Purpose: Drug can be hazardous for people if misused although they are useful for their indication. In urgent incidences such as overdose, proper treatment for intoxication can save patients' lives. In emergent case regarding drug overdose, pharmacists should know how to provide correct information including antidote to other healthcare providers. However, in Korea, there is neither regular class nor education material regarding detoxification. Therefore, the object of our study is to investigate the perception of pharmacists about it. Method: We surveyed hospital pharmacists by means of self-reporting questionnaire in order to investigate the pharmacists' perception of detoxification treatments from May $12^{th}$ 2012 to August $10^{th}$ 2012. The questionnaire comprised of demographic information, interest in detoxification treatment (inquiry from patients and education about antidote), knowledge of antidote, effective drug search route and perception of the need for education and stocking materials about antidotes). Results: It included total 281 hospital pharmacists from 30 hospitals in S. Korea. Of them, only 16.7% have been questioned about drug overdose from patient or representative and 35% have learned about antidotes in case of drug overdose through education program of Korean association of hospital pharmacist or university. About 98% thought that education and stocking materials about frequently overdosed drug and antidotes are helpful for patients in emergent case. Also, the percentage of correct answer of each questions about antidotes were higher in educated group ($p{\leq}0.001$). The more work years are, the percentage of correct answer of each questions are higher ($p{\leq}0.001$). Conclusion: In conclusion, it will be helpful for reducing damage by drug overdose that pharmacists take regular education about antidote for all pharmacist and pharmacy student. In addition, preparing and keeping booklet for Korean Style-antidote in pharmacy is needed currently for protecting public health.
The induction of detoxification enzymes by benzyl isothiocyanate (BITC) and its synthetic N-acetyl-L-cysteine (NAC) conjugate (NAC-BITC) was examined in Hepa1c1c7 murine hepatoma cells. BITC and NAC-BITC inhibited Hepa1c1c7 cell growth in a dose-dependent manner. Cell growth was 4.5~57.2% lower in Hepa1c1c7 cells treated with $0.1{\sim}1.0{\mu}M$ BITC than in control-treated Hepa1c1c7 cells. The NAC-BITC treatment had a similar inhibitory pattern on Hepa1c1c7 cell growth; $0.5{\mu}M$ and $10{\mu}M$ NAC-BITC decreased cell growth by 13.6% and 47.4%, respectively. Treatment of Hepa1c1c7 cells with $0.1{\sim}2.0{\mu}M$ BITC also elicited a dose-response effect on the induction of quinone reductase quinone reductase (QR) activity and QR mRNA expression. Treatment with $1{\mu}M$ and $2{\mu}M$ BITC caused 1.8- and 2.8-fold inductions of QR mRNA, respectively. By comparison, treatment with $1{\mu}M$ and $2{\mu}M$ NAC-BITC caused 1.6-and 1.9-fold inductions of QR mRNA, respectively. Cytochrome P450 (CYP) 1A1 and CYP2E1 induction were lower in $0.1{\sim}2{\mu}M$ BITC-treated cells than in control-treated cells. CYP2E1 activity was 1.2-fold greater in $0.1{\mu}M$ NAC-BITC-treated cells than in control-treated cells. However, the CYP2E1 activity of cells treated with higher concentrations (i.e., $1{\sim}2{\mu}M$) of NAC-BITC was similar to the activity of control-treated cells. Considering the potential of isothiocyanatesto prevent cancer, these results provide support for the use of BITC and NAC-BITC conjugates as chemopreventive agents.
The ecosystems of certain abandoned mines contain arsenic-resistant bacteria capable of performing detoxification when an ars gene is present in the bacterial genome. The ars gene has already been isolated from Pseudomonas putida and identified as a member of the membrane transport regulatory deoxyribonucleic acid family. The arsenite-oxidizing bacterial strains isolated in the present study were found to grow in the presence of 66.7 mM sodium arsenate($V;\;Na_2HAsO_4{\cdot}7H_2O$), yet experienced inhibited growth when the sodium arsenite($III;\;NaAsO_2$) concentration was higher than 26 mM. Batch experiment results showed that Pseudomonas putida strain OS-5 completely oxidized 1 mM of As(III) to As(V) within 35 h. An arsB gene encoding a membrane transport regulatory protein was observed in arsenite-oxidizing Pseudomonas putida strain OS-5, whereas arsB, arsH, and arrA were detected in strain OS-19, arsD and arsB were isolated from strain RW-18, and arsR, arsD, and arsB were found in E. coli strain OS-80. The leader gene of arsR, -arsD, was observed in a weak acid position. Thus, for bacteria exposed to weak acidity, the ars system may cause changes to the ecosystems of As-contaminated mines. Accordingly, the present results suggest that arsR, arsD, arsAB, arsA, arsB, arsC, arsH, arrA, arrB, aoxA, aoxB, aoxC, aoxD, aroA, and aroB may be useful for arsenite-oxidizing bacteria in abandoned arsenic-contaminated mines.

The present study investigates the possible regulatory role of exogenous nitric oxide (NO) in antioxidant defense and methylglyoxal (MG) detoxification systems of wheat seedlings exposed to salt stress (150 and 300 mM NaCl, 4 days). Seedlings were pre-treated for 24 h with 1 mM sodium nitroprusside, a NO donor, and then subjected to salt stress. The ascorbate (AsA) content decreased significantly with increased salt stress. The amount of reduced glutathione (GSH) and glutathione disulfide (GSSG) and the GSH/GSSG ratio increased with an increase in the level of salt stress. The glutathione S-transferase (GST) activity increased significantly with severe salt stress (300 mM). The ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), catalase (CAT) and glutathione peroxidase (GPX) activities did not show significant changes in response to salt stress. The glutathione reductase (GR), glyoxalase I (Gly I), and glyoxalase II (Gly II) activities decreased upon the imposition of salt stress, especially at 300 mM NaCl, with a concomitant increase in the $H_2O_2$ and lipid peroxidation levels. Exogenous NO pretreatment of the seedlings had little influence on the nonenzymatic and enzymatic components compared to the seedlings of the untreated control. Further investigation revealed that NO pre-treatment had a synergistic effect; that is, the pre-treatment increased the AsA and GSH content and the GSH/GSSG ratio, as well as the activities of MDHAR, DHAR, GR, GST, GPX, Gly I, and Gly II in most of the seedlings subjected to salt stress. These results suggest that the exogenous application of NO rendered the plants more tolerant to salinity-induced oxidative damage by enhancing their antioxidant defense and MG detoxification systems.
Objectives: The goal of this project was to measure the antibacterial effectiveness of Hwangryunhaedok-tang (barberry root decoction for detoxification) and its constituents. All of the active ingredients including the final product were prepared using high-pressure sterilization for use as medicinal eye drops. The varying forms of Hwangryunhaedok-tang (barberry root decoction for detoxification) were used to treat Staphylococcus epidermidis and Enterococcus faecalis which are found in keratitis. Methods: The antibacterial effect was measured by observing the presence or absence of antimicrobial activity when treated with varying concentrations of Hwangryunhaedok-tang extract. The tests were performed using a dosage of $70{\mu}l$ dosages of 100%, 50%, 10% and 1% the extracted solution by the minimum growth inhibitory concentration measurement. Antimicrobial activity was measured by examining the correlation between dosage strength and bacterial activity from $70{\mu}l$ to $10{\mu}l$ at the same concentration. Results: 1. Hwangryunhaedok-tang (barberry root decoction for detoxification), Phellodendri Cortex, and gardenia didn't show any antimicrobial effects against S. aureus, S. epidermidis, or E. faecalis. 2. Barberry root showed antimicrobial effects against S. aureus and S. epidermidis depending on the levels of concentration but didn't show any antimicrobial effects against E. faecalis. 3. Skullcap showed antimicrobial effects against S. aureus and S. epidermidis when a dosage of 100% extract $70{\mu}l$ was used. However, did not show any antimicrobial effects at all against E. faecalis. Conclusions: Hwangryunhaedok-tang (barberry root decoction for detoxification) and its constituents such as barberry root, phellodendri cortex, skullcap, and gardenia, can be used as an alternative to antibiotic medicinal eye drops to treat keratitis. However, further research on effective uses of and efficient extraction methods are needed.
McW의 간 해독효과를 조사하였다. 실험군은 총 4개군 즉, 증류수급여군으로 대조군(DC)과 증류수를 3주간 섭취케 한 후 희생직전에 BB를 처리한 군(DB), McW 급여군으로는 McW 대조군(MC)과 McW를 3주간 섭취케 한 후 희생직전에 BB를 처리한 군(MB)의 4개 군으로 구분하였다. DC군과 MC군의 혈청 ALT 및 AST 활성은 뚜렷한 차이를 보이지 않았으나 MB군에서는 DB군에 비하여 유의하게 낮았다. 간 AH 활성은 모든 실험군에서 유의한 변동이 없었으나 GST 활성은 MC군이 DC군에 비하여, MB군이 DB군에 비하여 각각 높았다. 간 조직의 LPO 함량은 DC 및 MC군이 유사하였으며, DB 및 MB군 모두 DC군보다 유의하게 증가하였으나 그 증가율은 MB군이 DB군에 비하여 낮았다. McW의 전자공여능은 DW에 비하여 유의하게 높았다. 이상의 결과 McW는 간 해독효소의 일종인 GST의 활성을 증가시킴과 동시에 McW의 지속적인 섭취에 의한 항산화 작용에 의해 해독작용을 나타내는 것으로 생각되나 추후 계속적인 연구검토가 행해져야할 것이다.
돼지고기 섭취가 카드뮴에 중독된 쥐의 대사에 어떻게 영향을 주는가를 규명하고자 이 연구를 수행하였다. 카드뮴(0, 25, 50, 100, 250 ppm)을 사료에 섞어 8주 동안 급여하여 일단 중독을 유발시킨 후 또 다시 8주 동안 돼지고기에 의한 해독 효과를 대조구와 비교하였다. 8주 동안 카드뮴 중독용 실험 식이를 급여한 결과, 카드뮴 중독 현상이 나타났다. 해독기간 중 돼지고기를 급여한 경우 체중 회복이 빠른 경향을 보였으나 유의적 차이는 없었다. 해독에 의해 헤모글로빈과 헤마토크릿은 완전히 정상치와 같은 수준으로 향상되었다. 체중 100g당 간의 무게는 250 ppm 카드뮴 중독 후 해독시 돼지고기를 섭취한 처리구에서 가장 높게 나왔다 카제인을 섭취한 처리군보다 돼지고기를 섭취한 처리군에서 카드뮴의 축적이 더 낮게 나타났으며 특히 신장에서의 카드뮴 축적 농도에 영향을 미치는 인자로서 카드뮴뿐만 아니라 카드뮴과 돼지고기 섭취 여부의 교호작용$(Cd^{***},\;Cd^*\;Pork^{**})$등이 나타났다. 해독시 metallothionein(MT) 합성은 돼지고기에 의해 영향을 받지 않는 것으로 보여졌다.

And then after 1 examined all the details on qi and taste of Lacca sinica exsiccata, its efficacy, detoxification, lacquer poison, and its effects on body symptom, 1 got the following results. The order of frequency that Lacca sinica exsiccata is used in Dongeuibogam is pressure-feeling, blood circulation, and insect biting. Its way of intake is not so much through herb-boiling or powdered medicine as through hand-made pills. When medicine is used in the form of pills, the Lacca sinica exsiccata is more included among other ingredients. When old doctors treated pressure-feeling in the chest, they mixed up other herbs, with not putting more emphasis on the efficacy of lacquer 010 doctors believed that toxicity of Lacca sinica exsiccata is not having its own poison, but having biased dominance in the use of its qE and taste. The way or detoxification or Lacca sinica exsiccata is used in the order of crab-boiled water, egg, Xanthoxylum piperitum, Perilla frutescens, Astar tataricus, a weeping willow, iron-tempered water, and Allium toberosum. Special point in detoxificating lacquer poison is that they used medicines for well-ciruculating pulmonary stream, medicines for promoting to urinate or discharge by helping large colons to move, medicines for making the lacquer scar small, medicines for helping digest, and medicines for improving vessel function in the poisoned area. With the above results, the more profound study, based on the crab-boiled water and egg, is expected to go on to increase the effect on the one hand, and to make the new way of lessening or removing the toxicity of lacquer with more safe use on the other hand.
생체에 투입되는 이물질(xenobiotics)들이 돌연변이를 일으키고 나아가서는 암을 유발시키게 되는 것은 이물질들이 생체내에서 어떤 양상으로 대사되느냐, 즉 이물질이 독성화되는 과정과 해독되는 과정에 있어서의 활성도의 차이에 따라 좌우되는 것으로 알려져 있다. 따라서 인삼이 이물질 대사 및 해독작용에 어떤영향을 미치는가를 밝혀보기 위하여 동물 실험을 통하여 발암물질로 알려진 Benzopyrene과 그 외 다른 유독한 화학약물들의 대사과정에 미치는 영향을 조사하여 보았다. 인삼투여군에 있어서 cyt. P-450와 관련된 Monooxy-genase system이 선택적으로 유도되었고 또 해독작용에 필수적인 Conjugation도 현저히 상승되었다. 이와 같은 이물질 해독작용의 선택적 유도는 인삼이 이물질 대사과정에 영향을 미쳐 독성물질 해독 항진효능이 있음을 제시해 주는 것으로 해석되어진다.
Background: Previous studies have shown the insecticidal efficacy of ginsenosides. In the present study, we aimed to investigate the metabolic mechanism related to the inhibitory effect of panaxadiol saponins (PDSs) against the Asian corn borer Ostrinia furnacalis (Guenee). Methods: Third instar larvae of O. furnacalis were fed normal diets with different concentrations of PDSs for 4 days. The consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food were recorded. A targeted gas chromatographye-mass spectrometry assay was performed to detect the profiles of amino acids, fatty acids, and carbohydrates in larvae of O. furnacalis. In addition, the activity of detoxification-related enzymes was determined. Results and Conclusions: PDSs decreased the consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food in the 3rd instar larvae of O. furnacalis in a dose-dependent manner. PDSs decreased 15 free amino acids, 16 free fatty acids, and 5 carbohydrates and increased the levels of palmitoleic acid, palmitic acid, and 9-octadecenoic acid in the 3rd instar larvae. The activity of detoxification-related enzymes, such as acetylcholinesterase, glutathione S-transferase, cytochrome P450, carboxylesterase, trehalase, acid phosphatase, and alkaline phosphatase, was reduced in a dose-dependent manner in the 3rd instar larvae exposed to PDSs. These data confirmed the inhibitory effect of PDSs against growth, food utilization, and detoxification in the 3rd instar larvae of O. furnacalis and the potential for using PDSs as an efficient tool for insect pest management for O. furnacalis larvae.
