생쥐의 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-유도 신경독성에 대한 대황의 보호효과 (Protective Effect of R. palmatum on 1-Methyl-4-phenyl-l,2,3,6-tetrahydropyridine (MPTP)-induced Neurotoxicity in Mice)

  • The protective efficacy of Rheum palmatum water extract on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism was studied in C57BL/6 mice. In order to demonstrate neuroprotective effect of R. palmatum extract, animals were administered intraperitoneally with the water extract (100 or 200 mg/kg/day) for 14 days, and MPTP (10 mg/kg/day) was injected subcutaneously into the mice for the first 6 consecutive days from the beginning 1 hr before R. palmatum extract treatment. All animals were measured the several neurobiochemical markers such as dopamine level and monoamine oxidase B (MAO-B) activity in various regions of brain. The treatment of mice with R. palmatum extract was confirmed recovery effect on MAO-B activity in the cerebellum and the cerebral cortex. R. palmatum extract was attenuated the MPTP-induced depletion of substantia nigra dopamine. The contents of MDA, a marker of lipid peroxidation, in brain tissues (cerebellum and cerebral cortex mitochondria) were decreased significantly by R. palmatum extract. These results suggest that R. palmatum water extract plays an effective role in attenuating MPTP-induced neurotoxicity in mice. This protective effect of R. palmatum might be estimated the result from the inhibitory activity on monoamine oxidase B and the enhancement of antioxidant activity.

A study AGN activity on environmental dependence in the SDSS late-type galaxies

  • We explore the role of various environments in triggering star formation (SF) and narrow-line active galactic nucleus (AGN) in SDSS spiral galaxies and the SF-AGN connection, using a volume-limited sample with $M_r$ < -19.5 and 0.02 < z < 0.055 selected from the SDSS Release 7. To avoid the dependency of AGN activity on bulge mass, the central velocity dispersion of the sample galaxies is limited to have a narrow range of $130{\leq}{\sigma}{\leq}200km\;s^{-1}$. We note that in gas sufficient galaxies, AGN feeding lags behind starburst, whereas as the gas exhausts, the SF slows down and AGN seems to even prevent the SF, and thus divide the high-${\sigma}$ sample into two subsamples according to their cold gas content at central region traced by fiber star formation rate, $SFR_{fib}$. We find that a high density (cluster) environment causes a significant increase in AGN activity as well as gas depletion in host galaxies. However, the finding is only noticeable in the high-${\sigma}$ and low $SFR_{fib}$ sample. It seems that a galaxy interaction with the nearest neighbor directly affects the SF of the central region. However, it is unclear whether it directly affects AGN activity.

Antiestrogenic Effects of Marijuana Smoke Condensate and Cannabinoid Compounds

  • The antiestrogenic effects of marijuana smoke condensate (MSC) and three major cannabinoids, i.e., $\bigtriangleup^{9}$-tetrahydrocannabinol (THC), cannabidiol (CBD), and cannabinol (CBN), were evaluated using in vitro bioassays, viz., the human breast cancer cell proliferation assay, the recombinant human estrogen receptor (ER) competitive binding assay, and the reporter gene assay. The inhibitory effects on estrogen were also examined using the ethoxyresorufin-O­deethylase (EROD) assay, the aromatase assay, and the 17$\beta$-estradiol ($E_{2}$) metabolism assay. The results showed that MSC induced the antiestrogenic effect via the ER-mediated pathway, while THC, CBD, and CBN did not have any antiestrogenic activity. This suggests that the combined effects of the marijuana smoke components are responsible for the antiestrogenicity of marijuana use. In addition, MSC induced the CYP1A activity and the $E_{2}$ metabolism, but inhibited the aromatase activity, suggesting that the antiestrogenic activity of MSC is also related to the indirect ER-dependent pathway, as a result of the depletion of the in situ $E_{2}$ level available to bind to the ER. In conclusion, pyrogenic products including polycyclic aromatic hydrocarbons (PAHs) in the non-polar fraction, which is the most biologically active fraction among the seven fractions of MSC, might be responsible for the antiestrogenic effect.

Shifts in Protein Metabolism in Hemolymph and Fat Body of the Silkworm, Bombyx mori L. in Response to Fluoride Toxicity

  • Changes in protein metabolism were studied in hemolymph and fat body on days 1, 3, 5 and 7 of the fifth-instar silkworm, Bombyx mori, exposed to lethal, sublethal doses and prevailing levels of fluoride in groundwater in Karnataka and Andhra Pradesh States of India. The total protein content indicated a depletion followed by a concomitant increase in accumulation of free amino acids. Concurrently, the activity of protease in both of the tissues was also increased. A steady enhancement in the activities of alanine aminotransferase and aspartate aminotransferase paralleled the elevation of glutamate dehydrogenase activity in the tissues studied. It is presumed, on the basis of these results, that the fluoride toxicity causes major changes in protein metabolism of the silkworms.

낙동강 유역의 지속적인 이용을 위한 미래예측 (Simulating the Future of Nakdong River Basin for the Sustainable Use)

  • A macroeconomic minimodel was simulatedto suggest the public for sustainable us of Nakdong River Basin. The minimodel for the simulation shows the interrelationships between natural environment and economic activity. Topsoil, water, economic assets, and money stock are plotted for 300 years, beginning with 1996 in each simulation. The computer simulation runs suggest that the Nakdong River Basin system in the near future may strongly be influenced by the favorable availability of outside resources, while the economic assets and money stock may be declined by indigenous environmental stock depletion. The simulation run made under the constant decrease in systems purchased inputs with investment ratio of developed country and for sustainable use. The results of simulation shows the recover of natural environment and decrease of economic activity under these condition. Therefore, the economic structure of Nakdong River Basin should be transformed from the present industrial structure to the social-economic structure based on an ecological-recycling concept which depend on renewable resources rather than industrial structural which depend on outside resources.

  • To examine whether DGRF inhibits $cPLA_2$ activity in vitro, we purified a 100 kDa $cPLA_2$enzyme from porcine spleen and performed an inhibition study at two concentrations of 5.0 and 50.0 $\mu$M 1-stearoyl-2-[1-$^{l4C}$ ]arachidonoyl-sn -glycero-3-phosphocholine as a substrate to rule out an apparent inhibition due to "substrate depletion". Here we reported that DGRF inhibited $cPLA_2$activity with $ID_{50}$ of 3.2 $\mu$M and virtually complete inactivation of the enzyme occurred at 60 $\mu$M. Interaction experiment between enzyme protein and inhibitor by ultrafiltration method indicated that 1-desgalloylrugosin-F inactivates $cPLA_2$enzyme by an irreversible mechanism.

  • The alkaline phosphatase (K-ALPase) gene of Kluyveromyces fragilis has been cloned (1) and determined its base sequences (2) previously in our laboratory. When the K-ALPase gene was expressed in Escherichia coli and Saccharomyces cerevisiae, it showed a constitutive activity in E. coli, and a derepressed activity in S. cerevisiae in phosphate-limited medium. Northem hybridization experiment was performed to elucidate the transcription level of the K-ALPase gene. Northern experiment showed that transcription level of K-ALPase gene in S. cerevisiae was higher in phosphate depletion, but it was higher in high phosphate medium than in phosphate limited medium in K. fragilis. The transcription initiation site of the K-ALPase gene was determined by primer extension analysis. It matched nucleotide position - 169 in relation to the putative trnslational start site.

  • Yeast cells respond to condition of amino acid starvation by synthesizing GCN4, a typical eukaryotic transcriptional activator, which regulates the expression of many amino acids biosynthetic genes. By introducing point mutations in the DNA binding domain of GCN4, mutants with normal DNA binding activity but defective in transcriptional activity were isolated to identify unknown proteins that could suppress the mutant phenotype under an amino acid depletion condition. As a result, SSB(Stress-Seventy B) subfamily proteins were identified as suppressors of mutant GCN4. SSB proteins were known as a member of yeast hsp70 family that probably aids passage of nascent chain through ribosomes. Among them, the mechanism of suppression by SSB2 on the defective GCN4 mutant strains is under investigation. Gcn4p directly interacts with Ssb2p through the basic DNA binding domain of GCN4. It suggests the possibility that physical interaction might induce the transcriptional activation of Gcn4p.

Effect of Ginseng Components on the Potassium Depleted Cardiomyopathic Rats and it's Mechanism of Action

  • The effect of ginseng components on the potassium depleted cardiomyopathic rat heart was investigated. In the perfused heart experiment using Langendorff apparatus, the deterioration rate of contriactile force of potassium depleted rat heart (low potassium diet group) was faster than that of normal rat heart and ginseng components showed the ability to slow the deterioration rate of potassium depleted hearts. Both sialic acid contents in carcolemmal ghost and sialyltransferase activity of 40,000 * g subcellular fraction prepared from cardiac ventricular tissue of low potassium diet group were significantly decreased compared to those of normal group. The decrease of the sialic acid content and sialyltransferase activity in sarcolemma of low potassium diet group was inhibited when ginseng was concomitantly administered. Calcium uptake of sarcoplasmic reticulum prepared from low potassium diet group was significantly greaterthan that of normal group. Ginseng extract or total saponin showed the tendency to inhibit the increase of cacium uptake.

Statistical studies of trough at middle latitudes observed by DMSP F15

  • The middle-latitude ionization trough, the plasma density depletion in the subauroral region, has been extensively studied since its discovery in the 1960s. Our knowledge of the variability of the trough is mostly based on the observations in the northern hemisphere. Recently, the COSMIC observations enabled the investigation of the trough in both hemispheres at all local time. However, the investigation was limited to the period of the low solar activity. In this study, we investigate the variability of the trough location and morphology with local time, solar cycle, magnetic activity, and interplanetary magnetic field. For this purpose, we analyze the DMSP F15 data acquired during 2000-2010.
