Radish roots were salted in 4 different salt concentrations (5, 10, 15, 20%) for 3 months followed by desalting. The effects of desalting process on salt concentration, Ca content, texture and sensory characteristics were investigated during 3 days of desalting period at $25^{\circ}C$. NaCl content in danmuji salted in lower NaCl concentrations decreased rapidly and a linear relationship was found between the decrease in NaCl content and the logarithmic value of desalting time. Changes in Ca content of danmuji also exhibited a similar trend to that of NaCl content. Hardness of radish root tended to increase wit an increase in desalting time up to 2 days of desalting and then decreased slightly at the 3rd day of desalting. The results of sensory evaluation revealed that regardless of salt concentration used acidic, salty odor and taste of danmuji decreased significantly during desalting, however, there was very little change in yeast-moldy odor and taste with the intensity being very low in danmuji salted in higher salt concentrations. Among the textural properties, the crispness of danmuji salted in 5, 10 and 20% NaCl concentrations increased up to 2 days of desalting and decreased slightly thereafter, whereas that of radish salted in 15% NaCl concentration increase continously during 3 days of desalting. Total acceptibility was best in danmuji salted in 20% NaCl concentration and desalted for 3 days.
Outdoor iron artefacts are easily corroded by affection of environmental pollutants directly. Especially they need to be removed $Cl^-$ ions, but outdoor iron artefacts are excluded for desalination owing to their special features. Therefore this study contains the conservation processes of an iron stele of the Joseon Dynasty as the sample as well as desalination experiments that were carried out the desalting method using spill pads and the immersion desalting method together in order to compare. Desalting methods were compared by analyses such as an optical and metallurgical microscope, SEM-EDS, pH meter, Ion Chromatography and X-ray diffraction. As a result of the analysis, the optical and metallurgical microscopy show that the corrosion products are constituted by the layers and the metallurgical microstructure is a white cast iron. The SEM-EDS results of corrosion products detected mainly Fe, O, and especially Cl upto 2.48wt%. The results of pH and anion analysis for the washing solution, the desalting method using spill pads shows the similar effect to the immersion desalting method. As a result of XRD analysis before and after desalting corrosion products, goethite, magnetite, lepidocrocite and akaganeite are detected before desalting, but akaganeite is not detected after desalting at the desalting methode using spill pads, which indicates to have an effect on desalination. Therefore the results show that the desalting method using spill pads has an effect on desalination similar to the immersing desalting method.
Excavated archaeological cast iron objects in improper storage are quickly corroded and disintegrated into block and powder finally. Hence desalination treatment which is a way of removing internal corrosive factors, especially chloride ion, is an important process. But desalination is often omitted or objects are dehydrated by alcohol because the destruction of objects could occur during desalting. Although current desalting methods mostly use an aqueous alkali solution, $OH^-$ ions of water could accelerate corrosion and broaden internal cracks cause of high surface tension. Therefore this study experimented desalting using ethyl alcohol, which is low surface tension, to investigate an effect of desalination. As a result, desalting using ethyl alcohol showed the similar or more effective results of desalting using water. In addition, as aspects of desalting safety, ethyl alcohol desalting method was smaller destruction of objects and extraction of Fe from the objects than the aqueous alkali solution. However, this study explored the possibility of desalting methods using organic solvent in fieldwork, so the results would provide basic date for making the safe and effective desalting method for archaeological cast iron objects through further experiments.
Desalting methods for chloride ions from the excavated iron objects were studied. These methods were compared with room temperature and heating condition of treating solution. During the Desalting treatments the $Cl^-$concentration of the solution were measured regularly and the plotted for $Cl^-$concentrations the square root of time, heating methods showed that the $Cl^-$ ions are more extracted. For the desalting treatments these plots were found to be flattening line, it was showed the changing second solution. Also, the changing solution in the room temperation detected less $Cl^-$ ions of heating treatment solution. As the examination for the relative humidity has compared for desalting objects during 8 days, it was showed a re-corroded appearance of R.H90%. As it did not detected a $Cl^-$ ions of re-corroded section, these phenomena were showed the naturally occuring corrosion of the objects in the high relative humidity.

The diffusion phenomena of water, solid and reducing sugar in Chinese cabbage during salting (5$0^{\circ}C$, 25% salt solution) and desalting (5$0^{\circ}C$, distilled water) were investigated. Water loss and solid gain during salting were rapid in the first 6hrs and then almost leveled off. After 24hrs of salting, water loss and solid gain in 100g of initial wet Chinese cabbage were 33.35g and 6.26g respectively. Moisture content was changed from 94.29% to 83.11% during 24hrs of salting. The reducing sugar concentration was also changed from 29.2 mg/$m\ell$ to 6.5mg/$m\ell$, which was linearized as a function of the square root of salting time and showing that Y=30.1841-5.0269√t. After 24hrs salting, water gain and solid loss during desalting were rapid in the first 4hrs and then increased linearly. After 12hrs of desalting, the water gain and solid loss in 100g of initial wet Chinese cabbage were 20.82g and 9.14g respectively. The amount of solid loss after 12hrs desalting was higher than that of solid gain after 24hrs salting due to the diffusion of solute presented initially in the Chinese cabbage during salting and desalting. The concentration of salt in Chinese cabbage after 12hrs desalting was 2.98% which was a suitable salt concentration for the preparation of Kimchi. At this time, the concentration of reducing sugar was only 1.6mg/$m\ell$. The linear regression equation of reducing sugar concentration during desalting was Y=6.7854-1.5992√t.
The performance of an electrodialyzer for concentrating seawater is predicted by means of a computer simulation, which includes the following five steps; Step 1 mass transport; Step 2 current density distribution; Step 3 cell voltage; Step 4 NaCl concentration in a concentrated solution and energy consumption; Step 5 limiting current density. The program is developed on the basis of the following assumption; (1) Solution leakage and electric current leakage in an electrodialyzer are negligible. (2) Direct current electric resistance of a membrane includes the electric resistance of a boundary layer formed on the desalting surface of the membrane due to concentration polarization. (3) Frequency distribution of solution velocity ratio in desalting cells is equated by the normal distribution. (4) Current density i at x distant from the inlets of desalting cells is approximated by the quadratic equation. (5) Voltage difference between the electrodes at the entrance of desalting cells is equal to the value at the exits. (6) Limiting current density of an electrodialyzer is defined as average current density applied to an electrodialyzer when current density reaches the limit of an ion exchange membrane at the outlet of a desalting cell in which linear velocity and electrolyte concentration are the least. (7) Concentrated solutions are extracted from concentrating cells to the outside of the process. The validity of the computer simulation model is demonstrated by comparing the computed results with the performance of electrodialyzers operating in salt-manufacturing plants. The model makes it possible to discuss optimum specifications and operating conditions of a practical-scale electrodialyzer.
Benzotriazole (B.T.A) which has been mainly used for the stabilization processing method of excavated copper and bronze artifacts is vaporized within 2~3 years after the usage because it is unstable at the acid conditions and cannot protect the surface of artifacts. In this study, NaOH method which has been used for the steel artifacts was applied as a stabilization process for the method of copper and bronze artifacts to gush chlorine ion out. For the reproduction of excavated samples, copper and bronze plates were dipped in 0.1M HCl for 26 hrs to form CuCl, rusted at $70^{\circ}C$ with RH 75% for the formation of corrosion products, and desalted in 0.1 M NaOH solution. The concentration of chlorine ion was measured by using ionchromatography. During the desalting process, a large quantity of chlorine ions was gushed out in early period and corrosion products were not additionally generated through the re-corrosion experiment. This NaOH desalting process was found to be a method of stabilization process for copper and bronze artifacts from the formation of Tenorite (CuO) during desalting as a protection layer for corrosion.
Papermaking reconstituted tobacco is an important strategy for recycling the waste tobacco residues. To indentify the influences of the inorganic components on harmful components delivery in cigarette smoke, a self-made electrodialysis stack was assembled to desalt the tobacco extract. The influences of the applied current and extract content on the removal rate of the inorganic ions were investigated. Results indicated that the applied current was a dominant impact on the desalination performance. High currents lower than the limiting current density could accelerate the desalting efficiency but cause higher energy consumption. A current of 2 A, or current density of ${\sim}11mA{\cdot}cm^{-2}$, was an optimal choice by considering both the energy consumption and desalting efficiency. A 20% tobacco extract was an appropriate content for the electrodialysis process. More than 90% of inorganic ions could be removed under the optimum condition. The preliminary result indicated that removal of inorganic components was beneficial to decrease the harmful component delivery in cigarette smoke. Naturally, ED is an environmentally friendly and high-effective technology for desalting the tobacco extract.
Proteomics for biomarker validation needs high throughput instrumentation to analyze huge set of clinical samples for quantitative and reproducible analysis at a minimum time without manual experimental errors. Sample preparation, a vital step in proteomics plays a major role in identification and quantification of proteins from biological samples. Tryptic digestion a major check point in sample preparation for mass spectrometry based proteomics needs to be more accurate with rapid processing time. The present study focuses on establishing a high throughput automated online system for proteolytic digestion and desalting of proteins from biological samples quantitatively and qualitatively in a reproducible manner. The present study compares online protein digestion and desalting of BSA with conventional off-line (in-solution) method and validated for real time sample for reproducibility. Proteins were identified using SEQUEST data base search engine and the data were quantified using IDEALQ software. The present study shows that the online system capable of handling high throughput samples in 96 well formats carries out protein digestion and peptide desalting efficiently in a reproducible and quantitative manner. Label free quantification showed clear increase of peptide quantities with increase in concentration with much linearity compared to off line method. Hence we would like to suggest that inclusion of this online system in proteomic pipeline will be effective in quantification of proteins in comparative proteomics were the quantification is really very crucial.
The most ideal method to measure the water-soluble $Cl^-$ ion eluted from iron artifacts is conducting the analysis on desalting solution by Ion Chromatography. But most institutes related to cultural heritages use Cl meter by reason of lack of budget and experts. This study evaluated reliability and stability between Cl meter and Ion Chromatography by doing cross-validation with results from two methods to detect $Cl^-$ ion of desalting solution. From D.I water, extremely small quantities of $Cl^-$ ion was detected by the influence of remaining water-soluble $Cl^-$ ion at the electrode of Cl meter and water-soluble $Cl^-$ which remains in Sodium sesquicarbonate, components of reagent was detected as well. The first desalting solution had the most $Cl^-$ ions, $Cl^-$ ion slightly decreased from the second to the fourth desalting solution and tend to decrease again at the stage of dealkalified in D.I water. Each Cl meter has the standard deviation according to the measured numbers and the higher concentration of $Cl^-$ ion the desalting solution has, the wider the deviation is. But when the concentration of $Cl^-$ ion is low, it was stable to use Cl meter to detect the concentration of $Cl^-$ ion from iron artifacts because there is the small deviation, It is thought that conductivity meter method is not suitable for measuring $Cl^-$ ion, because the electrical conductivity of alkaline solution is too high to measure $Cl^-$ ion.
