Dibutyryl Cyclic AMP가 생쥐여포난자의 성숙에 미치는 억제효과에 관한 자기방사법적 연구 (Autoradiographic Studies on the Inhibitory Effect of Dibutyryl Cyclic AMP on Mouse Oocyte Maturation in Vitro)

  • This experiment was undertaken in order to localize the labeled dbcAMP (dibutyryl cyclic AMP) in oocytes whose development has been suppressed by cold dbcAMP for 6 or 19 hours in vitro. Mouse oocytes were obtained from the ovaries of 3-4 week old A strain female mice, by puncturing the Graafian follicles in the modified Krebs-Ringer bicarbonate salt solution under the dissecting microscope. Those oocytes which have intact germinal vesicle were cultured in the basic culture medium supplemented with 0.4% bovine serum albumin (BSA). Cultivation of the oocytes was carried out in a microtube developed by Cho (1974). The cultures were then incubated in a humidified 5% $CO_2$ incubator maintained at $37^{\circ}C$ for 6 or 19 hours (Donahue, 1968). DbcAMP was added to culture medium for a final concentration of 100ug/ml, and $^3H-dbc$ AMP (specific activity 13 Ci/mM) for a final concentration of $40{\mu}Ci/ml$ was also added to the medium. For electron microscopic autoradiography, those oocytes recovered from the culture were washed with phosphate buffer (pH 7.4), and immediately prefixed in a 2.5% glutaraldehyde overnight and postfixed for 2 hours at $4 ^{\circ}C$ in 1% osmium tetroxide in phosphate buffer with pH 7.4 (Palade, 1952). After fixation, the materials were dehydrated in graded alcohol series and embedded in Epon 812 mixture based on the standard procedures (Luft, 1961). The thin sections $600-700{\AA}$ thick were mounted on the grids of 200 meshes. The grids containing sections were coated with a nuclear emulsion Kodak NTB-3 and stored in a cold dark box (at $4^{\circ}C$) for 3 weeks. After exposure, the samples were developed with Kodak D-19 and stained with uranyl acetate and lead citrate. Routine observation was made with Hitachi HU-11E electron microsocope. The results of the observation were as followings: 1. It was found that the labeled dbcAMP penetrated the egg plasma membrane and dispersed at random in the cytoplasm. 2. It was also observed that most of the labeled dbcAMP was attached to microfibrillar lattices portion of the oocyte cytoplasm. There fore, it is presumed that the receptor of the dbcAMP is localized in the microfibrillar lattices of the oocyte. 3. It also seems that some other cell organells such as mitochondria, Golgi complex, cortical granules are not directly related to the action of the dbcAMP. 4. The labeled dbcAMP was neither observed in the membrane nor in the nucleus. Therefore, it seems that there is no relationship between the concentration of dbcAMP and the nuclear membranous permeability. 5. There was no difference in number of dbcAMP particles when oocytes were cultured for 6 hours and 19 hours. 6. However, it was observed that, in same of the oocytes suppressed in germinal vesicle by dbcAMP for 19 hours, cell organells were moved and concentrated to a small portion of the cytoplasm, and that the morphology of the organells greatly changed to an abnormal. form. Therefore, it is supposed that those oocytes were in the process of degeneration. From the above results, it is expected that dbcAMP penetrated the egg membrane and was bound to the receptor which seems to be located in the microfibrillar lattiees portion, and that this dbcAMP-receptor complex inhibited some enzyme system of the oocytes which are essential for the germinal vesicle breakdown.

돼지 난모세포의 체외 성숙 후 극체 방출 및 미방출란의 핵형과 배발달율 (Comparison of Nuclear Status and Developmental Potential between Polar Body Extruded Oocytes and Non-extruded Oocytes on in vitro Maturation and Development of Porcine Follicular Oocytes)

  • 본 연구에서 돼지 난포란에서 채취된 난모세포들을 체외 성숙 후 세포 손상이 없이 성숙 난모세포의 발생능을 알아낼 수 있는 마커로 극체의 방출이 효과적으로 활용될 수 있는지를 알아보았다. 난모세포를 48시간 성숙 배양 후 극체의 방출 유무를 검사하고, 핵염색하여 염색체의 형태를 검사하였다. 확인된 난모세포들을 $16{\sim}18$시간 추가 배양한 후 7% ethanol로 활성화시키고 $5{\mu}g/ml$ cytochalasin B에 5시간 노출 후 NCSU23 배양액으로 7일간 배양하였다. 극체 방출율은 반복에 따라 $9.9{\sim}52.4%$, 퇴행율은 $21.4{\sim}61.8%$로 변이가 크게 나타났다(p<0.01). 극체를 방출한 난모세포의 핵상은 모두 극체와 19개의 염색체를 가진 제 2 감수분열 중기 핵상을 보여주었으며, 극체를 방출하지 못한 난모세포의 핵상은 핵이 팽화된 상태인 핵형이 39.1%, PCC 형태의 핵상이 19.6%, MI 형태의 핵상이 10.9%, MII이지만 극체가 관찰되지 않거나 매우 작은 상태인 경우가 13%, 핵이 응축된 형태인 경우가 6.5%, 핵이 없는 경우가 8.7%로 나타났다. 퇴행란으로 판단한 난모세포들은 핵염색을 한 결과 역시 세포질 상태가 정상적이지 못한 염색 상태를 보여주었다. 극체 방출 유무를 확인하지 않고 활성화 처리 후 배양하였을 때 분할율은 45.0%, 배반포기까지 발달율은 11.3%였으나, 극체 방출란만을 모아서 활성화처리를 하였을 때 분할율은 94.2%, 배반포기까지 발달율은 42.5%로 급격하게 향상되었다. 이상의 결과로 퇴행란과 극체 미방출란을 제거하고 실험에 활용한다면 배양 효과를 확인하거나 배아 생명 공학에 활용할 때 좀더 유리 할 것으로 사료된다.

결장암 및 직장암에서 암유전자 산물의 발현 (Expression of Oncogene Product in the Colorectal Carcinoma)

  • 1983년 6월부터 1993년 5월까지 10년간 영남대학교 의과대학 부속병원에서 결장 및 직장암으로 절제되어, 병리조직학적으로 결장 및 직장암으로 확진된 예 중, 파라핀 포매조직의 상태가 양호한 선암종 67례를 대상으로 하여 면역조직화학적 방법을 이용해서 대장암에서의 $p62^{c-myc}$$p21^{ras}$의 발현양상을 검색함으로써 종양유전자 산물과 대장암의 유형, 분화도 및 Dukes stage에 따른 연관성 유무를 관찰한 바 다음과 같은 결과를 얻었다. 결장 및 직장암에서 $p62^{c-myc}$ 발현은 직장암에서 더 많았으며 그 양상은 주로 미만성 반응이 많았고(p<0.05), $p21^{ras}$ 발현은 여자에서 더 많았다(p<0.05). 분화도에 따라서는 고분화 선암종에서 미만성으로 나타났고 강양성 반응의 경향을 보였다. 수정된 Dukes stage와 두 종양유전자 산물의 발현은 비교적 초기에 미만성으로 나타났다. 종양유전자 $p62^{c-myc}$의 발현은 전이된 림프절에서 원발병소보다 미만성, 강양성으로 더 많이 관찰 되었고, $p21^{ras}$의 발현은 원발병소에서 더 양성반응이 많았고 주로 미만성, 강양성으로 나타나는 경향이었다. 환자의 나이, 종양의 육안소견과는 통계학적 유의성이 없었다. 이상의 연구를 요약하면 종양유전자 산물 $p62^{c-myc}$$p21^{ras}$의 발현은 대장암 초기 및 고분화 선암종에서 더 많이 나타나는 것으로 생각되었고 환자의 나이, 종양의 육안소견과는 관련이 없는 것으로 사료되었다.

$[^{11}C]Raclopride$ PET을 이용한 흡연에 의한 도파민 유리 영상 연구 (Smoking-Induced Dopamine Release Studied with $[^{11}C]Raclopride$ PET)

  • 목적: 흡연에 보상 강화 효과는 흡연 시에 담배 내에 포함되어 있는 니코틴 성분에 의한 선조체에서의 도파민 유리가 중심 역할을 할 것으로 생각된다. 지금까지의 여러 동물실험 에서 니코틴과 도파민의 상호 관계와 니코틴에 의한 도파민 유리가 연구되었다. 이 연구에서는 $[^{11}C]raclopride$ PET을 이용하여 생체에서 흡연에 의한 도파민 유리를 영상화 하고자 하였다. 대상 및 방법: 비흡연자이거나 과거 흡연력이 있으나 1년 이상의 금연을 시행한 5 명의 정상인 남자를 대상으로 하였고, 이들의 평균 연령은 24세 이었다. 도파민 D2 수용체 영상을 위한 방사성 리간드인 $[^{11}C]raclopride$를 볼루스+연속 주입법에 의하여 주사하면서 120 분간 30개의 동적 영상($3{\times}20$초, $2{\times}60$초, $2{\times}120$초, $1{\times}180$초, $22{\times}300$초)을 얻었다. 영상 촬영 시작 50분에 니코틴 함량 1mg의 담배를 피도록 하였으며, 담배를 피우기 직전과 흡연시작 5분 후부터 흡연에 의하여 흡수된 혈중 니코틴 농도를 측정하기 위하여 일정 간격으로 정맥혈 샘플을 획득하였다. 30 개의 프레임은 인접 프레임과의 정합에 의하여 움직임을 보정하였고, 움직임이 보정된 120분 간의 동적 영상을 합하여 평균 영상을 만든 다음, 뇌 MRI와 공간 정합을 하였다. 평균 영상과 MRI 의 공간 정합 정보를 이용하여, 각 프레임을 MRI 에 공간 정합시켜, 공간 정합된 동적 영상에서 선조체에 MRI 정보를 이용하여 좌우 각각 3 개의 관심 영역(ventral striatum : VST, precomissural dorsal caudate; caudate nucleus; precommissural putamen; anterior putamen)과 소뇌에 관심영역을 설정하였고, 동적 영상으로부터 각 관심 영역 별로 시간-농도 곡선을 구하였다. $[^{11}C]raclopride$주사 후 선조체에서의 리간드 특이적 결합에 의한 항정상태(statedy state) 에 도달한 후 흡연전 평형 상태(equilibrium state)인 30-50 분과 흡연 후 재평형에 도달한 70-90분 영상에서 각 관심 영역에서의 방사성 농도를 구하였고, 조직비 방법에 근거하여 기저상태 및 흡연 상태의 방사성 리간드의 수용체 결합능 (binding potential;BP)을 구하였다($BP=C_{ROI}/C_{cerebellum}-1$). 흡연에 의한 도파민의 유리는 흡연 전후의 $[^{11}C]raclopride$의 수용체 결합능의 변화율로 계산하였다. 흡연에 의한 혈중 니코틴의 상승은 흡연후 90 분간의 혈중 니코틴의 축적 농도로 계산되었으며, 흡연에 의한 $[^{11}C]raclopride$의 수용체 결합능의 감소율과 혈중 니코틴의 축적 농도와의 상관관계를 스피어만 상관분석법(Spearman's correlation)에 의하여 알아보았다. 결과: 흡연에 의한 선조체에서의 평균 $[^{11}C]raclopride$의 수용체 결합능의 변화는 미상핵에서 4.7%, 전피각에서 4.0%, 복측 선조체에서 7.8% 의 감소를 보여 흡연에 의한 선조체내 도파민 유리를 정량화 하였다. 특히 선조체에서의 도파민 유리에 의한 수용체 결합능의 감소는 흡연에 의한 혈중 니코틴의 축적 농도와 양의 상관관계를 보였다(rho=0.9, p=0.04). 결론: $[^{11}C]raclopride$ PET을 이용하여 비흡연 정상인에서 흡연에 의한 도파민 유리를 영상화 및 정량화 하였고, 흡연에 의한 선조체내 도파민 유리는 흡연시 흡수된 니코틴의 축적 농도와 상관관계를 가짐을 보였다. 이 연구에서의 확립된 방법과 결과는 앞으로 흡연자에서 니코틴에 의한 도파민 신경계의 활성화 연구에 기여할 것이다.

비소세포폐암에서 p53유전자의 구조적 이상 및 단백질 발현이 예후에 미치는 영향 (Correlation of p53 Protein Overexpression, Gene Mutation with Prognosis in Resected Non-Small Cell Lung Cancer(NSCLC) Patients)

  • 연구배경 : p53 유전자는 염색체 17p에 존재하는 종양억제유전자인데, 돌연변이가 있는 경우의 mutant protein은 그 반감기가 4~8시간으로 wild type protein의 반감기 6~20분에 비하여 현저한 증가를 보이게 된다. 결과적으로 돌연변이가 있는 경우 p53 단백질이 과축적되어 과발현이 유발되는데, 이러한 기전을 이용하여 p53 면역조직화학염색은 p53유전자의 돌연변이의 검색 방법으로 많이 이용되고 있다. 유방암에서는 p53 핵단백질의 과발현이 있는 경우에 없을 때보다 무병생존기간 및 전체 생존기간 모두에서 유의하게 나쁜 임상 경과를 취함이 보고되고 있지만 폐암에서는 p53 유전자의 돌연변이 유무, 또는 p53단백의 과발현 유무가 예후에 미치는 영향은 아직까지 확실하지 않다. 저자 등은 한국인의 폐암에서 p53 종양억제 유전자의 돌연변이 빈도를 확인하고 이들이 폐암 환자의 임상 경과에 미치는 영향을 분석하며, 면역조직화학염색 및 PCR-SSCP 분석의 감수성과 특이성을 염기서열분석과 비교하여 확인하고자 본 연구를 시행하였다. 방법 : 근치적 폐절제술 후 임상경과를 추적중인 원발성폐암환자 75예를 대상으로 하였으며, 면역조직화학검사 및 분자생물학적 연구를 위하여는 이들의 종양조직(paraffin block)을 이용하였다. p53 단백질의 면역조직화학검사를 위한 일차항체로는 DO7(Novocastra, U.K.)을 사용하였고, PCR-SSCP 및 염기서열분석 등을 위하여는 파라핀 포매조직에서 암조직을 선택적으로 박절하여 DNA를 추출하여 이용하였다. 결과: 1) 전체 75예 폐암환자의 면역조직화학염색 결과 27예(36%)에서 p53단백질의 과발현을 보였다(Table 2.3, Fig 1). 2) 전체 환자의 p53 핵단백질 과발현 여부에 따른 중앙전체생존기간은 p53 음성군과 양성군 모두 25개월, 무병생존기간은 두군 모두 13개월로 동일하였다(Fig. 2). 3) PCR-SSCP 분석에 의한 p53 유전자의 mobility shift는 시험한 58예중 16예(27.6%)에서 확인되었다(Fig 3). Mobility shift 유무에 따른 중앙전체생존기간은 shift가 있는 군과 없는 군에서 각각 27개월과 20개월, 무병생존기간은 각각 8개월, 10개월로 유의한 차이가 없었다. 4) 염기서열분석을 통한 p53 유전자의 돌연변이는 시험한 29예중 10예(34.5%)에서 확인되었다(Fig 4). 염기서열분석 결과 돌연변이 유무에 따른 중앙전체생존기간은 돌연변이가 있었던 군과 없었던 군에서 각각 27개월, 22개월이었으며, 무병생존기간은 각각 20개월과 10개월로 유의한 차이는 없었으나 돌연변이가 있는 경우 조기재발을 하는 경향을 보였다(Fig 5, 6). 5) 면역조직화학염색은 PCR-SSCP 결과를 기준으로 할 때 민감도 67.0%, 특이도 74.0%, 그리고 일치도는 62.5% 이었다. PCR-SSCP의 결과는 염기서열분석 결과를 기준으로 할 때 민감도 91.8%, 특이도 96.2%, 일치도는 95.3% 이었다. 결론 : p53 핵단백질의 과발현 정도, PCR-SSCP 및 염기서열분석상 돌연변이 유무는 비소세포폐암환자의 근치적수술후 예후의 예측지표로서는 그 효용성이 적었으나, 돌연변이가 있는 경우 조기재발을 하는 경향을 보였다. 또한 p53유전자의 돌연변이 검색법으로는 면역조직화학염색보다는 PCR-SSCP법이 우수하였다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. The system based on PC-486DX was designed to acquire medical images produced by medical imaging instruments and related audio information, that is, doctors' reporting results. Input information was processed and analyzed, then the results were presented in the form of graph and animation.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.
