Non-inverted Meniscus식 모세관 구조물을 이용한 소형 루프히트파이프에 관한 실험적 연구 (A Study on the Miniature Loop Heat Pipe with Non-inverted Meniscus type Capillary Structure)

  • Experimental study was conducted to evaluate the performance of a miniature loop heat pipe (MLHP) with non-inverted meniscus type capillary structure. All parts of MLHP in this study were made of copper including the capillary structure and the distilled water was used as a working fluid of MLHP. The outer diameter of evaporator was 9 mm and its length was 119 mm. The effective pore size of the capillary structure was 30 micron and its porosity was 60%. The vapor transport line, the liquid transport line and the condenser were consisted of single 4.0 mm copper tube. The distance between the evaporator and the condenser region was 200 mm and the length of the loop was 969 mm. This MLHP was operated successfully at any orientation but the gravity highly influenced the thermal performance of the MLHP. The maximum thermal load was 130 watts at the bottom heat mode and the 20 watts at the top heat mode.

Optimization of Extraction Conditions and Quantitative Analysis of Isoquercitrin and Caffeic Acid from Aster scaber

  • To determine the optimum extraction conditions that give the highest yield of isoquercitrin and caffeic acid from Aster scaber, the effects of four extraction variables (solvent concentrations, extraction time, number of repeated extraction, and solvent volumes) on isoquercitrin and caffeic acid yield was examined via HPLC-UV. Our results showed that the highest extract and isoquercitrin yield were observed when A. scaber was extracted with 450 mL distilled water for 8 hr repeatedly for three times. In case of caffeic acid, the content was higher in the two repeated extracts. Also, content analysis of isoquercitrin in Aster species was performed in which A. fastigiatus, A. ageratoides, and A. scaber exhibited the highest isoquercitrin content at 6.39, 5.68, and 2.79 mg/g extract, respectively. In case of caffeic acid, the highest content of A. scaber and A. glehni was 0.64 and 0.56 mg/g extract, respectively. This study reports an optimized method for extraction of isoquercitrin and caffeic acid from A. scaber and evaluates potential sources of the compounds.

Simultaneous Determination of the Seven Phenylpropanoids in Xanthii Fructus Using a HPLC-PDA and LC-MS

  • Xanthii Fructus has been traditionally used for the treatment of rhinitis, rheumatoid arthritis, and eczema. In this study, a high-performance liquid chromatography-photodiode array (HPLC-PDA) method was developed and then used for the simultaneous analysis of eight phenylpropanoids in Xanthii Fructus. The analytical column used for this separation was a $SunFire^{TM}$ $C_{18}$ column, maintained at $40^{\circ}C$. The mobile phase used was 1.0% acetic acid in distilled water and 1.0% acetic acid in acetonitrile with gradient elution. For identify of each component, the mass spectrometer (MS) was used a Waters triple quadrupole mass spectrometer requipped with electrospray ionization (ESI) source. The HPLC-PDA method showed good linearity: correlation coefficients were ${\geq}0.9996$. The limits of detection and quantification of the eight compounds were 0.02 - 0.04 and $0.06-0.14{\mu}g/mL$, respectively. The extraction recoveries ranged from 97.51 to 108.67%. The relative standard deviation values of intra- and inter-day precision were 0.06 - 1.55 and 0.09 - 1.68%, respectively. The validated HPLC-PDA method was applied to simultaneously analyse the amounts of eight phenlypropanoids in Xanthii Fructus.

Re-induction of Embryogenic Tissue from the Cryopreserved Somatic Embryo in Japanese Larch (Larix leptolepis Gordon)

  • The study aimed to develop a cryopreservation method for long-term storage using mature somatic embryo of Japanese larch. In this study, desiccation treatments significantly affected re-induction rates of embryogenic tissue (ET) from dried somatic embryos. In the effect of different dehydration temperature and duration on the re-initiation ET. the highest frequency was shown when somatic embryos were dehydrated at $25^{\circ}C$ for 2 (45.5%) or 1 day (43.3%), respectively. In addition, low temperatures [$4^{\circ}C$, 2 days (44.2%) or 3 days (43.5%)] were marked higher ET initiation. After that, the initiation value was declined with dehydration duration. For comparison of different relative humidity on re-induction frequency of ET, the best re-induction (43.5%) was obtained from somatic embryos pre-dried at $(NH_4)_2SO_4$ (RH 79%). Both $Na_2HPO_4$ (RH 97%) and $Na_2CO_3$ (RH 88%) treatments were showed the similar rate, 34.6, 34.2%, respectively. However the lowest rate (19.6%) was observed in distilled water (RH 100%). In comparison of the various storage temperatures and duration of the dried somatic embryos, the highest frequency (66.9%) of re-initiation was obtained when somatic embryos were cryopreserved for one day. However, the frequency was gradually decreased as the time length of storage increased regardless of types of storage. None of ET re-initiated when stored at $4^{\circ}C$ for 1, 2 and 84 days.

산조인의 수치에 따른 Spinosin 함량 비교 (A Comparison of Spinosin Content in Zizyphi Semen and Its Processed Products by Roasting)

  • The aim of this study was to compare the amount of spinosin in the 70% ethanol extracts of non-processed Zizyphi Semem (ZS) and processed ZS by roasting using a high-performance liquid chromatography equipped with photodiode array detector. Separation of the spinosin was used $SunFire^{TM}$ $C_{18}$ analytical column ($5{\mu}m$, $4.6{\times}150mm$) using two mobile phase consisting of distilled water and acetonitrile, both with 1.0% (v/v) acetic acid. The flow rate was 1.0 mL/min and injection volume was $10{\mu}L$. Calibration curve of the spinosin was y = 22339.45x+483.99 in tested concentration range ($1.28-20.00{\mu}g/mL$) and correlation coefficient was 1.0000. In non-processed ZS sample, the amount of the spinosin was 0.94 m/g, while, the amount of the marker compound in processed ZS samples were 0.66-1.10 mg/g.

Characterization of an Anti-gout Xanthine Oxidase Inhibitor from Pleurotus ostreatus

  • We selected Pleurotus ostreatus from among several edible mushrooms because it has high anti-gout xanthine oxidase (XOD) inhibitory activity. The maximal amount of XOD inhibitor was extracted when the Pleurotus ostreatus fruiting body was treated with distilled water at $40^{\circ}C$ for 48 hr. The XOD inhibitor thus obtained was purified by Sephadex G-50 gel permeation chromatography, ultrafiltration, $C_{18}$ solid phase extraction chromatography and reverse-phase high-performance liquid chromatography with 3% of solid yield, and its XOD inhibitory activity was 0.9 mg/mL of $IC_{50}$. The purified XOD inhibitor was a tripeptide with the amino acid sequence phenylalanine-cysteine-histidine and a molecular weight of 441.3 Da. The XOD inhibitor-containing ultrafiltrates from Pleurotus ostreatus demonstrated dose-dependent anti-gout effects in a Sprague-Dawley rat model of potassium oxonate-induced gout, as shown by decreased serum urated levels at doses of 500 and 1,000 mg/kg, although the effect was not as great as that achieved with the commercial anti-gout agent, allopurinol when administered at a dose of 50 mg/kg.

Development of Analytical Method of Biotin in Complex Drugs and Dietary Supplements Using HPLC-UV

  • Recently, Korean Food and Drug Administration (KFDA) has focused on developing quality control guidelines for all commercial products in Korea to enforce regulations, improve the quality control, and protect consumers by developing prevalently used and efficient analytical tools to determine and quantify target compounds. Because the Korean Pharmacopeia (KP) presents microbiological assays for biotin, which is laborious and time-consuming, this study is focused on applying HPLC-UV to detect and quantify biotin in complex drugs and dietary supplements like multi-vitamin. Biotin in complex drugs was extracted from methanol and analyzed using mobile phase with 10 mM potassium phosphate (monobasic, pH=3.0) in distilled water and acetonitrile. Gradient condition was used to successfully detect and quantify biotin within 20 minutes. Validation result for linearity was significant that average $r^2$ was 0.999 (n=3) and its relative standard deviation (RSD) was 0.0578% which was less than 2%. Using this method, quantification of biotin in complex drugs was completed successfully and recovery tests were finished that recovery percentage greater than 95% with relative standard deviation less than 2%.

  • Zingiberis Rhizoma (ZR, Zingiberaceae) is one of the foods for a long time and contains various biological activities including anti-cancer, anti-inflammatory, anti-oxidant, and anti-platelet activities. The present study was to compare the amount of 6-gingerol in the 70% ethanol extracts of non-processed ZR and processed ZR by roasting using a high-performance liquid chromatography equipped with photodiode array detector. The 6-gingerol was separated on a Gemini $C_{18}$ analytical column ($5{\mu}m$, $4.6{\times}250mm$) using two mobile phase consisting of distilled water and acetonitrile. The flow rate was 1.0 mL/min and injection volume was $10{\mu}L$. In non-processed ZR sample, the amount of the 6-gingerol was 0.43% and the amount of the marker compound in processed ZR samples by roasting were 0.16-0.55%.

피리도스티그민 정제의 함량 측정을 위한 HPLC 분석법 (Quantitative Analysis of Pyridostigmine Bromide in Tablets by HPLC)

  • A reverse-phase, ion-pair high performance liquid chromatographic (HPLC) method for the simultaneous quantative determination of pyridostigmine and its hydrolytic product, 3-hydroxy-N-methylpyridinium (HMP), is descrihed, The assay of pyridostigmine and HMP was linear in the range of amount from 24 to 60 mg/tablet and from 2.4 to 12.0 mg/tablet, respectively, with coefficient of variation (C.V.) of 0.05-0.12% (n=7) and 0.25-0.52% (n=5), respectively, and applicable conveniently even in the case of the mixture of pyridostigmine and HMP. Meanwhile, the conventional UV method gave inaccurate results for the aged pyridostigmine tablets. In the extraction of pyridostigmine from tablets prior to be assayed by HPLC, methanol was found to be more effective than ethanol or distilled water. Multiple extraction (four times) with methanol resulted in the full recovery of pyridostigmine, whereas ethanol gave 95% recovery even after four times extraction. Based on these results. the present method would be very useful for the accurate determination of pyridostigmine in the aged pyridostigmine tablets.

Polygala japonica houttuyn의 성분연구 (Studies on the constiuents of polygala japonica houttuyn)

  • The finely powdered entire harbs of Polygala japonica Houttuyn, a drug known as Yong Shin-Cho in Korea, were boiled with methanol and from the filtered extract the methanol was distilled off under diminished pressure and the aqueous solution of the residue was evaporated to dryness after being mixed with ignited magnesia. The dried mass was boiled with absolute alcohol, the filtered clear liquid was evaporated to a small volume and the precipitate Saponin produced by mixing with ether was filtered off. When the filtrate was again evaporated to the thickness of a syrup and allowed to cool for a few days in an ice box, crystalls were separated out in about 5% yield, which formed colorless columns, M. P. 142.deg., from methanol and had the formula $C_{6}$ $H_{12}$ $O_{5}$. On heating it with acetic acid anhybride and sodium acetate, its tetraacetyl derivative $C_{6}$ $H_{8}$ $O_{5}$(C $H_{3}$CO)$_{4}$ was obtained and which formed colorless needles, M. P. 62-5.deg., from ethanol. Their melting points, results of elementar analysis and other characteristics agreed with that of Polygalitol and its derivative. Finally they were proved to be identical with Polygalitol and its derivative, respectively, through determination of mixed melting points with the samples. Polygalitol was isolated from several plants of genus polugala e. g. P. amara, P. vulgaris, P. teunifolia, P. senega etc. The authors added to them another instance of identifying Polygalitol from the plant of genus polygala.olygala.
