Variability in Ash, Crude Protein, Detergent Fiber and Mineral Content of Some Minor Plant Species Collected From Pastures Grazed by Goats

  • This study was conducted to determine the protein content, cell wall fractions, and mineral concentrations of some minor plant species collected for one year in pastures grazed by goats in the Philippines. An assessment of nutrient variability and a comparison of forage protein and mineral concentrations to the critical value of protein and minerals based on animal needs were also studied. The plant species were the following: grasses(Axonopus compressus, Eleusine indica, Rottboellia exaltata); legumes (Aeschynomene indica, Calopogonium muconoides, Desmodium tortousum); and herbs (Corchorus olitorius, Ipomea aquatica, Sida acuta, Synedrella nodiflora). The two seasons (dry and wet) were subdivided into Dry-1 (December to February, 132 mm total rainfall), Dry-2 (March to May, 25 mm total rainfall), Wet-1 (June to August, 1,138 mm total rainfall), and Wet-2 (September to November, 1,118 mm total rainfall). Results showed that significant differences were obtained on various nutrient fractions including those mineral concentrations across species. Across season, acid detergent lignin (ADL) had higher (p < 0.05) value at Dry-1. Legumes and herbs were higher in crude protein (CP) especially Sida acuta. Grasses showed the highest neutral detergent fiber (NDF) and acid detergent fiber (ADF) with the addition of Sida nodiflora (herb) for it contained high NDF. Aeschynomene indica contained the highest amount of ADL and the herbs (Ipomea aquatica and Sida acuta) had exceptionally high concentration of minerals. Coefficient variation of the various nutrient values ranged from 27.3 to 136.7%. Some forage minerals appeared to be deficient (sodium, phosphorus and copper) or excess (molybdenum) for the whole or part of the year. This study shows that some minor plant species could extend the range of concentration of some nutrients (i.e., CP and minerals) beyond that normally found in conventional pasture species.

Acute Toxicity of Synthetic Detergent on the Larvae of Loach, Misgurnus angillicaudatus (미꾸리 자어에 대한 합성세제의 급성독성)

  • Short-term acute toxicity of synthetic detergent(LAS) to larvae of loach, Misgurnus angillicaudatus was examined by static bioassay. The larvae were exposed to 15 different concentration of synthetic detergent for 16, 48, 72, 96 and 120 hours in order to determine median lethal concentration($LC_{50}$). The $100\%$ mortarlity of larvae was showed within 120, 96, 48 and 16 hours for 6, 18, 30 and 38 ppm, respectively. The median lethal concentration values of the larvae were 12.59 ppm for 48 hours, 4.00 ppm for 96 hours and 1.02 ppm for 120 hours. The permissible toxicant concentration of acute toxicity to larvae was $0.37{\sim}0.43$ ppm, and application factor of the synthetec detergent was $0.093{\sim}0.108$. The median lethal time($LT_{50}$) for different concentration also was determined. The $LT_{50}$ of 0.2 ppm was found within 165.1 hours and 2 ppm was 106.2 hours, while the $LT_{50}$ of 8 ppm was 60.3 hours and that of 38 ppm was 23.5 hours.

AMOUNT AND DISTRIBUTION OF DIETARY MINERALS IN SELECTED PHILIPPINE FORAGES

  • Eight Philoppine forages were studied to obtain the following: 1) nutrient concentrations and digestibility, 2) distribution of the various minerals in fiber fractions through mineral analyses of neutral detergent fiber(NDF) and acid detergent fiber(ADF) residues, and 3) correlation coefficients among the factors affecting forage quality and mineral concentrations. These Philippine forages were paragrass [Brachiaria mutica (Forsk.) Stapf], stargrass (Cynodon plectostachyum Pilger), napiergrass (Pennisetum purpureum Schumach.) calopo (Calopogonium muconoides Desv.), centrocema (Centrocema pubescens Benth.), gliricidia [Gliricidia sepium (Jacq.) Walp.] leucaena [Leucaena leucocephala (Lam.) de Wit.] and sesbania [Sesbania grandiflora (L.) Poir]. Species differences(p<0.01) were observed on various nutrient fractions including mineral composition and digestibility. The cell wall(NDF) fraction, prepared by boiling in neutral detergent solution, contained the following proportions of the total mineral originally present (%): calcium (Ca), 0.7; phosphorus(P), 14.3; magnesium(Mg), 1.9; potassium(K), 3.7; copper(Cu), 16.4; zinc(Zn), 2.9; molybdenum(Mo), 9.3; cobalt(Co), 16.2; manganese(Mn), 5.6, and iron(Fe), 81.3. The ligno-cellulose(ADF) fraction, prepared by boiling in acid detergent solution, contained the following proportions of the total mineral originally present(%): Ca, 0.2; P, 4.4; Mg, 0.7; K, 2.8; Cu, 32.3; Zn, 1.1; Mo, 8.9; Co, 4.7; Mn, 5.4; and Fe, 36.8. Correlation coefficients among the factors affection forage quality and mineral concentrations were also observed. Evidently, 75 and 45% of the minerals in grasses and legumes was positively correlated to CP and IVDMD, respectively. Moreover, 55, 80 and 75% of the forage minerals was negatively correlated to NDF, ADF and ADL fraction, respetively, implying that most of the minerals reside in the non-structural cell components.

The Apparent Digestibility of Corn By-products for Growing-finishing Pigs In vivo and In vitro

  • Two trials in vivo and in vitro were conducted, in vivo to determine the apparent digestibility of gross energy, crude protein, dry matter, acid detergent fiber, neutral detergent fiber and apparent digestible energy in 10 corn by-products. In vivo the diets included one basal corn diet, four corn gluten meal diets, four corn distillers dried grains with solubles diets and two corn distillers dried grains diets using the different methods, 12 crossbred barrows weigh $40{\pm}$1.6 kg were allocated into individual metabolic crate, according to a $6{\times}6$ Latin square design. In vitro using flask technique, filter bag technique and dialysis tubing technique, the digestibilities of gross energy, crude protein and dry matter in corn gluten meal and corn distillers dried grains with solubles were investigated. Pepsin, pancreatin, intestinal fluid, rumen fluid and cellulase were used in incubation. The results showed that correlation coefficient was 0.73 in corn distillers dried grains with solubles between the digestibility of crude protein and acid detergent fiber in vivo (p<0.01); and correlation coefficient was 0.68 in corn distillers dried grains with solubles between the digestibility of gross energy and neutral detergent fiber in vivo (p<0.01). Apparent digestible energy (DE) of corn by-products in pig total tract was predicted by the percentage of crude protein (CP) and the content of gross energy (GE) in feedstuff. The equation: DE=5,601.09+26.69$\times$CP %-0.5904$\times$GE, ($R^2=0.72$). In vitro, filter bag technique was more convenient; furthermore, the digestibility for the treatments (pepsin+pancreatin+rumen fluid and pepsin+pancreatin+cellulase) was better.

An Efficient Method for the Release of Recombinant Penicillin G Amidase from the Escherichia coli Periplasm (대장균의 periplasm으로부터 재조합 PGA 단백질의 효율적이고 간단한 방출 방법)

  • In this study, we report on a simple, efficient method for obtaining penicillin G amidase (PGA) from recombinant Escherichia coli using a formulation mixed with detergent and lysozyme. Research was conducted on the extraction efficiency of PGA from the periplasmic space in cells in terms of the type of detergent, detergent concentration, pH, reaction time, and temperature of permeabilization. The extraction yield of PGA in the formulated surfactant/lysozyme treatment was increased by approximately (55-65 U/ml) in comparison with that in the single surfactant treatment. The released PGA solution was concentrated and exchanged with buffer using an ultrafiltration (U/F) system. The yields of diatomite filtration, membrane filtration (M/F), and U/F were 69.7%, 93.8%, and 77.3%, respectively. A total of 212 KU of PGA was recovered. At the 25-L culture scale, the overall yield of extraction using the mixed surfactant/lysozyme method was 49.2%. The specific activity of extracted PGA was 11 U/mg in protein. The concentrated PGA solution was immobilized on microporous silica beads without further purification of PGA. The total immobilization yield of PGA on the resin was 48.7%, while the enzyme activity was 101 U/g. The immobilized PGA was successfully used to produce 6-APA from penicillin G. Our results indicated that a simple extraction method from periplasmic space in E. coli may be used for the commercial scale production of ${\beta}-lactam$ antibiotics using immobilized PGA.

The Optimum Levels of Alkaline Hydrogen Peroxide Treatment of Rice Straw for Feed (볏짚 사료가치 증진을 위한 알카리성 과산화수소의 적정 처리수준)

  • These studies were conducted to investigate the chemical composition changes in in vitro digestibility for the improvement of nutritive value of rice straw by alkaline hydrogen peroxide. The content of neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose, cellulose and lignin in rice straw was decreased with higher level of $H_2O_2\;(pH 11.5)$. The content of ADF, cellulose and ash of the rice straw washed after $H_2O_2\;(pH 11.5)$ treatment tended to be increased but NDF, hemicellulose and lignin were decreased with higher concentration of $H_2O_2\;(pH 11.5)$. In the rice straw washed after alkaline hydrogen peroxide treatment the decomposition of cellulose and lignin was effective in $pH\;11.5{\sim}12.5$, in smaller cutting size and $55^{\circ}C$. The in vitro organic matter digestibility was increased with higher $H_2O_2$ concentration and smaller cutting size of rice straw.

Effects of Application Times and Dilution of Cattle Slurry with Water on Dry Mattter Yield and Feed Values of Orchardgrass(Dactylis glomerata L.) (액상우분뇨의 시용시기와 희석이 오차드그라스의 건물수량과 사료가치에 미치는 영향)

  • The aim of this experiment was to investigate the effects of applying times and dilution rates of cattle slurry with water on dry matter yields and feed values of orchard grass. Cattle slurry was applied at the rates of average nitrogen fertilizer equivalent to 150kg/ha/year in 3 cutting frequency. Significantly higher dry matter yields than that of no fertilization (7.36 ton DM/ha) were recorded in the application of diluted cattle slurry (10.74~13.54 ton DM/ha) (p<0.05). Especially, this tendency was shown with higher annual dry matter yields at the partitioned dressing times, such as at the applications for 1st and 3rd growth, 2nd and 3rd growth, and 1st, 2nd and 3rd growth respectively. The yields of annual dry matter at fertilizing phosphorus, potassium and nitrogen were higher than no fertilization as 12.42 tons per ha. However, with application of diluted cattle slurry, dry matter yields per year (11.96 ton DM/ha) were significantly higher than those of fertilizing phosphorus and potassium (7.52 ton DM/ha) (p<0.05). The efficiencies of dry matter production with mineral and cattle slurry nitrogen application (kg DM/kg N) were 32.7 and 13.4~26.9kg DM/kg N respectively. Especially, these tendencies were higher in diluted application plots than in no-diluted application plots of cattle slurry. The contents of crude protein (CP, %), neutral detergent Eber (NDF, %), acid detergent fiber (ADF, %), annual yields of CP (kg/ha) and total digestible nutrient (TDNY, kg/ha) of orchardgrass were significantly higher at the application of diluted cattle Slurry than those at non-fertilizer application (p<0.05).

Nutrient composition and in vitro fermentability of corn grain and stover harvested at different periods in Goesan, a mountainous area

  • With South Korea's limited capability of feed production because of its relatively small cultivable area, the country is pushed to depend on foreign feed imports despite the immensely fluctuating price of corn. Hence, intensive efforts to increase the total cultivable area in Korea like extending of farming to mountainous area is being practiced. Corn was planted in Goesan County, a mountainous area in the country. Grain and stover were harvested separately in three harvest periods: early-harvest (Aug 8), mid-harvest (Aug 18), and late-harvest (Aug 28). The nutrient composition such as dry matter (DM), crude protein (CP), crude fat (EE), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), and non-fibrous carbohydrates (NFC) was determined after harvest. Effective degradability (ED) of the major nutrients (DM, NDF, ADF, and CP) were measured through in vitro fermentation of rumen fluid from Hanwoo (Korean cattle). pH, ammonia-N concentration, volatile fatty acid (VFA) concentration, and gas production were periodically measured at 0, 3, 6, 12, 24, 48, and 72 h. Corn grain showed higher nutrient content and ED than stover. It also had higher gas production but its pH, ammonia-N, and total VFA concentration were lower than corn stover. The best nutrient composition of corn grain was observed in early-harvest (high CP, EE, NDF, OM, NFC, and low ADF). Early-harvest of corn grain also had high effective degradability of dry matter (EDDM), effective degradability of neutral detergent fiber (EDNDF), effective degradability of acid detergent fiber (EDADF), and total VFA concentration. On the other hand, the best nutrient composition of stover was observed in mid-harvest (high DM, CP, NDF, and low ADF). EDDM, EDNDF, and EDADF were pronounced in early-harvest and mid-harvest of stover but the latter showed high total VFA concentration. Hence, early and mid-harvested corn stover and grain in a mountainous area preserved their nutrients, which led to the effective degradation of major nutrients and high VFA production.

Synergistic antibacterial effect of disinfectants and microbubble water to Salmonella Typhimurium

  • Salmonella is a pathogenic bacterium that has long been important industrially because it has a wide host range and can be transmitted to humans through direct contact as well as indirect contact such as food contaminated with animal waste. Understanding how to reduce Salmonella contamination in pig farms is important for public health and the livestock industry from an economic perspective. In the swine industry, high concentrations of disinfectants have been applied because it is difficult to effectively control Salmonella in environments contaminated with organic substances. In order to evaluate the synergetic effect of disinfectants, the efficacy of two commercial disinfectants diluted in hard water and microbubble water (MBW) were compared under the laboratory condition. Different concentrations of both disinfectants combined with 1% detergent diluted in the two diluents were evaluated for their antibacterial effect. In the case of monopersulfate-based disinfectant groups, the growth of Salmonella was not observed at 1:200 dilution with both the hard water and MBW combined with 1% detergent. In the case of citric acid-based disinfectant, the bacterial growth was not observed at 1:800 dilution with MBW combined with 1% detergent. Our results show that the use of MBW as a diluent might improve the biological activities of acid-based disinfectant.

Predicting standardized ileal digestibility of lysine in full-fat soybeans using chemical composition and physical characteristics

  • Objective: The present work was conducted to evaluate suitable variables and develop prediction equations using chemical composition and physical characteristics for estimating standardized ileal digestibility (SID) of lysine (Lys) in full-fat soybeans (FFSB). Methods: The chemical composition and physical characteristics were determined including trypsin inhibitor activity (TIA), urease activity (UA), protein solubility in 0.2% potassium hydroxide (KOH), protein dispersibility index (PDI), lysine to crude protein ratio (Lys:CP), reactive Lys:CP ratio, neutral detergent fiber, neutral detergent insoluble nitrogen (NDIN), acid detergent insoluble nitrogen (ADIN), acid detergent fiber, L* (lightness), and a* (redness). Pearson's correlation (r) was computed, and the relationship between variables was determined by linear or quadratic regression. Stepwise multiple regression was performed to develop prediction equations for SID of Lys. Results: Negative correlations (p<0.01) between SID of Lys and protein quality indicators were observed for TIA (r = -0.80), PDI (r = -0.80), and UA (r = -0.76). The SID of Lys also showed a quadratic response (p<0.01) to UA, NDIN, TIA, L*, KOH, a* and Lys:CP. The best-fit model for predicting SID of Lys in FFSB included TIA, UA, NDIN, and ADIN, resulting in the highest coefficient of determination (R2 = 0.94). Conclusion: Quadratic regression with one variable indicated the high accuracy for UA, NDIN, TIA, and PDI. The multiple linear regression including TIA, UA, NDIN, and ADIN is an alternative model used to predict SID of Lys in FFSB to improve the accuracy. Therefore, multiple indicators are warranted to assess either insufficient or excessive heat treatment accurately, which can be employed by the feed industry as measures for quality control purposes to predict SID of Lys in FFSB.
