Cytotoxic Activities of Green and Brown Seaweeds Collected from Jeju Island against Four Tumor Cell Lines

  • Methanolic and aqueous extracts from 37 seaweed species (10 green and 27 brown seaweeds) collected from Jeju Island coast were prepared at high ($70^{\circ}C$) and room ($20^{\circ}C$) temperatures and examined for cytotoxic activity against 4 tumor cell lines: U937 (human monoblastoid leukemia cell line), HL60 (human promyelocytic leukemia cell line), HeLa (woman cervical carcinoma cell line) and CT26 (mouse colon carcinoma line). Both MeOH extracts of Desmarestia tabacoides and Dictyota dichotoma possessed strong cytotoxic activities against all the tumor cell lines tested, but the aqueous extract exhibited no activity. On the other hand Ecklonia cava showed strong cytotoxic activities for the $20^{\circ}C$ aqueous extract against the three tumor cells except HeLa cell. Sagassum coreanum and Sagassum siliquastrum $20^{\circ}C$ aqueous extracts also exhibited strong cytotoxic activities against U937, HL60, HeLa cells. Even though green seaweeds showed less activity than brown seaweeds, $20^{\circ}C$ aqueous extracts of Codium contractum and Codium fragile exhibited strong cytotoxic activities against HL60 or CT26 cells, respectively.

  • The cytotoxic activities of Pinellia ternate, Aconitum carmichaeli, Arisaema amurense, Aconitum kusnezoffii and Scutellaria baicalensis on monkey kidney cell (Vero) and human liver cell (WRL68) were evaluated by Sulforhodamine B Protein (SRB) and Tetrazolium-based (MTT) colorimetric assay methods The results were as fellows : 1 The Pinellia ternate and Arisaema amurense did not show the cytotoxic activities at any concentration without processing or natural drugs. 2 The extracts of Aconitum carmichaeli, Aconitum kusnezoffii and Scutellaria baicalensis showed cytotoxic activities. However, the cytotoxic activities of processing drugs were less effective than the natural drugs. 3. The cytotoxic activities on monkey kidney cell (Vero) and human liver cell (WRL68) of Aconitum carmichaeli was determined by MTT assay. The Kyungpo(京?) of Aconitum carmichaeli showed less concentrate than that of the Dangpo(唐?). The $IC_{50}$ value on monkey kidney cell (Vero) and human liver cell (WRL68) of Kyungpo was $937{\pm}29\;and\;731{\pm}31{\mu}g/ml$, respectively 4. The cytotoxicity on monkey kidney cell (Vero) and human liver cell (WRL68) of Scutellaria baicalensis showed strong activities without processing or natural drugs.

Comparative Cytotoxic Activities of Various Ginsengs on Human Cancer Cell Lines

  • Comparative cytotoxic activities of petroleum ether soluble fraction from various ginsengs of Panax species were evaluated using A549 (human lung adenocarcinoma) and SK-OV-3(human ovary carcinoma) cancer cell lines. Korean red ginseng, Korean white ginseng, American ginseng and Canadian ginseng were found to show more potent cytotoxicitles on A549 and SK-OV-3 cell lines than Chinese red ginseng, Japanese red ginseng and Sanchi ginseng. It is noteworthy that especially, red ginseng prepared from the root of Panax ginseng cultivated in Korea shows relatively stronger cytotoxic activities than those cultivated in China and Japan.

Antibacterial, cytotoxic and neuropharmacological activities of Cerbera odollam seeds

  • The MeOH extract of seeds of Cerbera odollam Gaertn. (Apocynaceae) was screened for its antibacterial, cytotoxic and neuropharmacological activities. The extract showed moderate anti-bacterial activity against Salmonella typhi, Streptococcus saprophyticus, and Streptococcus pyogenes. It exhibited high level cytotoxicity against brine shrimp ($LC_{50}$: $3\;{\mu}g/ml$). The extract potentiated pentobarbital induced sleeping time in mice which was further supported by the exploratory behavior test at dose of 25 mg/kg. The overall results tend to suggest the antibacterial, cytotoxic and CNS depressant activities of the extract.

  • Some coumarins were sythesized for the screening of their cytotoxic activities against L1210 cell. Of the conmarins sythesized, 6, 7-dihydroxycoumarin (esculetin) and 7, 8-dihydroxycoumain (dephnetin) as coumarins with dioxygenated A-ring, and 6-acetoxy-5, 7-dimethoxycoumarin and5, 7-dimethoxy-6-hydroxycoumarin as trioxygenated ones, show considerable cytotoxic activities, ED 50 being 4. 3, 8. 8, 17.2 and 5.5 $\mu$g/ml in the same other as the substances. THe extent oxygenation of the A-ring and the positions of the oxygen functions eventually play an important role for the cytotoxic activity.

  • Cancer is the second leading cause of death in the world. According to the 2018 reports, one in six people worldwide is reported to die as a result of cancer. The discovery of anticancer drugs has been utilized extensively, but there has been no report on excellent selective activity in cancer cells. The discovery of bioactive substances from marine sponges has been the limelight in the pharmaceutical field over the past decade owing to the production of many bioactive compounds from the sponges to protect themselves against the environment. On top of that, marine sponges also produced cytotoxic compounds such as terpenoids, alkaloids, steroids, and peptides which suggests that marine sponges have high potential in the development of anticancer drugs. Thus, this study aimed to obtain new cytotoxic compounds from S. siphonella in Egypt and Arenoscelra sp. and Gelliodes sp. in Vietnam, and further investigation of the extract from these marine sponges led to isolation of ten new compounds and 21 known compounds. Chapter 1 will discuss about the isolation and structure elucidation of eight new polyacetylene derivatives from S. siphonella and their cytotoxic activities. The isolation and structural elucidation of one new polybrominated iododiphenyl ether from Arenosoclea sp. as well as cytotoxic activities of the isolated compounds will be reported in chapter 2. Finally, isolation and structure elucidation of new compounds from the marine sponge Gelliodes sp. and their cytotoxic activities will be discussed in chapter 3.

Preliminary antioxidant, antibacterial and cytotoxic activities of Momordica charantia Linn leaf

  • We undertook the present study to evaluate different pharmacological as antioxidant, antibacterial and cytotoxic activities of the crude ethanolic extract of Momordica charantia (Family: Cucurbitaceae) leaves. The antioxidant property of the extract was assessed by 1, 1-diphenyl-2-picryl hydrazyl free radical scavenging assay. The extract showed antioxidant activity where $IC_{50}$ was about ${\sim}500{\mu}g/ml\;and\;IC_{50}$ was about ${\sim}10{\mu}g/ml$ for standard drug ascorbic acid. The extract showed a broad spectrum of antibacterial activity against all the tested gram positive and gram-negative bacteria where Staphylococcus epidermidis, Staphylococcus aureus, Salmonella typhi and Shigella dysenteriae were prominent. And the zones of inhibition were ranging from 8-15 mm for all the tested bacteria. Its cytotoxic property was evaluated by brine shrimp lethality bioassay. The extract showed significant lethality and the $LC_{50}$ value was $20{\mu}g/ml$.

Antimicrobial and Cytotoxic Activities of Some Malaysian Flowering Plants

  • A total of 43 extracts from 21 species of Malaysian flowering plants were screened for antimicrobial and cytotoxic activities. Antimicrobial activity was tested against fungi, including yeast and candida, as well as Gram-positive and Gram-negative bacteria, and cytotoxicity was assayed using the CEM-SS and HT-29 cell lines. The methanol extracts of the roots and stems of Plumeria acutifolia showed the highest antimicrobial activity, i.e. against Saccharomyces cerevisiae and Candida lipolytica, and mostly moderate activity against the other microbes such as Aspergillus ochraceous and Saccharomyces lipolytica. Phyllanthus emblica also showed moderate activity especially that of the methanol extracts. The methanol extract of the roots of Plumeria acutifolia showed the strongest cytotoxic activity $(CD_{50}=\;3\;{\mu}g/ml)$. Most of Zingiberaceae species gave negative results for antimicrobial activity and showed low cytotoxic activity.

  • 4-year-old fresh ginseng was stored for 12 weeks at 4$^{\circ}C$ under the CA storage and samples were withdrawn after every 1, 2, 3, 4, 6, 8 and 12 weeks for processing of red ginseng. Antloxidative and cytotoxic activities of steamed red ginseng for the quality evaluation are summarized as follows: Reducible activity of water extract to 1, 1-dipenyl-2-picryl hydrazyl increased with increasing storage period. Antioxldative activity of red ginseng did not change in petroleum ether fraction, while it had a tendency to increase In the fractions of ethyl ether and ethyl acetate with increasing storage period. in contrast to fresh ginseng, malonaldehyde formation In heparin mlcrosome of red ginseng processed from fresh ginseng stored In CO2:O2:Nl (6:4:90) with a 0.5% DF-100 immersion condition decreased but was not changed significantly by the period of storage. Cytotoxic activity against cancer cell at a 40 mg/ml concentration of red ginseng slightly increased with an increase in storage period but no effect was observed at the concentration less than 10 mg/ml.

인체암세포주에 대한 천연자원의 세포독성 검색 (Ⅱ) (Cytotoxic Activities of Herbal Drugs Against Human Cancer Cell Lines (Ⅱ))

  • In our continuing search for new antineoplastic agents from natural products, one hundred and thirty-five herbal drugs were extracted with petroleum ether/ether (1:1), ethyl acetate and methyl alcohol, successively and their cytotoxicities were evaluated against A549 (human lung carcinoma) and SK-OV-3(human ovary adenocarcinoma) cell lines. Among them, fifteen kinds of ether extracts, eighteen kinds of ethyl acetate extracts and seven kinds of methanol extracts showed significant cytotoxic activities (above 70% inhibition) against A549 cell lines at a concentration of $40\;{\mu}g/ml,$ while ten kinds of ether extracts, thirteen kinds of ethyl acetate extracts and six kinds of methanol extracts demonstrated significant cytotoxic activities against SK-OV-3 cell lines at the above same concentration.
