Coculture of HSC with bone marrow-derived mesenchymal stem cells (BM-MSCs) is one of used methods to increase cell numbers before transplant to the patients. However, because of difficulties to purify HSCs after coculture with BM-MSCs, it needs to develop a method to overcome the problem. In the present study, we have examined whether a culture insert placed over a feeder layer might support the expansion of HSCs within the insert. $CD34^+/ $ cells isolated from the umbilical cord blood by using midiMACS were divided into three groups. A group of 1 $\times$$10^5$ cells were grown on a culture insert without feeder layer (Direct). The same number of HSCs was directly cocultured with BM-MSCs (Contact). The third group was placed onto an insert below which BM-MSCs were grown (Insert). To distinguish feeder cells from HSCs, BM-MSCs was pre-labeled fluorescently with PKH26 and 1 $\times$$10^5$ cells were seeded in the culture dishes. After culture for 13 days, the expansion factor (x) of HSCs that were grown without feeder layer (Direct) was $26.6 \pm 8.4.$ In contrast, the number of HSCs directly cocultured with feeder layer was 59.6 $\pm$ 0.5 and that of HSCs cultured onto an insert was $46.9 \pm 8.4.$ The percentage of BM-MSCs cells remained being fluorescent was $97.9 \pm 0.3%$ after culture. Immune-phenotypically large proportion of cultured cells were founded to be differentiated into myeloid/monocyte progenitor cells. The ability of BM-MSCs, fetal lung, cartilage and brain tissue cells to support ex vivo expansion of HSCs was also examined using the insert. After 11 days of coculture with each of these cells, the expansion factor of HSCs was 15.0, 39.0, 32.0 and 24.0, respectively. Based upon these observations, it is concluded that the coculture method using insert is very effective to support ex vivo expansion of HSCs and to eliminate the contamination of other cells used to coculture wth HSCs.
This experiment was carried out to elucidate the effect of the midsummer drainage times and some growth regulators on lodging characters, lodging and yield in puddled-soil drill seeding in rice. Dongjinbyeo, the mid-late maturing rice variety was seeded at May 11 by seeding machine with 4cm of furrow depth. Experimental plots were divided two main treatment without midsummer drain and two time drains (30 and 50 days after seeding), Inabenfide(IBF) was applied 40days before heading(DBH) and IBP was applied at 30DBH, respectively. Culm length was shorten, the wall of N$_4$ was thicken, and the breaking weight was increased at two time drainage and growth regulators applied in order of Inabenfide, IBP, and Control. Lodging wasn't occurred at two time drainage but it was occurred at none drainage in the order of Control, IBP, and Inabenfide applied. Yield was higher at two time drainage compared with none drainage and higher in order of Inabenfide, IBP and Control in none drainage but wasn't significantly different among growth regulators applied in two time drainage. Therefore, two times midsummer soil drying is recommendable management method for puddled -soil drill seeding of rice. Recent abnormal weather, especially continued rainfall during sowing season causes difficulty in proper sowing of wheat and delayed sowing after November 15 is concerned about freezing damage during winter, resulting in reduction of wheat yield. To correspond government policy of crop sufficiency improvement and produce and supply raw wheat and barley steadily, expansion of cultivation area is necessary and spring sowing of wheat is required. To obtain basic information on the improvement of spring sown wheat and barley production, comparison and path coefficients analysis was conducted for yield and yield related components from autumn and spring sown wheat and barley. Path analyses were known as very useful in clarifying the effects of yield components on grain yield formation, which were not accurately reflected in simple correlation anaylses. Most cultivated 5 wheat and 9 barley cultivars were sown on October and February at Cheon-ju province according to standard sowing method. For the spring sowing of wheat and barley, the varieties having vernalization degree I~III are seeded in the mid of February and seeding rate is 200~250kg/ha which is increased by 25% than autumn sowing. N-fertilizer of 95 kg/ha and the same amount of P, K dressed in autumn are applied at once as basal fertilizer. The magnitude of direct effect in each yield components on yield was in sequence as follows. In autumn wheat, grain number per $spike{\geq}$ the number of spike per $m^2$>1000-grain weight and in spring wheat, grain number per $ spike{\geq}the$ number of spike per $m^2$> 1000-grain weight. In autumn naked barley, 1000-grain weight> the number of spike per $m^2$, grain number per spike and in spring barely, the number of spike per $m^2$> grain number per spike > 1000-grain weight. In autumn covered barley, grain number per spike>the number of spike per $m^2$ and in spring coverd barley, the number of spike per $m^2$> grain number per spike, 1000-grain weight. In autumn malt barley, the number of spike per $m^2$>1000-grain weight and in spring malt barley, the direct effects of three yield components were similar. According to the path analysis of yield components for spring sown wheat and barley, it was suggested that adequate number of spike per $m^2$ was most important factor for yield increase.
The golden apple snail was introduced to the Korea from Japan as a potential food for people in 1983. It is one of the pests of direct-seeded rice in korea. This study was conducted to investigate rice(Oriza sativa) consumption and the extent of damage by the golden apple snail(Pomacea canaliculata) in direct-sown paddy fields. Food consumption of the golden apple snail was highest at $30^{\circ}C$. When the emergence of the 1st, 3rd, and 5th leave of rice supplied food, the consumption of young(shell height 15mm) and adult golden apple snail(over shell height 30mm) at $30^{\circ}C$ were 19.0/79.8, 11.0/54.5, 5.5/18.5, and 0.0/2.0 individuals, respectively. Young rice seedings(before 5th leaves of rice) are very susceptible to golden apple snail damage because the young, tender leaves and stems favor the snail's feeding habits. The field experiment with four treatments (0, 2, 5 and 10 individuals respectively per 10 $m^2$) was carried out. The most serious yield loss was 16% from the plot with 10 adults released at emergency stage of rice.

The research was conducted to determine a seed germination and seedling establishment of rice plant under seed coating materials such as iron, silicate, and phyllite and under covered with silicate and iron coated & silicate covered in the puddled wet hill seeding and wet line seeding methods. The seedling establishment was high in silicate and untreated control of 100%>phyllite coating of 91.5%>silicate coating of 88%>iron coating and silicate covered of 86%>silicate covered of 75.5% in the puddled wet hill seeding method, respectively. At 35days after treatment there was high in seedling height at silicate covered of 23.8cm>control of 23.6cm>silicate coating of 21.4cm>phyllite coating of 20.2cm>iron coating and silicate covered of 16.8cm>iron coating of 15.4cm. In puddle wet line seeding method rice seedling establishment was high at control and silicate covered of 100%>iron coating and silicate covered by 97.5%>phyllite coating by 94.8%>iron coating by 86%. Seedling height was high in silicate covered of 22.1cm>control of 21.2cm>silicate coating of 20.0cm>phyllite coating of 19.0cm>iron coating of 17.7cm>iron coating and silicate covered of 17.0cm, respectively.
Wet direct-seeding (WDS) is an important method for improving the competitiveness of rice production in South Korea. We analyzed the optimum direct-seeding date to establish the rice standing rate in each area and selected suitable areas for WHS by considering the heading stage limit date for improving cultivation safety. As a result, the rice direct-seeding date to control weedy rice was around 5.15, 5.17-5.19, and after 5.20 in southern Youngnam, southern Honam, and the Middle Coast areas, respectively. However, the optimum seeding date for good standing rice was in late March in most areas. Analyzing by area, most of the southern plains and parts of the central inland plain are suitable for WHS. However, most parts of Gwangwon-do, and the northern parts of Chungbuk, Gyeongbuk, and Yeonghonam areas are not suitable for WHS, and should therefore avoid WHS.
Comprehensive studies on seedling production and transplanting cultivation of waxy corn are necessary to fulfill the comsumer's preference for the high quality whole ear waxy corn and its year-round supply for higher price, and to escape marketing in a short period at harvest season. Two waxy corn hybrids (cv. Chalok# 1 and Chalok# 4) were used to study the effects of seeding date, nursery days and plug cell size on growth and yield of waxy corn, and to clarify the reliable transplanting date at the Experimental Field of the National Institute of Crop Science in 2007. The number of days from seeding to silking was increased as nursery days were extended, but the number of days from transplanting to silking was shortened comparing to the direct seeding. Number of days from seeding to silking was shorter by 9~12 days for Chalok# 1 than for Chalok# 4, while the number of days from seeding to silking was shorter by 12~15 days in the second cropping than in the first cropping. Culm length of transplanted waxy corns was decreased as the nursery days were extended. Culm length of seeded Chaok# 1 and Chaok# 4 were decreased by 17% and 24%, respectively, in the second cropping compared to those in the first cropping. Ear length was somewhat decreased as the nursery days were extended. Chalok# 1 in the second cropping the degree of decrease was much higher in second cropping than in first cropping of Chalok# 1. Comparing to the first cropping, number of marketable ears per 10a of Chalok# 1 decreased as high as 64%, while that of seeded Chalok# 4 decreased mere 12% in the second cropping.
The bird repellent, methiocarb 50% WP, has been used to reduce the bird damage in the crop field in the European countries. The bird damage occaisonally would occur in the wet direct seeded rice and in soybean field, and resulted in decreasing the crop productivity by the reduction of seedling emergence rate. In this experiment, rice seeds, Hwasungbyeo(Oryza sativa), were coated with the different application rate of methiocarb, 5, 10 and 15 per kg seed, and soybean, Taegwangkong(Glycine max), 2.5, 5.0 and 7.5g. In rice, the seeds coated with 10 and 15g of methiocarb were not lost by bird, while those with 5g and control were lost to 37 and 50% of total seeds, respectively. No damage by birds was observed in rice seedling when the coleoptile and radicle of rice were emerged at 7 days after the water seeding. The crop injury of methiocarb reducing the emergence rate of seedlings, shortening the shoot length, and decreasing the leaf number was occurred at the treated of methiocarb 15g per kg seed. In soybean, the loss of the sprouting by birds was lower in the treatment of methiocarb 7.5g per kg seed than that in both the control and the treatments of methiocarb, 2.5 and 5.0g per kg seed. No crop injury by the treatment of methiocarb was observed in all application rate.
Seven formulations of oxyfluorfen selected from the previous studies(4. 5) were tesed to evaluate crop injury and herbicidal efficacy on two rice cultivars and several annual and perennial weeds in a greenhouse. Each formulation at two different rates was applied to rice transplanted with 8-, 22- and 32-day old seedlings and to direct-seeded rice. Among the formulations, Elvan, Bentonite B. Chitosan and Coal Slag gave lower injury than a control formulation, Sand-coated oxyluorfen, and they did not have a problem with excessive release if active ingredient at once. Especially, the formulations of Elvan, Chitosan and Bentonite B controlled annual weeds (Echinochloa crus-galli, Monochoria vaginalis, Cyperus difformis., and Scirpus juncoides) and perennial weeds (Sagittaria pygmaea, and Cyperus serotinus). The surface structure of the formulations indicate the different possibilities of releasing of oxyfluorfen by different cracking and hole sizes, namely retention capacity.
For developing the site-specific fertilizer management strategies of crop, it is essential to know the spatial variability of soil factors and to assess their influence on the variability of crop growth and yield. In 2002 and 2003 cropping seasons within-field spatial variability of rice growth and yield was examined in relation to spatial variation of soil properties in the· two paddy fields having each area of ca. $6,600m^2$ in Suwon, Korea. The fields were managed without fertilizer or with uniform application of N, P, and K fertilizer under direct-seeded and transplanted rice. Stable soil properties such as content of clay (Clay), total nitrogen (TN), organic mater (OM), silica (Si), cation exchange capacity (CEC), and rice growth and yield were measured in each grid of $10\times10m$. The two fields showed quite similar spatial variation in soil properties, showing the smallest coefficient of variation (CV) in Clay $(7.6\%)$ and the largest in Si $(21.4\%)$. The CV of plant growth parameters measured at panicle initiation (PIS) and heading stage (HD) ranged from 6 to $38\%$, and that of rice yield ranged from 11 to $21\%$. CEC, OM, TN, and available Si showed significant correlations with rice growth and yield. Multiple linear regression model with stepwise procedure selected independent variables of N fertilizer level, climate condition and soil properties, explaining as much as $76\%$ of yield variability, of which $21.6\%$ is ascribed to soil properties. Among the soil properties, the most important soil factors causing yield spatial variability was OM, followed by Si, TN, and CEC. Boundary line response of rice yield to soil properties was represented well by Mitcherich equation (negative exponential equation) that was used to quantify the influence of soil properties on rice yield, and then the Law of the Minimum was used to identify the soil limiting factor for each grid. This boundary line approach using five stable soil properties as limiting factor explained an average of about $50\%$ of the spatial yield variability. Although the determination coefficient was not very high, an advantage of the method was that it identified clearly which soil parameter was yield limiting factor and where it was distributed in the field.
