Chemosensitization of Fusarium graminearum to Chemical Fungicides Using Cyclic Lipopeptides Produced by Bacillus amyloliquefaciens Strain JCK-12

  • Fusarium head blight (FHB) caused by infection with Fusarium graminearum leads to enormous losses to crop growers, and may contaminate grains with a number of Fusarium mycotoxins that pose serious risks to human and animal health. Antagonistic bacteria that are used to prevent FHB offer attractive alternatives or supplements to synthetic fungicides for controlling FHB without the negative effects of chemical management. Out of 500 bacterial strains isolated from soil, Bacillus amyloliquefaciens JCK-12 showed strong antifungal activity and was considered a potential source for control strategies to reduce FHB. B. amyloliquefaciens JCK-12 produces several cyclic lipopeptides (CLPs) including iturin A, fengycin, and surfactin. Iturin A inhibits spore germination of F. graminearum. Fengycin or surfactin alone did not display any inhibitory activity against spore germination at concentrations less than 30 ug/ml, but a mixture of iturin A, fengycin, and surfactin showed a remarkable synergistic inhibitory effect on F. graminearum spore germination. The fermentation broth and formulation of B. amyloliquefaciens JCK-12 strain reduced the disease incidence of FHB in wheat. Furthermore, co-application of B. amyloliquefaciens JCK-12 and chemical fungicides resulted in synergistic in vitro antifungal effects and significant disease control efficacy against FHB under greenhouse and field conditions, suggesting that B. amyloliquefaciens JCK-12 has a strong chemosensitizing effect. The synergistic antifungal effect of B. amyloliquefaciens JCK-12 and chemical fungicides in combination may result from the cell wall damage and altered cell membrane permeability in the phytopathogenic fungi caused by the CLP mixtures and subsequent increased sensitivity of F. graminearum to fungicides. In addition, B. amyloliquefaciens JCK-12 showed the potential to reduce trichothecenes mycotoxin production. The results of this study indicate that B. amyloliquefaciens JCK-12 could be used as an available biocontrol agent or as a chemosensitizer to chemical fungicides for controlling FHB disease and as a strategy for preventing the contamination of harvested crops with mycotoxins.

Analysis of Membrane Integrity and Removal Efficiency Considering Membrane Defect and Pore Size (막 파단 및 공극크기에 따른 막 완결성 및 제거효율 분석)

  • Microfiltration (MF) and ultrafiltration (UF) processes for removal of particulate materials (i.e., turbidity, microorganisms and viruses) have been used to produce drinking water with higher quality. As membrane filtration technique has become widely applied for drinking water treatment, the importance of membrane integrity test (MIT) has also been increasingly emphasized. The results of pressure decay test (PDT) were presented in the paper to monitor membrane integrity. In this paper the PDT was carried out with deliberately-defected membrane fibers to evaluate the sensitivity of PDT on membrane fiber damage. Variation of pressure decay rate and removal rate were investigated to evaluate the impact of defection (defection ratio) and pore size of membrane. The membrane integrity could be successfully monitored by the PDT. The pressure decay rate varied from $0.002{\sim}0.189kg_f/cm^2hr$ with the initial pressure ranged from 0.2 to $1.0kg_f/cm^2hr$. Higher initial pressure which provided with higher pressure decay rate was preferred to evaluate the defection of membrane fiber. As for the particle removal rate, the Log Removal Rate (LRV) of kaolin solution decreased significantly from 3.78 to 2.31 when one fiber out of 3,200 fibers was cut. The membranes with different pore size were tested to evaluate virus removal efficiency. The virus removal rate of the MF membrane ($0.1{\mu}m$) was about 30% although the poliovirus was smaller than the pore size of the MF membrane, indicating that the removal rate was much lower than Korea Water Works Association (KWWA) certificate LRV of 1.5.

  • Uncertainty is central to energy and climate policy. A growing number of literature show that almost all components of energy and climate models are, to some extent, uncertain and that the effect of uncertainty on the model outputs, in turn policy recommendations, is significantly large. Most existing energy and climate-economy models developed and used in Korea, however, do not take uncertainty into account explicitly. Rather, many models conduct a deterministic analysis or do a simple (limited) sensitivity analysis. In order to help social planners to make more robust decisions (across various plausible situations) on energy and climate change issues, an uncertainty analysis should be conducted. As a first step, this paper reviews the theory of decision making under uncertainty and the method for addressing uncertainty of existing probabilistic energy and climate-economy models. In addition, the paper proposes a strategy to apply an uncertainty analysis to energy and climate-economy models used in Korea. Applying the uncertainty analysis techniques, this paper revises the FUND model and investigates the impacts of climate change in Korea.

  • Recentincreasing awareness of the environmental damage caused by the $CO_2$ emission of fossil fuelsstimulated the interest in alternative and renewable sources of energy. Fuel cell is a representative example of hydrogen energy utilization. In this study, Molten Carbonate Fuel Cell system is simulated by using $Aspen^{TM}$. Stack model is consisted of equilibrium reaction equations using $ACM^{TM}$(Aspen Custom Modeler). Balance of process of fuel cell system is developed in Aspen $Plus^{TM}$ and simulated at steady-state. Analysis of performance of the system is carried out by using sensitivity analysis tool with main operating parameters such as current density, S/C ratio, and fuel utilization and recycle ratio.In Aspen $Dynamics^{TM}$, dynamics of MCFC system is simulated with PID control loops. From the simulation, we proposed operation range which generated maximum power and efficiency in MCFC power plant.

Immuno-chromatographic Analysis for HPV-16 and 18 E7 Proteins as a Biomarker of Cervical Cancer Caused by Human Papillomavirus

  • Among the more than 120 different types of human papillomavirus (HPV), types 16 and 18 have been known to be high risk agents that cause cervical cancer. We examined, in an immuno-chromatographic analysis, the potential of using the early gene product, E7 protein, as a diagnostic marker of cervical cancer caused by HPV. We developed monoclonal antibodies specific to HPV-16 and 18 E7 proteins that were produced from bacterial cells using gene recombinant technology. For each E7 protein, the optimal antibody pair was selected using the immuno-chromatographic sandwichtype binding system based on the lateral flow through membrane pores. Under these conditions, this rapid testing assay had a detection capability as low as 2 ng/mL of E7 protein. Furthermore, since viral analysis required the host cell to be lysed using chemicals such as detergents, it was possible that the E7 protein was structurally damaged during this process, which would result in a decrease in detection sensitivity. Therefore, we examined the detrimental effects caused by different detergents on the E7 protein using HeLa cells as the host. In these experiments, we found that the damage caused by the detergent, nonylphenylpolyethylene glycol (NP-40), was minimal relative to Triton X-100 commonly used for the cell lysis. Temperature also affected the stability of the E7 protein, and we found that the E7 protein was stabilized at 4$^{\circ}C$ for about 2 h, which was 4 times longer than at room temperature. Finally, a HPV-infected cervical cancer cell line, which was used as a real sample model, was treated using the optimized conditions and the presence of E7 proteins were analyzed by immuno-chromatography. The results of this experiment demonstrated that this rapid test could specifically detect HPV-infected samples.

Assessment of Flood Vulnerability to Climate Change Using Fuzzy Model and GIS in Seoul (퍼지모형과 GIS를 활용한 기후변화 홍수취약성 평가 - 서울시 사례를 중심으로 -)

  • The goal of this study is to apply the IPCC(Intergovernmental Panel on Climate Change) concept of vulnerability to climate change and verify the use of a combination of vulnerability index and fuzzy logic to flood vulnerability analysis and mapping in Seoul using GIS. In order to achieve this goal, this study identified indicators influencing floods based on literature review. We include indicators of exposure to climate(daily max rainfall, days of 80mm over), sensitivity(slope, geological, average DEM, impermeability layer, topography and drainage), and adaptive capacity(retarding basin and green-infra). Also, this research used fuzzy model for aggregating indicators, and utilized frequency ratio to decide fuzzy membership values. Results show that the number of days of precipitation above 80mm, the distance from river and impervious surface have comparatively strong influence on flood damage. Furthermore, when precipitation is over 269mm, areas with scare flood mitigation capacities, industrial land use, elevation of 16~20m, within 50m distance from rivers are quite vulnerable to floods. Yeongdeungpo-gu, Yongsan-gu, Mapo-gu include comparatively large vulnerable areas. This study improved previous flood vulnerability assessment methodology by adopting fuzzy model. Also, vulnerability map provides meaningful information for decision makers regarding priority areas for implementing flood mitigation policies.

Enhanced Sensitivity and Long-Term G2/M Arrest in Adriamycin-treated DNA-PK-null Cells are Unrelated to DNA Repair Defects (DNA-PK-null 세포주의 adriamycin 처리에 의한 G2/M 세포주기 변화)

  • While the DNA-protein kinase (DNA-PK) complex, comprised of DNA-PKcs and Ku80, is primary involved in the repair of DNA double-strand breaks, it is also believed to participate in additional cellular processes. Here, treatment of embryo fibroblasts (MEFs) derived from either wild-type (Wt) or DNA-PKcs-null (DNA-$PKcs^{-/-}$) mice with various stress inducing agents revealed that adriamycin was markedly more cytotoxic for $Ku80^{-/-}MEFs$ and led to their long-term accumulation in the $G_2$/M phase. This differential response was not due to differences in DNA repair, since adrimycin-triggered DNA damage was repaired with comparable efficiency in both Wt and $Ku80^{-/-}MEFs$, but was associated with differences in the expression of important cell cycle regulatory genes. Our results support the notion that Ku80-mediated cytoprotection and $G_2$/M-progression are not only dependent on the cell's DNA repair but also may reflect Ku80's influence on additional cellular processes such as gene expression.

A Study on Vulnerability Assessment to Climate Change in Siheung-si (시흥시 기후변화 취약성 평가 연구)

  • This study has purpose to minimize the impact of climate change of Siheung. Vulnerability assessment was carried out for establishing the Siheung Climate Change Master Plan. Climate change vulnerability assessment analyzed using climate exposure, sensitivity and adaptive capacity indicators. A proxy variable is selected from each indicator. Meteorological data uses the RCP scenarios provided by the Meteorological Administration, and this study assumes that the same trend will continues in the future. Siheung are vulnerable to heavy rains in the flooded roads and farmland. Also, it is necessary to be careful heat wave in summer. The size and scale of the damage depends on the city's ability to respond to the impacts of climate change. It is necessary to make a adaptation plan for climate change impact assessment and vulnerability analysis. This study will be used to make Siheung Climate Change Master Plan and to determine the priority of the policy as guideline. It is expected that this study is helpful to pursue climate change vulnerability assessment of other local governments.

The Functional Role of Lysosomes as Drug Resistance in Cancer (항암제 내성에 대한 라이소좀의 역할)

  • Lysosomes are organelles surrounded by membranes that contain acid hydrolases; they degrade proteins, macromolecules, and lipids. According to nutrient conditions, lysosomes act as signaling hubs that regulate intracellular signaling pathways and are involved in the homeostasis of cells. Therefore, the lysosomal dysfunction occurs in various diseases, such as lysosomal storage disease, neurodegenerative diseases, and cancers. Multiple forms of stress can increase lysosomal membrane permeabilization (LMP), resulting in the induction of lysosome-mediated cell death through the release of lysosomal enzymes, including cathepsin, into the cytosol. Here we review the molecular mechanisms of LMP-mediated cell death and the enhancement of sensitivity to anticancer drugs. Induction of partial LMP increases apoptosis by releasing some cathepsins, whereas massive LMP and rupture induce non-apoptotic cell death through release of many cathepsins and generation of ROS and iron. Cancer cells have many drug-accumulating lysosomes that are more resistant to lysosome-sequestered drugs, suggesting a model of drug-induced lysosome-mediated chemoresistance. Lysosomal sequestration of hydrophobic weak base anticancer drugs can have a significant impact on their subcellular distribution. Lysosome membrane damage by LMP can overcome resistance to anticancer drugs by freeing captured hydrophobic weak base drugs from lysosomes. Therefore, LMP inducers or lysosomotropic agents can regulate lysosomal integrity and are novel strategies for cancer therapy.

Evaluation of Applicability of Perovskite Dosimeter based on CsPbBr3 Material to Quality Assurance in Radiation Therapy (CsPbBr3을 기반으로 한 Perovskite 선량계의 방사선치료 Quality Assurance에 대한 적용가능성 평가)

  • In radiation therapy, accurate Quality Assurance (QA) is required to irradiate tumor tissue while minimizing damage to normal tissue. Therefore, a dosimeter that can accurately measure radiation is needed. The purpose of this study is to develop a highly efficient radiation dosimeter with high sensitivity by applying a particle in binder method that can reduce manufacturing costs and simplify processes to perovskite materials that are cheaper and simpler to manufacture. By evaluating the response characteristics to high-energy photon, the applicability of QA dosimeter to radiation therapy was evaluated. As a result of reproducibility evaluation, RSD at 6 MV energy was presented as 1.178% and 15 MV energy was presented as 1.141%. As a result of linearity evaluation according to linear regression analysis, R2 values of 0.9999 were presented under each condition of 6 MV and 15 MV energy. It was found that the CsPbBr3 dosimeter manufactured based on the results of reproducibility and linearity evaluation is highly applicable as a QA dosimeter in the field of therapeutic radiation. The CsPbBr3 dosimeter manufactured in this study presented more than the standard performance in the evaluation of reproducibility and linearity, and it can be used as a radiotherapy QA dosimeter through improvement.
