Intracellular Localization and Developmental Changes of Purine Catabolic Enzymes during Germination in Cotyledons of Rape Seedlings (발아중 유채자엽 퓨린 분해효소의 활성변화 및 세포내 위치)

  • Intracellular localization and the developmental changes in activities of uricase and allantoinase during germination were investigated with the cotyledons of rape(Brassica napus L.) seedlings. The development anddisappearance of uricase activity took place independently of light, but allantoinase activity was increased by light. The temporal pattern of uricase activity showed that uricolysis was actively taking place in the cotyledons during their early stages of germination. While uricase can be localized in the microbody fraction isolated from crude organelle extracts of the cotyledons by density gradient centrifugation, most of the allantoinase activity found in the microbody fraction did not appear to be an integral part of the microbody.

  • Multi-phase extraction of LNAPLs not only floating on groundwater but also captured in soil pores was studied in a simulated soil system. The extraction rate of LNAPL under the MPE conditions was much higher than the rate under usual hydraulic gradient conditions. The WE might be especially useful for recovery of oils in fine-textured soils such as clay-dominant soils. The vacuum-enhanced transport of LNAPL in soils also depended on the properties of LNAPL such as viscosity and density as well as soil textures.

  • Apolipophorin-III (ApoLp-III) was purified from adult haemolynph of Hyphantriu cuneo and their molecular weight and synthetic place were investigated. ApoLp-III purification was performed by KBr-density gradient ultracentrifugation followed by gel permeation chromatographv (Sephadex G-1001 and ion-exchange chromatography (CM-52) and their purity was confirmed on 10% SDS-PAGE. ApoLp-III has the molecular weight of 18 ItDa and is synthesized by fat body.

  • Gap junction channels were reconstituted into unilamellar liposomes using immunoaffinity purified connexin 32 gap junction protein from rat liver. Vesicles containing open channels and close channels were separated by means of iso-osmolar sucros density gradient sedimentation. The open channels formed in lipid vesicles were permeable to a fluorescent dye molecule, lucifer yellow of which the hydrodynamic size is similar to pore size of gap junctions in vivo. (omitted)

A New Constant Modulus Algorithm based on Maximum Probability Criterion

  • In this paper, as an alternative to constant modulus algorithm based on MSE, maximization of the probability that equalizer output power is equal to the constant modulus of the transmitted symbols is introduced. The proposed algorithm using the gradient ascent method to the maximum probability criterion has superior convergence and steady-state MSE performance, and the error samples of the proposed algorithm exhibit more concentrated density functions in blind equalization environments. Simulation results indicate that the proposed training has a potential advantage versus MSE training for the constant modulus approach to blind equalization.

  • Total RNA was isolated from two months old wheat, rice, tobacco and sweet potato. The procedure used was simple and provided pure RNA preparation. Lysis of plant tissue in a buffer with guanidine thiocyanate and CsCl density gradient centrifugation separated RNA from the rest of the cellular components. Subsequent cholroform/1-butanol extraction and ethanol precipitation were necessary to ensure contaminant-free RNA preparation. Storage of the lysed plant tissue in the buffer with guanidine thiocyanate preserved the sample for two months without noticeable RNA degradation.

  • Transdermal iontophoresis is a physical enhancement technique to facilitate the delivery of primarily charged molecules across the skin. Principal mechanism of iontophoresis is electrorepulsion experienced by the charged solutes under the application of a potential gradient. In this work, we have investigated several factors (concentration of NADPH, current density) that can affect the iontophoretic flux.   • The influences of density variation on stress and displacement fields at a propagating Mode-III crack tip in orthotropic functionally graded materials (OFGMs) are studied. The crack propagates dynamically at a right angle to the gradient of physical properties. Three kinds of elasticity and density gradients are analyzed in this study. They are as follows: (1) the density varies without elasticity variation, (2) the directions of the density and elasticity gradients are opposite to each other, and (3) same. For these cases, the stress and displacement fields at the crack tip are developed and the dynamic stress intensity factors for propagating cracks are also studied. When the crack speed is low, the influence of density variation on the stresses and displacement is low. However, when the crack speed is high, this influence is very high.

Signaling Protein Complex Formation in Detergent Resistant Membrane of Bovine Photoreceptor Rod Outer Segments

  • We have recently found that a detergent-resistant raft like membrane (DRM) can be prepared from bovine rod outer segment membranes as a low-density buoyant fraction in sucrose density gradient ultracentrifugation. G protein (transducin) and its effector enzyme (phosphodiesterase: PDE) drastically change their affinities to DRM in the process of phototransduction. We report here that the recruitment of transducin and/or $^2$PDE to DRM has close relationship with their states in signal transduction. Active T$\alpha$/PDE-complex has a high affinity to DRM, whereas inactive transducin, or inactive PDE are excluded from DRM. Active T$\alpha$/PDE-complex seems to bind to a GTPase activating protein (GRS9) in multi- protein complexes localized on DRM. Physiological significance of the multi-protein complex on the raft-like membrane in vertebrate phototransduction would be discussed.
