Changes of Water Activity and Physico-chemical Properties of Burley Tobacco as Affected by Storage and Moisture Contents (저장조건과 수분함량에 따른 버어리종 잎담배 수분활성도 및 이화학적 특성 변화)

  • The study was carried out to investigate water activity, physicochemical properties and color degree of burley tobacco stored in shelf with different moisture content for 56 days. The study was consist of two experimental designs, that is, moisture content in burley tobacco was kept to 13 to 15 %, 17 to 19 %, and 21 to 23 % under ambient temperature of $20^{\circ}C$ with relative humidity of 24 %, and another group of in burley tobacco was only kept in ambient temperature of $3^{\circ}C$ with relative humidity of 50 % for 56 days with same water content of former treatment. Normal leaves were kept for 56 days under 21 to 23% of moisture content with increased water activity and TSNA(tobacco-specific nitrosamine content. It was considered that favorable leaves conditions are due to favorable microbial growth resulting in increased TSNA content. The total nitrogen content has not changed in two experimental groups for 56 days, otherwise the nitrate form nitrogen and total alkaloid contents were only increased by 21 to 23 % of water content. Degree of pH change was slightly decreased in both storage and water treatment. It was more decreased in 21 to 23% of moisture content than that of other moisture content. In stored leaf tobacco, higher moisture content caused lower brightness, yellow degree, and carbonization quotient.

  • This study was conducted to investigate the effects of topping time and degree on the characteristics of three tobacco varieties, and to establish the early topping (Button stage) method at Eumseong Experiment Station, Korea Tobacco Research Institute in 1979-1980. The results are summarized as follows; yield increased in the plots of By 104, topping at button stage and under first leaf from floral axis. Quality was good in the plot topped at button stage and under first leaf from floral axis. Price per l0a was highest in the plot topped at button stage and under first leaf from floral axis.

Effects of 2-Chloroethylphosphonic Acid on Ripening and Total Alkaloid Content of Tobacco Leaves. (잎담배의 성숙촉진 및 Alkaloid 함량에 미치는 2-Chloroethylphosphonic Acid의 효과)

  • The present experiments were carried out to find out the proper concentration of CEPA(2-chloroethy-lphosphonic acid) for the purpose of accelerating the leaf maturity and of reducing the nicotine content of tobacco. Varried levels of CEPA were sprayed Yellow Special A right after topping. The effects of each level CEPA treatment on leaf ripening and total alkaloid contents were periodically observed. The results are summarized as follows; 1. The higher the CEPA concentration was, the more the leaf maturity was accelerated. During the period from six to 11 days after treatment, the differences of leaf maturity among levels were prominent. 2. Treatment with CEPA only on the upper surface of the tobacco leaf, accelerated the maturity of that particular part treated, but not apparently the other parts of leaf. 3. The higher the CEPA concentration was, the more the accumulation of the total alkaloid was reduced. The reduction of alkaloid accumulation was evident after acceleration of leaf maturity. 4. Distinctive acceleration of leaf maturity was observed in the fully developed lower leaves, while reduction of alkaloid accumulation in the growing upper leaves. The degree of total alkaloid content reduced in the ripened leaves, however, were similer to all the leaves at different positions. 5. Yields of tobacco leaves were not significantly affected by CEPA treatment. 6. In the present experiments, it may be concluded that CEPA2, 000ppm is the most applicable level for accelerating leaf maturity and decreasing total alkaloid content. In the view point of the practical use, however, the applicable level is assumed to be properly choosed between 500 to 2, 000ppm, depending on the situations. 7. The mechanism of accelerating maturity and reducing alkaloid accumulation of tobacco leaves by CEPA, is further to be explored.

  • Seasonal climatic factors associated with tobacco quality grade and production rate were analyzed. The degree of influence on yield distribution rate in high guality tobacco leaues was highly positive with the average temperature in early May, but negatively related to those in late May and early June. Positive correlations were noticed between the degree of influence and sunshine hours in Middle June, late June and late May in decrease order, while negative degree of influence was higher in early May than in late May, The order influenced by recipitation in a positive direction was early May, late May and middle May. Negative influence was noticed in middle and early June with a great degree.

  • Ten anther-derived doubled haploid (ADH) and 10 maternally-derived doubled haploid fines (MDH) from NC 2326 were evaluated in the field to compare their performance. Both anther-derived doubled haploid and maternally-derived doubled haploid lines were agronomically inferior to selfed parental variety for many parameters, but the degree of inferiority of the MDH lines were not significantly different from the selfed parental variety for any character investigated. The inferiority of the ADH lines were, however, significantly different from the selfed parental variety for yield, leaf length and width , and days to flower.

Influence of Maturity on the Plastid Pigments and Chromatic Characteristics of Paddy Tobacco (연초 수확엽의 숙도가 색채 및 색소물질 함량에 미치는 영향)

  • Field experiment was conducted to get the basic information about the difference in color and plastid pigments at the different stage of harvest of flue-cured tobacco cultivated in paddy field, and compared to upland ones. All tested crops were primed in lots of 3 to 4 at each priming, beginning at the bottom of the plant. Leaf pigments and chromatic values of tobacco leaves were determined in samples collected at weekly intervals, and according to analyses obtained from 4 stalk positions. By the degree of maturity, leaves harvested were separated with visual characters into four classes of immature, mature, ripe and mellow. regardless of stalk position, chlorophyll contents was higher in the order of immature > mature > ripe > mellow, respectively. In fresh leaves, chlorophyll levels was higher with ascending stalk position. chlorophyll a and b in cured leaves were less than 10% of the amount present at harvest, and it was estimated that maturity condition also influenced the rates of degradation of plastid pigments. Fully mature leaves of upland diverted from paddy field showed slightly higher in chlorophyll contents than in those of upland tobacco ones but somewhat lowered in $L^*$ and $b^*$ values, and there was no difference in $a^*$ value.

  • This study was conducted to determine effect of fruits removal on the CO2 exchange rates (CER) and growth of ginseng plant. Fruit of 2, 4 age plant removed at 7, May. The results of these investigations are as follows. 1. The net photosynthetic rates of the ginseng bearing fruits increased to a considerably greater degree than that of the ginseng without fruit in each ages. 2. The total dry matter per plant in bearing fruit (40.24g) had produced more dry matter than that of non-fruiting plant (38.13g) , but the root 4.y matter in fruiting plant (26.2g) had produced less dry matter than that of non-fruiting plant (27.1g) in 4 age. 3. The ginseng plant in bearing fruit did not influence the dry matter of stem and leaf. 4. The maximum RGR of root (17, June) was slower than that of fruit (4, June) .

  • Silkworm is known to be damaged under the unfavorable environmental conditions. Recently, it has been often argued that nicotine is a main factor to affect the silkworm nutritional balance and to result in decrease of quantitative characters, and unbalance of larval nutrition. Especially in spring season, there is tendency to produce nicotine damage to silkworm fed with the mulberry leaf near to tobacco field early cultivated in mulching system. In these connections, the trial was made in the different pots placing by the different distance 10rn, 20m, 40m, and 60m in the direction of east-north from the tobacco field. Location of pot near to the tobacco fold is 40m below the tobacco Held and gradually higher than the tobacco field by 10 degree of slope. In addition, when the mulberry field is divided into two areas from the east-south to west-north, southern east zone is lowering down below 10 degree of slope and has 36% of east-south wind, 21% of south-east wind and 16% of south wind, etc. Pots are left on the mulberry field from the completion of lower part leaf harvest to just before upper part leaf harvest is completed for 32 days. Then silkworm at 4th-5th stage was fed with the leaf grown during the period and larval characters are statistically analyzed. The results are summarized as follows: 1. 10-40m distant pot statistically at the same level has the longest larval duration and 60m distant pot is. not significant at 5% level. Whole larval duration has same tendency as in the 5th larval duration. 2. Percentage of delayed molting larvae at the 5th stage is increased in 10m distant pot and there is no significance statistically in other pots, as compared with that of control. 3. It is not showed that there is no damage to mortality of the old larvae in 60m distant pot. 4. 10-20m distant pot produces a low pupation and others are at the same level statistically at 5% significance, 5. It is showed that nicotine damage is occurred in single cocoon freight and cocoon shell weight up to 60m distant pot, rather higher damage in cocoon shell weight than in single cocoon weight. 6. It is showed that cocoon yield from 10, 000 molted larvae including double cocoon is decreased up to 40m distant pot far from the tobacco field in which 40% of cocoon yield in 10m distant pot by 7kg, 26% of cocoon yield in 20m distant pot by 3.9kg, 24% of cocoon yield in 40m distant pot by 3.6kg, and 12% of cocoon yield in 60m distant pot by 1.8kg, as compared with control. However, there is no significance statistically at 5% level in 60m distant pot. With these above results, it is concluded that nicotine da mage could be occurred in cocoon yield and cocoon quality up to 60m distant far from the tobacco field.

  • Six orient varieties of N.tabacum and 15 $F_1$ hybrids, not including reciprocal crosses, were evaluated for the degrees of heterosis and the combining abilities of 6 characteristics in Suweon, Korea, 1981. The values 9f general combining abilities tiers greater than those of specific combining abilities in all traits but significant SCA was also observed for Plant height, days to flowering and yield. In general, CCA effect was large in Samsun, Kavala and Izmir varieties. Large SCA values were observed for number of leaves in Samsun x Izmir, days to flowering in Samsun x Serves, leaf shape in Basma x Xantlli, and yield in Basma x Kavala and Xanthi x Izmir. Heterosis of favorable directions in $F_1$ was generally observed for all traits. Especially the $F_1$ of Samsun x Kavala flowered earlier than its midparents. Large heterosis effects were manifested for the number of leaves in Samsun x Izmir and yield in Samsun x Izmir as well as Xanthix Izmir.

Cosuppression and RNAi induced by Arabidopsis ortholog gene sequences in tobacco

  • The Arabidopsis ${\omega}$-3 fatty acid desaturase (AtFAD7) catalyzes the synthesis of trienoic fatty acids (TA). A transgenic tobacco line, T15, was produced by a sense AtFAD7 construct and showed a cosuppression-like phenotype, namely extremely low TA levels. The sequence similarity between AtFAD7 and a tobacco ortholog gene, NtFAD7, was moderate (about 69%) in the coding sequences. AtFAD7 siRNAs accumulated at a high level, and both AtFAD7 and NtFAD7 mRNAs are degraded in T15 plants. The low-TA phenotype in T15 was dependent on a tobacco RNA-dependent RNA polymerase6 (NtRDR6). We also produced tobacco RNAi plants targeting AtFAD7 gene sequences. The AtFAD7 siRNA level was trace, which was associated with a slight reduction in leaf TA level. Unexpectedly, this RNAi plant showed an increased NtFAD7 transcript level. To investigate the effect of translational inhibition on stability of the NtFAD7 mRNAs, leaves of the wild-type tobacco plants were treated with a translational inhibitor, cycloheximide. The level of NtFAD7 mRNAs significantly increased after cycloheximde treatment. These results suggest that the translational inhibition by low levels of AtFAD7 siRNAs or by cycloheximide increased stability of NtFAD7 mRNA. The degree of silencing by an RNAi construct targeting the AtFAD7 gene was increased by co-existence of the AtFAD7 transgene, where NtRDR6-dependent amplification of siRNAs occurred. These results indicate that NtRDR6 can emphasize silencing effects in both cosuppression and RNAi.
