This study aims to present new frequency-related seismic intensity parameters (SIPs) based on the Hilbert-Huang Transform (HHT) analysis. The proposed procedure is utilized for the processing of several seismic accelerograms. Thus, the entire evaluated Hilbert Spectrum (HS) of each considered seismic velocity time-history is investigated first, and then, a delimited area of the same HS around a specific frequency is explored, for the proposition of new SIPs. A first application of the suggested new parameters is to reveal the interrelation between them and the structural damage of a reinforced concrete frame structure. The index of Park and Ang describes the structural damage. The fundamental frequency of the structure is considered as the mentioned specific frequency. Two statistical methods, namely correlation analysis and multiple linear regression analysis, are used to identify the relationship between the considered SIPs and the corresponding structural damage. The results confirm that the new proposed HHT-based parameters are effective descriptors of the seismic damage potential and helpful tools for forecasting the seismic damages on buildings.
This study was undertaken to determine free radical scavenging capacity and oxidative DNA damage protecting activity of methanol extract of red tea stem. The extract was subjected to assess their antioxidant potential using various in vitro systems such as $DPPH^{\bullet}$, $ABTS^{{\bullet}+}$, super oxide and nitric oxide free radicals and it exhibited $IC_{50}$ values of $68.88{\pm}1.1$, $12.08{\pm}0.65$, $404.38{\pm}1.6$, $93.6{\pm}2.7{\mu}g/mL$ respectively. Red tea extract also showed ferric reducing ability (FRAP) with 2606.85 mmol Fe (II)/g of extract. Furthermore, Methanol extract of red tea stem showed significant DNA damage protecting activity in concentration dependent manner against $H_2O_2+UV$ induced photolysis on pUC19 plasmid DNA. Results of this study showed that the methanol extract of Red Tea stem has strong antioxidant potential along oxidative DNA damage protecting capacity that would be the significant sources of natural antioxidants, which might be helpful in preventing the progress of various oxidative stress generated diseases. Further study is necessary for isolation and characterization of the active antioxidants, which may serve as a potential source of natural antioxidant.
The single cell gel electrophoressis(SCGE) assay, also known as the comet assay, is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakage in mammalian cells. The SCGE or comet assay is a promising test for the detection of DNA damage and repair in individnal cells. It has widespread potential applications in DNA damage and repair studies, genotoxicity testing and biomonitoring. In this microgel electrophoresis technique, cells are embedded in agarose gel on microscope slides, iysed and electrophoresed under alkaline conditions. Cells with increased DNA damage display increased migration of DNA from the nucleus towards the anode. The length of DNA migration indicates the amount of DNA breakage in the cell. The comet assay is also capable of identifying apoptotic cells which contain highly fragmented DNA. Here we review the development of the SCGE assay, existing protocols for the detection and analysis of comets, the relevant underlying principles determining the behaviour of DNA and the potential applications of the technique.
The current study explores the soil-structure interaction (SSI) effect on the potential seismic damage of mid-rise non-seismically designed reinforced concrete frames retrofitted by Fibre Reinforced Polymer (FRP). An 8-storey reinforced concrete frame poorly-confined due to transverse reinforcement deficiency is selected and then retrofitted by FRP wraps to provide external confinement. The poorly-confined and FRP retrofitted frames with/without SSI are modelled using hysteretic nonlinear elements. Inelastic time history and damage analyses are performed for these frames subjected to different seismic intensities. The results show that the FRP confinement significantly reduces one or two damage levels for the poorly-confined frame. More importantly, the SSI effect is found to increase the potential seismic damage of the retrofitted frame, reducing the effectiveness of FRP retrofitting. This finding, which is contrary to the conventionally beneficial concept of SSI governing for decades in structural and earthquake engineering, is worth taking into account in designing and evaluating retrofitted structures.
The main objective of this paper is to develop an improved model for the analysis of liquefaction potential and to predict excess pore pressure (EPP) using the proposed model that can simulate behavior of saturated sand under earthquake loading conditions. The damage concept is adopted for the development of the proposed model. For the development of the model, a general formulation based on experimental results and damage potential using cumulative absolute velocity (CAV) is proposed for a more realistic description of dynamic responses of saturated sand. Undrained dynamic triaxial tests are conducted using earthquake loading conditions. Based on test results, the NCER-NCW function in terms of $w_d$ and CAV is developed. Procedure for the evaluation of EPP and determination of model parameters for the proposed model is presented as well. For the determination of initial liquefaction, the minimum curvature method using the NCS-NCW curve is proposed. It is observed that predicted initial liquefaction using the proposed method agrees well with measured initial liquefaction. From results of additional undrained dynamic triaxial tests, it is seen that predicted EPP generation using the proposed model agrees well with measured results for earthquake loading cases.
The potential damage region of a WC-Co cemented carbide die is investigated for cold forging process of a wheel-nut by numerical simulation with its chemical composition considered. Numerical simulation is utilized to calculate internal stress, especially for the WC-Co die, during the forging process. Finite element model is established, in which the elasto-plastic properties are applied to the work-piece of bulk steel, and elastic properties are considered for the lower die insert of the WC-Co alloy. This stress analysis enables to distinguish the potential damage regions of the WC-Co die. The regions from calculation are comparatively analyzed along with the crack area observed in the die after repetitive manufacturing. Effect of chemical composition of the WC-Co is also evaluated on characteristics of potential damage region of the die with variance of mechanical properties considered. Derived from Mohr-Coulomb fracture model, furthermore, a new stress index is presented and used for die stress analysis. This index inherently considers hydrostatic pressure and is then capable of deducing wide range of its distribution for representing stress state by modification of its parameter implying pressure sensitivity.
The spontaneous frequency of genetic damage and the possible relationship of this damage to total radical-trapping antioxidant potential(TRAP) and antioxidant vitamins, including plasma levels of $\alpha$-carotene, $\beta$-carotene, cryptoxanthin, retinol, $\alpha$-tocopherol and ${\gamma}$-tocopherol in humans were investigated in 57 subjects using two indices of genetic damage, SCE & HFC frequency. The mean of SCE and HFC frequencies were weakly correlated with plasma TRAP(r=-0.305, p<0.1 for SCEs: r=-0.297, p<0.1 for HFCs, respectively), but those were strongly negatively correlated with plasma $\beta$-carotence(r=-0.385, p<0.01 for SCEs : r=-0.392, p<0.01 for HFCs) and cryptoxanthin(r=-0.312, p<0.05 for SCEs : r=0.335, p<0.05 for HFCs, respectively) levels in the subjects. However, those DNA damage markers including SCE and HFC did not correlate with either plasma $\alpha$-carotene, $\alpha$-tocopherol or retinol concentrations. The mean of SCE and HFC frequencies were positively correlated with plasma ${\gamma}$-tocopherol level(r=0.421, p<0.01 for SCEs : r=0.399, p<0.01 for HFCs, respectively). These findings indicate that increased cytogenetic DNA changes, as determined by SCE and HFC frequencies are possibly associated with generation of free radicals in lymphocytes and decreased plasma antioxidant vitamin($\beta$-carotene and cryptoxanthin) status in the subjects. Detection of structural damage is an inverse problem in structural engineering. There are three main questions in the damage detection: existence, location and extent of the damage. In concept, the natural frequency and mode shapes of any structure must satisfy an eigenvalue problem. But, if a potential damage exists in a structure, an error resulting from the substitution of the refined analytical finite element model and measured modal data into the structural eigenvalue equation will occur, which is called the residual modal forces, and can be used as an indicator of potential damage in a structure. In this study, a useful damage detection method is proposed and compared with other two methods. Two degree-of-freedom system and Cantilever beam are used to demonstrate the approach. And the results of three introduced method are compared.
Objectives: The aim of this study was to analyze and classify the characteristics of potential damage from hazardous chemical accidents in 229 administrative units in South Korea by reflecting the social and environmental characteristics of areas where chemical accidents can occur. Methods: A number of indicators were selected through preceding studies. Factor analysis was performed on selected indicators to derive factors, and cluster analysis was performed based on the factor scores. Results: As a result of the cluster analysis, 229 administrative units were divided into three clusters, and it was confirmed that each cluster had its own characteristics. Conclusions: The first cluster, "areas at risk of accident occurrence and spread of damage" was a type with a high potential for accident damage and a high density of hazardous facilities. The second cluster, "Urban infrastructure damage hazard areas" appeared to be a cluster with high urban development characteristics. Finally, the third cluster 'Urban and environmental damage hazard areas' appeared to be a cluster with an excellent natural environment. This study went further from the qualitative discussion related to existing chemical accidents to identify and respond to accident damage by reflecting the social and environmental characteristics of the region. Distinct from the previous studies related to the causes of accidents and the response system, it is meaningful to conduct empirical research focusing on the affected areas by analyzing the possibility of accident damage in reflection of the social and environmental characteristics of the community.
