Micro-CT analysis of LPS-induced Alveolar Bone Loss in Diabetic Mice

  • Periodontal disease induces an increased incidence of tooth loss, particularly in cases with an associated loss of alveolar bone and periodontal ligaments. In this study, alveolar bone loss was detected by micro-computed tomography (CT) following exposure to E. coli lipopolysaccharide (LPS) in a streptozotocin (STZ)-induced diabetic mouse model. A 10 mg/ml dosage of E. coli LPS was applied between the first, second and third molars of the mice three times a week for 10 weeks. The loss of periodontal ligaments and alveolar processes was then evaluated by micro-CT using two and three dimensional microstructure morphometric parameters. In the diabetic mice, E. coli LPS induced the destruction of periodontal ligaments and loss of alveolar process spaces. The distances between periodontal ligaments were significantly widened in the STZ-LPS group compared with the untreated STZ group. The 10 mg/ml exposure to E. coli LPS in the STZ mice also resulted in a significant decrease in the alveolar bone volume fraction. The results of our study suggest that alveolar bone loss can be readily detected by volumetric micro-CT analysis as an increase in the distance between periodontal ligaments and in the alveolar process length.

고식이섬유쌀의 급여가 KK 당뇨 마우스의 체내 지질 대사에 미치는 영향 (Supplementary Effect of the High Dietary Fiber Rice on Lipid Metabolism in Diabetic KK Mice)

  • This study was conducted to investigate the supplementary effects of Suwon 464 developed by Rural Development Administration, which has over two times of dietary fiber content compared with normal rice (Ilpum), on lipid metabolism in diabetic mice. We supplied 5 kinds of experimental diets (com starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight, organ weights, and lipids levels of serum, liver and feces were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, 1.4, and 2.0% respectively. Body weight and liver and epididymal fat pad weights were lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentrations of serum triglyceride was lower in SP and SB groups than CO and IP groups. The levels of hepatic total lipid and total cholesterol were significantly lower in SP and SB groups than CO group, and the level of hepatic triglyceride was lower in IB, SP and SB groups than CO group. The levels of total lipid and triglyceride excreted in feces were higher in IB, SP and SB, and the level of total cholesterol in feces was higher in SP and SB groups than CO group. These results suggested that the high dietary fiber rice (Suwon 464) decrease the triglyceride or total cholesterol concentrations of serum and liver by increasing of fecal lipid excretion in diabetic mice. 혈액은행의 혈소판농축액을 이용한 당뇨생쥐의 창상치유 (Treatment of Diabetic Mouse Wounds using Blood Bank Platelet Concentrates)

  • Purpose: Many clinical trials have shown the effectiveness of platelet releasate on chronic wounds. However, a large volume of blood must be aspirated from a patient and a platelet separator is required. Here, we hypothesized that platelet concentrate obtained from a blood bank (PCBB) would be also effective at stimulating wound healing. The purpose of this study was to investigate the effectiveness of PCBB on accelerating healing of diabetic wounds in vivo. Methods: Round wounds of 5 mm diameter were made at four sites(two wounds on the left and two on the right side) on the backs of nine diabetic mice. Three hundred million platelets suspended in 0.05 ml fibrinogen were dispersed on each wound on left sides. Same amount of fibrinogen without platelets was dispersed on right side control wounds. Thereafter, 0.05 ml thrombin was applied to the each wound. Ten days after wound treatment, healed wounds were excised and the extent of wound healing in each group was compared. Results: Quantitative histologic analysis of epithelial gap distances revealed that PCBB treatment had greatly accelerated wound healing. Mean epithelial gap distances for PCBB treated and control wounds were $2.5{\times}0.6mm\;and\;3.6{\times}0.5mm$, respectively(p<0.05). Conclusion: Our results suggest that PCBB has potential to accelerate the healing of diabetic wounds.

A Vinegar-processed Ginseng Radix (Ginsam) Ameliorates Hyperglycemia and Dyslipidemia in C57BL/KsJ db/db Mice

  • Having idea to develop more effective anti-diabetic agent from ginseng root, we comprehensively assessed the anti-diabetic activity and mechanisms of ginsam in C57BL/KsJ db/db mice. The db/db mice were divided into 4 groups; diabetic control (DC), ginsam at a dose of 300 or 500 mg/kg (GS300 or GS500) and metformin at a dose of 300 mg/kg (MT300). Ginsam was orally administered for 8 weeks. GS500 reduced the blood glucose concentration and significantly decreased an insulin resistance index. In addition, GS500 reduced the plasma non-esterified fatty acid, triglyceride, and increased high density lipoprotein-cholesterol as well as decreased the hepatic cholesterol and triglyceride. More interestingly, ginsam increased the plasma adiponectin level by 17% compared to diabetic control group. Microarray, quantitative-polymerase chain reaction and enzyme activity results showed that gene and protein expressions associated with glycolysis, gluconeogenesis, and fatty acid oxidation were changed to the way of reducing hepatic glucose production, insulin resistance and enhancing fatty acid $\beta$-oxidation. Ginsam also increased the phosphorylation of AMP-activated protein kinase and glucose transporter expressions in the liver and skeletal muscle, respectively. These changes in gene expression were considered to be the mechanism by which the ginsam exerted the anti-diabetic and anti-dyslipidemic activities in C57BL/KsJ db/db mice.

γ-Oryzano1의 급여가 KK 당뇨 마우스의 혈당에 미치는 영향 (Effect of γ-Oryzanol on Blood Glucose in Diabetic KK Mice)

  • 본 연구는 쌀겨 및 쌀눈 기름에 들어 있는 ${\gamma}$-oryzano의 혈당 저하효과를 구명하기 위하여 수행하였다. 실험동물은 인슐린 비의존형 당뇨병 모델인 KK 당뇨마우스를 이용하였고, 실험식이의 0.2% 수준에서 ${\gamma}$ -oryzanol 첨가 유무에 따라 CO군과 GO군을 선정 후 8주 동안 실험식이를 급여하였다. 실험결과 실험식이 섭취량, 체중변화,장기의 무게는 실험군 사이에 차이가 없었으나, GO군에서 실험식이 급여 5주부터 공복혈당의 감소를 보였고, 내당능이 개선되어 혈당면적이 유의하게 낮은 수준을 보였다. 장기간의 혈당상태 지표인 당화혈색소가 실험군 사이에 유의한 차이를 보이지 않았으나 GO군에서 감소하는 경향을 보였고, 인슐린 수준은 ${\gamma}$-oryzanol을 섭취한 GO에서 높은 경향을 보였다. 따라서 ${\gamma}$ -oryzanol은 혈청 인슐린 수준을 높이고 혈당 농도 감소 효과가 있는 것으로 보이나, 실험식이 급여 기간의 변화 및 ${\gamma}$-oryzano의 첨가 수준에 따른 혈당 저하 효과 검토 등이 보완되어야 할 것으로 생각된다.

Anti-diabetic effects of blue honeyberry on high-fed-diet-induced type II diabetic mouse

  • BACKGROUND/OBJECTIVE: The blue honeysuckle berry (Lonicera caerulea var. edulis L.) is a small deciduous shrub belonging to the Caprifoliaceae family that is native to Russia, China, Japan, and Korea. The berry of this shrub is edible, sweet and juicy and is commonly known as the blue honeyberry (BHB). This study examined the anti-diabetic potential of BHB on high-fat-diet-induced mild diabetic mice. The hypoglycemic, and nephroprotective effects of the 12-week oral administration of blue honeyberry extract were analyzed. MATERIALS/METHODS: The hypoglycemic effects were based on the observed changes in insulin, blood glucose, and glycated hemoglobin (HbA1c). Furthermore, the changes in the weight of the pancreas, including its histopathology and immunohistochemical investigation were also performed. Moreover, the nephroprotective effects were analyzed by observing the changes in kidney weight, its histopathology, blood urea nitrogen (BUN), and serum creatinine levels. RESULTS: The results showed that the high-fat diet (HFD)-induced control mice showed a noticeable increase in blood glucose, insulin, HbA1c, BUN, and creatinine levels. Furthermore, growth was observed in lipid droplet deposition related to the degenerative lesions in the vacuolated renal tubules with the evident enlargement and hyperplasia of the pancreatic islets. In addition, in the endocrine pancreas, there was an increase in the insulin-and glucagon-producing cells, as well as in the insulin/glucagon cell ratios. On the other hand, compared to the HFD-treated mice group, all these diabetic and related complications were ameliorated significantly in a dose-dependent manner after 84 days of the continuous oral administration of BHBe at 400, 200 and 100 mg/kg, and a dramatic resettlement in the hepatic glucose-regulating enzyme activities was observed. CONCLUSIONS: By assessing the key parameters for T2DM, the present study showed that the BHBe could act as a potential herbal agent to cure diabetes (type II) and associated ailments in HFD-induced mice.

Glucose-lowering Effect of Powder Formulation of African Black Tea Extract in $KK-A^y/TaJcl$ Diabetic Mouse

  • We observed the suppressive effect of a powder formulation of African black tea extract prepared from the leaves of Camellia sinensis on type 2 non-insulin dependent diabetic mice, $KK-A^y/TaJcl$. Black tea extract significantly showed suppressive effect of the elevation of blood glucose on oral glucose tolerance test of 8 week-old $KK-A^y/TaJcl$ mice (p<0.05). Long-term treatment with black tea extract showed significant suppression of post-prandial blood glucose and obesity (p<0.05). The weight of the intestine of mice treated with black tea extract was significantly reduced (p<0.05). From these results, African black tea used in this study showed a suppressive effect on the elevation of blood glucose during food intake and the body weight.

스트렙토조토신으로 유도된 당뇨 마우스에서 Monascus purpureus을 이용한 발효 쑥의 기억력 개선 효과 (Memory improvement effect of Artemisia argyi H. fermented with Monascus purpureus on streptozotocin-induced diabetic mice)

  • 본 연구는 쑥 추출물(AA)과 홍국균(Monascus purpureus)으로 발효된 쑥(AAFM)을 이용하여 STZ로 인한 당뇨성 인지장애 개선효과를 확인하고자 수행되었다. C57BL/6L 마우스에 streptozotocin(STZ)을 복강 주사하여 당뇨를 유발시킨 후 4개의 군으로 분리하였고, 6주간 24시간 간격으로 AA 10 mg/kg BW와 AAFM 10 mg/kg BW을 경구투여 하였다. 7일 간격으로 혈당을 측정하였으며, 혈당 측정 결과 AA와 AAFM을 섭취한 군에서 STZ군 대비 유의적인 혈당감소를 나타내었다. Passive avoidance, Morris water maze 및 Y-maze test를 통해 STZ로 유도한 당뇨 동물모델에서 기억력 장애를 확인하였으며, 발효 쑥(AAFM)군에서 학습에 의한 기억력과 운동능력이 더 회복된 것을 확인하였다. 혈청분석 결과 STZ군에서는 GOT, GPT, BUN와 혈중 지질 함량이 높게 나타난 반면 발효 쑥(AAFM)군에서는 GOT, GPT 및 BUN이 STZ그룹에 비해 유의적으로 감소하였으며, HTR이 높게 나타났고 TG와 LDLC가 낮게 나타나 혈중 지질 성분이 개선됨을 확인할 수 있었다. 마우스 뇌 조직에서 발효 쑥(AAFM)은 아세틸콜린에스터레이스 활성을 저해시키고 아세틸콜린 함량을 증가시킴으로써 STZ로 인한 당뇨병에서의 신경전달시스템을 개선효과를 나타내었다. 발효 쑥(AAFM)의 경우, 마우스의 뇌 조직에서 SOD 함량을 증가시키고, 뇌와 간에서의 MDA 함량을 감소 시킴으로써 STZ와 같은 산화적 스트레스 인자에 대한 산화방지 효과를 나타내었다. 이러한 결과들을 종합해볼 때, 홍국균을 활용한 발효 쑥(AAFM)은 STZ로 유도된 당뇨 모델에서 당뇨성 인지장애에 대한 기억력 개선효과를 나타내었다. 특히, 쑥 추출물(AA)보다 마우스의 운동능력을 개선시켰으며, 혈중 지질 개선효과를 나타냈다. 더불어 산화방지 효과를 나타내 고혈당으로 발생될 수 있는 산화적 스트레스로부터 뇌 조직 등을 보호할 수 있음을 확인하였다. 결국 고혈당으로 인한 인지장애의 예방과 치료에도 효과를 나타낼 것으로 사료되어 고부가가치 식품으로 활용될 수 있는 산업적 가능성이 있다고 판단된다.

수풍순기환 투여가 고지방, 고탄수화물 식이로 유발된 비만형 제2형 당뇨병 동물모델에 미치는 영향 (The Effects of Supungsunki-hwan on High Fat, High Carbohydrate Diet-induced Obese Type 2 Diabetic Mouse Model)

  • Objective : Obesity is an important cause of diabetes, and lipotoxicity causes insulin resistance. Recently a lot of research is being done on PPAR-${\alpha}$. TNF-${\alpha}$. adiponectin, and leptin, which are important obesity related factors. In this study, we investigated the effects of Supungsunki-hwan on high fat. high carbohydrate diet-induced obese type 2 diabetic mouse models. Methods: Diabetes was induced in ICR male mouse (30${\pm}$5g) with Surwit's high fat, high sucrose diet. Mice were divided into 4 groups(n=10) of Normal. Control. Supungsunkj-hwan group. and acarbose group. The Supungsunki-hwsn group was given 10% Supungsunkj-hwan in their diet. and the acarbose group was given 0.5% acarbose in their diet. After 6 weeks. body weight. food intake, FBS and OGTT. lipid profile and liver enzymes, epididymal fat weight, and gene expression of leptin, adiponectin, TNF-${\alpha}$ and PPAR-${\alpha}$ were measured. Leptin. adiponectin. tumor necrosis factor(TNF)-${\alpha}$ and peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ were evaluated by reverse transcription-polymerase chain reaction. Results : Supungsunkj-hwan increased the gene expression of PPAR-${\alpha}$, which reduces lipotoxicity and insulin resistance. Supungsunkj-hwan also significantly reduced triglyceride. AST. ALT serum levels. and 1 hour oral glucose tolerance levels. Conclusion : These results show that Supungsunkj-hwan improves insulin resistance in the liver and muscles, by reducing triglyceride levels and lipotoxicity through increased PPAR-${\alpha}$ gene expression. This is supported by the fact that Supungsunkj-hwan significantly reduces 1 hour oral glucose tolerance levels. Therefore we suggest that Supungsunkj-hwan would be an effective treatment for obese type 2 diabetic patients.

피세아테놀과 레스베라트롤의 혈당조절 및 TLR4-NF-κB 경로 조절 작용 (In Vitro and In Vivo Effects of Piceatannol and Resveratrol on Glucose Control and TLR4-NF-κB Pathway)

  • 본 연구에서는 팔미틴산을 처리한 지방간질환 간세포 모델과 비만/당뇨 동물 모델인 KK/HlJ 마우스를 이용하여 피세아테놀과 레스베라트롤 투여가 염증조절에 주는 영향을 알아보고자 하였다. 4주간의 피세아테놀과 레스베라트롤 섭취는 공복혈당과 경구당부하 검사 2시간 후 AUC를 감소시켜 혈당 조절 개선 효과를 보였다. 또한, 팔미틴산을 처리한 지방간질환 간세포 모델에 피세아테놀과 레스베라트롤을 처리한 결과 염증조절 경로인자인 TLR4와 $NF-{\kappa}B$의 발현을 유의적으로 감소시켰다. 이를 in vivo 비만/당뇨 동물 모델인 KK/HlJ 마우스의 간 조직에서 확인한 결과 피세아테놀 섭취는 NLRP3와 $NF-{\kappa}B$의 간 조직에서의 발현을 유의적으로 감소시켰고, IL-1 발현을 감소시키는 경향을 보였다. 하지만 동량의 레스베라트롤 섭취는 이와 같은 항염증 효과를 보이지 않았다. 결론적으로 혈당 조절 개선 효과와 항염증 효과에 있어 피세아테놀이 레스베라트롤보다 우수한 효과를 가지고, 피세아테놀의 항염증 효과는 혈당 조절 개선 효과에 부분적으로 기여할 것으로 제안한다.
