Histological changes was investigated in the 4 weeks old rat brain using NMDA (N-methyl-D-asparate) which is capable of mediating excitotoxic events. The changes were occured when the injected NMDA solved in PBS was over $1.0{\mu}g/g$(about 90nM). The necrosis of Purkinje cells in cerebellum and the increasement of coloidal plexus cell number were prevalent. The Purkinje cell number of necrosis were increased according to increasement of amount of injected NMDA. In spite of increasement of degenerated Purkinje cell number, differentiation of new Purkinje cell was not identified because total number of Purkinje cell was not changed. The change of cell number was observed in coloidal plexus cell rather than degeneration of cell. About 5 time increasement was occured. This change may cause increasement of cerebrospinal fluid and the makes mophorogy of brain more round than nomal.
Automatic segmentation of brain MRI data usually leaves some segmentation errors behind that are to be subsequently removed interactively, using computer graphics tools. This interactive removal is normally performed by operating on individual 2D slices. It is very tedious and still leaves some segmentation errors which are not visible on the slices. We have proposed to perform a novel 3D interactive correction of brain segmentation errors introduced by the fully automatic segmentation algorithms. We have developed the tool which is based on 3D semi-automatic propagation algorithm. The paper describes the implementation principles of the proposed tool and illustrates its application.
The purpose of the present study was to explore the proper method for animal stroke model of Oriental medicine To this end, brain ischemia was induced by distal middle cerebral artery occlusion(dMCAO) and proximal middle cerebral artery occlusion(pMCAO) and evaluated with the method of Triphenyl Tetrazolium Chloride (TTC) staining and Swimming Behavior Test. Results demonstrated that first, infarct size and volume of pMCAO group were significantly bigger that those of dMCAO group. Second, analysis of swimming behavior test revealed that the percentage of left turning angles of pMCAO was significantly bigger than that of dMCAO. Third, during swimming behavior test, there were peculiar traces of small successive circles that represent motor dysfunction and conscious disturbance among dMCAO group. The results of the study thus indicate that non-invasive intraluminal method of pMCAO was the appropriate animal stroke model for Oriental medicine in the light of brain ischemia as hemiplesia and conscious disturbance.
We developed an experimental model of brain death using dogs. Brain death was caused by increasing the intracranial pressure[ICP suddenly by injecting saline to an epidural Foley catheter in five female mongrel dogs[weight, 20-25Kg .Hemodynamic and electrocardiographic changes were evaluated continuously during the process of brain death. 1. Abrupt rise of ICP after each injection of saline followed by a rapid decline to a new steady-state level within 15 minutes and the average volume required to induce brain death was 7.6$\pm$0.8ml.2. Body temperature, heart rate, mean pulmonary arterial pressure, left ventricular[LV enddiastolic pressure and cardiac output was not changed significantly during the process of brain death, but there was an increasing tendency.3. Mean arterial pressure and LV maximum +dP/dt increased significantly at the time of brain death.4. Hemodynamic collapse was developed within 140 minutes after brain death.5. Marked sinus bradycardia followed by junctional rhythm was seen in two dogs and frequent VPB`s with ventricular tachycardia was observed in one dog at the time of brain death. Hyperdynamic state develops and arrhythmia appears frequently at the time of brain death. Studies on the effects of brain death on myocardium and its pathophysiologic mechanism should be followed in the near future.
목적: 3T 장비의 신생아 뇌자기공명영상에서 T1 강조 고속 역전회복기법 (fast inversion recovery, FIR)과 자기화 삼차원 경사에코기법, (magnetization-prepared three dimensional gradient echo sequence, 3D GRE)을 스핀에코기법 (SE)과 비교하여 유용성을 알아보는 데 있다. 대상 및 방법: 20명의 신생아에서 T1 강조 SE, FIR, 그리고 3D GRE의 신호소음비 (SNR)와 대조소음비 (CNR)를 측정하고 시각적으로 회백질-백질 구별, 수초화 인식, 인공음영 발생을 점수화하여 비교하였다. 각 영상기법의 CNR 비교에는 Wilcoxon signed ranked test를 사용하였다. 결과: 세가지 영상기법 중 3D GRE가 가장 우수한 SNR을 보였고 CNR은 FIR과 3D GRE 모두 SE보다 우수하였으며 FIR보다 3D GRE가 더 우수하였다. 회백질-백질의 구분과 수초화 유무 역시 스핀에코보다 FIR과 3D GRE에서 더 잘 보였다. 그러나 3D GRE는 움직임에 의한 인공음영이 많았고 FIR에서 혈류에 의한 혈관의 고신호강도가 자주 발견되었다. 결론: 3T 장비에서 신생아 뇌영상을 얻을 때 FIR과 3D GRE 기법은 SE보다 좋은 T1 강조영상을 제공할 것으로 기대된다.
The effects of the protein fraction of Panax ginseng on primary cultured chicken embryonic brain cells and DRG cultured with a deficient medium were studied. The protein fraction was further fractionated into four groups according to the molecular weight; larger than 10,000 dalton(fraction A), between 5,000 and 10,000 daltons(fraction B), between 1,000 and 5,000 daltons(fraction C), between 500 and 1,000 daltons(fraction D). All four protein fractions at the concentration of $100\;{\mu}g/ml$ significantly increased the number of the brain cells which promoted the neurite outgrowth. The activity of PDHC in the brain cells was elevated significantly by the protein fraction B at the concentration of $100\;{\mu}g/ml$. It was noted that $100\;{\mu}g/ml$ protein fraction C and D significantly enhanced the synthesis of protein in the brain cells. At the concentration of $100\;{\mu}g/ml$, the protein fraction B enhanced RNA synthesis and the protein fraction A significantly enhanced DNA synthesis in the brain cells. The protein fractions B, C, and D significantly promoted the neurite outgrowth of DRG at the concentration of $100\;{\mu}g/ml$.
BACKGROUND/OBJECTIVES: Brain aging is a major risk factor for severe neurodegenerative diseases. Conversely, L-histidine and L-carnosine are known to exhibit neuroprotective effects. The aim of this study was to examine the potential for L-histidine, L-carnosine, and their combination to mediate anti-brain aging effects in neuronal cells subjected to D-galactose-induced aging. MATERIALS/METHODS: The neuroprotective potential of L-histidine, L-carnosine, and their combination was examined in a retinoic acid-induced neuronal differentiated SH-SY5Y cell line exposed to D-galactose (200 mM) for 48 h. Neuronal cell proliferation, differentiation, and expression of anti-oxidant enzymes and apoptosis markers were subsequently evaluated. RESULTS: Treatment with L-histidine (1 mM), L-carnosine (10 mM), or both for 48 h efficiently improved the proliferation, neurogenesis, and senescence of D-galactose-treated SH-SY5Y cells. In addition, protein expression levels of both neuronal markers (β tubulin-III and neurofilament heavy protein) and anti-oxidant enzymes, glutathione peroxidase-1 and superoxide dismutase-1 were up-regulated. Conversely, protein expression levels of amyloid β (1-42) and cleaved caspase-3 were down-regulated. Levels of mRNA for the pro-inflammatory cytokines, interleukin (IL)-8, IL-1β, and tumor necrosis factor-α were also down-regulated. CONCLUSIONS: To the best of our knowledge, we provide the first evidence that L-histidine, L-carnosine, and their combination mediate anti-aging effects in a neuronal cell line subjected to D-galactose-induced aging. These results suggest the potential benefits of L-histidine and L-carnosine as anti-brain aging agents and they support further research of these amino acid molecules.
CT는 인체 내 장기의 해부학적 구조를 정확하게 나타낼 수 있으며, 영상의 분해능이 우수하다. Brain CT 검사 시 수정체의 방사선 감수성이 높아서 피폭의 영향을 많이 받는다. 본 연구는 수정체에 피폭선량을 감소하기 위해 비스무스와 텅스텐 필라멘트 차폐물질을 사용하여 Non-shield와 Shield에 대한 피폭선량의 변화와 차폐율을 비교 하고자 한다. 본 연구에서는 3D printing으로 제작한 비스무스, 텅스텐 필라멘트 차폐물질을 사용하여, 차폐물질 두께와 slice에 따라 피폭되는 선량을 측정하였다. 헤드팬텀을 고정시켜 안구에 Magicmax universal 선량계를 위치시켜 차폐 물질을 놓지 않았을 경우와 그 위에 차폐 물질을 놓았을 때 차폐율을 각각 비교하기 위해 두 물질을 1mm ~ 5mm 두께로 각각 측정하였다. 1 mm 두께의 필라멘트에서 비스무스 필라멘트는 26.8 %, 텅스텐 필라멘트는 43.1 % 차폐율이 나타났다. 따라서 비스무스 필라멘트보다 텅스텐 필라멘트에서 더 큰 차폐효과가 나타났다. 차폐체의 종류, 두께, 슬라이스 간격에 따라 선량을 측정한 결과, 비스무스 필라멘트 보다 텅스텐 필라멘트가 더 큰 차폐효과가 나타났다.

Depression is one of the most common neuropsychiatric disorders and is associated with dysfunction of the neuroendocrine system and alterations in specific brain proteins. Boophone disticha (BD) is an indigenous psychoactive bulb that belongs to the Amaryllidacae family, which is widely used in Southern Africa to treat depression, with scientific evidence of potent antidepressant-like effects. The present study examined the antidepressant effects of BD and its mechanisms of action by measuring some behavioural parameters in the elevated plus maze, brain content of corticosterone, brain derived neurotropic factor (BDNF), and neuroblast differentiation in the hippocampus of Balb/c mice exposed to the five day repeated forced swim stress (5d-RFSS). Male Balb/c mice were subjected to the 5d-RFSS protocol to induce depressive-like behaviour (decreased swimming, increased floating, decreased open arm entry, decreased time spent in the open arms and decreased head dips in the elevated plus maze test) and treated with distilled water, fluoxetine and BD. BD treatment (10 mg/kg/p.o for 3 weeks) significantly attenuated the 5d-RFSS-induced behavioural abnormalities and the elevated serum corticosterone levels observed in stressed mice. Additionally, 5d-RFSS exposure significantly decreased the number of neuroblasts in the hippocampus and BDNF levels in the brain of Balb/c mice, while fluoxetine and BD treatment attenuated these changes. The antidepressant effects of BD were comparable to those of fluoxetine, but unlike fluoxetine, BD did not show any anxiogenic effects, suggesting better pharmacological functions. In conclusion, our study shows that BD exerted antidepressant-like effects in 5d-RFSS mice, mediated in part by normalizing brain corticosterone and BDNF levels.
We have developed a 3-D image processing and display technique that include image resampling, modification of MIP, and fusion of MIP image and volumetric rendered image. This technique facilitates the visualization of the three-dimensional spatial relationship between vasculature and surrounding organs by overlapping the MIP image on the volumetric rendered image of the organ. We applied this technique to a MR brain image data to produce an MRI angiogram that is overlapped with 3-D volume rendered image of brain. MIP technique was used to visualize the vasculature of brain, and volume rendering was used to visualize the other structures of brain. The two images are fused after adjustment of contrast and brightness levels of each image in such a way that both the vasculature and brain structure are well visualized either by selecting the maximum value of each image or by assigning different color table to each image. The resultant image with this technique visualizes both the brain structure and vasculature simultaneously, allowing the physicians to inspect their relationship more easily. The presented technique will be useful for surgical planning for neurosurgery.
