Corynespora cassiicola에 의한 토마토 갈색무늬병(가칭)의 발생 (Occurrence of Target Spot of Tomato Caused by Corynespora cassiicola in Korea)

  • 토마토잎에 발생한 갈잭의 둥근 윤문병반에서 분리한 Carynespara sp.는 C. cassiicola로 동정되었으며 이 균을 접종 실험한 결과 토마토에 병원성을 나타내었다. 이 균의 생장적온은 $27^{\circ}C$였으며 $19^{\circ}C$$35^{\circ}C$에서는 균총생장이 저조하였다. 포자형성은 potato dextrose malt agar에 서 매우 양호하였으며 V-8 juice agar와 potato dextrose agar에서는 중간정도였다. 병원균을 인공배양하여 토마토 건전식물체에 분무접종하였을 때 자연상태에서와 유사한 갈색의 윤문을 형성하는 병징을 나타내었다.

  • 콩 잎에 발생한 갈색점무늬병반에서 분리한 Corynespora sp.는 C. cassiicola로 동정되었으며 이 균을 접종 실험한 결과 콩에 병원성을 나타내었다. 이 균의 생장적온은 $27^{\circ}C$였으며 $19^{\circ}C$$35^{\circ}C$에서는 균총생장이 저조하였다. 포자형성은 potato dextrose malt agar에서 매우 양호하였으며 V-8 juice agar와 potato dextrose agar에서는 중간정도였다. C. cassiicola에 대한 콩 품종의 감수성 정도를 비교하였던바 공시한 9개 품종 중 황금 콩이 저항성을 나타내었고 기타 품종은 저항성 및 감수성을 나타내었다.

  • Eight Korean native black goats were used, $10m{\ell}$ of blood was collected from the Jugular vein into heparinized tubes a week interval. The heparinized blood was centrifuged for separation to blood plasma and corpuscles. The hematocrit, per centage of blood that is red blood cells, was reschuffuled of 10 %, 20 %, 30 %, 40 % and 50 % using blood plasma, 0.9 % NaCl solution and 5.4 % dextrose solution. The sedimentation rates of red blood cell obtained at $7^{\circ}C{\pm}1^{\circ}C$ and $27^{\circ}C{\pm}1^{\circ}C$ are summarized as follows. 1) The sedimentation rates of red blood cell were more increased by lower PCV%, i.e. there was a reverse relationship between the sedimentation rates and PCV% at any condition of these experiments. 2) The RBC were sedimented the most quickly in the NaCl solution and slower in the plasma compare with the dextrose solution at the same PCV%. 3) There was no temperature effect on the sedimentation rates between the two groups of $7^{\circ}C{\pm}1^{\circ}C$ and $27^{\circ}C{\pm}1^{\circ}C$(at PCV 20% and 10%), even though the temperature difference is $20^{\circ}C$.

The Effect of Dilution with 10% Dextrose Solution on the Potency of Botulinum Toxin A

  • Purpose: The aim of this study was to compare the potency-stabilizing effects of two different diluents of botulinum toxin A (10% dextrose solution and 0.9% saline). Methods: A mouse lethality bioassay was undertaken. Ninety mice were divided into experimental and control groups which received varying dosages in subgroups of 10. The experimental group was injected with botulinum toxin A diluted with 10% dextrose solution and the control group was injected with botulinum toxin A diluted with 0.9% saline. A 72 hours after intraperitoneal injection, the number of dead mice was counted to confirm median lethal dose ($LD_{50}$) of each group. Results: The value of $LD_{50}$ in the experimental group was approximately 0.131 mL (1.31 U) and the value of $LD_{50}$ in the control group was approximately 0.107 mL (1.07 U). The potency preservation rate of the experimental group was estimated to be 93.5% and that of the control group was estimated to be 76.3%. Conclusions: Dilution with 10% dextrose solution displayed less potency loss than 0.9% saline.

  • Purpose: The purpose of this study was to examine the pain reducing effects of the dextrose-coated pacifier on venipuncture in premature infants. Methods: The design of this study is a nonequivalent control group pretest-posttest design and a crossover trial. The analysed cases were 40 premature infants (20 in experimental group and 20 in control group) in neonatal intensive care unit of a University Hospital, Gyeongnam Province, Korea. The data were collected from April to October, 2009. The experimental treatment was carried out nursing 20% dextrose-coated pacifier on venipuncture for IV injection. The instruments were $O_2$ saturation and heart rate on pulse oxymeter monitor to measure physiologic pain responses, and NIPS to measure behavioral pain responses. Collected data were analyzed with $x_2$ test, t-test using SPSS program. Results: The effects of the 20% dextrose-coated pacifier were found in the physiologic (only heart rate) and behavioral pain response on venipuncture. Conclusion: These finding is suggested that the dextrose-coated pacifier could be an effective nursing intervention for reducing pain on venipuncture in premature infants.

포도당 수액요법을 받는 당뇨병 환자와 비당뇨병 환자의 양쪽 손가락 끝 혈당치 비교 (Comparison of Blood Glucose Levels from the Fingertips of Both Patients with and without Diabetes Who are Receiving Dextrose Fluid Therapy)

  • Purpose: The aim of this study was to identify the effect of dextrose intravenous fluid (IVF) on the blood glucose levels taken from both fingertips of patients with diabetes mellitus (DM) and those without DM. Methods: From October 2012 to February 2013, 21 DM and 25 non-DM patients were recruited. Blood glucose levels taken from the both fingertips of patients at 7AM while IVF was not infusing and at 11AM, 5PM, and 9PM while IVF was infusing. Results: The differences between the mean values of blood glucose from the fingertips at four different times was not statistically significant in DM patients as well as non-DM patients. Also intra-class correlation for blood glucose levels from the fingertips with and without IVF infusion in both groups was shown over about 0.95 at each time (p<.001). Conclusion: Blood glucose levels measured in the same fingertips of both arms were the same regardless of dextrose IVF infusion. The results indicated that IVF with dextrose may not have any significant effect on the fingertip results of blood glucose level. This study may suggest that patients' both arms and any fingertips can be used for blood glucose monitoring even when the patients are on dextrose IVF infusion.

화경버섯의 배양조건에 따른 균사생장 및 섬유질분해효소 활성에 관한 연구 (Studies on the Mycelial Growth and Cellulolytic Enzyme Production of Lampteromyces japonicus at Various Cultral Conditions)

  • 본 연구는 섬유소분해능이 우수한 L. japonicus의 균사생육에 미치는 중요 인자에 대한 배양학적 특성을 규명하여 인공재배 및 균사체 배양을 이용한 섬유소분해효소의 생산을 높일 수 있는 기초자료를 얻고자 실험을 수행하였다. 그 결과, 균사체 배양의 최적온도는 $30^{\circ}C$이었으며, 최적 pH 는 6.0이었다. 균사생장에 적합한 배지는 glucose peptone 배지, malt yeast extract 배지, yeast malt peptone 배지, potato dextrose 배지, Hennerberg 배지 등으로 조사되었다. 균사생장에 가장 양호한 탄소원은 dextrose 였으며, 최적농도는 1.2%이었다. 또한 질소원으로서 균사생장에 가장 양호한 유기, 무기질소원, 아미노산의 종류 및 최적농도는 각각 yeast extract(1.7%), $(NH_4)_2HPO_4(0.2%)$, glutamine(0.2%) 이었다. 균사생장에 양호한 무기염류는 $Al_2(SO_4)_3{\cdot}14H_2O$인 것으로 조사되었으며 최적농도는 0.1 M 이었다.

  • 수종 곤충기생균의 균사생장 및 포자형성에 미치는 탄소원 (dextrose, starch, sucrose, sorbitol)과 실소원(Yeast extract 농도별) 배지 및 pH의 영향을 조사하였다. Metarrhizium ftavoviride var. minus와 Hirsutella strigosa는 dextrose, starch, sucrose 원배지에서 균사생장이 많은 반면 한국에서 발견된 Hirsutella sp.(Korea)는 dextrose률 제외한 배지에서 좋은 생장을 보였다. 사용한 실소원은 균사의 생장에 필수적이나 기생균의 종류에 따라 생장의 차이가 있었다. 그러나 Nomurea riteyi는 배지원에 관계없이 까다로운 균사생장을 나타냈으며 공시균 모두 pH 부근의 배지에서 균사생산량이 많았다. 한편 2종 기생균을 공시하여 포자형성을 조사한 결과, M. flaνoviride var. minus는 공시실소원과 dextrose원 배지에서 포자생산이 많아 기생균별 배지 선택성의 차이를 나타냈다.

인산가용화 사상균, Penicillium sp. PS-113 균주의 분리 및 배양특성 (Isolation and Cultural Characteristics of a Phosphate-solubilizing Fungus, Penicillium sp. PS-113)

  • A fungus with high phosphate solubilizing activites was isolated from soil using potato dextrose agar-calcium phosphate medium and identified to Penicillium sp. PS-113, based on the morphological characteristics of conidiophore and conidia; flask shape of phialide, simple branching type of conidiophore, and columnar shape of conidial head, in malt extract agar and potato dextrose agar media. The optimum temperature ad initial pH to solubilize rock phosphate in potato dextrose broth-rock phosphate medium were 30$^{\circ}C$ and pH 8.0, respectively. In these conditions phosphate solubilizing activities of Penicillium sp. PS-113 against four types of insoluble phosphate like tricalcium-phosphate, aluminium phosphate, hydroxyapatite and rock phosphate were quantitatively determined. As results, this fungus highly produced free phosphates to the culture broth with the concentrations of 1,283 ppm against tricalcium-phosphate, 585 ppm against rock phosphate, 528 ppm against aluminium phosphate, and 242 ppm against hydroxyapatite, respectively.

  • When starch is hydrolyzed either by acid or by the enzymes maltase or diastase, contained in germinating barley, a yield of 80% of maltose is obtained. Maltose is built of two molecules of ${\alpha}$-glucose, bound in the position 1:4 i.e., carbon atom 1 of one glucose molecule is bound in a glucosidic bond to carbon atom 4 of the second molecule. Until around 1960, dextrose and glucose syrups were prepared from starch exclusively by acid hydrolysis. The process was corrosive, and the dextrose yield low. It was, therefore, a great step forward when pure glucoamylase in combination with bacterial ${\alpha}$-amylase made possible a complete enzymatic hydrolysis of starch to dextrose. Today several enzymatic processes are used in the industry.
