Effects of Dietary Oligosaccharide on the Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 혈당 및 혈중 지질조성에 미치는 Oligosaccharide의 영향)

  • This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.

Effects of Dietary Supplementation with Allium hookeri Root on Hepatic Enzyme Contents in Streptozotocin-induced Diabetic Rats (삼채 보충식이가 당뇨흰쥐 간의 항산화효소 함량에 미치는 영향)

  • The purpose of this study was to examine the effect of Allium hookeri (AH) root on hepatic antioxidative enzyme contents in streptozotocin (STZ)-induced rats. Diabetes mellitus was induced in male Sprague-Dawley rats through injection of STZ dissolved in citrate buffer into tail veins at a dose of 45 mg/kg body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet, and the experimental groups were fed a modified diet containing 5% and 10% of AH root powder for 4 weeks. The experimental groups were divided into four groups: a normal control (N-control), STZ-control, STZ-AH 5%, and STZ-AH 10% supplemented groups. The STZ-AH 5% group showed a significant increase in liver glycogen compared to the STZ-control group. Muscle glycogen and liver protein contents significantly increased in the AH-supplemented groups compared to the STZ-control group. The liver malondialdehyde content of the AH-supplemented group was significantly lower than that of the STZ-control group. Xanthine oxidase content was significantly reduced in all experimental groups. Glutathione-S-transferase content was significantly elevated in the AH-treated groups compared to the STZ-control group. Superoxide dismutase content was not significantly different among the experimental groups. Catalase content was significantly higher in the STZ-AH 10% group compared to the STZ-control group. These results show that supplementation with AH root may be useful for diabetic therapy and damage from oxidative stress.

Effects of Enzymatic Hydrolysates from Hamcho (Salicornia herbacea L.) on Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (함초(Salicornia herbacea)의 효소적 가수분해물이 스트렙토조토신-유발 당뇨쥐의 혈당 강하 및 혈청 지질 개선효과)

  • This study was designed to investigate the effects of enzymatic hydrolysates (EH) from Hamcho (Salicornia herbacea L.) on blood glucose and serum lipid status in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into normal and 5 diabetic groups. The diabetic groups were fed enzymatic hydrolysate-free control (DM) diets or diets supplemented with 0.02% (DM-2), 0.04% (DM-4), 0.08% (DM-8), and 0.16% (DM-16) of enzymatic hydrolysate for 4 weeks. Body weight gains were lower in five diabetic groups than that of the normal group. Blood glucose was decreased in EH-supplemented groups as compared to the normal group, and especially the lowest blood glucose levels were found in DM-4 and DM-8 groups. Activities of three disaccharidase in the middle part of the intestine, such as maltase, sucrase and lactase, in EH-supplemented groups were significantly lower than those of DM group. There was no significant differences in the activities of glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) among all experimental groups. Serum triglyceride in DM group was significantly increased as compared to the normal group, but those of EH-supplemented groups were decreased to the normal level. Total cholesterol level in DM group was higher than EH-supplemented groups and normal group, but that of DM-16 group was significantly decreased to the normal level. HDL cholesterol level in DM group was significantly decreased compared to the normal group, but that of EH-supplemented groups was increased to the normal level. These results suggest that enzymatic hydrolysate from Hamcho has hypoglycemic and hypolipidemic effects on STZ-induced diabetic rats and may be useful as a dietary supplement for the treatment of diabetes.

Analysis for Compatibility of Daehwangmokdan-tang and Its Pharmacological Activities (대황목란탕(大黃牧丹湯)의 배오(配伍)분석과 그 약리활성)

  • Objectives : I analysed daehwangmokdan-tang's compatibility principle by the system of chief, deputy, assistant, and envoy and investigated pharmacological activities by categorizing with chemical components, molecular level, cellular level, animal level and human level based on Korean and Chinese studies for this formula. Methods : Daehwangmokdan-tang's compatibiltity principle was examined by the system of chief, deputy, assistant, and envoy. I looked into studies that presented in Korea from 1956 to 2016 about daehwangmokdan-tang through KOREA INSTITUTE OF ORIENTAL MEDICINE, Korean medicine information system (OASIS) and in Chinese for 20 years about daehwangmokdan-tang through China National Knowledge Infrastructure, CNKI. Then classify into chemical components, molecular level, cellular level, animal level and human level to analyse. Results : According to the system of chief, deputy, assistant, and envoy, chief herb is Rhei Radix et Rhizoma and Moutan Cortex, deputy herb is Natrii Sulfas and Persicae Semen, assistant and envoy herbs are Trichosanthis Semen. The amount of extraction of paeonol, total anthraquinone, and conjugated anthraquinone from daehwangmokdan-tang with the formulation of the system of chief, deputy, assistant, and envoy was the highest, and in the formulation of chief herb and deputy herb, the extraction amount of paeonol and conjugated anthraquinone was the lowest. With other formulations, the amount of extraction of total anthraquinone and conjugated anthraquinone was improved, although the degree was different. In particular, when it is blended with Persicae Semen as a deputy herb, the extraction amount of total anthraquinone and conjugated anthraquinone of Rhei Radix et Rhizoma as a chief herb is greatly increased, and the extraction amount of paeonol is rather different, but it is lowered. It was found that the amount of Mg Ca K Na in daehwangmokdan-tang was the highest. Pharmacological activities can be detected in inflammatory mediators and enzymes for molecular level. For cellular level, it can be determined in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell line. In mouse and rats for animal level and human level, in inflammatory diseases (acute appendicitis, acute pancreatitis, acute cholecystitis, acute abdominal disease, ect.), pharmacological activities was caught. Conclusions : From the above results, daehwangmokdan-tang is composed in line with the system of chief, deputy, assistant, and envoy, suggesting that there is certain rationality and scientific. Pharmacological activities of daehwangmokdan-tang are effective to anti-inflammation, improvement of sepsis, analgesic, muscle relaxation, and improvement of intestinal flora and its metabolites. Daehwangmokdan-tang is consistent with the action of diuresis and anti-inflammation in terms of the content of elements, suggesting that there is action of purging fire and removing blood stasis, defecation detoxification, cooling blood and clearing heat and activating blood and dispersing stasis.

Effect of Fermented Red Ginseng Extracts on Physiological Activity and Blood Glucose Level in Streptozotocin Induced Diabetic Rats (홍삼발효 추출물의 생리활성 및 streptozotocin으로 유발된 당뇨쥐의 혈당강하에 미치는 영향)

  • The purpose of this study was investigated hypoglycemic effects of fermented red ginseng extracts. We prepared non-fermented red ginseng extracts(R), fermented with Lactobacillus plantarum(RL) extracts, Saccharomycescerevisiae(RS) extracts, and L. plantarum mixed S. cerevisiae(RLS) extracts, examined composition of ginsenosides, SOD-like activity, and $\alpha$-glucosidase inhibitory activity. Ginsenoside Re was highest contents in all extracts, second was ginsenoside Rc and then ginsenoside Rb1. Concentration of these ginsenoside was showed higher in RS than in other extracts. SOD-like activity and $\alpha$-glucosidase inhibitory activity were shown higher in fermented red ginseng extracts than non fermented extracts. And activities of mixed fermentation extracts(RLS) higher than single fermentation extracts(RL, RS). Effects of blood glucose level, serum lipid profile and metabolic variables were evaluated in streptozotocin(STZ) induced diabetic rat. Experimental group was divided into 7 groups: normal control group(hereafter NC group), diabetes control group(DC group), positive control group treated with 50 mg/kg body weight of acarbose(PC group), treated with 300 mg/kg body weight of R, RL, RS and RLS extracts groups, respectively. Blood glucose level of DC group was maintained high level in all experimental period, but treated with red ginseng extracts groups was reduced the glucose level by R group 18.00%, RL group 28.07%, RS group 29.03%, RLS group 42.42%, respectively. The concentration of total cholesterol and triglyceride of fermented red ginseng extracts treated groups (RL, RS, RLS) was lower than non- fermented extracts group(R) DC and PC groups. The activity of ALT, AST in RLS treated groups were lower than other groups.

Allium Hookeri Extract Enhances Glucose Uptake through GLUT4 Up-regulation in 3T3-L1 Cells (GLUT4 상향조절을 통한 Allium hookeri 추출물의 3T3-L1 세포 내 포도당 흡수 증진 효과)

  • Diabetes mellitus is associated with insulin resistance, which leads to down-regulation of insulin signaling and the decreased glucose uptake. Adipocytes are sensitive to insulin, and closely implicated in insulin resistance and diabetes. Insulin stimulates differentiation of preadipocytes to adipocytes, and increases glucose transport. Allium species have been used as traditional medicine and health-promoting foods. Allium hookeri (A. hookeri) is reported to improve the pancreatic ${\beta}-cell$ damage and exhibit pancreatic anti-inflammatory activity in streptozotocin-induced diabetic rats. We investigated whether A. hookeri extract (AHE) may stimulate glucose uptake in adipocytes through increasing insulin sensitivity. AHE enhanced fat accumulation, a differentiation biomarker, under the partial induction of differentiation by insulin. $PPAR{\gamma}$, a transcription factor highly expressed in adipocytes, promotes adipocyte differentiation and insulin sensitivity. AHE increased the differentiation of preadipocytes through up-regulation of $PPAR{\gamma}$. The activation of $PPAR{\gamma}$ increases the GLUT4 expression during adipocyte differentiation. GLUT4 is responsible for glucose uptake into the adipocytes. AHE increased the expression of GLUT4 in adipocytes, and subsequently enhanced the insulin-stimulated glucose uptake. These results suggest that AHE promotes adipocyte differentiation through activation of $PPAR{\gamma}$, and leads to enhance glucose uptake in adipocytes along with GLUT4 up-regulation. Thus, AHE may be effective for the insulin-sensitizing and anti-diabetic activities.

Inhibitory Effect of Combination with Korean Red Gnseng and Morus Alba in High Fructose-induced Vascular Inflammation and Steatohepatitis (고과당식이 투여 랫드모델에서 홍삼과 상엽 복합투여에 대한 혈관염증 및 지방간염 억제 효과)

  • This study was designed to elucidate whether combination with Korean red ginseng and Morus alba L. (MPM), traditional treatment for diabetes, ameliorates on high fructose-induced steatohepatitis and vascular inflammation. Animals were divided into four groups; Control receiving tap water, fructose-fed, rosiglitazone-treated fructose-fed rats, and MPM-treated fructose-fed rats both receiving supplemented with 60% fructose (n=10). The MPM or rosiglitazone groups initially received a high-fructose diet alone for 8 weeks, with supplementation with MPM or rosiglitazone, peroxisome proliferators-activated receptor gamma ($PPAR{\gamma}$) agonist, occurring during the final 6 weeks. Treatment with MPM significantly prevented the increase in c-reactive protein (CRP) levels in the high fructose group. MPM suppressed high fructose diet-induced vascular inflammation marker expression such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. MPM also reduced intima/media thickness of thoracic aorta. Histologic observation and oil red O staining demonstrated hepatic tissue damage and lipid accumulation were severe in high fructose group. Treatment with MPM ameliorated hepatic tissue morphology with minimized steatosis. In addition, MPM attenuated hepatitis by inhibition of monocyte chemoattractant protein-1 (MCP-1) expression. MPM-fed group showed lower serum GOT and GPT levels comparing with high fructose group. MPM and rosiglitazone (positive control) significantly decreased the size of epididymal adipocytes. Taken together, the administration of MPM inhibited high fructose-induced steatohepatitis and vascular inflammation. These results suggested that MPM is useful in the prevention or treatment of metabolic syndrome-related disorders such as fatty acid metabolism and vascular homeostasis.

Pharmacological Evaluation of Bamboo Salt (죽염의 약리작용 평가)

  • Bamboo salt has been used for the purpose of precaution and treatment of certain diseases including cancer. Therefore, present study was carried out to ascertain the effects of bamboo salt upon anti-cancer, anti-hypertensive, and anti-diabetic activities as well. To examine the anti-cancer activity of bamboo salt, ICR mice implanted with 1$\times$l0$^{6}$ cells of sarcoma 180 intraperitoneally had been treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 60 days using adriamycin (2 mg/kg) as a positive control. Neither survival rate nor body weight had been significantly influenced by all the treatments indicating that bamboo salt A did not exert the anti-cancer effect on ICR mice. Anti-hypertensive activity was examined in spontaneously hypertensive rats (SHR) which had been administered with bamboo salt A, crude salt, and reagent-grade NaCl (0.1, 0.5, and 1.0% in drinking water) for 28 days using hydralazin (2 mg/kg) as a positive control. Blood pressure and heart rate were measured at 1, 3, and 4 weeks after the starting date. Significant anti-hypertensive activity was not observed in any treated group compared to the positive control group. In order to determine if bamboo salt had anti-diabetic activity, rats in which diabetes had been induced by streptozotocin (45 mg/kg, i.m.) were treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 28 days using insulin (50 U/kg, s.c..) as a positive control. Blood samples were taken and analyzed at 1,2, and 4 weeks after the starting date. Bamboo salt did not cause any decreasing effect on the blood glucose levels. These results clearly demonstrated that bamboo salt A did not exert anti-cancer, anti-hypertensive, or anti-diabetic activities in the present experimental animals.

Effects of Chinese Medical Material Extract on Plasma Lipids and Glucose in Male Rats (한약재 추출물이 흰쥐의 혈장지질 및 혈당농도에 미치는 영향)

  • Chinese medical materials of Astragalas manbranaceus Bunge, Angelica gigas Nakai, Cnidium officinale, Glycyrrhiza uralensis Fisch, Lycium Chinese Mill were mixed, mixing ratio was 28:20: 18:18:16(w/w). Yield and concentration of extract obtained through extraction- concentration process of the mixed Chinese medical materials were 37.28%(w/w) and 50Brix, respectively. Animal feed(AF) used for animal test was manufactured from both mixing of general AF and this extract(0∼ 5%, w/w) and freeze dried. After the male rats were fed experimental diets in order to test during 4 weeks, the results of plasma TG, total cholesterol, phospholipid content changes and plasma, pancreas and femur insulin concentration changes were followed. The more the amount of treatment of Chinese medical material extract are, the higher the effect of concentration decline are in plasma lipid concentration. And at 3% treatment plasma TG, total cholesterol and phospholipid contents are decreased respectively 8.62%, 2.81% and 2.11%. Plasma glucose concentration has also similar effect that those things are decreased at 3% treatment. Insulin concentration of plasma, pancreas and femur are difficult to prove the effects due to respective increase and decrease. We guess that these results comes from not experimenting intentioned rat of diabetes mellitus but normal rat.

A Study on the Treatment of SD rat Menopausal Obesity Utilizing Fermentation Techniques (발효기법을 이용한 SD rat의 갱년기 비만 치료효과에 관한 연구)

  • In our study, osteoporosis was induced by ovariectomized in female Sprague-Daweley (SD) rats, and the prevention and treatment efficacy of the climacteric disease the ostmenopausal type I pattern was examined by using the experimental substance soybeans and arowroot. No clinical signs and mortality after continuous oral administration of experimental substances for 14 weeks were not observed, however, the weight of experimental animals underwent ovariectomized was increased significantly in comparison with control group (p<0.01). The atrophy of the uterus due to T3 group was 23.64% after 7 weeks, 24.44% after 14 weeks (in comparison with N.C. group), and it was statistically significantly increased. In regard to the change of blood cells, it was observed that platelets were statistically significantly reduced in the ovariectomized group, and administered orally experimental substances continuously for 7 weeks, in all groups administered experimental substances, it was found that platelets had the tendency to be increased more than N. C. group. In regard to He change of blood biochemistry, removed the ovary, the concentration of ALP showed the tendency to be increased than control group, and particularly in T3 group, it was increased significantly. In regard to the concentration of cholesterol, in comparison with negative control group, it was reduced 68% at 7 weeks and 35% at 14 weeks. After ovariectomized, the amount of estrogen was found to be reduced by 21.37% in comparison with control group, it showed the tendency to be increased by 4.49% in T1 group and 7.62% in T2 group, the concentration of estrogen in each group showed the tendency to be increased than negative control group, and in T3 group, it was increased to 100.46% and 117.65% in T4 group, and it was increased more than control group. Based on the above experimental results, in the experimental animals female rats, because of the hormonal imbalance induced by ovariectomized, a large mount of fat is accumulated in the body and due to it osteoporosis, obesity, hypertension, hyperlipidemia, fat of the liver, arteriosclerosis, diabetes, and other metabolic diseases were developed. Hence, when the experimental substance Extraction of ferment arrowroot was orally administered continuously for 14 weeks, it was thought that a certain proportion of the hormonal balance was maintained that functioned as a substance interfering the accumulation of fat, and it was considered to be of help in the treatment of not only osteoporosis Type I, but also for the prevention and treatment of various endocrinal diseases.
