Fluorine Labeling in Biosynthetic Studies (I) : Synthesis of Fluorfarnesols

  • The Synthesis of E, E, E-12-fluorofarnesol and E, Z-6-fluorofarnesol which are key intermediates for the study of biosynthesis of some sesquiterpenes, is described. E, E-Farnesyl acetate is treated with selenium dioxide to give E, E, E-12-hydroxy farnesyl acetate, whih is transformed by DAST into E, E, E-12-hydroxy farnesyl acetate, which is transformed by DAST into E, E, E,-12-fluorofarnesylacetate. The latter compound is hydrolyzed to E, E, E,-12-fluorofarnesol. The reformatsky reaction of 6-methyl-5-hepten-2-one with ethyl bromofluoroacetate affords ethyl 2-fluoro-3-hydroxy-3, 7 dimethyl-6-octanoate. This ester is acetylated and eliminated to give ethyl (Z)-2-fluoro-3, 7-dimethylocta-2, 6-dienoate, which is transformed to allyl bromide via allylic alcohol. The allyl bromide is treated with dianion of methyl acetate to give-keto ester. The $\beta$-keto ester is converted to diethyl phosphoryloxy compound. The conjugate addition of lithium dimethylcuprate to the latter compound gives fluoro ester, which is treated with DIBAL to afford E, Z-6-fluorofarnesol.

발효콩 추출물의 항돌연변이원성 및 세포독성 효과 (Antimutagenic and Cytotoxicity Effects of Fermented Soybean Extract)

  • This study was carried out to determine the antimutagenic and anticancer effects of fermented soybean using Ames test and cytotoxicity, respectively. The ethyl acetate fraction (200 g/plate) of fermented soybean in the Salmonella typhimurium TA100 strain showed 86.6% of inhibition rate against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). In addition, the suppression of ethyl acetate fraction with same concentration of fermented soybean in the Salmonella typhimurium TA98 and TAI00 strains showed 82.4% and 90.8% inhibition against 3-amino-l,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-l), respectively. The cytotoxicity effects of fermented soybean against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL ethyl acetate fraction of fermented soybean showed strong cytotoxicities of 71.6%, 91.5% and 80.7% against A549, AGS and MCF-7, respectively.

신규 Squarylium Dyes 증감제에 의한 Poly(vinyl cinnamoyl acetate)의 고감도화(I) (Photosensitive Effect of Poly(vinyl cinnamoyl acetate) by New Squarylium Dyes(I))

  • Some photopolymer, poly(vinyl cinnamoyl acetate)(PVCiA) was synthesized by esterification of polymer(vinyl alcohol(PVA) with monochloroacetic acid, followed by reaction poly(vinyl monochloroacetate)(PVChA) and potassium cinnamate. When esterification of PVA with monochloroacetic acid was reacted in the dimethyl sulfoxide(DMSO), in the synthesis of PVChA, it is very good yield and the successive cinnamoyl acetoxyl esterification of PVCiA can be successfully synthesized. But PVCiA is low photosensitive polymer if not added photosensitizing dyes. Here, we synthesized photosensitizing dyes. trifluoromethylindol squarylium dye derivatives(TFSQ). Indolization of 3-trifluoromethylphenyl-hydrazine formed 4- and 6-substituted indoles, the rate of 4- and 6-substituted indoles is close to unity. Absorption's coefficient of 1,3-bis(4-trifluoromethyl-1,3,3-trimethylindol) squarylium dye(TFSQ) is 7.57$\times$10\ulcorner, PVCiA added with TFSQ(3%) was highly sensitized six times than not added.

전통(傳統) 약주(藥酒)의 향기성분(香氣成分) 비교(比較) (Comparison of the Aroma Components in the Korean Traditional Yakjus.)

  • 전통약주(傳統藥酒)인 백하주(白霞酒), 녹파주(綠波酒), 동동주(酒), 청명주(淸明酒)를 양조(讓造)하여 휘발성(揮發性) 향기성분(香氣成分)을 head space gas로 포집(捕集)하여 분석(分析)하였다. 향기성분(香氣成分)으로 formaldehyde, acetone, diacetyl, acetaldehyde, ethyl acetate, ethanol, propanol, iso-butanol, iso-amyl alcohol, dimethylsulfide가 분리(分離) 정량(定量)되었으며 acetaldehyde, ethyl acetate, n-propanol은 각주품(各酒品)에 따라 $17.84{\sim}73.74ppm$으로 acetaldehyde는 동동주(酒)가 62.04ppm, ethylacetate는 녹파주(綠波酒)가 41.26ppm, n-propanol은 녹파주(綠波酒)가 73.74ppm으로 가장 많았으며 iso-butanol과 iso-amyl alcohol은 청명주(淸明酒)가 각각(各各) 194.21ppm, 925.04ppm으로 가장 높았으며 백하주(白霞酒)는 72.12ppm 및 418.14ppm으로 가장 낮았다. Formaldehyde와 acetone은 $0{\sim}2.15ppm$으로 백하주(白霞酒)에서는 acetone이 정량(定量)되지 않았다. Dimethyl sulfide의 함량(含量)은 주품(酒品)에 따라 $5{\sim}65ppb$로써 동동주가 65ppb로 가장 많았으며 diacetyl은 $0.032{\sim}1.012ppm$으로 주품(酒品)에 따라 차(差)가 컸다.

  • PBT (polybutylene terephthalate)는 저흡수율, 치수 안정성, 내마모성 등 기계적 특성이 우수하며, 전기전자 부품, 자동차 부품, 각종 정밀 부품에 사용된다. DMT (dimethyl terephthalate)와 BD (1,4-butandiol)를 사용하여 PBT의 원료단량체인 BHBT (bis-hydroxybutyle terephthalate)를 생산하는 에스테르 교환 반응 반응에 대해 연구 하였다. 촉매로는 zinc acetate가 사용되었다. 기존의 연구에서는 반응 중 생성 메탄올이 제거되는 반회분식 반응기를 통한 kinetics 연구가 이루어져 역반응이 고려되지 않음에 따른 모델의 부정확함이 있었다. 본 연구에서는 회분식 반응기를 사용하고 반응 중 DMT와 메탄올 양을 정량하여 생성되는 MHBT (methyl hydroxylbutylene terephthalate)와 BHBT를 추정할 수 있도록 하고, 이 반응들에서 역반응들을 고려할 수 있도록 하여 보다 정확한 모델을 제안하였다. 다양한 반응속도 모델을 제시하였고, 이 모델들이 예측한 값들이 실험 데이터와 잘 일치함을 보였다.

  • DMT(dimethylterephthalate)와 DEG(diethylene glycol)의 에스테르 교환반응을 통하여 BHEET(bis-hydroxyethoxyethyl-terephthalate)을 생성하는 반응에 대하여 조사하였다. BHEET는 polyurethanr foam의 원료인 폴리에스터 폴리올의 단량체이다. DMT를 이용한 기존의 에스테르 교환 반응은 반응 중 생성 MeOH이 제거되는 반회분식 반응기를 사용하였다. 따라서 이러한 kinetics 연구에서는 역반응이 고려되지 않는다. 본 실험에서는 촉매로는 zinc acetate를 사용하였고, 소형 회분식 반응기를 통하여 DMT와 MeOH의 양을 정량하여 생성되는 MHEET와 BHEET를 추정하였다. 이 반응들에서 역반응을 고려한 보다 정확한 반응 kinetics를 조사하였다. 제안된 모델의 예측 값들이 실험값들과 잘 일치함을 보였다.

폴리아크릴로니트릴/셀룰로오스 아세테이트/N,N-디메틸포름아미드 용액의 유연학적 특성 (The Rheological Properties of Poly(acrylonitrile)/Cellulose Acetate Blend Solutions in N,N-Dimethyl Formamide)

  • 폴리아크릴로니트릴(PAN)과 셀룰로오스 아세테이트(CA) 블렌드/디메틸포름아미드(DMF) 용액의 유변학적 특성을 온도와 블렌드 조성비에 대하여 조사하였다. 모든 고분자 용액이 온도 변화에 따라 매우 특징적인 유변학적 거동을 보였다. 8 wt% 용액의 경우 $20{\sim}60^{\circ}C$ 온도범위에서 온도의 증가와 더불어 용액의 점도가 증가하고 손실탄젠트값이 감소하였다. $20^{\circ}C$에서는 용액의 물성이 블렌드의 조성에 영향을 받았으나 40 및 $60^{\circ}C$에서는 조성비가 용액 물성에 미치는 영향이 크게 감소하였다. 더 높은 온도에서의 더 긴 분자 이완시간으로부터 온도가 증가함에 따라 분자간력에 의한 물리적 구조 형성이 촉진된다는 것을 알 수 있었다. 이러한 유변학적 특성에 대한 원인은 묽은 용액의 특성으로부터 유추할 수 있었다. 고분자 용액의 고유점도가 온도가 증가함에 따라 감소하였다.

면섬유에 번식하는 Aspergillus niger H-18과 Aspergillus fumigatus E-29에 대한 방미제의 항균효과 (Studies on the Maildew-Proofing of Cotton Fabrics by Growth of Aspergillus niger H-18 and Aspergillus fumigatus E-29)

  • A. niger H-18 and A. fumigatus E-29 wrer selected for their strong abilities to produce cellulase. The λd numerical values of the cotton fabrics inoculated with A. niger H-18 and A. fumigatus E-29 were 580 nm for the both strains of molds. By the growth of molds, lightness, original color scale, and grey scale of the fabrics gradually decreased while chroma increased. The minimum inhibitory concentrations of mold-proofing agents, such as Leperon WL, 8-hydroxyquinoline copper acetate, trimethylol melamine and dimethyl ethylene urea was 50 ppm. Glycoxale was not effective at the above mentioned concentration. Since Leperon WL, trimethylol melamine and dimethyl ethylene urea effectively inhibited the growth of A. niger H-18 and A. fumigatus E-29, tensile strength and elongation of the fabrics were not changed. however, cotton fabrics treated with glycoxale of the fabrics were not changed. However, cotton fabrics treated with glycoxale and inoculated with A. niger H-18 and A. fumigatus E-29 showed decreased in tensile strength by 31.1% and 33.9%, respectively.

Maximization of Extracted Condition of Pro-angiogenic Components in Citrus unshiu Peels using Dimethyl Sulfoxide

  • Aqueous extraction of Citrus unshiu peels (AECUP) is mainly comprised with pro-angiogenichesperidin and narirutin. In this study, we report approaches to increasing the yields of extracted hesperidin and narirutinfrom Citrus unshiu peels using proper solvents. Significantly improved yields of both compounds were obtained using methanol and dimethyl sulfoxide (DMSO) compared to acetonitrile, ethyl acetate, ethanol, and isopropyl alcohol. Especially, effect of DMSO was by far the better of the two solvents in extraction of hesperidin. In addition, the DMSO extracted hesperidin significantly induced the pro-angiogenic effects of human umbilical vein endothelial cells (HUVECs) and markedly up-regulated phosphorylation of the ERK1/2 signaling pathway. These results demonstrate that pro-angiogenic inducer; hesperidin and narirutin can be simply, easily, and effectively extracted from Citrus unshiu peels.

Identification of Impurities in a Sample of Illicitly Synthesized Methamphetamine

  • Analysis of illicit methamphetamine samples seized in Korea is discussed. The samples are extracted with the small portion of ethyl acetate under neutral conditions and the extracts are analyzed by GC-MS. Several impuritiy peaks are found in each chromatrogram. Eight compunds (1,2-Dimethyl-3-phenylaziridine, amphetamine oxime, ephedrine, N-formylmethamphetamine, N-acetylmethamphetamine, acetylephedrine, 3,4-dimethyl-5-phenyl-2-oxazolidone, methamphetamine dimer) are identified impurities in illicit methamphetamine and the identity of the impurity is conformed synthesis.
