Serratia marcescens catabolic threonine dehydratase는 streptomycin sulfate treatment, Sephadex G-200 gel filtration, AMP-Sepharose 4B affinity chromatography 등의 방법으로 정제하였는데, 최종 단계에서 회수율은 15.5%이었으며 50배 정제되었다. Native 분자량은 native pore gradient polyacrylamide gel electrophoresis(PAGE) 방법으로는 120,000이었다. SDS-PAGE에 의한 subunit의 분자량은 30,000이었고, 즉 S. marcescens 효소는 4개의 동일한 subunit으로 구성된 homo-tetrameric protein임이 판명되었다. S. marcescens 효소의 L-threonine에 대한 Km값은 AMP가 있는 조건에서 7.3 mM, AMP가 없는 조건에서 92 mM이었다. S. marcescens 효소는 효소 1 mole 당 각각 2 mole의 pyridoxal 5'-phosphate(PLP), 16개의 free-SH group을 가지고 있었다. S. marcescens 효소는 AMP의 존재 하에서 $\alpha$-ketobutyrate, pyruvate, glyoxylate, phosphoenol pyruvate(PEP)에 의해 효소 활성이 억제되었으며, cAMP와 ADP에 의해서는 효소 활성이 증가되었다. 효소학적 성질면에서 S. marcescens 효소는 E. coli 효소보다는 S. typhimurium 효소와 유사하였다. 한편, E. coli 효소는 cAMP에 의하여 효소 활성이 증가되고, S. typhimurium 효소는 ADP에 의해 효소 활성이 증가되는 것과 다르게, S. marcescens 효소는 cAMP와 ADP 모두 효소 활성이 증가되었다. 따라서 이상의 연구 결과들은 세 enteric bacteria의 catabolic threonine dehydratase가 서로 작은 차이점이 있다는 것을 반영하며, 이러한 사실을 규명하기 위해서는 향후 보다 심층적인 연구를 수행하여야 할 것으로 사료된다.
UK-58,852의 생합성에 관여하는 유전자를 분리하기 위해 Actinomadura roseorufa의 genomic DNA와 E. coli-Streptomyces shuttle cosmid vector인 pOJ446이 genomic library를 만들었다. Genomic library는 dehydratase PCR product와 eryA 유전자를 probe로 하여 sugar 생합성 유전자와 polyketide typel 유전자가 집단으로 존재하는 cosmid pHD54를 분리하였고, 이를 제한 효소인 BamHI, SmaI와 Sonicater를 이용해서 subcloning 하였다. 이들의 염기서열을 부분 분석한 결과, polyketide 생합성에 관여하는 ketoacyl synthase, methylmalonyl acyltransferase, ketoreductase, enolreductase 그리고 PKS loading domain 등 polyketide synthase type I 임을 보여주고 있고, BLAST 분석된 결과를 보면 polyketide synthase 유전자는 rifamycin 생합성 유전자와 유사성이 높다. 그리고 sugar 생합성에 관여하는 유전자로는 oxidoreductase, dTDP-D-glucose 4,6 dehydratase, dTDP-D-glucose synthase 그리고 dTDP-4-keto-6-deoxy-D-glycose 3,5-epimerase으로 구성된 gene cluster를 확인하였다. 그리고 염기서열 분석된 유전자중 dTDP-D-glucose synthase를 발현하여 유전자의 기능을 확인하였다.
Biodegradative threonine dehydratase was purified to homogeneity from Serratia marcescens ATCC 25419 by streptomycin sulfate treatment, Sephadex G-200 gel filtration chromatography followed by AMP-Sepharose 4B affinity chromatography. The molecular weight of the purified enzyme was 118,000 by fast protein liquid chromatography using superose 6-HR. The enzyme was determined to be a homotetrameric protein with subunit molecular weights of 30,000 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The enzyme was inhibited by ${\alpha}-Keto$ acids and their derivatives such as ${\alpha}-ketobutyrate$, pyruvate, glyoxlyate, and phosphoenol pyruvate, but not by ${\alpha}-aminobutyrate$ and ${\alpha}-hydroxybutyrate$. The inhibition of the enzyme by pyruvate and glyoxylate was observed in the presence of AMP. The inhibitory effect of glyoxylate was decreased at high enzyme concentration, whereas the inhibition by pyruvate was independent of the enzyme concentration. The kinetics of inhibition of the enzyme by pyruvate and glyoxylate revealed a noncompetitive and mixed-type inhibition by the two inhibitors with respect to L-threonine and AMP, respectively.

A novel 6-deoxyhexose biosynthetic gene cluster different from the one for the biosynthesis of streptomycin was isolated from Streptomyces griseus using specifically designed PCR primers to compare the sequence of known dTDP-glucose synthase genes. We cloned a 5.8-kb DNA from Streptomyces griseus ATCC10137, which contained the 4-ketoreductase homologue (grsB), dTDP-glucose synthase (grsD), and dTDP-glucose 4, 6-dehydratase (grsE) genes. Escherichia coli cultures containing plasmid of the PCR product which encoded the grsE region under the controUed T7 promoter were able to catalyze the formation of dTDP-4-keto-6-deoxy-D-glucose from TDP-glucose. The enzyme showed high substrate specificity, being specific to only dTDP-glucose that is known to be incorporated into secondary metabolites such as antibiotics.

알려진 dTDP-D-glucose 4,6-dehydratase의 amino acid 서열로 부터 primer를 제작하여 내열성 균주인 Thermus caldophilus GK24에서 colony hybridization 과정을 거쳐 dTDP-D-glucose 4,6-dehydratase를 포함하는 cosmid DNA를 얻었다. 유전자 분석을 위해 cosmid DNA를 subclone 하여 작은 크기로 분리하였다. 분리된 cosmid를 pSMTC-1 으로 명명하고 pSMTC-1를 BamHI으로 반응시켜 BamHI 단편 모두를 pGEM 7(+)를 이용하여 subclone 하였다. 각각의 이름은 크기에 따라 pKCB10(1.2kb-BamHI), pKCB20(1.6kb-BamHI), pKCB30(2.Ikb-BamHI), pKCB40(2.5kb-BamHI), pKCB50(2.5kb-BamHI), pKCB60(2.7kb-BamHI), pKCB70(3.4kb-BamHI), pKCB80(4.4kb-BamHI), pKCB90(7.0kb-BomHI) 으로 명명하였다. 각각의 subclone된 유전자를 분석하기 위해 Erase-a-base 방법을 이용하여 template를 준비하였고 이를 자동 염기서열 분석기를 이용하여 염기서열을 분석하였다. 염기서열분석 결과 pKCB80(4.2kb)에 dTDP-D-glucose synthase(orfA) 유전자를 비롯하여 dTDP-D-glucose 4,6-dehydratase(orfB), orfC (dTDP-4-keto-L-rhamnose reductase) 그리고 orfD(dTDP-4-keto-6-deoxy-D-glucose 3,5-epimerase)와 유사한 유전자들이 있음이 확인 되었고 dTDP-L-rhamnose의 생합성 과정을 예상할 수 있었다.
Many antibiotics contain partially deoxygenated sugar components that are usually essential for biological activity, affinity, structural stability, and solubility of antibiotics. Gene probes of the biosynthetic genes related with the deoxysugar were obtained from PCR. Primers were designed from the conserved peptide sequences of the known dTDP-D-glucose 4,6-dehydratases, which are the key step enzymes in the biosynthesis of deoxysugar. The primers were applied to amplify parts of dehydratase genes to 27 actinomycetes that produce the metabolites containing deoxysugar as structural constituents. About 180 and 340 bp DNA fragments from all of the actinomycetes were produced by PCR and analyzed by Southern blot and DNA sequencing. The PCR products were used as gene probes to clone the biosynthetic gene clusters for the antibiotic mithramycin, rubradirin, spectinomycin, and elaiophyrin. This method should allow for detecting of the biosynthetic gene clusters of a vast array of secondary metabolites isolated from actinomycetes because of the widespread existence of deoxysugar constituents in secondary metabolites.
Dehydroquinate dehydratase (DHQD) catalyzes the conversion of 3-dehydroquinic acid (DHQ) into 3-dehydroshikimic acid in the mid stage of the shikimate pathway, which is essential for the biosynthesis of aromatic amino acids and folates. Here, we report two the crystal structures of type II DHQD (CgDHQD) derived from Corynebacterium glutamicum, which is a widely used industrial platform organism. We determined the structures for CgDHQDWT with the citrate at a resolution of 1.80Å and CgDHQDR19A with DHQ complexed forms at a resolution of 2.00 Å, respectively. The enzyme forms a homododecamer consisting of four trimers with three interfacial active sites. We identified the DHQ-binding site of CgDHQD and observed an unusual binding mode of citrate inhibitor in the site with a half-opened lid loop. A structural comparison of CgDHQD with a homolog derived from Streptomyces coelicolor revealed differences in the terminal regions, lid loop, and active site. Particularly, CgDHQD, including some Corynebacterium species, possesses a distinctive residue P105, which is not conserved in other DHQDs at the position near the 5-hydroxyl group of DHQ. Replacements of P105 with isoleucine and valine, conserved in other DHQDs, caused an approximately 70% decrease in the activity, but replacement of S103 with threonine (CgDHQDS103T) caused a 10% increase in the activity. Our biochemical studies revealed the importance of key residues and enzyme kinetics for wild type and CgDHQDS103T, explaining the effect of the variation. This structural and biochemical study provides valuable information for understanding the reaction efficiency that varies due to structural differences caused by the unique sequences of CgDHQD.
식이 셀렌이 납섭취에 따른 흰주의 혈액 및 간장중의 $\delta$-Aminolevulinic Acid Dehydratase(ALAD)활성 저해에 미치는 영향을 알아보기 위하여 체중이 141$\pm$5g이 되는 Sprague-Dawley 종 숫쥐를 식이내 납함량을 0(대조군), 200, 1,000, 2,000, 5,000ppm으로 달리하고, 다시 0.5ppm의 셀렌을 첨가한군과 비첨가군등 9군으로 나누어 4주간 사육한 후 체중증가, 식이효율, 혈중hemoglobin, hematocrit치, 혈액 및 간장중의 ALAD 효소활성, 그리고 뇨증 aminolevulinic acid(ALA)의 배설량을 측정한 결과는 다음과 같다. 1. 식이섭취량은 대조군과 납식이군과 별 차이가 없었고, 식이효율 및 체중증가량은 2,000, 5,000 ppm 납식이군이 유의적으로 낮았으나, 셀렌을 첨가했을 때는 대조군과 차이가 없었다. 2. 장기의 무게는 200, 1,000ppm 납식이군에서 신장의 무게가 대조군에 비해 비대했으나, 셀렌을 첨가했을때는 타실험군과 차이가 없었다. 3. Hemoglobin 함량과 hematocrit치는 납함량이 높을수록 대조군에 비해 감소하였으나, 납함량이 1,000, 2,000ppm군에서는 셀렌을 첨가했을때 비첨가군에 비해 증가되었다. 4. 혈액중 ALAD효소 활성은 납함량이 증가될수록 대조군에 비해 ALAD활성이 현저히 감소되었고 200, 1,000, 2,000ppm 군에서는 셀렌을 첨가했을때 비첨가군에 비해 그 활성이 증가되었다. 간장중의 ALAD효소 활성도 혈액에서와 같은 경향으로 납함량이 즐가될수록 감소되었으나, 혈액에서 보다는 ALAD 활성 감소가 적었으며 셀렌을 첨가한 경우 모든 식이군에서 비첨가군에 비래 ALAD 활성이 증가되었다. 5. 뇨중의 ALA 배설량은 ALAD활성과는 반대로 식이내 납함량이 높을수록 그 배설량이 증가되었고, 셀렌 첨가군에서는 그 배설량이 감소되었다.
