 갈변 억제)

  • This study was conducted to inhibit the browning of fresh-cut lotus roots by blanching in a Glycyrrhiza glabra L. and Astragalus membranaceus Bunge extract solution (0.5%). Lotus roots were cut to 1 cm using a knife. The fresh-cut lotus roots were blanched at $50^{\circ}C$ in distilled water (DW, Cont), Glycyrrhiza glabra L. extract solution (GE), and Astragalus membranaceus Bunge extract solution (AE). After treatment, they were packaged with 0.04 mm PE bags and were stored at $10^{\circ}C$ for 12 days. Their color, hardness, DPPH radical scavenging activity, pH, and total soluble solids were then investigated. During storage, the samples treated with GE and AE had high $L^*$ values. They were also found to have higher values compared to the control group in the analysis of the browning index. No difference was found, however, in the hardness analysis. With regard to the DPPH radical scavenging activity, the sample treated with Glycyrrhiza glabra L. extract had the highest activity. In the pH analysis, all the groups were found to have a decrease tendency for all the storage periods. The total soluble solids decreased in the control group and slightly increased in the AE and GE groups. These results show that blanching in Glycyrrhiza glabra L. or Astragalus membranaceus Bunge extract is effective for preventing the browning of fresh-cut lotus roots.

Suprathreshold Taste Intensities for Sucrose, NaCl, Citric Acid, and Quinine HCl in Young Koreans and the Influence of Sex, Taste Preference, and Smoking (청년 한국인의 초역치 미각강도에 대한 성, 미각기호 및 흡연의 영향)

  • The aim of this study was to measure the suprathreshold taste intensity for sucrose, NaCl, citric acid, and quinine HCl in Korean young people using a whole-mouth, sip-and-spit procedure, employing the method of magnitude matching. The results were analysed in terms of sex, taste preference, and smoker or nonsmoker. One hundred twenty three subjects (61 male and 62 female, mean age of 25.2$\pm$2.1 years) were included for the study. Subjects were instructed to give nonmodulus magnitude estimates to the intensities of five concentrations each of sucrose, NaCl, citric acid, and quinine HCl; distilled water; 6 loudness levels of a 1,000-Hz tone, using the same 9-point intensity scale. Each of the 21 taste stimuli and 6 auditory stimuli are presented in random order twice. The auditory function is used to assess the absolute intensity function of the subject's taste system. The results were as follows; 1. The perceived taste intensity for sucrose, NaCl, citric acid, and quinine HCl increased progressively as the concentrations of taste solutions were increased. Most of the subjects are standing within the zone of mean value $\pm$1standard deviation in their total perceived intensity score, and nobody is outside the limit of mean value - 2standard deviation. 2. There were not significant difference in total perceived intensities for sucrose, NaCl, citric acid, and quinine HCl between males and females. 3. There were not significant difference in total perceived intensities for sucrose, NaCl, citric acid, and quinine HCl according to the difference in taste preference. 4. There were not significant difference in total perceived intensities for sucrose, NaCl, citric acid, and quinine HCl between smokers and non-smokers in males.

Effects of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) Treatment on the Monosodium Iodoacetate-induced Early Stage Osteoarthritis in Rats (가감소속명탕이 Monosodium Iodoacetate로 유발된 골관절염의 초기변화에 미치는 영향)

  • Objectives: This study was performed to investigate the effects of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) on the monosodium iodoacetate(MIA) induced early state osteoarthritis in rats. Methods: Osteoarthritis was induced by injection of MIA(0.25 mg) into knee joints of rats. Osteoarthritis rats were divided into control(n=8) and treated=8 group respectively, Control group was taken distilled water and treated group was taken extracts of Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) by orally for 20 days. Body weight was measured at 0, 5, 10, 15, 20 days after MIA injection. At the end of experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Proteoglycan(PG) content of articular cartilages were analysed by safranine O staining method. The content of tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta})$ in synovial fluids were analysed by enzyme-inked inmunosorbent assay(ELISA) method. And also cycloxygenase-2(COX-2), matrix metalloproteinase 3(MMP-3), inducible nitric oxide synthase(iNOS), calpain immunochistochemical examination on the knee joints were performed. Results: PG content in articular cartilages of the treated group was significantly increased compared with control group. Histopathological osteoarthritic score of the treated group was significantly decreased compared with control group. $TNF-{\alpha}$ content in synovial fluids, expression of iNOS and calpain in synovial membrane of the treated group were significantly decreased compared with control group. But body weight, $1L-1{\beta}$ content in synovial fluids, expression of iNOS and MMP-3 of the treated group were not significantly changed compared with control group. Conclusions: On the basis of these results, we conclude that Gagamsosokmyeong-tang(Jiajianxiaoxuming-tang) has anti-arthritic effects on the MIA induced early stage osteoarthritis in rats.

A STUDY ON THE EFFECT OF THERMOCYCLING TO THE PHYSICAL PROPERTIES OF DENTURE LINERS (열 순환에 따른 의치이장채의 물리적 성질의 변화에 관한 연구)

  • For the improvement of denture fitness of changed residual ridge, denture liner can be used. Denture liner should be very stable physically in various environments of the mouth as well as be bonded strongly with denture resin. In this study. the specimens bonded with four kinds of soft denture liner and three kinds of hard denture liner were used to test the physical properties of the liners. All experimental groups were stored in $37{\pm}1^{\circ}C$ distilled water for 24hours, followed by thermocycling between $15^{\circ}C$ and $45^{\circ}C$ with 15 second dwell time. 1000, 2000, 3000 cycles of thermocycling were excuted and physical properties were measured by Instron Universal Testing Machine. The obtained results were as follows : 1. In tensile bond test of sea liners, it was shown that both of $Molloplast-B^{(R)}$ specimens before and after thermocycling had the highest tensile strength, and in case of hard liners, Dura-Liner $II^{(R)}$ specimen had the highest tensile strength before and after thermocycling. Depending on thermocycling, $Soft-Relining^{(R)}$, $Denture-Relining^{(R)}$, $Molloplast-B^{(R)}$, $Coe-Soft^{(R)}$ and $Kooliner^{(R)}$ specimen showed significant difference(p<0.05). 2. In strain test of soft liners, it was shown that $Molloplast-B^{(R)}$ specimen before thermocycling and the $Coe-Soft^{(R)}$ after thermocycling showed highest results, and in case of hard liners, the Dura-Liner $II^{(R)}$ specimen before and after thermocycling had the highest result. Depending on thormocycling, $Denture-Relining^{(R)}$, $Molloplast-B^{(R)}$ and Dura-Liner $II^{(R)}$ specimen showed significant difference(p<0.05). 3. In maximum distance test of soft liners. the $Molloplast-B^{(R)}$ specimen before thermocycling and the $Coe-Soft^{(R)}$ after thermocycling showed highest results. and in case of hard liners, the Dura-Liner $II^{(R)}$ specimen before and after thermocycling showed highest result. Depending on thermocycling, $Denture-Relining^{(R)}$, $Molloplast-B^{(R)}$ specimens showed significant difference(p<0.05). 4. In elasticity test of soft liners, the $Molloplast-B^{(R)}$ specimen before and after thermocycling showed highest result. and in case of hard liners, the Dura-Liner $II^{(R)}$ specimen before thermocycling and the $Tokuso-Rebase^{(R)}$ after thermocycling showed highest results. Depending on thermocycling, $Soft-Relining^{(R)}$ $Molloplast-B^{(R)}$ specimens showed significant difference (p<0.05).

THE EFFECTS OF DENTURE CLEANSERS AND DISINFECTANTS ON THE COLOR, SURFACE HARDNESS, SURFACE ROUGHNESS OF DENTURE BASE RESINS (의치세정제와 소독제가 의치상 레진의 색조, 표면경도, 표면조도에 미치는 영향에 대한 연구)

  • The purpose of this study is to compare effects of denture cleansers and disinfectants on the color. surface hardness, and surface roughness of reinforced acrylic resin using polyhedral oligosilsesquioxane molecules(POSS resin) to those of common resins. According to manufacturer's instructions, 45 specimens were made from three denture resins(Luciton 199, Paladent 20, POSS resin), and polished. Five denture cleansers(distilled water, glutaraldehyde, alkaline hypochlorites, chlorhexidine, alkaline peroxides) in combination with three denture resins were evaluated before and after immersion for 7 days. Color data in $L^*a^*b^*$ system were measured with a colorimeter. Surface hardness data were measured with a microhardness tester. Surface roughness data were measured with a 3-dimensional surface analyzer. Data were analyzed with two-way ANOVA, one-way ANOVA, and t-test. The results were as follows : 1. All resins(Luciton 199, Paladent 20, POSS resin) showed significant differences in color after immersion in hypochlorites(p<0.05). 2. POSS resin showed significant differences in color compared with Paladent 20 in all denture cleansers, but no statistically significant differences with Luciton 199(p<0.05). 3. Luciton 199 showed significant differences of surface hardness in chlorhexidine, Paladent 20 showed significant differences in glutaraldehyde and chlorhexidine. POSS resin showed a little change of surface in all denture cleanser, but no statistically significant differences(p<0.05). 4. Luciton 199 and Paladent 20 showed significant differences of surface roughness in hypochlorites and glutaraldehyde, and POSS resin showed no statistically significant differences in all denture cleansers(p<0.05).

A Study on Consolidation Characteristics of Remolded Clay due to the Liquid Limit (액성한계에 따른 재성형 점토의 압밀특성에 관한 연구)

  • In this study, a constant rate of strain test (CRS) and oedometer test was performed in order to study the consolidation characteristics to the liquid limit using a re-shaped clay which was preconsolidated at a constant pressure. Consolidation samples were made of kaolinite which was mixed with bentonite of 6 %, 9 %, 12 % and 15 % of it by weight and the test value of liquid limit of samples were appeared in 77 %, 84 %, 88 % and 91 % respectively. And then consolidation samples which were agitated sufficiently adding distilled water 2 times of liquid limit were preconsolidated in the condition of a constant pressure of 0.2 MPa. The oedometer test which is commonly used recently and the consolidation test of constant rate of strain which were applied in 0.001 %/min, 0.004 %/min, 0.01 %/min rate of strain according to ASTM, D4186-82 were performed and the preconsolidation pressures were compared and analyzed at 0.2 MPa preconsolidated pressure. As a result, in the case of low value of liquid limit, preconsolidation pressure was appeared same as 0.2 MPa preconsolidated pressure at the high speed strain rate, and in the case of high value of liquid limit, preconsolidation pressure was appeared same as 0.2 MPa preconsolidated pressure at the low speed strain rate.
