The Relationship Between Green Stem Disorder and the Accumulation of Vegetative Storage Protein in Soybean

  • Green stem disorder (GSD) of soybean (Glycine max (L.) Merr.) is characterized by delayed senescence of stems with normal pod ripening and seed maturation (Hobbs, 2006). GSD complicates harvesting of soybeans by significantly increasing the difficulty in cutting the affected plants. There is also the potential for moisture in the stems to be scattered on the seed, reducing the grade and storability of the seed. Not only the cause of GSD is yet unknown, but also GSD cannot be evaluated until maturity, therefore the method to evaluate GSD in early growth stage with high sensitivity is necessary. In previous studies, it has been reported that vegetative storage protein (VSP) accumulates and the syndrome of GSD appears in soybean after depod treatment (Fischer, 1999). Soybean VSP is a storage protein which is abundant in young sink leaves and degraded during seed fill (Wittenbach, 1982). Hence, we have established a system to quantify VSP of high sensitivity by using standard protein made by genetically transformed E. coli and specific antibody against VSP, and studied the relationship between VSP and GSD, by depod experiment and drought/excess wet experiments. The result of depod experiment with the cultivar 'Yukihomare' was the same with the previous studies, VSP accumulated much more than control and the syndrome of GSD appeared in soybean in depod treatment. Drought and excess wet had different impact on GSD. Excess wet caused GSD of the cultivar 'Tachinagaha (GSD susceptible)', while drought caused a little syndrome of GSD in the cultivar 'Touhoku 129 (GSD resistant)'. The accumulation of VSP differed between the two cultivars over time. In conclusion, the accumulation of VSP came along with the emergence of GSD. Different cultivars showed different response to drought and excess wet. In the future, it is expected that the dynamics of VSP will be elucidated in detail, leading to the development of early diagnosis technology for green stem disorder and the elucidation of mechanism of soybean GSD.

Studies on Analysis of Growth Characteristics, Ability of Dry Matter Production, and Yield of Panax ginseng C. A. Meyer at Different Growth Stages with Different Cultivars and Shading Nets in Paddy Field (논토양에서 해가림 유형 및 품종별 인삼의 생육시기에 따른 생육특성, 건물생산 능력 및 수량성 비교)

  • This study was conducted to examine major growth responses, the production and partitioning of dry matter on different growth stages, and yields and to select the optimal shading material in both quality and productivity of ginseng. Two cultivars of ginseng, Cheonpoong and Geumpoong, were cultivated in the paddy soil with three different shading materials; three-layered blue and one-layered black polyethylene (TBP), blue polyethylene sheet (BPS), and aluminium-coated polyethylene sheet (APS). Plant heights were linearly increased until June 24 and then maintained with showing higher height in Cheonpoong than that in Geumpoong cultivar. Root lengths were gradually increased until October 16. They were longer in Cheonpoong than that in Geumpoong cultivar, showing slightly longer with APS compared to TBP and BPS. The ability of producing dry matter of leaves was much higher from April to June compared to those of other growth periods, whereas its ability of root was concentrated from the end of June to the end of August. Among the shading materials, the ability of producing dry matter of shoot was higher with TBP than those with BPS and APS, while its ability of root was not appeared certain tendency unlike the shoot. The yield of ginseng roots was the highest with TBP among three shading materials and it was higher in Cheonpoong than that of Geumpoong cultivar. The shading materials which affect the light intensity and the temperature would be considered as an important factor to get better quality and productivity of Korean ginseng.

'Joseong', a New Early-Heading Forage Triticale Cultivar for Paddy Field of Double Cropping (이모작 재배에 적합한 조숙성 조사료용 트리티케일 신품종 '조성')

  • 'Joseong', a winter forage triticale cultivar (X Triticosecale Wittmack), was developed by the breeding team at the Department of Rice and Winter Cereal Crop, NICS, RDA in 2010. The cultivar 'Joseong' was selected from the cross FAHAD_5/RHINO1R.1D 5+10 5D'5B'//FAHAD_5 by CIMMYT (Mexico) in 1992. Subsequent generations were handled in pedigree selection program at Mexico from 1993 to 1998, and a line 'CTSS92Y-A-4Y-0M-5Y-0B' was selected for earliness and good agronomic characteristics. After preliminary and advance yield test in Korea for 2 years, the line designated as a line name of 'Iksan26'. The 'Iksan26' was subsequently evaluated for earliness and forage yield in 7 different locations such as Jeju, Iksan, Cheongwon, Yesan, Naju, Daegu, and Jinju from 2008 to 2010 and finally named as 'Joseong'. The cultivar 'Joseong' has characteristics of dark green leaf, yellow culm and spike, and large grain of yellowish brown color. The heading date of cultivar 'Joseong' was April 24, which was 5 days earlier than that of check cultivar 'Shinyoung'. It showed better tolerance or resistance to lodging, wet injury, powdery mildew, and leaf rust than those of the check cultivar 'Shinyoung'. The average forage dry matter yield of cultivar 'Joseong' at milk-ripe stage was 14.5 MT $ha^{-1}$, which was lower than 16.5 MT $ha^{-1}$ of the check cultivar 'Shinyoung'. The silage quality of 'Joseong' (5.3%) was lower to the check cultivar 'Shinyoung' (6.0%) in crude protein content, while was 2.1% superior to the check cultivar 'Shinyoung' (61.9%) in TDN (total digestible nutrients). It showed 5.1 MT $ha^{-1}$ of grain yield, which was 40% higher than that of the check cultivar 'Shinyoung' (3.65 MT $ha^{-1}$). This cultivar is recommended for fall sowing crop in the area where daily minimum mean temperatures are averaged higher than $-10^{\circ}C$ in January, and as a winter crop using whole crop forage before planting rice in around Korea.

Changes in pasting properties and free fatty acids of different brown rice cultivar during storage

  • Paddy rice is typically stored during postharvest until rice grain is processed into brown rice and milled rice by hulling and milling procedure, respectively. Recently, instead of storing paddy rice, storage of brown rice has been in the spotlight because it is more convenient and economically feasible. Different brown rice cultivars with varying amylose contents including waxy rice, medium-waxy rice, and non-glutinous rice were stored in room temperature storage for four months, and the changes in grain qualities of brown rice were evaluated. Amylose content significantly affected pasting properties in which rice cultivar with higher amylose content showed longer pasting time and higher peak viscosity. Storage also affected pasting viscosities, showing an increase in peak viscosity, but a decrease in breakdown viscosity. The changes in pasting viscosity during storage could be an important starch property for aged brown rice utilization. Waxy brown rice showed the weakest aging property in terms of free fatty acids (FFA) accumulation, whereas non-glutinous rice was more substantial grain quality against aging. The FFA values of two months storage were not significantly different from the initial FFA contents, suggesting that brown rice stored in room temperature for two months could be feasible for direct consumption of brown rice.

Comparison of NERICA and Asian rice among traits relevant to drought resistance in the field and the effects of compost

  • Recently NERICA (New Rice for Africa) was developed by a crossing of African rice (Oryza glaberrima Steud.) and Asian rice (Oryza sativa L.) in West Africa, and is considered to be drought resistant, but drought resistance of NERICA and differences between Asian rice are not clarified enough. In this research, NERICA (four cultivars and two lines), Asian rice (three cultivars and sativa parent of NERICA) and African rice (glaberrima parent of NERICA) were cultivated in the field in Shizuoka University under drought and traits of each cultivar and line relevant to drought resistance, stomatal conductance by porometer, soil water content of individual depths by TDR method, SPAD values by SPAD meter and leaf thickness by micrometer, were measured and compared with dry matter production and yield. Effects of compost were also compared among sativa parent, one NERICA cultivar and two NERICA lines. Glaberrima parent showed highest top dry weight. One NERICA line, one drought resistant Asian rice cultivar and sativa parent, showed higher top dry weight and yield (ear weight) than other Asian rice cultivars and NERICA cultivars and line tested. Compost tended to increase top dry weight and yield in one of NERICA line and sativa parent. But in one NERICA cultivar and line, top dry weight and yield were not increased. In one of Asian rice, one of NERICA line and sativa parent that showed high top dry weight and yield, stomatal conductance was high. On the contrary the glaberrima parent and in other NERICA cultivars and line it was low. In sativa parent compost increased stomatal conductance but in NERICA cultivar and lines it was not. Among cultivars and lines that showed high top dry weight and yield sativa parent and one of NERICA line SPAD value and leaf thickness were high but in one of Asian rice and glaberrima parent they were low. Cultivar and line differences in yield and top dry weight among Asian rice and NERICA were significantly correlated with those in stomatal conductance ($r=0.778^{**}$ and $r=0.654^*$, respectively) and those in leaf thickness ($r=0.600^*$ and $r=0.640^*$, respectively). In Asian rice cultivars average soil water content was significantly correlated with yield ($r=0.886^*$) but in NERICA cultivars and lines it was not significant correlated (r= -0.256). Cultivar and line differences in leaf thickness were significantly correlated with SPAD value ($r=0.773^{**}$). In Asian rice cultivars it was significantly correlated ($r=0.962^{**}$), but in NERICA cultivars and lines it was not significantly correlated (r=0.559). Asian rice cultivars tended to consume soil water to increase yield but in NERICA cultivars and lines the tendency was not clear. Correlation between SPAD value and leaf thickness was different between Asian rice and NERICA cultivars and lines, and in Asian rice cultivars it was significantly correlated but in NERICA cultivars and lines it was not significant. Importance of maintaining high stomatal conductance by high leaf thickness was clarified.

Characteristics and breeding of a new cultivar Pleurotus eryngii var. ferulae, 'Beesan No.2' (아위느타리 신품종 '비산2호'의 육성 및 자실체 특성)

  • To develop a new cultivar of King oyster mushroom white variety (Pleurotus ferulae), GW10-68 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2798-24 derived from ASI 2798 and dikaryotic strain ASI 2803. The GW10-95(ASI 2803 ${\times}$ ASI 2798-24) was shown the best cultural characteristics, selected to be a new cultivar and designed as 'Beesan No.2'. The 'Beesan No.2' was formed incompatibility line distinctly in the confrontation growth of parental strains ASI 2803, ASI 2798 and Beesan No.1. Analysis of the genetic characteristics of the new cultivar 'Beesan No.2' showed a different DNA profile as that of the control strains, ASI 2803, ASI 2798 and 'Beesan No.1', when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about 123.3 g. And also the stipe was long. This new cultivar 'Beesan No.2' of Pleurotus ferulae was characterized white-like variety of King oyster mushroom in the color of stipe, that was long and low yield compared to that of other cultuvar 'Beesan No.1'. We therefore expect that this new strain will increase of export and substitute cultivar of King oyster mushroom.

Characteristics and breeding of a new cultivar Pleurotus eryngii, Seolsong (큰느타리버섯 신품종 '설송'의 육성 및 그 특성)

The Pe21-53(G09-21-10 x ASI 2844-9) was shown the best cultural characteristics, selected to be a new cultivar and designated as 'Seolsong'. The 'Seolsong' was distinctly formed incompatibility line in the confrontation growth of parental strains Keunneutari No. 2, Aeryni 3 and ASI 2844. Analysis of the genetic characteristics of the new cultivar 'Seolsong' showed a different DNA profile as that of the control strains, Keunneutari No. 2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25{\sim}30^{\circ}C$ and pH 5~8, respectively. This new cultivar 'Seolsong' of fruiting body production per bottle was about $131.4{{\pm}}43.1$ g which is about 102% quantity compared to that of other cultivar Keunneutari No. 2. And also the stipe is thick and long, but the number of available stipe is few. Particularly, it was tolerant of high moisture above 90% during the growth period after primodia formation. We therefore expect that this new strain will save of labor and cost of cultivation by without culling work.

Antimicrobial and Antioxidant Activities in Different Parts and Cultivars of Broccoli (브로콜리 품종 및 부위에 따른 항균활성과 항산화 효과)

  • The antimicrobial and antioxidant activities of different cultivars and parts of broccoli were investigated. The screening of antimicrobial activities for the floret and leaf extracts of 11 cultivars against selected gram positive (Staphylococcus aureus and Listeria monocytogenes) and gram negative bacteria (Escherichia coli O157:H7 and Salmonella enteritidis) were conducted. The AMG cultivar showed the highest antimicrobial activity against S. aureus with the floret and leaf extracts (12.83 and 13.00 mm). Antibacterial activity against L. monocytogenes was strongest with floret extract of NJ cultivar (13.58 mm) and leaf extract of YDR cultivar (13.92 mm). Moreover, the size of inhibition zone against L. monocytogenes was bigger than those of 4 kinds of pathogenic bacteria. Both floret and leaf extracts of Grd cultivar showed the highest antimicrobial activity against E. coli O157: H7, but there was no difference between floret and leaf extracts. Floret extract o f NJ cultivar and leaf extract of NY and 0c cultivars were effective against S. enteritidis whereas leaf extract exhibited better antimicrobial effect than floret extract. These results showed that floret extract of NJ and leaf extract of 0c had the highest antioxidant activity which was 39.90 and 43.64%, respectively. The antioxidant activity of leaf extract was 1.5 times higher than that of floret extract. All cultivars except NJ showed that electron donating ability of leaf extract was higher than that of floret extract.

Effects of Ozone on $CO_2$ Assimilation and PSII Function in Two Tobacco Cultivars with Different Sensitivities

  • Two tobacco cultivars (Nicotiana tabacum L.), Bel-B and Bel-W3, tolerant and sensitive to ozone, respectively, were grown in a greenhouse supplied with charcoal filtered air and exposed to 200 ppb ozone for 4 hr. Effects on chlorophyll fluorescence, net photosynthesis, and stomatal conductance are described. Quantum yield was calculated from chlorophyll fluorescence and the initial slope of the assimilation-light curve measured by the gas exchange method. Only the sensitive cultivar, Bel-W3, developed visual injury symptoms on up to 50% of the $5^{th}$ leaf. The maximum net photosynthetic rate of ozone-treated plants was reduced 40% compared to control plants immediately after ozone fumigation in the tolerant cultivar; however, photosynthesis recovered by 24 hr post fumigation and remained at the same level as control plants. On the other hand, ozone exposure reduced maximum net photosynthesis up to 50%, with no recovery, in the sensitive cultivar apparently causing permanent damage to the photosystem. Reductions in apparent quantum efficiency, calculated from the assimilation-light curve, differed between cultivars. Bel-B showed an immediate depression of 14% compared to controls, whereas, Bel-W3 showed a 27% decline. Electron transport rate (ETR), at saturating light intensity, decreased 58% and 80% immediately after ozone treatment in Bel-B and Bel-W3, respectively. Quantum yield decreased 28% and 36% in Bel-B and Bel-W3, respectively. It can be concluded that ozone caused a greater relative decrease in linear electron transport than maximum net photosynthesis, suggesting greater damage to PSII than the carbon reduction cycle.

Inoculum Sources to Generate High Mechanical Transmission of Barley yellow mosaic virus

  • Mechanical transmission of barley seedlings with barley yellow mosaic virus (BaYMV) is generally inefficient and is the major constraint for testing cultivar resistance to the virus. To explore mechanical transmission, BaYMV-infected barley plants were grown at different conditions and used as inoculum sources to seedlings of susceptible barley cultivar Baegdong. Extracts prepared from BaYMV-infected Baegdong plants at 47, 53, 74, and 90 days after symptom appearance (DASA) and grown at 10 and $12^{\circ}C$ gave 10, 30, 68 and 76% infection, respectively on inoculated susceptible barley cv. Baegdong seedlings. While Jinyangbori, another susceptible cultivar obtained 95% infection rate inoculated with extracts from 90 DASA disease source and grown at $10/12^{\circ}C$. However, low infection rates were obtained when the virus sources were grown in a greenhouse at $15-18^{\circ}C$. Our results indicate that longer incubation period and lower temperature are required for virus accumulation and stability.
