제2 언어습득과 외국어 학습 - 상황과 조건에 알맞은 교수법 개발을 위하여 (Zweitsprachenerwerb und Fremdsprachenlernen - Zur Entwicklung der lernerangemessenen Lern- und Lehrmethode -)

  • Um von der 'kritischen' Situation des Faches DaF rauszukommen, werden viele Versuche untemommen, durch die Verbesserung des Untenichts in der Richtung nach der Entwicklung der kommunikativen $F\"{a}higkeit$ Deutsch als Fremdsprache wieder attraktiv und $konkurrenzf\"{a}hig$ zu machen. Aber leider sieht die Zwischenbilanz nicht so erfolgreich aus. Eine der Ursachen $daf\"{u}r\;l\"{a}sst$ sich mE. darin sehen, dass die $Ans\"{a}tze,\;die\;haupts\"{a}chlich\;f\"{u}r$ den Unterricht von Deutsch als Zweitsprache entwickelt wurden, ohne $gen\"{u}gende$ Berucksichtigung auf die kulturellen und institutionellen Unterschiede zwischen den Lemsituationen im zielsprachigen Land und im Heimatland der Schuler eingesetzt werden. In diesem Kontext $w\"{a}re\;es\;n\"{o}tig$, auf die Unterschiede zwischen DaF und DaZ und ihre Wirkung auf die Lehrstrategien im DaF-Unterricht zuruckzugreifen, Dabei sind auf einige Faktoren besonders zu achten, die DaF-Untenicht vom DaZ-Untenicht unterscheiden: die Mangel an den $M\"{o}glichkeiten$ der authentischen Kommunikation, die $Homogenit\"{a}t\;der\;Sch\"{u}ler$, die Lemkultur in der schlulischen Institution, das Alter, in dem die Sch\"{u}ler$ mit dem Lemen der Zielsprache beginnen. Daraus lassen sich die folgenden unterrichtlichen Konsequenzen ergeben: Die Lehrer sollten sich bemtihen, den $Sch\"{u}lern\;die\;M\"{o}glichkeiten\;f\"{u}r$ die authentischen Kommunikation besorgen, besonders $au{\ss}erhaib$ des Unterrichts, z.B. durch das Chatting- Projekt, $w\"{a}hrend$ im Unterricht mehr $Anl\"{a}sse\;f\"{u}r$ die Reflexion gegeben sollten. Und dabei sollten die unterrichtlichen Vorteile-die gemeinsame Muttersprache und Kultur, das Alter usw. - aktiv benutzt werden. Die meisten Koreaner beginnen $n\"{a}mlich$ mit dem Lernen der zweiten Fremdsprache erst in oder nach der $Pubert\"{a}t$. In diesem Zusammenhang sollte es auch betont werden, dass sich die Autonomie der Lerner durch den Unterricht richtig entwickelt, damit sie $sp\"{a}ter$ ihre Fertigkeiten autonom entwickeln $k\"{o}nnen$.

Virtual Screening for Potential Inhibitors of NS3 Protein of Zika Virus

  • Zika virus (ZIKV) is a mosquito borne pathogen, belongs to Flaviviridae family having a positive-sense single-stranded RNA genome, currently known for causing large epidemics in Brazil. Its infection can cause microcephaly, a serious birth defect during pregnancy. The recent outbreak of ZIKV in February 2016 in Brazil realized it as a major health risk, demands an enhanced surveillance and a need to develop novel drugs against ZIKV. Amodiaquine, prochlorperazine, quinacrine, and berberine are few promising drugs approved by Food and Drug Administration against dengue virus which also belong to Flaviviridae family. In this study, we performed molecular docking analysis of these drugs against nonstructural 3 (NS3) protein of ZIKV. The protease activity of NS3 is necessary for viral replication and its prohibition could be considered as a strategy for treatment of ZIKV infection. Amongst these four drugs, berberine has shown highest binding affinity of -5.8 kcal/mol and it is binding around the active site region of the receptor. Based on the properties of berberine, more similar compounds were retrieved from ZINC database and a structure-based virtual screening was carried out by AutoDock Vina in PyRx 0.8. Best 10 novel drug-like compounds were identified and amongst them ZINC53047591 (2-(benzylsulfanyl)-3-cyclohexyl-3H-spiro[benzo[h]quinazoline-5,1'-cyclopentan]-4(6H)-one) was found to interact with NS3 protein with binding energy of -7.1 kcal/mol and formed H-bonds with Ser135 and Asn152 amino acid residues. Observations made in this study may extend an assuring platform for developing anti-viral competitive inhibitors against ZIKV infection.

  • Das Ziel dieser Arbeit liegt darin, den Bestand der wissenschaftlichen Web-Publikationen in der deutschen Linguistik darzustellen. Das Internet bietet heute $f\"{u}r$ die Forschung bereits zwei der wichtigsten produktiven $M\"{o}glichkeiten;n\"{a}mlich$ Information und Kommunikation. Akademische Kreise haben diverse Homepages entwickelt. Der schnelle Zugang zu aktuellen bibliographischen Daten und Forschungsergebnissen hat $f\"{u}r$ koreanische Germanisten einen besonders hohen Stellenwert. Wissenschaftliches Publizieren in Form von Fachzeitschriften ist ein gutes Modell $daf\"{u}r$. Fachzeitschriften erscheinen weltweit und relativ schnell, erreichen aber nur geringe Auflagen. Der Leserkreis ist fast identisch mit der Gruppe der potentiellen Autoren und Herausgeber. Ein Vorteil des elektronischen Publizierens ist die M\"{o}glichkeit$ multimeiale Dokumente und $weiterf\"{u}hrende$ Hyperlinks zu integrieren. Aber die $Qualit\"{a}t\;der\;Aufs\"{a}tze$ kann man kaum objektiv ermitteln und nur schwer beurteilen. Elektronische Zeitschriften $k\"{o}nnen$ sich in der Wissenschaft nur dann etablieren, wenn es gelingt, als wissenschaftliche Arbeiten von den wissenschaftlichen Kreisen oder von der Univerwaltung anerkannt zu werden. 데이터통신 기반 차세대 기지방어작전 모델 개발 (Development of the Data Communication based Future Air Base Defence Operation Model)

  • 기지방어, 대공방어 그리고 화생방방호 모델은 공군의 지상 작전 모델로써 참가 노드간 음성통신 중심의 체계로 설계되어 있어 정확한 의사전달에 문제가 있다. 이러한 문제를 해결하기 위해 최근 국방 분야에서는 데이터통신 기술을 이용하여 지휘통제 능력을 향상시키고 있다. 본 논문에서는 데이터통신에 기반한 차세대 기지방어작전 모델을 통해 기지방어 임무 성능향상을 극대화할 수 있는 새로운 모델을 제안한다. 현재까지 연구된 논문을 참고하여 새로운 모델을 개발하였고, 제안한 기지방어작전 모델을 비행기지내에 기존 정비통제용으로 구축되어 있던 와이브로 네트워크상에서 적용하여 실제실험을 하였다. 새로운 기지방어작전 모델의 분석 및 실험 결과 본 논문에서 제안한 모델이 성능개선에 효과가 있다는 결론에 도달한다.

정수처리에서 응집제 종류와 분리공정이 조류 제거에 미치는 영향 (Effect of Coagulants and Separation Methods on Algal Removal in Water Treatment Process)

  • 본 연구에서는 정수공정에서 응집제 종류와 고액분리방법이 조류제거에 미치는 영향을 조사하였다. 따라서, 원수의 특성을 탁도, $KMnO_4$ 소비량, UV-254, chlorophyll-a 등으로 파악하고 이들 수질인자간의 상관관계를 분석하였다. 또한 시판중인 Alum계열 응집제(Alum, PAC, PACS)가 조류제거에 미치는 영향을 실험실 쟈테스트를 이용하여 탁도 및 유기물제거, pH 저하특성 및 알카리도 소모특성 및 개체수 제거율 등으로 연구하였다. 상관관계 분석에 의하면 유기물항목인 UV-254, $KMnO_4$ 소비량, chlorophyll-a와 탁도의 상관계수는 각각 0.775, 0.674 및 0.623으로 상관성이 매우 높은 것으로 나타났다. 조류발생시 응집제 종류별 응집침전에서 유기물 제거특성은 조류의 농도가 적을 경우에는 유사한 효율을 보였으나 조류농도 증가에 의한 높은 유기물 농도($KMnO_4$ 소비량 20mg/l 이상)와 높은 탁도(100NTU 이상)에서는 PAC, PACS와 같은 무기고분자 응집제가 Alum보다 제거효율이 우수하였다. 조류번성시 응집공정을 거친 후 침전과 부상공정의 비교실험에서 부상분리공정을 사용할 경우 Alum이 모든 분석항목에서 고분자 응집제인 PAC, PACS보다 우수하였으며, 95% 이상의 제거율을 나타내었다. 반면에 PAC와 PACS의 경우는 침전공정이 부상공정보다 효율이 높았으며, $KMnO_4$ 소비량의 경우 부상법 제거율의 2배 이상 효율을 보여주었다. 따라서 조류제거를 향상시키기 위해서는 Alum을 사용할 경우 부상분리, PAC, PACS를 사용할 경우 침전공정이 바람직한 것으로 나타났다. 대상원수에서는 남조식물문의 Microcystic와 녹조식물문의 pediastrum simplex 등 두 가지의 속이 다수 속으로 출현하였으며, Alum, PAC, PACS의 투입후 침전실험에서 27%, 45%, 22%의 조류개체수의 제거율을 보였으나, 용존공기부상법(DAF)을 적용한 경우에는 모든 응집제에서 동, 식물플랑크톤 모두가 100%에 가까운 제거율을 보여주었다.

H2O2 처리된 LLC-PK₁세포에서 Redox Status 및 NF-κB Signaling에 대한 하고초(夏枯草)의 효과 (Effects of Prunellae Herba on the H2O2-Treated LLC-PK Cell's Redox Status and NF-κB Signaling)

  • This study was to investigate the anti-inflammatory effects of Prunellae Herba(PH). The generation of superoxide anion radical (․O2-), nitric oxide (NO), peroxynitrite (ONOO-) and Prostaglandin E₂(PGE2) were measured in the H2O2-Treated renal epithelial cells(LLC-PK1 cell) of mouse. And the effects of Prunellae Spica on the expression of NF-κB (p50, p65), IKK-α, phospho-IκB-α and inflammation-related proteins, COX-2, iNOS, IL-1β and VCAM-1, were examined by western blot. The fluorescent probes, 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein (DAF-2) and dihyldrorhodamine 123 (DHR 123) were used to estimate the scavenging effect of Prunellae Spica on ․O2-, NO, ONOO-. Western blot was conducted to assess the protein expression levels of NF-κB (p50, p65), IKK-α, phospho-IκB-α, inflammation-related proteins, COX-2, iNOS, IL-1β, VCAM-1. PH inhibited H2O2-treated cell death dose-dependently. It reduced the generation of ·O2-, NO, ONOO- and PGE₂ in the H2O2-treated renal epitheial cells(LLC-PK1 cell) of mouse in vitro. PH reduced the expression of NF-κB, IKK-α, phospho-IκB-α, COX-2, iNOS, IL-1β and VCAM-1 genes through means of decreasing activation of NF-κB signaling as well. According to these results, PH has an antioxidative activity and anti-inflammatory effect by regulating the NF-κB pathway. This suggest that PH is expected to be used to regulating inflammatory process and treating inflammation-related disease.

  • Objectives : The aims of this study were to investigate the cytoprotective, antioxidative and inflammation genes inhibitory effects of Patriniae Radix on the mouse LLC-$PK_1$ cells (renal epithelial cells). Methods : The cytoprotective effect of Patriniae Radix was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The antioxidative effect was measured in terms of generation amount of superoxide anion radical (${\cdot}{O_2}^-$) by 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), nitric oxide (NO) by 4,5-diaminofluorescein (DAF-2), peroxynitrite ($ONOO^-$) by dihyldrorhodamine 123 (DHR 123) and prostaglandin $E_2$ ($PGE_2$) by $PGE_2$ immunoassay on $H_2O_2$-treated LLC-$PK_1$ cells. For measuring of inflammation genes inhibitory effects, western blot was performed to detect IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, NF-${\kappa}B$ (p50, p65), COX-2, iNOS, IL-$1{\beta}$ and VCAM-1 protein level in cytosol fractions from LLC-$PK_1$ cells. Results : Patriniae Radix extract reduced the $H_2O_2$-induced cell death and inhibited the amount of $H_2O_2$-induced ${\cdot}{O_2}^-$, NO, $ONOO^-$, $PGE_2$ generation dose-dependently on the mouse LLC-$PK_1$ cells in vitro. Also Patriniae Radix extract inhibited the expression of IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, COX-2, iNOS, IL-$1\beta$ and VCAM-1 genes dose-dependently by means of decreasing activation of NF-${\kappa}B$. Conclusions : According to above results, it was identified that Patriniae Radix had the cytoprotective, antioxidative and inflammation genes inhibitory effects. So it was suggested that Patriniae Radix would be effective to the treatment for the inflammatory process and inflammation-related diseases.

자금정(紫金錠)이 간암세포주 HepG2의 세포고사 및 세포주기에 미치는 영향 (Induction of Apoptosis and Cell Cycle Arrest by Jageum-Jung in HepG2 Hepatoma Cells)

  • Objectives : Jageum-Jung is used as an anti-cancer agent in oriental medicine, but the mechanism by which it induces cell death in cancer cells is still unclear. The purpose of this study was to investigate the effects of Jageum-Jung on apoptosis and cell cycle arrest in HepG2 hepatoma cells. Methods : Various cancer cell lines including HepG2, C6 glioma, SH-SY5Y, PANC-1, and MCF-7 cells, were used. Apoptosis was determined by DAPI nuclei staining and flow cytometry in HepG2 cells treated with various concentrations (from 25 to 200 ${\mu}g/ml$) of $H_2O$ extract of Jageum-Jung (JGJ) for 48 hrs. Expression of cell cycle arrest mediators including Rb, p53, p21, cyclin B1, cdk4, and cyclin E proteins were measured by Western blot analysis. To estimate intracellular hydrogen peroxide levels and intracellular nitric oxide levels, HepG2 cells were stained with DCFH-DA dye and DAF dye, subjected on flow cytometric analysis. Results : 1. Jageum-Jung decreased the viability of HepG2 cells in a dose-dependent manner. 2. Jageum-Jung induced the catalytic activation of caspase-3 in HepG2 cells. 3. Jageum-Jung increased the intracellular hydrogen peroxide and NO in HepG2 cells. 4. Jageum-Jung increased the expression of Rb, p53 and p21 in HepG2 cells. 5. Jageum-Jung induced the expression of cyclin B1, cdk4, and cyclin E in HepG2 cells. Conclusions : Taken together, we suggest that Jageum-Jung exhibits cytotoxic effects on HepG2 cells, causing apoptosis and cell cycle arrest. The results showed that Jageum-Jung may do so by regulating the expression of specific target molecules that promote efficient apoptotic cell death following $G_2$/M phase arrest in a dose-dependent manner.

청혈산(淸血散)이 Redox Status 및 NF-${\kappa}$B 의존성 단백질에 미치는 영향 (Effects of Cheonghyul-San on the Generation of Redox Status and on the Expression of NF-${\kappa}$B Dependent Proteins)

  • The aim of this study was to investigate the effects of Cheonghyul-san on the generation of peroxynitrite ($ONOO^-$), nitric oxide (NO) and superoxide anion radical ( ${\cdot}\;O_2^-$), and on the expression of NF-${\kappa}$B-dependent proinflammatory proteins in ob/ob mice. Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57BL/6J black mice) and control obese (ob/ob mice) groups have received the standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein-2 (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blot was performed using anti-IKK-${\alpha}$, anti-phospho I${\kappa}$B-${\alpha}$, anti-NF-${\kappa}$B (p50, p65), anti-COX-2, anti-iNOS, anti-VCAM-1 antibodies, respectively. Cheonghyul-san prevented $H_2O_2$-induced cell death. Cheonghyul-san inhibited the generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ in the $H_2O_2$-treated LLC-$PK_1$ cells. The generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ were inhibited in the Cheonghyul-san-administered ob/ob mice groups. The GSH/GSSG ratio was decreased in the ob/ob mice, whereas the ratio was improved in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the protein expression levels of phospho-I${\kappa}$B-${\alpha}$, IKK-${\alpha}$, NF-${\kappa}$B (p50, p65), COX-2, iNOS and VCAM-1 genes. These results suggest that Cheonghyul-san is an effective scavenger of $ONOO^-$, ${\cdot}\;O_2^-$ and NO, and has an inhibitory effect on the expression of NF-${\kappa}$B-dependent inflammatory genes in ob/ob mice. Therefore, Cheonghyul-san might be used as a potential therapeutic drug against the diabetes- and obesity-related proinflammatory diseases.

  • Peroxynitrite ($ONOO^-$), superoxide anion radical (${\cdot}\;{O_2}^-$) and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging processes, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer, diabetes, obesity and atherosclerosis. The aim of this study was to investigate the effects of Ojunghwan on the generation of peroxynitrite ($ONOO^-$), nitric oxide (NO) and superoxide anion radical (${\cdot}\;{O_2}^-$), and on the expression of $NF-{\kappa}B$-dependent inflammatory proteins in ob/ob mice. Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57/BL6J black mice) and control obese (ob/ob mice) groups have received the standard chow. The experimental groups were fed with a diet of chow supplemented with 30 and 90 mg Ojung-hwan per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blot was performed using anti-phospho $I{\kappa}B-{\alpha}$, $anti-IKK-{\alpha}$, $anti-NF-{\kappa}B$ (p50, p65), anti-COX-2, anti-iNOS, anti-VCAM-1 and anti-MMP-9 antibodies, respectively. Ojunghwan inhibited the generation of $ONOO^-$, NO and ${\cdot}\;{O_2}^-$ in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondrial fraction in vitro. The generation of $ONOO^-$, NO, ${\cdot}\;{O_2}^-$ and $PGE_2$ were inhibited in the Ojunghwan-administered ob/ob mice groups. The GSH/GSSG ratio was decreased in the ob/ob mice, whereas that were improved in the Ojunghwan-administered groups. Ojunghwan inhibited the expression of $phospho-I{\kappa}B-{\alpha}$, $IKK-{\alpha}$, $NF-{\kappa}B$ (p50, p65), COX-2, iNOS, VCAM-1 and MMP-9 genes. These results suggest that Ojunghwan is an effective scavenger of $ONOO^-$, ${\cdot}\;{O_2}^-$, NO and $PGE_2$, and has an inhibitory effect on the expression of $NF-{\kappa}B$-dependent inflammatory genes in ob/ob mice.
