  • Hydroxyapatite powders were synthesized with Ca/P=1.67 and pH=11 by precipitaton method, after characterization of these specimen, behavior in Ringer's solution and biocompatibility of hydroxyapatite, such as cytotoxity test and implantation test, were investigated. The hydroxyapatite ceramics had structural stability in Ringer's solution, and hydroxyapatite ceramics did not prevent cell growing and exhibit any cytotoxic effects. In implantation of hydroxyapatite ceramics into muscle under abdome of guinea pig, hydroxyapatite ceramics did not show any symptom of rejection for cellular texture.

  • Cerarmide acts as a lipid second messenger in the cellular signal transduction and is involved in mediating a variety of cell functions such as proliferation, differentiation, growth arrest, and apoptosis. In the present study, we have investigated the effect of ceramide on cellular cytotoxity and reactive oxygen species (ROS) to understand the relationship between them.   • The purpose of this study was to evaluate drug-loaded biodegradable membranes for guided tissue regeneration(GTR). The membranes were made by coating mesh of polyglycolic acid(PGA) with polylactic acid(PLA) containing 10% flurbiprofen or tetracycline. The thickness of membrane was $150{\pm}30{\mu}m$, and the pore size of surface was about $8{\mu}m$ in diameter. The release of drugs from the membrane was measured in vitro. Cytotoxity test for the membrane was performed by gingival fibroblast cell culture, and the tissue response was observed after implant of membrane into the dorsal skin of the rat for 8 wks. Ability to guided tissue regeneration of membranes were tested by measuring new bone in the calvarial defects(5mm in diameter) of the rat for 5 weeks. The amount of flurbiprofen and tetracycline released from membrane were about 30-60% during 7 days. Minimal cytotoxity was observed in the membrane except 20% drug containing membrane. In histologic finding of rat dorsal skin, many inflammatory cells were observed around e-PTFE, polyglactin 910 and PLAPGA membrane after 1 or 2 weeks. PLA-PGA membrane was perforated by connective tissue after 4 or 6 weeks, and divided as a segment at 8 weeks.   • The morphlogical characteristics and cytotoxic acitivity for14 local varieties fo Codonopsis lanceolata collected from Korea were examined to get basic data for breeding and selection of good cultivars. Plant height ranged from 197.2 cm to 253.2 com, that from Beakunsan(No.11) was the highest and and that form Samcheok(No.5) was the lowest. Generally, plant height of those form northern area of Korea tends to be shorter than those from Southern area. Leaf width and length of that form Husando (No. 12) was the largest While that form chejudo(No.4) was the smallest and those from other areas showed no significant difference. As to the number of branches, that form Yanggu(No.1) was theleast and those from other areas did not show Chejudo(No.14) were the heaviest. Among 14 varieties tested, Heongseong(No.3) and Huksando(No.6) exhibited strong cytotoxicity , which showed 45.0 an d62.0$\mu\textrm{g}$. $m\ell$ respectively.

  • The oxidation behavior and Biocompatibility of pure Ti and Ti-8Ta-3Nb alloy were studied in dry air atmosphere. The specimens showed that Ti-8Ta-3Nb alloy had higher oxidation resistance than pure Ti at $650^{\circ}C$. Cytotoxicity test also revealed that moderate oxidation treatment lower cell toxicity and Ti-8Ta-3Nb alloy showed better results compared with pure Ti. The weight gains during the oxidation increase rapidly at temperature above $600^{\circ}C$.

  • As part of our program to isolate bioactive compounds from korean natural sources, we have screened traditional medicinal plants to cytotoxity on human tumor cells. Of them, the MeOH extract from rhizome of Coptis japonica Makino was found to be active against five cultured human tumor cell lines. So, the MeOH extract was subjected to successive solvent partitioning to give n-hexane, chloroform and BuOH. The activity was concentrated into the chloroform extract. The extract was chromatographied on a silica gel column and resulted in the isolation 5 alkaloids. (omitted)

The Effects of Pulsatilla Koreana for Anti - Inflammatory and Cellular Activity of Periodontal Tissue (백두옹 추출물의 치주조직 세포에 활성도 및 항염 효과에 관한 연구)

  • This study was performed to define the cytotoxicity and the anti-inflammatory action of Pulsatilla koreana extracts. To analyze cytotoxic effects, gingival and periodontal ligament fibroblasts were used, and anti-inflammatory actions related to reduction of $IL-1{\beta}$ and $PGE_2$ production were performed in vitro, for the suggestion of efficacy and safety on periodontal therapeutic use of Pulsatilla koreana extracts. We extracted ethylacetate and butylalcohol from well-dried and ground Pulsatilla koreana throughout multiple processing, then used different concentration solution(0.1 %, 0.2 %, 0.4 %, 0.01 %, 0.02 %, 0.04 %, 1 %, 2 %) of ethylacetate and butylalcohol extracts to examine eytotoxic effects and anti-inflammatory actions Cytotoxic effects were examined by ELISA reader using MTT(Methyl Thiazol-2-YL-2, 5-diphenyl Tetrazolium bromide)solution following culture of human gingival and periodontal ligament fibroblasts. Synthesis of $IL-1{\beta}$was examined by $IL-1{\beta}$ enzyme-immunoassay(EIA)system after separation and culture of monocyte, and $PGE_2$ was examined by $PGE_2EIA$ system after culture of gingival fibroblasts. The results were as follows: 1. In the MTT test of gingival fibroblasts, the change of optical density was decreased significantly at 2 % of butylalcohol extracts and 0.04 %, 0.1 %, 0.2 %, 0.4 %, 1 %, 2 % of ethylacetate extracts.(p<0.05) 2. In the MTT test of periodontal ligament cells, the change of optical density were not differ significantly. but butylalcohol and ethylacetate extracts except from butylalcohol 0.01 % showed high cell cytotoxity. 3. Both ethylacetate and butylalcohol extracts from Pulsatilla koreana inhibited the synthesis of $IL-1{\beta}$and inhibition effect of ethylacetate extracts were higher than butylalcohol extracts. 4. Both ethylacetate and butylalcohol extracts from Pulsatilla koreana inhibited the synthesis of $PGE_2$, and ethylacetate extracts were higher than butylalcohol extracts. In conclusion, ethylacetate and butylalcohol extracts from Pulsatilla koreana showed little cell cytotoxity for gingival and periodontal ligament fibroblasts, and the inhibition of $IL-1{\beta}$ and $PGE_2$ sysnthesis, therefore it is considered that these extracts can be developed as the therapeutics of the periodontal disease.

Anti-Angiogenic and Anti-Cell Adhesion Effect of the Camellia japonica Flower Extract (동백꽃 추출물의 신생혈관생성 및 세포부착 억제 효과)

  • The Camella japonica flower(CJF) extract was studied for their anti-angiogenenic and anti-cell adhesion effect. CJF-extract inhibited the tube formation on human umbilical vein endotherial cells(HUVEC) with butanol extract by 70.2%, acetone extract by 54.2%, ethyl acetate extract by 37.0%, chloroform extract by 21.2%. Cell adhesion molecules were effectively suppressed at different concentration of CJF at 50, 100, 200 ug/well such as for intercellular adhesion molecule(ICAM) by 5.9%, 29.4% and 52.9%, for vascular cell adhesion molecule(VCAM) by 12.5%, 43.8% and 62.5%, for E-selectin by 7.1%, 21.4% and 35.7%, respectively. Signal molecules of vascular endotherial growth factor receptor 2(VEGFR2), ${/beta}$-catenin and PI3K are inhibited by different concentration of CJF at 10, 20 and 30 ${\mu}g/mL$ with western blot. Angiogenesis will be inhibited with suppressing NF-kB molecule resulted in signal molecules blocked by CJF. CJF will be useful materials for treatment of angiogenesis related diseases such as cancer, metastasis, rheumathioid arthritis and obesity.

Antioxidant Activity and Cytotoxicity on Human Cancer Cells of Anthocyanin Extracted from Black Soybean (검정콩 안토시아닌의 항산화 및 암세포독성)

  • Anthocyanin pigments in soybean seed coat were D3G (Delphinidin-3-glucoside), C3G (Cyanidin-3- glucoside) and Pt3G (Petunidin-3-glucoside), which have been known potential roles in the prevention and treatment of chronic diseases. Anthocyanin contents in seed coat of blck soybean were significantly different according to soybean variety, C3G content showed the highest value in all materials and its variation was also wide. Antioxidant activity of each pigment was analyzed by DPPH and TEAC methods in which D3G and C3G showed high activity. And this study was carried out to investigate the effects of anthocyanin to human cancer cells. Cytotoxity were analyzed by MTT assay after anthocyanin pigments treated on leukemia (Jurkat T) and adenocarsinoma (MCF-7) cells. It showed decrement of cell numbers as anthocyanin concentration is increasing. ${EC}_50$ range of anthocyanin concentrations were $100{\sim}250\;ug/mL$ and $100{\sim}250\;ug/mL$ in Jurkat T and MCF-7 cell, respectively. D3G showed higher cytotoxicity than other pigments in Jurkat T cell whereas activity of C3G was high in MCF-7 cell. It is believed that supplementation of human diets with soybean anthocyanin markedly reduces human cancer mortality rates.

Inhibitory Effects of Asadisulphide on the TEA-induced Adherence of HL-60 Cells (TPA로 야기된 HL-60 세포의 기질부착에 대한 Asadisulphide의 억제 효과)

  • Asadisulphide were purified from Ferrula assafoetida by organic solvent extraction and chromatography. Its inhibitory effects on the TPA-induced adherence of HL-60 cells was analyzed. Since ethyl acetate extracts of F. assafoetida has the strongest inhibitory effect on adherence of HL-60 cells, it was re-extracted with ethyl acetate, hexane, and ethyl ether and chromatographed three times to isolate asadisulphide. At the minimum concentration of 2 $\mu\textrm{g}$/ml, asadisulphide inhibited adherence of 98% of HL-60 cells that have been treated with TPA. It also showed anti-cancer effect with no cytotoxity in the ED$_{50}$ value of 20 $\mu\textrm{g}$/ml.
