  • In the production of a low cost bacterial insecticide, it is important to produce a high spore concentration using low price substrates. Experiments were carried out to investigate the effects of the addition of mineral salts and glucose, and of dissolved oxygen concentration on the cell growth and spore formation of Bacillus thuringiensis var aizawai using a cheap wheat and soybean meal in the batch culture. The maximum viable cell number was 1.2${\times}$109 CFU/mL at 12 hr culture and spore yield was 54.2% at 74 hr culture using an industrial medium containing 20 g/L wheat meal and 30 g/L soybean meal under 1.0 vvm aeration and 200 rpm agitation. The cell growth and the spore formation were not enhanced by the addition of mineral salts in industrial medium, whereas th addition of 10g/L glucose decreased the cell growth and spore formation. We could obtain a maximum viable cell number of 2.2${\times}$109 CFU/mL and spore number of 1.9${\times}$109 CFU/mL at the dissolved oxygen concentration of 60% of saturation. The spore concentration was enhanced approximately by 2 times as compared to the dissolved oxygen concentration of 50%. In the bench-scale culture, the maximum viable cell and spore number were 2.5${\times}$109 CFU/mL, and 2.2${\times}$109 CFU/mL, respectively under 1.0 vvm aeration and 400 rpm agitation. The spore yield was 88% based on the maximum viable cell number. As a result, it was confirmed that the production of high spore concentration could be obtained by a bench-scale culture using an industrial medium.

  • Distribution of suspended particulate matter, dissolved oxygen and major inorganic nutrients along a meridional section ($126^{\circ}$ 33' E) in the Cheju Strait is described along with the hydrographic and current data obtained during April 25-27, 1995. The current measurements was conducted using a vessel-mounted Acoustic Doppler Current Profiler (ADCP). Repeated coverage along an ADCP transect during 25 hours allows to calculate the daily mean along- and cross-strait currents. Measured material concentrations and the mean current speed were used to estimate the flux density (cencentration times current speed) of materials. Two types of depth distibution of flux densities were observed. for nitrate and suspended particulate matter, the depth distribution pattern of materials determines those of flux densities. However, flow patterns determine those of flux densities for dissolved oxygen, phosphate and silicic acid. The total along-strait water volume transport is about 0.3 Sv (1Sv $10^{6}$ $m^{3}/s^{-1}$). The total along-strait material transports are estimated to be 3.1 $${\times} $10^{5}$ $g/s^{-1},$ 2.4 ${\times}$ $10^{6}\;g/s^{-1},$ 7.I ${\times}$ $10^{2}\;mol/s^{-1},$ 3.I ${\times}$ $10\;mol/s^{-1},$ 1.7 ${\times}$ $10^{3}\;mol/s^{-1}$ for suspended particulate matter, dissolved oxygen, nitrate ion, silicic acid and phosphate ion, respectively.

  • Experiments were carried out to examine the effects of the different levels of oxygen, ammonia and hydrogen sulfide on survival, specific growth rate (SGR) and feed conversion ratio (FCR) of juvenile shrimp, Fenneropenaeus chinensis. Survival of the shrimp exposed to the hypoxia with $\leq$2.5 mg/L dissolved oxygen for 24 days were significantly affected. SGR and FCR of the shrimp exposed to$\leq$3.0 mg/L dissolved oxygen were significantly reduced than those of shrimp reared at 6.8 mg/L dissolved oxygen concentration. Survival of the shrimp exposed to $\geq$2.0 mg/L of ammonia levels for 24 days was significantly affected. Moreover, compared to the control group significant decrease of SGR and FCR of the shrimp has been observed with $\geq$1.0 mg/L and $\geq$0.5 mg/L of ammonia concentrations. In case of hydrogen sulfide, $\geq$0.5 mg/L was the critical level showed its significant negative effect on survival rate of shrimp exposed for 24 days. While, the group exposed with $\geq$0.07 mg/L and $\geq$0.05 mg/L hydrogen sulfide levels had a lower SGR and FCR values than did the control group in the same stipulated time of exposure.

  • The purpose of this research was to investigate the effects of low concentration of oxygen on reduction of perchlorate, especially low perchlorate influent concentrations in a biofilm reactor, as well as the effect of flow pattern in a biofilm reactor. Dissolved oxygen averaging 1 mg/L did not inhibit reduction of influent perchlorate from 23 to $426\;{\mu}g/L$ in the biofilm reactors when sufficient acetate was added, probably due to limitation of oxygen diffusion into the biofilm. Influent perchlorate ranging from 23 to $426\;{\mu}g/L$ was reduced to below detection level ($4\;{\mu}g/L$) in the presence of 1 mg/L dissolved oxygen (DO). Chloride was produced in a ratio of $0.37gCl^-/g{ClO_4}^-$ and $0.35gCl^-/g{ClO_4}^-$ in plug flow and recirculation biofilm reactor which is similar to stoichiometric amount ($0.36gCl^-/g{ClO_4}^-$) indicating complete perchlorate reduction at $426\;{\mu}g/L$ of ${ClO_4}^-$ feeding. At $23\;{\mu}g/L$L influent perchlorate, total biomass solids were 3.18 g and 2.81 g in the plug flow and recirculation biofilm reactors. The most probable number(MPN) analysis for perchlorate-reducing bacteria showed $10^4$ to $10^5\;cells/cm^2$ in both biofilm reactors throughout the experiments. The effluent perchlorate concentrations were not significantly different in the two different flow regimes, plug flow and recirculation biofilm reactors.

  • An evaluation method for oxygen permeable characteristics of the membrane covering to each cathode of multiple cathode - single anode type dissolved oxygen sensor, which has high reproducibility and is capable of measuring multiple components in solutions. For this purpose, a measuring circuit for the multiple cathode type DO sensor was designed to lower the noise signal by adapting a digital LPF to readout the sensor output accurately. Digital LPF is designed by setting up the transfer function to set the cutoff frequency to 10Hz, and the transfer function is programmed by C language, and then the filtering characteristics are evaluated with the simulation and experiments. Using this LPF added measuring circuit for the multiple cathode type DO sensor, we have obtained the calibration factor for each cathode to calibrate the variation of the output signals. The calibration factor was obtained by measuring the sensor output signal followed by oxygen partial pressure, using the same oxygen permeable membrane at each cathode of the multiple cathode type DO sensor.

  • The effects of dissolved oxy-gen(DO) concentration and DO control modes on cell growth in a 4L computer-controlled bioreactor system were investigated using Phellinus linteus WI-001, a producer of protein-polysaccharides having potent anticancer activi-ties. When DO was controlled at about 20%, maximum cell concentration and specific growth rate of the strain were observed to increase 36% and 64%, respectively, as compared to the experiment performed without DO control. By adopting cascade automatic control of DO ar 20% in a mixed automatic control mode using computer-con-trolled program, 19.5g/L of maximum cell concentration was obtained. These results showed that the mixed auto-matic control mode was the effective method for enhancing cell growth of the shear sensitive Phellinus linteus.

  • A series of laboratory-scale corrosion experiments was carried out to observe the effect of dissolved oxygen (DO) in the presence of other water quality parameters, such as hardness, Cl-, and pH using various pipe materials. In addition, a simulated loop system was installed at a water treatment plant for pilot-scale experiment. Laboratory-scale experiment showed that corrosion rates for galvanized steel pipe (GSP), carbon steel pipe (CSP), and ductile cast iron pipe (DCIP) were decreased to 72%, 75%, and 91% by reducing DO concentration from 9${\pm}$0.5 mg/L to 2${\pm}$0.5 mg/L. From the pilot scale experiment, it was further identified that the average ionization rate of zinc in GSP decreased from 0.00533 to 0.00078 mg/$cm^2$/d by controlling the concentration of DO. The reduction of average ionization rate for copper pipe (CP) and stainless steel pipe (SSP) were 71.4% for Cu and 63.5% for Fe, respectively. From this study, it was concluded that DO could be used as a major parameter in controlling the corrosion of water pipes.

  • The purpose of this study is to develop the stochastic stream water quality model for the intake station of Chung-Ju city waterworks in the Han river system. This model was based on the theory of Box-Jenkins Multiplicative ARIMA(SARIMA) and the state space model to simulate changes of water qualities. Variable of water qualities included in the model are temperature and dissolved oxygen(DO). The models development were based on the data obtained from Jan. 1990 to Dec. 1997 and followed the typical procedures of the Box-Jenkins method including identification and estimation. The seasonality of DO and temperature data to formulate for the SARIMA model are conspicuous and the period of revolution was twelve months. Both models had seasonality of twelve months and were formulates as SARIMA {TEX}$(2,1,1)(1,1,1)_{12}${/TEX} for DO and temperature. The models were validated by testing normality and independency of the residuals. The prediction ability of SARIMA model and state space model were tested using the data collected from Jan. 1998 to Oct. 1999. There were good agreements between the model predictions and the field measurements. The performance of the SARIMA model and state space model were examined through comparisons between the historical and generated monthly dissolved oxygen series. The result reveal that the state space model lead to the improved accuracy.

  • Biosynthesis of polyhydroxybutyrate and copolymer consisting of 3-hydroxybutyrate and 3-hydroxyvalerate [poly(3HB-co-3HV)] by Bacillus thuringiensis R-510 grown with glucose or with mixtures of glucose and propionate was investigated. n-Alkanoic acids other than propionate were not precursors of 3HV units. The fraction of 3HV unit in the copolymer increased from 0 to 84 mol% of 3HV. Polymer yield decreased as the fraction of propionate was increased but the molecular weight distribution was not affected by the composition of carbon substrate. The minimum melting temperature (around 65.deg.C) of poly (3HB-co-3HV) copolymers was observed for the polymer bearing approximately 35 mol% of 3HV. Polyhydroxyalkanoates production by this organism was not dependent on nutritional limitation, but remarkably influenced by dissolved oxygen concentration in the culture medium. Low level of dissolved oxygen concentration prevented spore formation in the cells and stimulated the synthesis of polyhydroxyalkanoate. The composition of poly (3HB-co-3HV) produced by B. thuringiensis R-510 lyhydroxyalkanoate. The composition of poly(3HB-co-3HV) propduced by B. thuringiensis R-510 varied according to the growth time. However, there was no evidence that polymers isolated from cells were mixtures of immiscible polymers.
