Tuberous sclerosis complex (TSC, MIM#191100) is an autosomal dominant neurocutaneous syndrome caused by mutation or deletion of TSC1 encoding hamartin or TSC2 encoding tuberin and characterized by seizure, mental retardation, and multiple hamartomas or benign tumors in the skin, brain, retina, heart, kidney, and lungs. The TSC2 gene on chromosome 16p13.3 lies adjacent to the PKD1 gene which is responsible for autosomal dominant polycystic kidney disease (MIM#173900). The TSC2/PKD1 contiguous gene syndrome (TSC2/PKD1 CGDS, MIM#600273) is caused by deletion of both TSC2 and PKD1 gene. We recently experienced a 15 month-old boy and a 26 month-old girl with TSC2/PKD1 CGDS confirmed by multiplex ligation-dependent probe amplification (MLPA) analysis. They showed not only typical neurologic manifestations of TSC such as epilepsy, subependymal nodules, and subcortical tubers, but also polycystic kidney disease. The contiguous gene syndrome involving PKD1 and TSC2 should be suspected in children with enlarged polycystic kidneys and TSC. MLPA analysis is a useful method for the genetic confirmation of TSC2/PKD1 CGDS.
We report the case of a 46-year-old Chinese male patient who visited our clinic complaining of infertility. Semen analysis revealed azoospermia, and azoospermia factor c region partial deletion (b1/b3) was detected using Y chromosome microdeletion analysis. Testicular sperm extraction was performed after genetic counseling. The bilateral ductus deferens and a portion of the epididymis were absent, whereas the remaining epididymis was expanded. Motile intratesticular spermatozoa were successfully extracted from the seminiferous tubule. On histopathology, nearly complete spermatogenesis was confirmed in almost every seminiferous tubule. To our knowledge, this is the first case report of b1/b3 deletion with a congenital bilateral absence of the vas deferens and almost normal spermatogenesis.

Genome analysis of a model filamentous fungus, Aspergillus nidulans, revealed that there are six putative forkhead genes. Among them, fkhF (AN8949.2) showed A. nidulans-specific. fkhF gene is located in chromosome VII and composed of 2,337 bp coding region for 778 amino acid. Since little is known about the involvement of the forkhead proteins in the developmental process of the filamentous fungi, including A. nidulans, we generated a deletion mutant of fkhF gene and analyzed. Deletion of fkhF resulted in less-dense conidiophore formation in a solid culture. However, the sexual developmental process or cleistothecia formation was normal. Furthermore, fkhF deletion mutant produced conidiophores and conidia under the submerged culture, suggesting that the fkhF gene is involved in repression of inappropriated induction and maturation of asexual developmental process but not in sexual development.

With the popularization of digital devices in the era of the 4th industrial revolution and the increase in cyber crimes targeting them, the importance of securing digital data evidence is emerging. However, the difficulty in securing digital data evidence is due to the use of anti-forensic techniques that increase analysis time or make it impossible, such as manipulation, deletion, and obfuscation of digital data. Such anti-forensic is defined as a series of actions to damage and block evidence in terms of digital forensics, and is classified into data destruction, data encryption, data concealment, and data tampering as anti-forensic techniques. Therefore, in this study, anti-forensic techniques are categorized into data concealment and deletion (obfuscation and encryption), investigate and analyze recent research trends, and suggest future anti-forensic research directions.
1. The values of Cronbach's alpha for the Taeyang, Taeum, Soyang and Soeum questionnaire were 0.7955, 0.7776, 0.8545, and 0.8601 respectively. These results indicate a highly satisfactory level of internal consistency for the questionnaire. 2. If the deletion of an item increases Cronbach's alpha then what that means is that the deletion of that item improves reliability. Therefore, any items that result in substantially greater values of alpha than the overall alpha may need to be deleted from the questionnaire to improve its reliability. 3. Factor analysis was performed on the 81 questionnaires. Based on the scree plot and the number and decrement of eigen values greater than one, three to four factor solution was most significant. 4. The hierarchical cluster analysis was performed on the 81 Sasang constitution questionnaire. These results suggested that two or four clusters identified with homogeneous groups 5. Furthemore, there were statistically significant difference among the each group by ANOVA(P<0.0001).
The cyclic AMP (cAMP) pathway plays a major role in growth, sexual differentiation, and virulence factor synthesis of pathogenic fungi. In Cryptococcus neoformans, perturbation of the cAMP pathway, such as a deletion in the gene encoding adenylyl cyclase (CAC1), causes defects in the production of virulence factors, including capsule and melanin production, as well as mating. Previously, we performed a comparative transcriptome analysis of the Ras- and cAMP- pathway mutants, which revealed 163 potential cAMP-regulated genes (38 genes at a 2-fold cutoff). The present study characterized the role of one of the cAMP pathway-dependent genes (serotype A identification number CNAG_ 06576.2). The expression patterns were confirmed by Northern blot analysis and the gene was designated cAMP-regulated gene 1 (CAR1). Interestingly, deletion of CAR1 did not affect biosynthesis of any virulence factors and the mating process, unlike the cAMP-signaling deficient cac1$\Delta$ mutant. Furthermore, the car1$\Delta$ mutant exhibited wild-type levels of the stress-response phenotype against diverse environmental cues, indicating that Car1, albeit regulated by the cAMP-pathway, is not essential to confer a cAMP-dependent phenotype in C. neoformans.
We present a statistical analysis of Korean phonological variations using automatic generation of phonetic transcription. We have constructed the automatic generation system of Korean pronunciation variants by applying rules modeling obligatory and optional phonemic changes and allophonic changes. These rules are derived from knowledge-based morphophonological analysis and government standard pronunciation rules. This system is optimized for continuous speech recognition by generating phonetic transcriptions for training and constructing a pronunciation dictionary for recognition. In this paper, we describe Korean phonological variations by analyzing the statistics of phonemic change rule applications for the 60,000 sentences in the Samsung PBS(Phonetic Balanced Sentence) Speech DB. Our results show that the most frequently happening obligatory phonemic variations are in the order of liaison, tensification, aspirationalization, and nasalization of obstruent, and that the most frequently happening optional phonemic variations are in the order of initial consonant h-deletion, insertion of final consonant with the same place of articulation as the next consonants, and deletion of final consonant with the same place of articulation as the next consonants. These statistics can be used for improving the performance of speech recognition systems.

Purpose : The p53 tumor suppressor gene is one of the most frequently altered genes in human malignancies. We try to explore the implication of p53 alteration in Ewing's sarcoma. Materials and Methods : We analyzed 35 paraffin blocks to explore the deletion and sequence alterations of p53. Results : Quantitative PCR analysis showed that 2 tumors showed a homozygous deletion of the gene. Mutational analysis of exons 4 to 9 of p53 by PCR-SSCP revealed that 3 tumors carry sequence alterations in exons 5 or 8, and DNA sequencing analysis identified missense point mutations. Conclusion : Taken together, our data demonstrate that p53 is genetically altered in a small fraction of Ewing's sarcoma.

Double-stranded RNA (dsRNA) induces gene silencing in a sequence-specific manner by a process known as RNA interference (RNAi). The RNA-induced silencing complex (RISC) is a multi-subunit ribonucleoprotein complex that plays a key role in RNAi. VIG (Vasa intronic gene) has been identified as a component of Drosophila RISC; however, the role VIG plays in regulating RNAi is poorly understood. Here, we examined the spatial and temporal expression patterns of VIG-1, the C. elegans ortholog of Drosophila VIG, using a vig-1::gfp fusion construct. This construct contains the 908-bp region immediately upstream of vig-1 gene translation initiation site. Analysis by confocal microscopy demonstrated GFP-VIG-1 expression in a number of tissues including the pharynx, body wall muscle, hypodermis, intestine, reproductive system, and nervous system at the larval and adult stages. Furthermore, western blot analysis showed that VIG-1 is present in each developmental stage examined. To investigate regulatory sequences for vig-1 gene expression, we generated constructs containing deletions in the upstream region. It was determined that the GFP expression pattern of a deletion construct (${\Delta}-908$ to -597) was generally similar to that of the non-deletion construct. In contrast, removal of a larger segment (${\Delta}-908$ to -191) resulted in the loss of GFP expression in most cell types. Collectively, these results indicate that the 406-bp upstream region (-596 to -191) contains essential regulatory sequences required for VIG-1 expression.

Purpose: To summarize the results of chromosomal microarray analysis (CMA) for copy number variants (CNVs) detection and clinical utility in a single tertiary hospital. Materials and Methods: We performed CMA in 46 patients over the course of two years. Detected CNVs were classified into five categories according to the American College of Medical Genetics and Genomics guidelines and correlated with clinical manifestations. Results: A total of 31 CNVs were detected in 19 patients, with a median CNV number per patient of two CNVs. Among these, 16 CNVs were classified as pathogenic (n=3) or likely pathogenic (LP) (n=11) or variant of uncertain significance (n=4). The 16p11.2 deletion and 16p13.11 deletion classified as LP were most often detected in 6.5% (3/46), retrospectively. CMA diagnostic yield was 24.3% (9/37 patients) for symptomatic patients. The CNVs results of the commercial newborn screening test using next generation sequencing platforms showed high concordance with CMA results. Conclusion: CMA seems useful as a first-tier test for developmental delay with or without congenital anomalies. However, the classification and interpretation of CMA still remained a challenge. Further research is needed for evidence-based interpretation.
