Gonadal Development, Spawning and Plasma Sex Steroid Levels of the Indoor Cultured Grunt, Hapalogenys nitens

  • The gonadosomatic index (GSI), gonadal development and changes in hormones in plasma level of the indoor cultured grunt (Hapalogenys nitens) were investigated by histological study from August 2011 to October 2012. The GSI showed similar trends with gonad developmental stages during the culture periods. Changes in plasma level of estradiol-$17{\beta}$ of female H. nitens reached the highest value before the spawning period, and seasonal changes in plasma level of estradiol-$17{\beta}$ were similar in trends of oocyte developments and GSI changes. Testosterone levels of male H. nitens reached the highest value before and after the spent stage. Ovarian developmental stages of H. nitens could be classified into early growing stage, late growing stage, mature stage, ripe and spawning stage, recovery and resting stage. The testicular developmental stages could be divided into growing stage, mature stage, ripe and spent stage, and recovery and resting stage.

  • An experiment was conducted to establish a large scale production method of anti-serum against chicken IgA and to profile the developmental changes of serum IgA levels during the feeding period(from hatching to 7 weeks of age) in broiler chicks. Blood samples were taken from Hubbard chicken at the age of hatching, 3 days of age, and weekly thereafter till to 7 weeks of age. The pure IgA was isolated from ammonium sulfate treated chicken bile juice by gel filtration chromatography ( Sepharose GL-6B) - The quantitative assay of serum IgA were carried by RID method. Developmental changes of serum IgA concentrations were 0.42 mg /mL at hatching, thereafter dicreased gradually, lowest at 1 week of age(0.17 mg /mL), and gradually increased to 7 weeks of age(2.73 mg /mL). There was no sexual difference in serum IgA level, but female chicks showed higher IgA levels than male chicks during the experimental period.

Effects of Theme-Based Garden Book Making in Capstone Design on Self-Determination and Social Skills of College Students with Developmental Disabilities

  • Background and objective: This study was conducted to examine the effects of the theme-based garden book (photo book) making integration program on social skills and self-determination of college students with developmental disabilities. Methods: We analyzed the changes in social skills and self-determination of college students with developmental disabilities. The first step was defining the theme-based garden book (photo book) making integration program, which was done by focus group interviews (FGI) comprised of specialists in the industry and academia. The second step was carrying out 8 sessions of the program. The third step was examining the effects of the program on social skills and self-determination of college students with developmental disabilities. The participants of this study were 36 college students with developmental disabilities attending N University, and 17 students were in the experiment group and the other 19 in the control group. Results: The control group showed no statistically significant changes in the three sub-domains of social skills, while the experimental group showed statistically significant differences across all three sub-domains, such as cooperation(P < .05), assertion(P < .05), and self-control(P < .05). The experimental group, that participated in the program showed statistically significant differences across all four sub-domains of self-determination skills, such as behavior (P < .05), cognition(P < .05), school support(P < .05) and home support(P < .05). This proved that the theme-based garden book (photo book) making program contributed to bringing positive effects to self-determination and social skills. Conclusion: These results suggest that the theme-based garden book (photo book) making integration program could improve self-determination and social skills of college students with developmental disabilities. These positive changes are related to the high level of interest in theme-based garden design programs among college students with developmental disabilities, and thus these programs can be used effectively to improve social adjustment.

  • The changes in calcium binding protein(CBP) of mouse epididymal sperm during their post-testicular differentiation were analyzed by two-dimensional SDS-PAGE. According to dpididymal maturation, capacitation and acrosome reaction of spermatozoa, both quantitative and qualitative changes of CBPs in the epididymal sperm was detected. It suggested that the development of fertilizing ability of epididymal sperm was closely related to the changes in the CBPs profiles of sperm during epidiyaml transit.

Expression of CyI Cytoplasmic Actin Genes in Sea Urchin Development

  • We present a study of evolutionary changes in expression of actin genes among closely related sea urchin species that exhibit different modes of early development. For this purpose, polyclonal antisera raised against peptides from the carboxyl terminus of the HeCyI cytoskeletal actin of Heliocidaris erythrogramma were used. H. erythrogramma is a direct developing sea urchin that proceeds from embryonic to adult stages without an intervening feeding larval stage. Expression patterns of the CyI actin isoform were compared with those of Heliocidaris tuberculata and to a related sea urchin Strongylocentrotus purpuratus, which both produce a feeding pluteus larval stage. The CyI actin of all three species is expressed in the same cell types. However, its expression patterns have been changed with reorganization of early cell lineage differentiation, which is apparent among the three species. Thus. evolutionary changes in CyI actin gene expression patterns are correlated with not only phylogenetic relationship, but developmental mode. The implication of this observation is that evolutionary changes in expression patterns of histospecific genes may underlie the emergence of novel developmental processes.

  • "Epigenetics" means the study of heritable changes in gene-activity without changes in DNA sequences. Methylation of the cytosine residue in a CpG dinucleotide sequence is a characteristic of the vertebrate genome. In vertebrates, methylation of DNA mainly occurs at the 5′-position of cytosine in a CpG dinucleotide forming 5-methylcytosine. Methylation of DNA plays a profound role in transcriptional repression of gene expression through several mechanisms. Generally, DNA of inactive genes is more heavily methylated than that of active ones; conversely demethylation of DNA reactivates gene expression in vivo and in vitro.

  • Intracellular localization and the developmental changes in activities of uricase and allantoinase during germination were investigated with the cotyledons of rape(Brassica napus L.) seedlings. The development anddisappearance of uricase activity took place independently of light, but allantoinase activity was increased by light. The temporal pattern of uricase activity showed that uricolysis was actively taking place in the cotyledons during their early stages of germination. While uricase can be localized in the microbody fraction isolated from crude organelle extracts of the cotyledons by density gradient centrifugation, most of the allantoinase activity found in the microbody fraction did not appear to be an integral part of the microbody.

Differential Inheritance Modes of DNA Methylation between Euchromatic and Heterochromatic DNA Sequences in Ageing Fetal Bovine Fibroblasts

  • To elucidate overall changes in DNA methylation that occurs by inappropriate epigenetic control during ageing, we compared fetal bovine fibroblasts and their aged neomycin-resistant versions using bisulfite-PCR technology. Reduction in DNA methylation was observed in euchromatic repeats (18S-rRNA/art2) and promoter regions of sing1e-copy genes (the cytokeratin/-lactoglobulin/interleukin-13 genes). Contrastingly, a stable maintenance of DNA methylation was revealed in various heterochromatic sequences (satellite I/IIalphoid and Bov-B). The differential inheritance modes of DNA methylation was confirmed through the analysis of individual neomycin-resistant clones. These global, multi-loci analyses provide evidence on the tendency of differential epigenetic modification between genomic DNA regions during ageing.

  • The seminiferous epithelium cycle and developmental stages of spermatids in Clethrionomys rufocanus were observed under a light microscope. The seminiferous epithelium cycle was divided into 8 stages. Type Ad spermatogonia appeared through all stages. Type Ap, In, and B spermatogonia appeared in stages I, II, III, and IV. In the first meiosis prophase, the leptotene spermatocytes appeared from stage V, the zygotene spermatocytes in stages I, VI, VII, VIII, the pachytene spermatocytes from stages II to VI, the diplotene spermatocytes in stage VII. The meiotic figures and interkinesis spermatocytes were observed in stage VIII. Developing spermatids were subdivided into 10 steps, based on the morphological characteristics such as the acrosome formation changes in spermatozoa, nucleus, cytoplasm, and spermiation changes. The C. rufocanus spermatocytogenesis and spermiogenesis results displayed similar results with Apodemus agrarius coreae and A. speciosus peninsulae. Considering all the results, the spermatogenesis may be useful information to analyze the differentiation of spermatogenic cells and the breeding season.
