  • Objectives: This study was designed to establish an accurate analytical method for asbestos in a biological sample for determining occupational asbestos-related diseases and relief of the health effects of environmental asbestos. Methods: Biological samples were obtained from lungs of rats following intratracheal instillation of asbestos(Chrysotile, anthophyllite) and were prepared according to digestion method(Wet digestion, high temperature ashing, low temperature ashing). The samples were then analyzed for asbestos fibers using a transmission electron microscope equipped with an energy dispersive X-ray spectrometer. Results: Low temperature plasma ashing removed more of the organic components and reduced fiber loss compared to the wet digestion method, making specimens so prepared more suitable for transmission electron microsocpy. Conclusions: The low temperature ashing technique is the most accurate method for analyzing asbestos in biological samples.

  • Ruminal digestion of dry matter (DM) and neutral detergent fiber (NDF) of processed (ensiled, deepstacked or composted) broiler litter (BL) was determined in situ and in vitro, and compared with rice straw (RS). DM disappearances at 24 and 48 h and digestion of differently processed BL were higher than those of RS. Compared with RS, processed BL was low in NDF disappearance at 72 h incubation, digestion rate ($K_dB$) and digestibility at 0.025 of passage rate; however, deepstacked BL was similar in these NDF characteristics. Processing of BL affected ruminal digestion of nutrients such as DM and NDF adversely. NDF of composted BL, especially, was the most indigestible. This in situ nutritional evaluation indicated that deepstacked BL, the most widely used form of BL, was superior in DM characteristics (fractions, ruminal disappearance and digestibility) and similar in NDF characteristics (ruminal disappearance and digestibility) to RS.

  • The purpose of this study was to investigate the effects of mechanical(ball-mill) solubilization of excess sludge especially focused on the TSS(total suspended solid) reduction during the conventional aerobic digestion of sewage sludges including primary and/or excess sludge, HRT was examined at the 10 days and 20 days. According to the results of this study, TSS removal efficiency of solubilized excess sludge was almost two times higher than that of non-solubilized excess sludge. And as the proportion of the primary sludge increased, TSS removal efficiency became worse because primary sludge rarely contained microbial cells which could be easily solubilized physically. It was also proved that by the application of proper solubilization techniques to the excess sludge, HRT for the aerobic digestion could be lessened(above 50%) dramatically keeping the same or better digestion performance. The fact that between primary and excess sludges, only the excess sludge is quite effective in the sludge solubilization and in it's reduction says that excess sludge releasing sources are key-point in the sludge cake reduction field as a source control.

  • Proteomics for biomarker validation needs high throughput instrumentation to analyze huge set of clinical samples for quantitative and reproducible analysis at a minimum time without manual experimental errors. Sample preparation, a vital step in proteomics plays a major role in identification and quantification of proteins from biological samples. Tryptic digestion a major check point in sample preparation for mass spectrometry based proteomics needs to be more accurate with rapid processing time. The present study focuses on establishing a high throughput automated online system for proteolytic digestion and desalting of proteins from biological samples quantitatively and qualitatively in a reproducible manner. The present study compares online protein digestion and desalting of BSA with conventional off-line (in-solution) method and validated for real time sample for reproducibility. Proteins were identified using SEQUEST data base search engine and the data were quantified using IDEALQ software. The present study shows that the online system capable of handling high throughput samples in 96 well formats carries out protein digestion and peptide desalting efficiently in a reproducible and quantitative manner. Label free quantification showed clear increase of peptide quantities with increase in concentration with much linearity compared to off line method. Hence we would like to suggest that inclusion of this online system in proteomic pipeline will be effective in quantification of proteins in comparative proteomics were the quantification is really very crucial.

  • Synthetic organic polyelectrolytes can be used to condition sludges to enhance their dewaterability. When conditioning biological sludges, the charge on the polymer has a significant impact on the effectiveness of the polymer as a conditioner. The objectives of this investigation were to determine the most effective type and dosage of polymer for conditioning digested sludge prior to dewatering, and to investigate the relationship between dewaterability and particle size. 3 types of digested sludge were used under the different digestion processes like anaerobic digestion, aerobic digestion and intermittent aerobic digestion. CST(capillary suction time), TTF(time-to-filter) and SRF(specific resistance to filtration) were tested as a dewaterability index and the number of particle distribution was analyzed using particle size analyzer. The results indicate that cationic polymer appears to be required for effective conditioning of these 3 types digested sludge and the optimal polymer dosage is about 0.6% of SS. CST and TTF are well correlated with mean particle diameter of which the dimension order is 1.7.

  • Waste activated sludge (WAS) was thermally pretreated to enhance hydrolysis and ultimately methane yield. Batch and semi-continuous anaerobic digestion were conducted to evaluate the performance of methane fermentation of the hydrolyzed sludge and to investigate the kinetics of sludge fermentation. Thermal pretreatment remarkably enhanced digestion performances particularly the methane fermentation with three times more methane production than before the pretreatment. Gas production and kinetic parameters in the semi-continuous anaerobic digestion were estimated using Chen Hashimoto model. The model simulation fitted well the experimental results and the model was shown to be suitable for evaluating the effects of disintegration of WAS in anaerobic digestion. Three parameters ($B_o$, K, and ${\mu}_m$) determined by model simulation were $0.0807L-CH_4/g-VS$, 0.453 and $0.154d^{-1}$ for control sludge, and $0.253L-CH_4/g-VS$, 0.835 and $0.218d^{-1}$ for thermally pretreated sludge, respectively.

  • To study on the assignment of the eight trigrams (八卦) for the process on intake, digestion, excretion of food. Being based BEOK-HEE's diagram of eight trigrams order(伏羲次序圖), provisional namely six entera are assigned eight trigrams(八卦). Eight trigrams(八卦) are attached to the air(GEON 乾), the month(TAE 兌), the stomach(LEE 離), the small intestine(JIN 震),the large intestine(SON 巽), the rectum(GAM 坎), the anus(GAN 艮), the excrements(GON 坤). The BEOK-HEE's diagram of eight trigrams order(伏羲次序圖) means immutable order naturally. The process of six entera's digestion is also unchangeable in region and program. Therefore we can set up the process of six entera's digestion as the category of observation on the basis of BEOK-HEE's diagram of eight trigrams order(伏羲次序圖), and then we can arrange eight trigrams(八卦) on the process of six entera's digestion.

  • The impacts of ultrasonic pretreatment on the biodegradability of domestic sewage sludge were evaluated through a series of anaerobic digestion experiments in batch system. The gas and methane production from the sludge samples pretreated by an ultrasonic tool with different durations were measured with time. Although the biogas production increased with the extent of sludge solubilization and the period of ultrasonic pretreatment, the enhancement of sludge biodegradability was much more sensitive to the pretreatment for the relatively short periods. Most of the enhanced biodegradability by the pretreatment was appeared in the early stage of anaerobic digestion, less than 6 days. The maximum biogas production per day was observed between 4 to 6 days when the sludge was pretreated less than 10 minutes while it was obtained in the beginning for the sludge pretreated longer periods. The results suggest that the repeated alternation of low strength ultrasonic pretreatment and anaerobic digestion may be more effective than the combination of one time pretreatment for a relatively long period and following anaerobic digestion.

  • This study was aimed at investigating a filtration bed system for the production of liquid fertilizer using wastewater generated after anaerobic digestion of animal manure. Slurry, Compost, & Biofiltration-Methane System, designed by Korean National Institute of Animal Science, is now a standard method to recycle anaerobic digestion wastewater. However, currently provided protocol to run the system needs continuous upgrades as the system is relatively new and can be more cost-effective if few adjustments are made. In this study, we tried to enhance the capability of the system to process anaerobic digestion wastewater by replacing the bed materials with the cheaper one, and determining the time point of operation from which filtrates can be utilized for fertilization.
