Immunofluorescence Microscopy and Biochemical Characterization of Two Nuclear Envelope Proteins of Amoeba proteus by Using a Monoclonal Antibody (단항체를 이용한 아메바(Amoeba proteus) 의 2종 핵막 단백질에 대한 면역형광현미경적 및 생화학적 특성 조사)

  • Distribution of the antigens during the cell cycle of amoebae was followed by immunof-luorescence microscopy using a monoclonal antibody against the nucleus as a probe. While the cells were in the interphase, the antigen was localized on the nucleus membrane. But it was dispersed all over the cytoplasm during mitosis and cytokinesis. The molecular weights of the immunoreacted antigens were 210 KD and 190 KD as determined by SDS PAGE and western blotting of the purified nuclei. The antigens were not soluble in non-ionic detergent, but were released from the nucleus by incubation with 0.05 M sodium carbonate, pH 10.6 or with 8 M urea at serial chemical extraction. Thus the antigens appeared to be peripheral proteins of the nurBeus envelope. The isoelectic point of both antigens was 7.64 as determined by 2 D PAGE and transfer blotting. Considering the peiipherd association with the nucleus membrane and the dispersed distribution during mitosis, the antigens could be lamin like proteins. Hourever, it appears also possible that they are the component molecules of the unusually structured aurous lamina of amoeba nucleus since they have the large molecular weight and the basic pl.

Influences of ethanol and temperature on sucrose-evoked response of gustatory neurons in the hamster solitary nucleus

  • Taste-responsive neurons in the nucleus of the solitary tract (NST), the first gustatory nucleus, often respond to thermal or mechanical stimulation. Alcohol, not a typical taste modality, is a rewarding stimulus. In this study, we aimed to investigate the effects of ethanol (EtOH) and/or temperature as stimuli to the tongue on the activity of taste-responsive neurons in hamster NST. In the first set of experiments, we recorded the activity of 113 gustatory NST neurons in urethane-anesthetized hamsters and evaluated responses to four basic taste stimuli, 25% EtOH, and 40℃ and 4℃ distilled water (dH2O). Sixty cells responded to 25% EtOH, with most of them also being sucrose sensitive. The response to 25% EtOH was significantly correlated with the sucrose-evoked response. A significant correlation was also observed between sucrose- and 40℃ dH2O- and between 25% EtOH- and 40℃ dH2O-evoked firings. In a subset of the cells, we evaluated neuronal activities in response to a series of EtOH concentrations, alone and in combination with 32 mM sucrose (EtOH/Suc) at room temperature (RT, 22℃-23℃), 40℃, and 4℃. Neuronal responses to EtOH at RT and 40℃ increased as the concentrations increased. The firing rates to EtOH/Suc were greater than those to EtOH or sucrose alone. The responses were enhanced when solutions were applied at 40℃ but diminished at 4℃. In summary, EtOH activates most sucrose-responsive NST gustatory cells, and the concomitant presence of sucrose or warm temperatures enhance this response. Our findings may contribute to elucidate the neural mechanisms underlying appetitive alcohol consumption.

Studies on the nuclear cytology of Pyricularia oryzae CAV (도열병균의 핵학적 연구)

  • Results obtained from the observation of 1,000 matured resting conidia of Pyricularia oryzae CAV. were as fellows. The percentage of a condium conitaining was 95.5, while that of a multinucleate conidium was 4.5. Formation of the three-celled mature conidium containing a nucleus in each cell, which was derived from the immatured conidium, could be recognized. Chromosomes could be observed in the conidium when the nuclear division took place. The number of chromosomes was n=3, 4, 5 or 6 but rile majority was 5. The nucleus was moved into the germ-tube from the conidium by following ways: (a) a resting nucleus divided into two nuclei in the conidium and one of them was moved into the germ-tube. The other nucleus remained in the conidium (43 per cent): (b) a nucleus was moved into the germ-tube from the conidium without nuclear division (57 Per cent). The appressorium without a nucleus was abundantly observed when stain was made. However, tile number of a mononucleate appressorium was 476 out of 500 appressoria which had nuclear sap. On the ground of this experiment, we could support the conidium of blast fungus contained mononucleus and also homokaryon seemed to b: appealed on the conidia even though those were multilocular. As the results, it could be concluded that use of inoculum derived from single spore isolate was reasonable for any experiments in the rice blast fungus.

  • To investigate the process of spermiogenesis of the Korean eastern Daubenton's bat, Myotis daubentonii ussuriensis, the testis obtained from mature male bats was studied by transmission electron microscope and were based on the variety and diagnostic characters of cell organells. The results obtained from the present study are as follows. According to the differentiation of the cell organells, the spermiogenesis of the Korean eastern Daubenton's bat, M. d. ussuriensis, was divided into Golg, cap, acrosome, maturation and spermiation phases. Besides, these Golgi, cap, acrosome, and maturation phase were subdivided into the steps of early and late phases repectively and matruation phase was subdivided into step of early, mid and late phases. Therfore, the spermiogenesisof M. d. ussuriensis has been divided into a total of 11 phases. The chromatin granules began to condense at the early cap phase, regularized at the acrosome phase, and a perfect nucleus of sperm was formed at the maturation phase. The chromatoid body was occurred in the upper cytoplasm of nucleus at the early Golgi phase, and it was accurred the posterior cytoplasm of the nucleus at the early maturatio phase. The formation of sperm tail began to be develop in the early golgi phase, and completed at the spermiation phase. The fiber structure of middle piece was consisted of nine outer doublets and two central singlet microtubules and Nos. 1, 5, 6 and 9 in the outer dense were larger than the others(2, 3, 4, 7, 8).

  • To investigate the degeneration process in the intervertebral disc, a three dimensional (3D) poroelastic finite-element (FE) model was developed. Disc was modeled as two different regions, such as annulus modeled with fiber reinforced 20 node poroelastic ground matrix and nucleus having large porosity. Excess Von Mises stress in the disc element assumed to be a possible source of degeneration under compressive loading condition. Recursive calculation was continued until the desired convergence was attained by changing the permeability and porosity of those elements, which could be predicted from the previous iteration. The degenerated disc model showed that relatively small compressive stresses were generated in the nucleus elements compared to normal disc. Its distribution along the sagittal plane was matched well with a previously reported experimental result. Contrasts to this result, pore pressures in the nucleus were higher than those in the normal disc. Total stress indicated similar values for two different models. This new approach using poroelastic modeling could provide the explanation of the interaction between fluid and solid matrix in the disc during the degeneration process.

Towards Designing Environmentally Stable Conjugated Polymers with very Small Band-Gaps

  • We have investigated substituent effect on the stabilization energies, and nucleus-independent chemical shifts of pentafulvalenes and on the electronic structures of the corresponding polypentafulvalenes to design environmentally stable semiconductive or conductive polymers. Geometrical optimizations of the molecules were carried out at the density functional level of theory with B3LYP hybrid functional and 6-311+G(d) basis set. Stabilization energies were estimated using isodesmic and homodesmotic reactions. As a criterion of aromaticity nucleus-independent chemical shifts of the molecules were computed using GIAO approach. For the polymers the geometrical parameters were optimized through AM1 band calculations and the electronic structures were obtained through modified extended Huckel band calculations. It is found that strong electronwithdrawing substituents increase isodesmic and homodesmotic stabilization energies of pentafulvalene, though it does not increase the aromaticity. Nitro-substituted pentafulvalene is estimated to have stabilization energy as much as azulene. However, substitution either with electron-donating groups or with electronwithdrawing groups does not significantly affect the electronic structures of polypentafulvalene and poly (vinylenedioxypentafulvalene).

Effect of Zizyphus jujuba Extract on Nicotine Sensitization (산조인 추출물의 니코틴 민감화에 미치는 효과)

  • Repeated administration of all addictive drugs, including nicotine, can produce sensitization of extracellular dopamine levels in the nucleus accumbens and behavioral sensitization in rat, as evidenced by an enhanced locomotor response and increased dopamine release in brain to a subsequent injection of the drug. In order to investigate the effect of Zizyphus jujuba extract on repeated nicotin-induced sensitization, rats were given repeated injection of saline or nicotine (0.4 mg/kg s.c., twice a day for 7 d), followed by one challenge injection on the 4th day after the last daily injection. Systemic challenge with nicotine (0.4 mg/kg s.c.) and a direct local challenge of 3 mM a larger increase in locomotor activity and extracellular dopamine release in the nucleus accumbens in nicotine-pretreated rats than in saline-pretreated rats, respectively. Zizyphus jujuba extract significantly decreases locomotor activitiy and dopamine release in the nucleus accumbens induced by a nicotine challenge. These results suggest that Zizyphus jujuba extract may attenuate nicotine-induced neurochemical and behavioral sensitization and may be effective in suppressing compulsive nicotine-seeking behavior.

Determination of metabolites of prostanozol in human urine by LC/ESI/MS and GC/TOF-MS (LC/ESI/MS와 GC/TOF-MS를 이용한 인체뇨시료에서의 Prostanozol 대사체 검출)

  • This research examined prostanozol and its metabolites in urine of women who took the medicine (prostanozol). Prostanozol and its metabolites were successfully separated and detected by using LC/ESI/MS and GC/TOF-MS. Mass spectrum of LC/ESI/MS estimated molecular weight of Prostanozol and its metabolites and that of GC/TOF-MS verified them. For M1, carbon number 17 of Prostanozol substituted to a keto group and it is called 17-keto-Prostanozol. M2 turned out to be hydroxy-17-keto-Prostanozol. It came from substitution of one hydroxyl group of pyrazole nucleus and A-ring of M1. Substitution of one hydroxyl group of B-ring or C-ring became M3, hydroxy-17-keto-Prostanozol. M4 was found to be a hydroxy-17-keto-Prostsnozol transposed from one hydroxyl group to a D-ring. M5 has a hydroxyl group of carbon number 17. One hydroxyl group is substituted from B-ring or C-ring and it is assumed to be hydroxy-17-hydroxy-Prostanozol. M6 was turned out to be dihydroxy-17-keto-Prostanozol transposed from one hydroxyl group to pyrazole nucleus or A-ring and to B-ring or C-ring. Like M6, M7 has a keto group at carbon number 17 and was identified as dihydroxy-17-keto-Prostanozol. M7 has one hydroxyl group at pyrazole nucleus or A-ring and also at D-ring. At last M8 was found to be dihydroxy-17-hydroxy-Prostanozol. Pyrazole nucleus or A-ring has got one hydroxyl group and other rings were substituted to another hydroxyl group. From above, M5, M7 and M8 were verified as new metabolites that were not discovered yet. Prostanozol and all of the 8 metabolites formed glucuronic conjugates as a result of conjugation reaction test in human body. Some of 8 metabolites were excreted without forming conjugates. Particularly M6 and M7 were excreted as sulfate conjugates.

Secretory and Sensory Receptor Cells in the Sucker of Korean Octopus minor II (한국산 낙지(Octopus minor) 흡반(Sucker)내 분비 및 감각수용세포 II)

  • Five kinds of sensory cells, called A1-, A2-, B-, C-, and D-type cell, respectively, are observed in the epithelial tissue of suker's infundibulum of Cephalopoda, Octopus minor. The A1-type cells lie side by with the B-type cell in the epithelium of sucker's infundibulum. In the A1-type, the nucleus shapes irregularly and the karyolymph appears dark due to its high electron density. The cytoplasm is filled with many vacuoles of various sizes ($0.04\sim0.4{\mu}m$ in diameter), which move to the apical portion of the cell to be secreted via glycocalyx. The A2-type cells are mainly found at the basal portion of the epithelium. The shape of its nucleus is similar to that in the A1-type cell, and the cytoplasm, filiform or in reticular form, shows high electron density. The B-type cell contains an ovoid nucleus and the cytoplasm where lots of vacuoles which resemble the endoplasmic reticulum and electron-dense round granules of various sizes $(0.25\sim0.6{\mu}m)$ are found. The vacuoles and granules are secreted into the free surface via glycocalyx. The C- and D-type cells in simple or stratified layer are observed at the folded portion of the sucker's epithelium. The C-type cell contains a low electron-dense elliptical nucleus, while the D-type cell has an irregular nucleus where beterochromatin is well developed.

  • The neurotropic psudorabies virus(PRV) to replicate within neurons is very useful pathogen for neuronal tracing. I carried out this study to investigate the parametric analysis on the viral infection in the rat circadian control center with two genetically engineered strains out of PRV. The two strains are isogenic with the attenuated Bartha strain of PRV ; in one strain a lacZ reporter gene was inserted into the gC locus (PRV-BaBlu ; $4.75\times10^8pfu/ml$) and the other strain contained a PRV envelope glycoprotein gene(PRV-D ; $2.5\times10^8pfu/ml$) theat is absent in PRV-BaBlu. simultaneous or temporally separated sequential injection of$2{\mu}l$ of each strain into the vetreous body of eye produced a course of transsynaptic infection of retinohypothalamic circuitry. The results were as follows; 1. PRV-BaBlu and PRV-D infected the suprachiasmatic nucleus in hypothalamus and intergeniculate leaflet in lateral geniculate nucleus of thalamus. 2. The rate of PRV infection was dependent upon PRV strain. 3. Pre-infected neurons by PRV-D were interfered with the replication of PRV-BaBlu. 4. Dual injection of PRV-D and PRV-BaBlu showed more virulent than the parental strain.
