Reproductive Cycle of Small Filefish, Rudarius ercodes (그물코쥐치, Rudarius ercodes의 생식주기)

  • The reproductive cycle of the small filefish, Rudarius ercodes was investigated based on the annual variations of gonadosomatic index(GSI) and hepatosomatic index(HSI) by electronic and photic microscophy. The specimens used were collected at the coastal area of Benden island, Sizuokagen, Japan, from September 1982 to August 1983. GSI began to increase from March, starting season of longer daylength and higher water temperature, and reached the maximum value between June and August. It began to decrease from September with the lowest value appearing between November and February without any evident variation. The annual variations of HSI were not distinct in male filefish and were negatively related to GSI in female : HSI decreased in the summer season when the ovary was getting mature and reached the maximum in the winter season when the ovary was getting retrogressive. The ovary consisted of a pair of saccular structure with numerous ovarian sacs branched toward the median cavity. Oogonia divided and proliferated along the germinal epithelium of the ovarian sac. Young oocytes with basophile cytoplasm showed several scattering nucleoli along the nuclear membrane. when the oocytes growing to about 300 ${\mu}m$, nuclear membrane to disappear with nucleus migrating toward the animal pole. The regions of protoplasm were extremely confined within the animal hemisphere in which most of cytoplasms were filled with yolk materials and oil drops. After ovulation, residual follicles and growing oocytes remaining in the ovarian sacs degenerated. But perinucleatic young oocytes without follicles formed were not degenerated, and growing continuously still in the next year. Mitochondria and endoplasmic reticula in the cytoplasm remarkably increased with oocytes maturing and yolk accumulating. Those were considered to be functionally related to the yolk accumulation. Five or six layers of possible vitellogenin, oval-shaped disc structures with high electron density, appeared in the apex of follicular processes stretching to the microvilli pits of mature oocytes. Testis consisting of a pair of lobular structures in the right and left were united in the posterior seminal vesicle, Cortex of testis was composed of several seminiferous tubules, and medulla consisting of many sperm ducts connected with tubules. Steroid hormone-secreting cells with numerous endoplasmic reticula and large mitochondria of well developed cristae were recognized in the interstitial cells of the growing testis. Axial filament of spermatozoon invaginated deeply in the central cavity of the nucleus and the head formed U-shape with acrosome severely lacking, mitochondria formed large globular paranuclei at the posterior head, and microtubular axoneme of the tail represented 9+9+2 type. The annual reproductive cycles could be divided into five successive stages : growth(March to July), maturation(May to September), Spawning(mid May to early October) and resting stages(October to February). The spawning peak occurred from June to August.

Effect of Blue Light on the Major Components of Soybean-sprouts (Blue광(光) 조사(照射)가 콩나물의 주요성분(主要成分)에 미치는 영향(影響))

  • Growth of soybean sprouts(Glycine Max L.) and amounts of some chemical components were measured when they were exposed to blue light (120lux, 3hrs/day) during their growth. Hypocotyl length of irradiated soybean sprouts exceeded slightly that of control (dark) soybean sprouts, but the tfresh weight of whole sprouts as well as each part of the sprout showed no difference between the two groups. Chlorophyll content of cotyledon under blue light increased significantly with the lapse of days (3.57 and $8.45\;{\mu}g/100g$ fresh weight on the 3rd and 7th day). Bluelight irradiated sprouts contained more vitamin C than control sprouts (21.7% and 30.8% higher for the cotyledon and hypocotyl). Total amount of protein was not affected. Hypocotyl protein content was 8% of that in original soybean. Blue light did not affect the activity of trypsin inhibitor of sprouts. Similar activity of the inhibitor was observed in the cotyledon whereas hypocotyl showed activity corresponding to 23.7% of original bean. Polyacrylamide gel electrophoretogram for the protein showed 10, 9, and 11 bands in the original bean. 5th day cotyledon and hypocotyl respectively. Especially, band 3 of low Rm value was major protein component for the hypocotyl. Band 5 and 11 could be seen only in the protein of hypocotyl from bluelight irradiated sprouts, whereas no effect of blue light on the electrophoretogram was observed for the cotyledon.

Taxonomic reconsideration of the Philadelphus schrenkii complex (고광나무분류군(Philadelphus schrenkii complex)의 실체에 대한 형태 고찰)

  • The Philadelphus schrenkii complex including P. schrenkii var. schrenkii, P. schrenkii var. jackii, P. schrenkii var. mandshuricus, P. tenuifolius, P. pekinensis, P. seoulensis, P. lasiogynus, and P. scaber, is distributed in China, Korea, japan, and far eastern Russia. Their taxonomic delimitations are obscure because quantitative characters, such as presence/absence and density of hair on leaf, petiole, style, disc, calyx-tube, and pedicel, are highly variable with no apparent characteristics that consistently distinguish from each other within the group. Thus, these morphological characters are reexamined using univariate analyses. P. pekinenesis is distinct from other taxa in having glabrous disk, calyx, pedicel, and leaf blade. In addition, the number of flowers in an inflorescence [(5)7-9(11) vs 5-7 in other taxa], smaller calyx tube [2.5-3 mm vs. (2.5)3-4(5.9) in other taxa], and slightly divided styles help distinguish P. pekinenesis from other related taxa in Korea. However, P. schrenkii var. schrenkii, P. schrenkii var. jackii, P. schrenkii var. mandshuricus, and P. lasiogynus show continuous variation in the pubescence of disk, calyx, pedicel, and leaf blade, and this may reflect a lack of divergence among them. Particular emphases are placed on P. tenuifolius and P. seoulensis, because of the high degree of morphological intergradation with respect to the degree of pubescence in disk, calyx, pedicel, and leaf blade and their inconsistent circumscription across various systematic treatments. P. scaber, distributed in the southern Korea, has often been treated as an independent species having exfoliated branchlets, distinct serrated leaf margin, and recurved styles. However, this current study suggests that P. scaber should be recognized as a form of P. tenuifolius because these distinguishing characters are continuous between P. tenuifolius and P. scaber. In geographical distribution, P. pekinensis is mainly distributed in the southern part of Korea, while P. tenuifoilus and P. schrenkii are commonly found all over the country.

Comparison of Bite Forces between Pre- and Post-Treatment in Patients with Temporomandibular Disorders (측두하악장애 환자의 치료 전후 교합력 비교)

  • The purpose of this study was to compare the maximum bite forces between pre- and post-treatment related to specific diagnostic groups of TMD including masticatory muscle disorder (MMD), disc derangement (DD), joint inflammation (JI) and osteoarthritis (OA). Bite force between pre- and post-treatment was compared in 36 patients with unilateral TMD, successfully-managed in the Department of Oral Medicine, Dankook University Dental Hospital, for this study. The ratio of men to women was 7:29 and their mean age of $28.1{\pm}13.7$ years. The patients were categorized, through clinical and radiographic examination, into aforementioned 4 groups; MMD (N=18), DD (N=6), JI (N=5) and OA (N=7). The maximum bite force measurements were done at the antagonizing canines and 1st molars using a bite force recorder. Paired t-test, ANOVA, Multiple Comparison t-tests were used for statistical analysis. The results of this study showed that the maximum bite force before treatment increased after TMD treatment, which was noticeable at the canines (p=0.001 and p=0.000 for the affected and unaffected sides, respectively). In comparison related to the diagnostic groups of TMD, patients with osteoarthritis of TMJ exhibited the lowest strength while those with inflammatory disorder of TMJ had the highest strength on the affected sides. Increase of bite force after treatment was also found in each group. Significant difference between pre- and post-treatment was found at canines on the affected sides in MMD (p=0.045) and DD groups (p=0.009) while on the unaffected sides in OA group (p=0.003). Conclusively, the reduced bite force due to TMD could be recovered by conservative TMD treatment and that the difference of bite forces between pre- and post-treatment was noticeable at the canines.

COMPARATIVE STUDY ON FLUORIDE RELEASE AND RE-UPTAKE CAPACITY OF SEVERAL FLUORIDE-RELEASING RESTORATIVE MATERIALS (수종의 수복재의 불소 적용법에 따른 불소 유리에 관한 비교 연구)

  • Fluoride released from dental restorative materials effectively declines the incidence and activity of dental caries and inhibits tooth demineralization. This study investigated the fluoride release and uptake characteristics of one composite resin $(Z-250^{TM})$ three glass ionomer-based restorative material ($Dyract^{(R)}$ AP, Fuji II $LC^{(R)}$, Fuji IX GP $Fast^{(R)}$) Forty discs(6mm diameter and 1mm height) were prepared for each material. Each disc was immersed in 5ml of distilled water within polyethylene vial and stored at $37^{\circ}C$. The distilled water was changed every 24 hours and the release of fluoride was measured for 31 days. At the end of this period, each specimen was subjected to one of four treatments : (A) no fluoride treatment (control), (B) application of a fluoride dentifrice (500ppm) for three minutes three times; (C) application of the 1.23% acidulated phosphate fluoride(APF) foam for one minute once, (D) the same regimen as (B), plus application of the APF foam for one minute once. Then, all samples were reassessed for an additional 7 days. For all samples, the greatest fluoride release was observed after the first day of the study but diminished with time. On the 7th day of the study, fluoride release level was stabilized. Fuji II $LC^{(R)}$ and Fuji IX GP $Fast^{(R)}$ released higher amount of fluoride than other materials ; however, no statistically significant difference was found from Fuji II $LC^{(R)}$ and Fuji IX GP $Fast^{(R)}$. The amount of fluoride of $Dyract^{(R)}$ AP, Fuji II $LC^{(R)}$ and Fuji IX GP $Fast^{(R)}$ was increased after fluoride treatment, and diminished with time.

Growth Inhibition against Contaminants in Aseptic Chocolate Milk Using Physicochemical Methods (물리.화학적 처리에 의한 멸균 초콜릿 우유 오염균의 생육억제 효과)

  • This study was conducted to investigate the cause of microbiological contaminants in aseptic chocolate milk and evaluate the effect of a physicochemical treatment on the growth inhibition of isolated bacterial strains. The bacterium isolated from aseptic chocolate milk was identified as Bacillus lentus and was named B. lentus M1. In the heat and pH treatment, the growth of B. lentus was inhibited at 110$^{\circ}C$ for >15 min and at pH's <5 and >10. An electrolyzed water treatment against B. lentus M1, revealed 5 mm growth past the inhibition zone. The effect of ozone gas on B. lentus M1 growth was evaluated using viable cell counts. When the initial number of B. lentus M1 was $10^2$ and $10^3$ CFU, the bacteria were completely suppressed by ozone gas treatment for 10 and 30 min, respectively. In a microwave treatment, B. lentus M1 was sterilized following microwave treatment for 1 min. As the result of ${\gamma}$-irradiation against B. lentus M1, numbers decreased as the ${\gamma}$-irradiation dosage increased. These results show the growth inhibition effects against contaminants in aseptic chocolate milk using physicochemical treatments.

Callus Initiation and Organ Formation from in vitro Culture of Pelargonium spp. (In Vitro Culture에 의한 Pelargonium spp.의 Callus 유도 및 기관분화에 관하여)

  • In order to investigate the micropropagation of Pelargonium, 2 cultivars of P. peltatum 'Pouletta' and P. zonale 'Pinto Red' were cultured in vitro on the MS basal medium supplemented with various concentrations of growth regulators. It attempted to study the induction of callus and the differentiation of organs from leaf disc, petiole segments, stem segments. hypocotyle segments and flower stalk segments. The results are summarized as follows; A. As for the initiation of callus, stem explant was proved to be the most suitable one among various explants of P. zonale 'Pinto Red'. The medium was supplemented with 1.0mg/1 BAP and 1.0mg/1 NAA. As NAA concentration increased, callus formation was enhanced, but higher concentration of NAA inhibited callus fromation. Leaf and hypocotyle explants showed less callus formation than stem and petiole explants. B. In P. zonale 'Pinto Red' petiole culture, the condition of cullus culture such as hormone concentration resulted in affecting shoots differentiation. The best result of shoots formation from the callus reculture were obtained from the combination of 0.5-1.0mg/1 BAP and 0.1-1.0mg/1 NAA when the callus was cultured in 1.0mg/1 BAP and 0.05mg/1 NAA. When the callus was cultured in medium without BAP, the shoot was not differentiated in subculture regardless to BAP and NAA concentration. and only callus was formed. C. Poly-phenol substance was observed in MS medium supplemented without PVP, in which callus was not formed from the leaf of P. peltatum 'Rouletta'. Polyphenol substance was not observed in MS medium supplemented with PVP, in which callus formation was increased. D. The callus formation of P. peltatum 'Rouletta' showed the stem explant being best result. The best result particularly in the stem explant among others. The optimal hormonal concentration was 0.1mg/1 NAA and 5.0mg/1 BAP. The shoot formation was observed at 0.05mg/1 NAA and 1.0mg/1 BAP, 0.1mg/1 NAA and 5.0mg/1 BAP. The shoot was malformed and the tissue recultured turned necrotic.

Antimicrobial Activity of Medicinal Herbs against Staphylococcus aureus (Staphylococcus aureus에 대한 한약재의 항균활성)

  • Antimicrobial activity of 18 different traditional medicinal herbs extracts against Staphylococcus aureus was determined by a paper disc method. The Prunella vulgaris, Caesalpinia sappan and Rhus javanica extracts in 5 mg/ml, Poncirus trifoliata, Lonicera japonica and Seutellaria baicalensis extracts in 10 mg/ml and Schizandra chinensis, Alpinia katsumadai, Siegesbeckia orientalis extracts in 30 mg/ml showed a significant antimicrobial activity against Staphylococcus aureus. Minimum inhibitory concentrations of medicinal herbs extracts were in the range of $1{\sim}34\;mg/ml$ and $1{\sim}46\;mg/ml$, in the case of MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with EtOAc layer. Optical density at 620nm after 24 hours incubation of Staphylococcus aureus in the presence of 100, 300 or 500 ppm of Caesalpinia sappan extract ranged from 0.02 to 0.03 compared to 0.4 in the absence of Caesalpinia sappan extract, indicating that growth of Staphylococcus aureus was significantly inhibited within 24 hours by the addition of at least 100 ppm of Caesalpinia sappan extract. Optical density at 620 nm after 24 hours incubation of Staphylococcus aureus in the presence of 300 ppm of Rhus javanica extract ranged from 0.02 to 0.03 compared to 0.4 in the absence of Rhus javanica extract, indicating that growth of Staphylococcus aureus was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Rhus javanica extract. Optical density at 620 nm after 24 hours incubation of Staphylococcus aureus in the presence of 300 ppm of Seutellaria baicalensis extract ranged from 0.02 to 0.07 compared to 0.4 in the absence of Seutellaria baicalensis extract, indicating that growth of Staphylococcus aureus was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Seutellaria baicalensis extract. In conclusion, these findings suggest that extracts from medicinal herbs may play important roles for antimicrobial activities against Staphylococcus aureus.

The fracture resistance of heat pressed ceramics with wire reinforcement (금속선 강화에 따른 열 가압 도재의 파절저항)

  • Statement of problem: Ceramics have been important materials for the restoration of teeth. The demands of patients for tooth-colored restorations and the availability of various dental ceramics has driven the increased use of new types of dental ceramic materials. Improved physical properties of theses materials have expanded its use even in posterior crowns and fixed partial dentures. However, ceramic still has limitation such as low loading capability. This is critical for long-span bridge, because bridge is more subject to tensile force. Purpose: The wire reinforced ceramic was designed to increase the fracture resistance of ceramic restoration. The purpose of this study was to evaluate the fracture resistance of wire reinforced ceramic. Material and methods: Heat pressed ceramic(ingot No.200 : IPS Empress 2, Ivoclar Vivadent, Liechtenstein) and Ni-Cr wire(Alfa Aesar, Johnson Matthey Company, USA) of 0.41 mm diameter were used in this study. Five groups of twelve uniform sized ceramic specimens(width 4 mm, thickness 2 mm, length 15 mm) were fabricated. Each group had different wire arrangement. Wireless ceramic was used as control group. The experimental groups were divided according to wire number and position. One, two and three strands of wires were positioned on the longitudinal axis of specimen. In another experimental group, three strands of wires positioned on the longitudinal axis and five strands of wires positioned on the transverse axis. Three-point bending test was done with universal testing machine(Z020, Zwick, Germany) to compare the flexural modulus, flexural strength, strain at fracture and fracture toughness of each group. Fractured ceramic specimens were cross-sectioned with caborundum disc and grinded with sandpaper to observe interface between ceramic and Ni-Cr wire. The interface between ceramic and Ni-Cr wire was analyzed with scanning electron microscope(JSM-6360, JEOL, Japan) under platinum coating. Results: The results obtained were as follows: 1. The average and standard deviation in flexural modulus, flexural strength and fracture toughness showed no statistical differences between control and experimental groups. However, strain was significantly increased in wire inserted ceramics(P<.001). 2. Control group showed wedge fracture aspects across specimen, while experimental groups showed cracks across specimen. 3. Scanning electron microscopic image of cross-sectioned and longitudinally-sectioned specimens showed no gap at the interface between ceramic and Ni-Cr wire. Conclusion: The results of this study showed that wire inserted ceramics have a high strain characteristic. However, wire inserted ceramics was not enough to use at posterior area of mouth in relation to flexural modulus and flexural strength. Therefore, we need further studies.

Evaluations on Deodorization Effect and Anti-oral Microbial Activity of Essential Oil from Pinus koraiensis (잣나무 정유의 소취효과 및 구강균에 대한 항균활성 평가)

  • Essential oils of various plants have been known for potential biological effects such as antibacterial, antifungal, spasmolytic, antiplasmodial activities and insect-repellent property. Recently, the essential oils have attracted considerable interest in oral disease therapy. This essential oil has been known as being effective on easing sick house syndrome, giving forest aroma therapy effect and acting as repellent against pest. The essential oil of Pinus koraiensi, a native plant from Hongcheon-gun, Gangwon-do, was obtained by hydrodistillation. In light of its medicinal importance, in this study its composition, antibacterial activity and the reducing effect of offensive odor have been analyzed. The composition of essential oil was determined by GC and GC-MS. We have identified 14 compounds, of which 1R-${\alpha}$-pinene (19.38 %), 3-carene (10.21 %), camphene (9.82 %), limonene (9.00 %), bicyclo[2,2,1] heptan-2-ol (8.76 %) and ${\beta}$-phellandrene (7.98 %) were the main components. Essential oils from P. koraiensis, Chamaecyparis obtusa, Abies holophylla and Pinus densiflora were compared in terms of alleviating effect of malodors caused from formaldehyde, ammonia, trimethylamine and methylmercaptan. P. koraiensis essential oil was found to decrease the amounts of ammonia and trimethylamine by 75.17 % and 77.36 %, respectively. Antibacterial activity against Streptococcus mutans and Streptococcus sobrinus, which were known as oral cavity inducer, was investigated using the paper disc agar diffusion method. The inhibition zone was observed against S. mutans (5.97 mm) and S. sobrinus (1.40 mm), respectively. P. koraiensis essential oil shown effective deodorization and inhibitory activity against oral cavity in this study might be potential material in oral sanitary industry.
