Extraction of Dimethyl-$\beta$-propiothetin from Enteromorpha intestinalis (창자파래(Enteromorpha intestinalis)로부터 Dimethyl-$\beta$-propiothetin 추출)

  • The DMPT produced by marine algae is the main biogenic precursor of oceanic DMS. Also, DMPT is an efficient stimulant for growth, feeding, and body movement of fish and striped prawn, and appears to play a physiologic role as an osmoprotectant in algae. This study was focused on the extraction of dimethyl-$\beta$-propiothetin as bioactive substance from green seaweed. Identification and quantification of dimethyl-$\beta$-propiothetin were measured by headspace gas chromatography after conversion to dimethyl sulfide by treatment with saturated NaOH solution. Dimethyl-$\beta$-propiothetin was extracted through various processes(solvent extraction, ultrasonication, boiling and autoclaving) from Enteromorpha intesinalis. The content of dimethyl-$\beta$-propiothetin extracted by autoclaving treatment showed higher than those of various extraction methods. Dimethyl-$\beta$-propiothetin content in extract of Enteromorpha Enteromorpha was 311,200ng/g after autoclaving at 121$^{\circ}C$ for 60min. Dimethyl-$\beta$-propiothetin in extract of Enteromorpha intestinalis was comparatively stable under low temperature. The retentions of dimethyl-$\beta$-propiothetin content in extract of Enteromorpha intestinalis were 75.8 ~99.8% by incubation at 10~6$0^{\circ}C$ for 2 hours. Chemical decomposition of dimethyl-$\beta$-propiothetin was observed under laboratory conditions at pH values higher than 9.5.

  • To investigate the effects of growth and body composition of spotted sea bass (Lateolabrix sp.) by the supplemented dimethyl-$\beta$-propiothetin(DMPT). fish was fed the diets each containing 3 mM, 5 mM, 7 mM and 10 mM for eight weeks. The weight gain, feed efficiency and feed intake were significantly increased with dietary DMPT level. Condition factor was reduced by dietary DMPT supplementation. Crude protein content of fish after eight weeks increased with dietary DMPT level, whereas crude lipid content deceased with dietary DMPT level. Amino acid content was higher in DMPT-added group than control group. Amino acid composition was not influenced by dietary DMPT supplementation. Fatty acid content deceased with dietary DMPT level.

  • This study was focused on the optimal extracting conditions of dimethyl-$\beta$-propiothetin as bioactive substance from green seaweed. Dimethyl-$\beta$-propiothetin in exract of Ulva pertusa was comparative stable under low temperature. The retentions of dimethyl-$\beta$-propiothetin content in extract of Ulva pertusa were 76.6~99.8% by incubation at 10~6$0^{\circ}C$ for 2 hours. Chemical decomposition of dimethyl-$\beta$-propiothetin was observed under laboratory conditions at pH values higher than 9.5.

  • This study was conducted to investigate the effects of different dietary dimethy-β -propiothetin (DMPT) levels on growth and body composition in Korean rockfish (Sebastes schlegeli) after a eight weeks feeding experiment. Body weight gain, feed efficiency and feed intake were significantly increased with dietary DMPT level. After eight weeks of feeding trial, moisture content decreased with supplemented DMPT level, while crude protein and lipid contents increased with dietary DMPT level. Amino acid content was elevated DMPT feeding groups compared to the control group. Amino acid composition was not significantly different in the dietary groups. Fatty acid content increased with dietary DMPT level.

The content of DMPT extracted from Ulva pertusa by auroclaving was 32.5mg%. Body weight gain, feed efficiency and feed intake were significantly increased with dietary DMPT level. After eight weeks of feeding trial, moisture content decreased with supplemented DMPT level, while crude protein and lipid contents increased with dietary DMPT level. Amino acid contents of muscle was elevated in the DMPT added groups compared to control group. Amino acid composition of muscle in fish fed the diets supplemented DMPT was not different to that of control group. Fatty acid contents of muscle in fish increased with dietary DMPT level.
