곡피(?皮)와 상목피(橡木皮) 추출물이 대식세포 RAW264.7 활성화에 미치는 영향 (Effects of Quercus dentata and Quercus acutissima Extracts on the Activations of Macrophage RAW264.7 Primed with $IFN-{\gamma}$)

  • Objectives : The effects of methanol extracts from the cortex of Quercus dentata Thunb. and Quercus acutissima Carruth, on the activation of macrophage were examined. Methods : Methanol extracts of Quercus dentata (QD) and Quercus acutissima (QA) were applied to cell line RAW264,7 (macrophage), and their effects were examined. Results : 1. Extracts from QD and QA had no specific influence on the cell growth. 2. Extracts from QD and QA did not activate macrophage independently, but the addition of $IFN-{\gamma}$ facilitated the generation of macrophage's nitric oxide(NO). 3. QD and QA extracts increased the manifestation of iNOS gene, when macrophage was activated by $IFN-{\gamma}$. 4. QD and QA extracts increased the manifestation of $TNF-{\alpha}$ in macrophage, which took 2 hours. 5. QD and QA extracts increased the generation of $PGE_2$ in macrophage. Conclusion : QD and QA activate macrophage in the presence of $IFN-{\gamma}$. After activation is primarily facilated by $IFN-{\gamma}$, it works on macrophage secondarily for the manifestation of iNOS gene and for the generation of $TNF-{\alpha}$.

씀바귀 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl acetate Fraction of Ixeris dentata (Thunb.) Nakai in Caenorhabditis elegans)

  • Through the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of Ixeis dentata was investigated. The ethyl acetate soluble fraction of the I. dentata methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain (CF1553). As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

환경구배처리에 따른 떡갈나무의 생육 반응 (The Growth Response of Quercus dentata Sapling to the Environmental Gradients Treatment)

  • 떡갈나무는 우리나라의 건조한 석회암 지역에 서식하는 주요한 교목이다. 본 연구는 떡갈나무의 생태적 특성을 밝히기 위하여 생육에 주요한 영향을 미치는 광, 수분 그리고 유기물 등의 환경요인을 각각 4구배로 하여 온실에서 유식물에 처리하여 생육반응을 관찰하고, 분석하였다. 광 구배에서 지상부, 지하부 그리고 식물체 무게는 높은 조건에서 잘 자랐고, 낮은 조건에서 못 자랐다. 지하부/지상부 비는 가장 높은 조건에서 제일 높았다. 수분 구배에서 모든 측정 항목에서 구배에 따른 생육의 차이가 없었다. 유기물 구배에서 지상부, 지하부 그리고 식물체 무게는 약간 높은 조건에서 잘 자랐고, 이보나 낮거나 높은 조건에서 못 자랐다. 이상으로 볼 때 떡갈나무의 생육은 수분 조건보다는 광 조건과 유기물 조건에 의하여 더 큰 영향을 받는 것으로 나타났다.

  • 천연림내의 참나무류와 자연 수분된 종자 및 인공적인 종간 교잡 종자를 양묘해서 키운 묘목의 종 및 잡종 간의 개아기(開芽期) 차이를 조사하였다. 금산연습림(禁山演習林)의 천연림에서 개아(開芽)는 Q. crispula > Q. anguste-lepidota > Q. takatorensis ${\fallingdotseq}$ Q. serrate > Q. dentata 순으로 진행되었다. 자연 수분된 종자로 양묘한 묘목에서는 Q. aliena ${\fallingdotseq}$ Q. crispula > Q. anguste-lepidota > Q. takatorensis ${\fallingdotseq}$ Q. serrate > Q. dentata 순으로 개아(開芽)하였다. 종간 교잡의 묘목에서 $F_1$의 개아기(開芽期)는 양친종의 사이에 속했지만, 개아기(開芽期)가 빠른 양친의 영향을 보다 크게 받는 것으로 나타났다. 종간교잡의 결과를 통해서 추정할때 참나무류의 개아기(開芽期)의 빠르고 느린 특성은 유전적인 지배를 받아, 후대에 유전되는 것으로 사료된다.

A Preliminary Study on Hypocholesterolemic and Hypoglycemic Activities of Some Medicinal Plants

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

Morphology and Molecular Phylogeny of Psilothallia dentata (Ceramiaceae, Rhodophyta)

  • Psilothallia is a ceramiaceous red algal. genus that includes three species worldwide: P. dentate, P. siliculosa, and P. striate. The latter two species are limited to Australian waters, and P. dentate occurs in Japan. We here report the detailed morphology of P. dentate, and also determined plastid protein-coding psbA in P. dentate and putative relatives. Psilothallia dentate is distinguished by compressed thalli with alternate-distichous determinate branchlets, six periaxial cells, rhizoidal filaments in axes, cystocarps with 7-8 involucral filaments, spermatangia on branched filaments, and tetrahedrally divided tetrasporangia on branched filaments. Psilothallia dentate is also unusual in that cystocarps, spermatangial clusters, and tetrasporangial tufts are formed on short adventitious indeterminate branches arising on axils of determinate branchlets. The phylogenetic trees of psbA sequences show that P. dentata was nested in a monophyletic Glade comprising Ptilota, Neoptilota, and Plumaria. This result suggests that the taxonomic position of P. dentate may be transferred from the tribe Rhodocallideae to the Ptiloteae.

씀바귀와 비름의 휘발성 풍미성분 (Volatile flavor components of Ixeris dentata and Amaranthus mangostanus)

  • 야생 식용 산채류중 씀바귀와 비름의 휘발성 풍미성분을 수증기 증류법으로 추출해서 GC와 GC-MS 조합에 의하여 분석하였다. 씀바귀에서는 alcohols류 13종, esters류 9종, aldehydes류 12종, ketones류 6종, hydrocarbons류 9종, acids류 6종, phenols류 3종 그리고 기타 3종의 총 61 성분이 확인 되었다. 비름에서는 alcohols류 15종, esters류 5종, aldehydes류 13종, ketones류 8종, hydrocarbons류 3종, acids류 7종, 기타 5종의 총 56 성분이 확인되었다.

Ixeris dentata (Thunb) Nakai Attenuates Cognitive Impairment in Scopolamine-induced Amnesic Mice

  • The purpose of the current study is to investigate the cognition-enhancing effects of Ixeris dentata (Thunb) Nakai in scopolamine-induced amnesic mice. Scopolamine (2 mg/kg, i.p.) was used to induce amnesia in mice. The cognitive-enhancing activity of the IDE (10, 20 and $40{\mu}g/mL$) was studied by passive avoidance response, elevated plus maze and Y-maze behavioral paradigm in normal and scopolamine-induced amnesic mice. Scopolamine-induced cognitive deficits were significantly reversed by IDE (P < 0.001 at 20 mg/kg) in a dose-dependent fashion in all the behavioral paradigms tested. IDE possesses cognitive-enhancing properties in amnesic mice due to its potent antioxidant action.

관능검사를 통한 씀바귀의 쓴맛 연구 (A Sensory Evaluation of the Bitter Compounds from Ixeris dentata Nakai)

  • Ixeris dentata Nakai (Compositae) is a perennial herb and the young shoots when prepared properly are edible as a bitter appetizing vegetable in the early spring. The whole plant has been used as a folk medicine in treating diabetes in Korea. As a part of our studies on bitter substances, we have investigated the bitterness of the plant through a taste-guided fractionation method. Bitter substances were extracted by the solvent fractionation and the bitterness was evaluated by 10 trained panelists consisted with graduate students of Dept. of Foods and Nutrition. It has been revealed that the bitter compounds were distributed throughout the extracting fractions and the higher intensity of the bitterness was seen in the n-BuOH fractions. The bitter compounds were subfractionated by the silica gel column chromatography and the bitterness was evaluated by the trained panelists. The subfractions which were indicated as the bitter compounds will be further studied to isolate the compounds. The H$_2$O fractions were grouped according to the molecular weight through the ultrafiltration and the group with 1,000 < m.w. < 30,000 was evaluated as the most intense bitter subfraction.

Morphological and Phylogenetic Characteristics of Tuber himalayense Collected from Rhizosphere of Quercus dentata in Korea

  • We collected the ascomata of Tuber species from the rhizosphere of Quercus dentata in Danyang, Korea. We observed the morphological characteristics of ectomycorrhizal roots and ascomata, and identified the species based on the results of the phylogenetic analysis conducted using the DNA sequences of an internal transcribed spacer, a large-subunit rDNA, translation elongation factor 1-α DNA (TEF1), and MAT. Finally, we identified the fungal species as Tuber himalayense B.C. Zhang & Minter, which has not been recorded previously in Korea. We evaluated the morphological characteristics and conducted phylogenetic analysis of the ascoma and mycorrhiza (associated with Q. dentata) of T. himalayense.
