식물세포분리효소를 이용한 수박과 참외의 단세포 함유 반응물 특성 (Characteristics of Suspension Containing Single Cells from Watermelon and Muskmelon Treated with Cell Separating Enzymes)

  • 수박과 참외의 황용도 증진을 위하여 조직을 개개의 세포로 분리할 수 있는 단세포화 기술을 적용하였다. 그 결과 이들을 시판 식물세포분리효소인 Macerozyme A와 Sumyzyme MC로 30분에서 120분간 반응시간별 단세포 함유 반응물의 특성을 조사하였다. 단세포화로 얻을 수 있는 수율, 반응률, 단세포 함유 반응물의 색도, 점도, 총당, 총펙틴, 총폴리페놀, 무기질, 유리 아미노산 함량을 조사하였으며 또한 분리된 단세포의 형태변화를 입도분포로 조사하였다. 전반적으로 효소반응시간이 경과 할수록 반응물의 성분 함량은 증가하였으나 세포벽에 의하여 내부 구성성분이 보호를 받아 안정하게 유지되는 것으로 나타났다. 수박과 참외의 무기질 함량은 주스보다는 단세포 함유 반응물에서 상대적으로 함량이 높게 나타났다. 참외의 경우 무기질인 K의 함량은 효소반응물과 주스에서 각각 240.8 mg%, 172.7 mg%였다. 본 연구 결과는 수박과 참외로부터 효소반응 물의을 이용하여 새로운 형태의 음료 개발을 위한 기초자료로 활용 가능할 것으로 판단되었다.

혐기성 소화효율 향상을 위한 초음파를 이용한 슬러지 전처리 및 ADM1 모의 (Ultrasonic Sludge Disintegration for Improving Anaerobic Digestion and Simulation of ADM1)

  • 본 연구에서는 초음파 cavitation을 이용한 슬러지의 전처리가 슬러지의 가용화와 혐기성 소화조의 운전효율에 미치는 영향을 바이오가스 생성을 중심으로 알아보고, 전처리의 효과를 ADM1 (Anaerobic Digestion Model No. 1)을 이용하여 시뮬레이션 한 결과와 비교하였다. 실험결과, 초음파 전처리는 하수슬러지의 탈수성을 감소시키며 SCOD의 생성을 증가시키는 것으로 나타났다. 실험실 규모의 혐기성 소화조는 초음파 미처리 슬러지(control)와 30분, 60분, 90분간 처리된 슬러지를 대상으로 4 계열로 운전되었으며, 그 처리효율은 TCOD의 경우, 각각 31.9%, 37.9%, 38.5%, 42.2%의 제거효율을 나타내었다. 또한 TS의 감량효율은 각각 15.9%, 20.8%, 21.5%, 24.1%로 평가되어 전처리에 따른 제거효율의 증가를 확인 할 수 있었고, 슬러지 부하와 전처리 시간의 증가에 따라서 바이오가스의 생성량도 증가하는 것으로 나타났다. 슬러지 전처리에 따른 혐기성 소화조의 운전결과와 시뮬레이션 결과의 추이는 전반적으로 일치하는 경향을 나타내었다.

Flucetosulfuron 수면부상형 입제의 최적화 및 제초효과 (Optimization of self-dispersible floating granule (UG) of flucetosulfuron and its herbicidal performance)

  • Flucetosulfuron 수면부상성 입제의 제형요소 및 효능을 최적화하기위한 일련의 실험을 실시하였다. SPA와 xanthan gum을 혼합하여 입제로 제제한 경우 조립성과 부상성이 우수하여 점착제로 선발하였으며 증량제는 조립성, 부상성 및 붕괴성이 우수한 KCl을 선발하였다. 또한 습윤 및 분산제로는 분산성이 가장 우수한 Surfynol 440을 선발하였다. 입제의 입경은 부상성과 분산성을 고려 1.0 mm로 결정하였다. 본 연구로 선발된 부제를 이용해 조제한 flucetosulfuron 수면부상성 입제를 실내 간이 포장 및 야외 포장에서 제초활성을 평가한 결과 잡초를 처리지점에서 15 m에 다다르는 지점까지 우수하게 방제하였다. 따라서 본 수면부상성 입제 기술은 flucetosulfuron을 비롯한 노동력 절감형 제초제 개발에 사용될 수 있을 것으로 기대된다.

현대 패션에 나타난 의복구성의 부분 변형 (The Partial Transformation of Clothing Construction in Modern Fashion)

  • Fashion have been expressed by a face through various a period, social change, and various culture changing. Human expresses as "Transformation" by applying these needs of changing to the fashion. A origin tribe expressed its own self by using direct body transformation and extreme decorations in the past. However, human express creative and esthetic desire based on shape, material, and transformation method in the present time characteristics of the body. Exceptional transformation breaking a existing fixed idea appeared frequently due to dissolution through transformation which is positive expression method in the fashion from 20th century. As a results, followings are obtained in this study. First, human body transforms by using a tool or intermediation in investigation of aesthetic meaning for human body. The object, Transformation, is stably transformed by disintegration, distortion, exaggeration and simplification reduction, as design's sensitivity. Second, transformation from relation of clothing composition is expressed by extension, reduction, simplification, and dissolution. In transformation from original tribe's sensitivity, past decorative desire lead to transformation of human body. To give variable change from past to present fashion, external formative will is introduced. Then, extreme expression is made by direct transformation of clothing type. It seems to be accomplished that human body's expression method is continuously changed into extension, exaggeration, reduction, and dissolution from transformation method as described before. Transformation of modem fashion is expression method by creative supervision. Extreme transformation substituted body's each part is based on immanent play and representative satisfaction. Through these transformation, it is judged that variety of creative type is achieved.

In Vitro Schistosomicidal Activity of Phytol and Tegumental Alterations Induced in Juvenile and Adult Stages of Schistosoma haematobium

  • There is renewed interest in natural products as a starting point for discovery of drugs for schistosomiasis. Recent studies have shown that phytol reveals interesting in vivo and in vitro antischistosomal properties against Schistosoma mansoni adult worms. Here, we report the in vitro antischistosomal activity of phytol against Schistosoma haematobium juvenile and adult worms and alterations on the tegumental surface of the worms by means of scanning electron microscopy. The assay, which was carried out with 6 concentrations (25, 50, 75, 100, 125, and $150{\mu}g/ml$) of phytol, has shown a promising activity in a dose and time-dependent manner. There was a significant decline in the motility of the worms and a mortality rate of 100% was found at 48 hr after they had been exposed to phytol in the concentration of $150{\mu}g/ml$. Male worms were more susceptible. On the ultrastructural level, phytol also induced tegumental peeling, disintegration of tubercles and spines in addition to morphological disfiguring of the oral and ventral suckers. This report provides the first evidence that phytol is able to kill S. haematobium of different ages, and emphasizes that it is a promising natural product that could be used for development of a new schistosomicidal agent.

미생물 발효 영지버섯 추출액의 다당체에 관한 연구 (Studies on the Polysaccharide of Ganoderma lucidium Extract by Microorganism Fermentation)

  • In order to investigate the totale yield, the content of soluble polysaccharide and the others of FEW extract from the yeast fermentated Ganoderma lucidium by supersonic method, the yeast strain was inoculated after pretreatment and subsequently followed fermentation and supersonic extract. The main construction of the extract method is composed of the main glucose and together with the xylose, fucose, galactose and mannose. Results show that because of the generated lactic acid and ethanol, pH value of extract decreases and the safety as well as the preservation is improved. The extract yield, the total soluble polysaccharide, SOD-like activity, cytotoxic effect and growth inhibitory effect against cancer cell line are much higher in FE method than RE method, especially FEW3 extracts fermented during 24hrs. It is concluded that yeast fermentation makes the extract yield increase because of the cell disintegration, the useful ingredients of the germ body, the metabolic products, the insoluble ingredients due to the generation of ethanol, and the cell fragmentation caused by the supersonic waves vibration. Content of generated ethanol, total soluble polysaccharide and extract yield all increase during the fermentation time from 24 to 72 hours and the optimum fermentation condition is at $27^{\circ}C$ for 72 hours. The bitter taste and smell of the Ganoderma lucidium extract is diminished, fragrant-bitter taste and smell is generated so that the whole functional quality is improved.

디젤기관의 분무선단 도달거리에 관한 모델링 (A Modeling about Penetration Behavior of Diesel Engine Liquid Fuel Spray)

  • 본 논문에서는 소형 고속 디젤 분무의 반경 방향의 연류 농도 분포를 고려한 비균질 분무의 분무 선단 도달 거리에 대한 관계식을 유도하고, 비정상 분무인 디젤 분무의 분사 차압-시간 곡선을 입력시킴으로써, 분사 초기 뿐 아니라 전 분사 기간의 분무 선단 도달 거리를 예측할 수 있는 새로운 모델을 제안하여 실험치와 비교 분석하였다. 그리고 분사각과 분열장의 무차원 실험식을 도출하여 계산에 사용하였고 분무 선단 도달 거리의 천이점과 분열장에 대하여 고찰하였다.

Calcium sulfate-Hydroxyapatite 혼합재의 물성 및 치근천공 치유효과에 관한 연구 (THE PHYSICAL PROPERTIES AND HEALING EFFECT OF CALCIUM SULFATE-HYDROXYAPATITE COMPOUND ON ROOT PERFORATION)

  • Treatment of root perforation elicits special considerations due to its blood-contaminated circumstances. It is known that conventional dental restorative materials are all leaking. Calcium sulfate is the material which react with water to become chemically set. This study, therefore, was performed to develop a new compound containing calcium sulfate and to evaluate its physical and biological characteristics. Three materials were used, IRM, calcium sulfate, calcium sulfate-hydroxyapatite compound. The composition of the calcium sulfate-hydroxyapatite compound was basically 50 % of calcium sulfate and 50 % of hydroxyapatite mixed with guajacol. The materials were mixed in conventional way and underwent four physical test procedures, setting time, solubility test, compressive strength, and marginal leakage test. All materials were evaluated under the scanning electron microscope to examine the marginal sealing ability. Animal experiment was also performed to test the materials' tissue response. Twenty-four dog's premolars were tested with either furcation perforations or apical retro-fillings. From the results, we found that calcium sulfate possess the good marginal sealing ability. However, calcium sulfate creates many voids which is caused by crystal thrusting action when it reacts with water. It seemed that the voids caused disintegration of the material which eventually lead to tissue reaction. By compounding calcium sulfate and hydroxyapatite, we were able to obtain the better physical properties but it showed larger marginal gap between the material and the root surface. Within the six weeks observation period, both IRM and calcium sulfate-hydroxyapatite compound showed good tissue responses in animal experiment. It is concluded that calcium sulfate would be the material of choice in root perforation repair, but the physical property needs to be further improved.

Optimization Study on the Formulation of Roxithromycin Dispersible Tablet Using Experimental Design

  • This study set out to improve the physical and pharmaceutical characteristics of the present formulation using an appropriate experimental design. The work described here concerns the formulation of the dispersible tablet applying direct compression method containing roxithromycin in the form of coated granules. In this study $2^3$ factorial design was used as screening test model and Central Composite Design (CCC) associated with response surface methodology was used as optimization study model to develop and to optimize the proper formulation of roxithromycin dispersible tablet. The three independent variables investigated were functional excipients like binder (X1), disintegrant (X2) and lubricant (X3). The effects of these variables were investigated on the following responses: hardness (Y1), friability (Y2) and disintegration time (Y3) of tablet. Three replicates at the center levels of the each design were used to independently calculate the experimental error and to detect any curvature in the response surface. This enabled the best formulations to be selected objectively. The effect order of each term to all response variable was X3> X2> Xl> X1*X2> X2*X2> X2*X3> X3*X3> Xl*X3> Xl*Xl and model equations on each response variables were generated. Optimized compositions of formula were accordingly computed using those model equations and confirmed by following demonstration study. As a result, this study has demonstrated the efficiency and effectiveness of using a systematic formulation optimization process to develop the tablet formulation of roxithromycin dispersible tablet with limited experiment.

Preliminary studies on the microplastic pollution in Dal lake, Kashmir (first report)

  • We provide the first study on the occurrence of microplastics in Dal lake, Kashmir, India. Microplastics act as catastrophe that trigger many environmental problems. The key origins of microplastics are larger plastics, which split into smaller plastics after UV light disintegration. There is relatively little work carried out on the existence of microplastics. The present work has been undertaken on the occurrence of microplastics at four pre-selected sites (surface water) in Dal lake, Kashmir. The samples were taken to the laboratory to dissolve organic matter by using H2O2 (6%). To speed up the organic digestion; the treated mixture was heated on a hot plate at 70℃. The mixture was then subjected to density separation. The supernatant obtained was observed under the microscope (10X) and measurements were taken. At site-I, the microplastics ranged from 2-3 mm, site-II 5-6 mm, site-III 3-4 mm, site-IV 4-5 mm in thickness, indicating the presence of microplastics in the lake. The presence of microplastics indicated that the lake has undergone an anthropogenic change over a period of time. Our research highlights the value of enhancing the quality of the drainage system and sewage disposal. This work can be helpful to recognize successful microplastic control management techniques and possible threats associated with the Dal lake. So far, no such data on the presence of microplastics in Kashmir lakes is available.
